Objective To investigate the correlation between ankle-brachial index and ultrasonography in lower ex- tremities vascular in type 2 diabetic patients.Methods 218 type 2 diabetic patients were enrolled,and ankle-brachial index(ABI)measurements and uhrasonography in lower extremities vascular were performed in all patients.Lower extremi- ties vascular disease(LEVD)were divided into six species according to the uhrasonography results and scored in accord- ante with its serious degree,then investigate the correlation between ABI and uhrasonography results score.All patients were divided into deferent groups according to uhrasonography results and ABI range,and then compared their deference. 2.Effect of Shuanghuanglian injection on cerebral nuclear factor-kappaB expression in mice with viral encephalitis.

OBJECTIVETo observe the effect of Shuanghuanglian injection on cerebral expression of nuclear factor-kappaB (NF-kappaB) in mice with viral encephalitis.
METHODSThe mice with experimental viral encephalitis received treatment with Shuanghuanglian injection at the dose of 0.2, 1.5, and 5 for 5, 10 or 20 consecutive days. The total RNA of the brain tissue was extracted to analyze the protein and mRNA expression of NF-kappaB using Western blotting and RT-PCR, respectively.
RESULTSCompared with the control group, the mice with experimental viral encephalitis showed significantly increased protein and mRNA expressions of NF-kappaB (P<0.01). Treatment with Shuanghuanglian injection at the doses of 0.2 and 1.5 mg/kg significantly lowered NF-kappaB protein and mRNA expressions in the brain of mice with viral encephalitis (P<0.05), and the effect was even more obvious at the dose of 5 mg/kg (P<0.01).
CONCLUSIONShuanghuanglian injection can reduce the expression of NF-kappaB in the brain of mice with viral encephalitis in a dose- and time-dependent manner.

3.Quantitative determination and pharmacokinetics of retinamido-ester in rat plasma by liquid chromatography-atmospheric pressure chemical ionization-tandem mass spectrometry

A highly sensitive, rapid and selective liquid chromatography-tandem mass spectrometry(LC-MS/MS) method for the quantitative determination of retinamido-ester in rat plasma was developed and validated. A simplified protein precipitation with acetonitrile was employed for the sample preparation.The separation was carried out on an Agilent TC C18 column ( 150 mm×4. 6 mm ID, 5 μm particle size)with the mobile phase consisted of methanol-water-formic acid (93 : 7 : 0.1). Simvastatin was used as internal standard. The detection was performed on a trap-quadrupele tandem mass spectrometer by selected reaction monitoring (SRM) scan mode via atmospheric pressure chemical ionization (APCI). The range of and inter-day precision values were between 95.97% and 104. 43%, and RSD was between 4. 63% and 10. 69%, respectively. This method was applied to determine the pharmacokinetic parameters. The main pharmacokinetic parameters of retinamido-ester after oral administration via gastric gavage of 2. 5, 5, 10mg·kg-1 were as follows,T1/2;(11.28±7.23),(8.90±3.82),(8.01±5.56)h; AUC0-∞:(103.41±61.46),(190.23±74.99),(421.66±299.20)ng·h·mL-1;MRT:(6.31±0.75),(5.98±0.71), (6.18±0.97) h; CL/F: (30. 10 ± 13.67), (29.58±10.59), (31.18 ±17.51)L·h-1·kg-1;Vd/F:(414.94±159.82),(356.16±139.85),(369.28±322.73)L·kg-1,respectively.
4.Strategy of prevention and management of intraoperative fractures of acetabulum during primary total hip arthroplasty.

OBJECTIVETo analyse the causes and resultes of intraoperative occurrence of an acetabular fracture in primary total hip arthroplasty and discuss propriate prevention and treatment of this problem.
METHODSBetween July 2004 and December 2006, 326 primary total hip arthroplasties were retrospectively reviewed and found that eleven patients (eleven hips) had sustained an intraoperative acetabular fracture involving 7 male and 4 female,aged from 37 to 71 years (mean 54 years). Analyzed the causes of fractures, the anatomic location, treatment and outcome of the fractures. Acetabular component designs were categorized and each design was analyzed for fracture risk.
RESULTSAmong 11 cases, the fractures occurrenced as setting the prosthesis in 6 cases, grinding and drilling in 3 cases, drawning hook in 2 cases. There were 5 cases of central type fracture, 3 of posterior-wall, 2 of anterior, 1 of posterior-upper. Nine of these patients (nine hips) had been followed-up for 12 to 29 months(mean 18 months). There were no heterotopic ossification, no dislocation, no deep venous embolism. All fractures were healing.
CONCLUSIONAcetabular fracture during primary total hip arthroplasty is a complication of acetabular fixation without cement, which should be think highly of. In the present series, retention of a stable cup is associated with uneventful osseous in growth and excellent early-term outcomes.

5.Global gene response to GSM 1800 MHz radiofrequency electromagnetic field in MCF-7 cells.

