Objective To observe the course of motor related activation cortex after unilateral subcortical ischemic stroke. Methods6 healthy volunteers and 3 patients with solitary lacunar infarction in subcortical region underwent blood-oxygen level dependent functional MRI (BOLD-fMRI) as a block design of sequential finger tapping task. The data were analyzed with Statistical Parametric Mapping (SPM2). The activated voxels and the laterality index (LI) were calculated. The patients were assessed with the Fugl-Meyer Assessment of upper limb after scanning, and were scanned again 8 months after stroke. ResultsThe activated cortices in patients were more extensive than that of volunteers. In the early stage of stroke, the activation cortex involved the bilateral sensory motor cortex (SMC) in all the patients, but various in other regions. In the late stage, the ipsilateral activations decreased while the contralateral activation increased in SMC. The LI of hemisphere, SMC, and M1 is higher than the early stage. ConclusionThe motor related activation gradually localized to the contralateral SMC with motor function recovery.

4.Absorption of extractive Polygonum orientale in rat everted gut sacs.

Using in vitro everted gut to investigate the intestinal absorption of the extracts from Polygonum orientale at different concentration. UPLC-MS/MS was used to detect the content of protocatechuic acid, isoorientin, orientin, vitexin, cynaroside, quercitrin, kaempferol-rhamnoside in different intestinal segments, then compared the results with the absorption of chemical components of extractive P. orientale in each intestinal segments, and calculated the absorption parameter. We took the statistic analysis with SPSS statistic software. The influence significance of each factors were analyzed to describe the character of absorption. The absorption of each component is linearity in different intestinal segments and different dose, and the square of coeficient correlation exceed 0.95, which consistent with zero order rate process. The K(a) increase along with the raised dosage of the extractive P. orientale (R2 > 0.95), indicated it is the passive absorption; different intestinal segments have different absorption. And the absorption trend in intestinal is duodenum, jejunum, ileum are greater than the colon. As ingredients are selectively absorbed in intestinal sac, the everted intestinal sac method is selected to assess the intestinal absorption charcteristics of ingredients of extractive P. orientale.

5.Researching on fingerprint of Inulacappa by HPLC.

OBJECTIVEThis study is to establish the fingerprint and find out the common chromatographic peaks of Inula cappa by HPLC.
METHODThe HPLC analysis was performed on an Agilent Eclipse Plus C18 column (2.1 mm x 150 mm, 1.8 μm) with 0.1% fomic acid aqueous solution-0.1% fomic acid acetonitrile solution as mobile phase at a flow rate of 0.3 · mL(-1) · min(-1); The detective wavelength is 325 nm; The column temperature is 45 °C.
RESULTThe results indicated that 5 of 17 common peaks were identified . The similarity about 10 groups of Inulacappais is over 0.95.
CONCLUSIONThis method is able to be a scientific basis of quality assessment according to its convenient and reliable.

6.Effects of clenbuterol on the hepatic flux of nitrogen, VFA and glucose in sheep.

AIMTo examine the liver mechanism with which clenbuterol (CL) is explained how to affect growth metabolism.
METHODSThe technique of chronic poly catheter was used to study the effects of CL (0.8 mg/kg b w) on the hepatic flux of nitrogen, VFA and glucose in 4 sheep.
RESULTSThe urea-nitrogen flux in CL-treated period always was lower than that in control during 24 h. The average flux of urea-nitrogen in hepatic and portal vein were decreased by 16.86% (P < 0.01) and 15.51% (P < 0.05), respectively, compared with that of control. The peptide level in hepatic vein was decreased with the treatment of CL, average flux of peptide was decreased by 38.71% (P < 0.01). But the peptide level of portal vein in CL treatment period was similar to control. Moreover, VFA level in the portal vein was enhanced by CL, the average flux of acetate in portal vein was increased by 19.49% (P < 0.01). No difference of VFA level in hepatic vein was noted between CL-treated period and control. In addition, the glucose flux in hepatic vein was obviously increased with CL treatment, the average flux of glucose was increased by 25.96% (P < 0.01). And glucose flux in portal vein was also elevated during CL-treated period.
CONCLUSIONCL can affect growth metabolism of animal with increasing nitrogen deposition, improving absorption and utilization of VFA and enhancing glucose synthesis in sheep liver.