OBJECTIVETo investigate whether GSM 1800 MHz radiofrequency electromagnetic field (RF EMF) can change the gene expression profile in MCF-7 cells and to screen RF EMF responsive genes.
METHODSSubcultured MCF-7 cells were intermittently (5-minute fields on/10-minute fields off) exposed or sham-exposed to GSM 1800 MHz RF EMF, which was modulated by 217 Hz EMF, for 24 hours at an average specific absorption rate (SAR) of 2.0 W/kg or 3.5 W/kg. Immediately after RF EMF exposure or sham-exposure, total RNA was isolated from MCF-7 cells and then purified. Affymetrix Human Genome U133A Genechip was applied to examine the change of gene expression profile according to the manufacturer's instruction. Data was analyzed by Affymetrix Microarray Suite 5.0 (MAS 5.0) and Affymetrix Data Mining Tool 3.0 (DMT 3.0). Quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to validate the differentially expressed genes identified by Genechip analysis.
RESULTSA small number of differential expression genes were found in each comparison after RF EMF exposure. Through reproducible and consistent analysis, no gene or five up-regulated genes were screened out after exposure to RF EMF at SAR of 2.0 W/kg or 3.5 W/kg, respectively. However, these five genes could not be further confirmed by RT-PCR.
CONCLUSIONThe present study did not provide clear evidence that RF EMF exposure might distinctly change the gene expression profile in MCF-7 cells under current experimental conditions, implying that the exposure might not affect the MCF-7 cell physiology, or this cell line might be less sensitive to the RF EMF exposure.

6.Changes in neural cell adhesion molecule mRNA expression and protein level in the CA1 region of the hippocampus during long term potentiation induction and maintenance.

It has been demonstrated that neural cell adhesion molecule (NCAM) is critical for the induction and maintenance of long term potentiation (LTP) in the CA1 region of rat hippocampus. In the present study, we investigated the changes in NCAM mRNA expression and NCAM protein level after the induction of LTP in vitro using the techniques of in situ hybridization and Western blot. The results showed that the number of NCAM mRNA positive labelled neurons significantly increased (76.6+/-11.5 neurons) 10 min after tetanus when the slope of fEPSP markedly increased. The level of NCAM protein also increased significantly (7.190+/-0.64 arbitrary unit/50 microg protein) 10 min after tetanus. The number of NCAM mRNA positive labelled neurons no longer changed (73.3+/-14.0) 1 h after tetanus, however, the NCAM protein level (9.031+/-0.71) at 1 h after tetanus was higher than that at 10 min after tetanus. Moreover, the NMDA receptor inhibitor AP-5, which blocked LTP, prevented the increase in NCAM mRNA expression and NCAM protein level. The results demonstrate that NCAM mRNA expression maintains a high level, whereas NCAM protein changes from a low level to a high level during induction and maintenance of LTP.

8.Efficacy and safety of paroxetine in treatment of post-stroke depression and anxiety

Objective To investigate the efficacy and safety of paroxetine in the treatment of post-stroke depression and anxiety.Methods Patients with post-stroke depression and anxiety (312 cases) were randomly divided into observation and control groups.All patients were given routine,symptomatic,supportive treatment and rehabilitation training,psychological intervention,in addition,the observation group was treated with paroxetine hydrochloride.The Hamilton Depression Scale (HAMD) score,anxiety scale (HAMA) score,plasma substance P (SP),neuropeptide Y (NPY),cortisol (Cor) content and side effects scale (TESS) score of both groups before and after treatment were analyzed and compared.Results After treatment,the HAMD and HAMA scores of two groups were significantly decreased,and those of the observation group was significantly lower than the control group (P < 0.05).The clinical curative effect of the observation group was better than that of the control group (P < 0.05).In both groups,the levels of SP and Cor were significantly decreased,NPY was significantly increased,and the improvement of the observation group was higher than that of the control group (P < 0.05).In the observation group,some patients had anorexia,nausea,dizziness,insomnia and other reactions,but they were tolerant,and there was no significant difference in the TESS score between the observation group and control group.Conclusion Paroxetine treatment of post-stroke depression/anxiety can reduce depression and anxiety symptoms,improve neuropeptide and cortisol levels,and have good curative effect and good safety.It is worthy of clinical application and further study.
9.Correlation of Epstein-Barr virus infection and childhood lymphoma.

OBJECTIVETo investigate the correlation of Epstein-Barr virus (EBV) infection and childhood lymphoma.
METHODSParaffin-embedded specimens of lymphoma collected between 1996 and 2005, including 36 Hodgkin lymphomas (HL) and 51 non-Hodgkin lymphomas (NHL), were included in this study. Paraffin-embedded specimens of reactive hyperplasia of lymph nodes (RL) collected during the same period were used as controls. Immunohistochemical (IHC) assay was used to detect EBV-LMP1 and in situ hybridization (ISH) to detect EBV-EBERs.
RESULTSEBV was detected in 72.2% (26/36) of the Hodgkin lymphomas, 15.7% (8/51) of the non-Hodgkin lymphomas and 33.3% (15/45) of the reactive hyperplasia of lymph nodes. There was a significant difference among Hodgkin lymphoma, non-Hodgkin lymphoma and RL (P = 0. 000).
CONCLUSIONChildhood Hodgkin lymphoma is closely correlated with Epstein-Barr virus infection. However, the low rate of EBV infection detected in childhood non-Hodgkin lymphoma might be due to heterogeneous distribution of pathological types in this study.