8.The significance and effects of liver X receptor alpha in nonalcoholic fatty liver disease in rats.

OBJECTIVETo explore liver X receptor alpha (LXR alpha) gene changes and their significance in nonalcoholic fatty liver disease (NAFLD) in rats.
METHODSA rat model of nonalcoholic fatty liver disease was produced with a fatty diet regime (feeding group, FG). Rats fed with normal diet served as controls (CG). The mRNA and protein expressions of LXR alpha in liver tissues were detected by reverse transcription and polymerase chain reaction (RT-PCR) and Western blot.
RESULTSThe concentration of free fatty acid (FFA) in the sera of GF rats started to increase to 0.33 mmol/L after 4 weeks of fat diet feeding, while the FFA of the CG was just 0.24+/-0.03 mmol/L, and the difference was significant (P<0.05). The concentration of ALT and AST in sera of the FG rats started to increase to 75.8 U/L and 138.9 U/L at the 8th week, much higher than those of the CG (P<0.01), and at the 12th week they increased further (P<0.01). Meanwhile, the mRNA and protein expressions of LXR alpha at the 2nd week was significantly increased to 0.62 (P>0.01) and its peak was reached at the 12th week (P<0.01). There was a significant positive correlation between the expression of LXR alpha and the degree of NAFLD.
CONCLUSIONThe changes of LXR alpha gene are closely related to the development of nonalcoholic fatty liver disease.

9.Adaptive Cardio Sequence Prospective ECG-gated Dual-source CT Coronary Artery Imaging:An Initial Study

Objective To investigate the feasibility of the adaptive cardio sequence prospective ECG-gated dual-source CT coronary angiography (CTCA).Methods Thirty one patients underwent prospective ECG-gated dual-source CTCA. The patients were divided into two groups: group A, heart rate ≤ 75 times / minutes, 70% of R-R interval was chose; group B, heart rate> 75 times / minutes, 40% of R-R interval was chose. Two experienced radiologists assessed the image quality of 15 segments of coronary arteries in a double blinded fashion in a four score scale. Student t test was used to compare the image quality between two groups. CT dose index(CTDIvol) and effective dose(ED) were also calculated for both groups. Results 437 segments of coronary arteries in 31 patients were included this analysis, 408 segments (93.4%) had assessable image quality, while 29 segments (6.6%) had non assessable image quality. Of 226 segments in group A, 12 segments (5.3%) had non assessable image quality; however, 17 segments (8.1%) of 211 segments in group B had non assessable image quality. The average score of image quality was 3.57±0.64 in 31 patients. There was no statistical difference for mean image quality of group A (3.65±0.39) and group B (3.41±0.43)( t=1.62,P=0.12 ) . There was a statistical difference for mean image quality for segment 10 on a per segment basis ( t = 3.8 ,P<0.05). CTDIvol and ED were (18.88±5.04) mGy and (4.31±1.05) mSv, respectively.Conclusion Adaptive cardio sequence prospective ECG-gated dual source CT can acquire assessable image quality of coronary artery with lower radiation dose to the patients, which has the potential to be a routine CTCA technique.
10.Clinical prospective study on maternal-fetal transmission of human bocavirus.

OBJECTIVETo investigate maternal-fetal transmission at human bocavirus (HBoV).
METHODSIgG antibody to HBoV in serum samples of 316 mothers were determined with ELISA and HBoV DNA was determined with real time PCR in the sera of the mothers and their infants.
RESULTSHBoV-IgG was positive in 40.20 percent (127/316) of the mothers, while it was positive in 29.43 percent (93/316) of the cord blood specimens of the infants. The difference between the two groups was significant (X2=8.12, P less than 0.005); 93 samples of both the mothers and the infants were positive for HBoV-IgG.
CONCLUSIONHBoV-IgG can cross the placenta to the fetuses through placenta. Further study is needed to answer the question whether vertical maternal-fetal transmission occurs.
