Objective To clone human beta-2 adrenoceptor gene from human bladder smooth muscle and to construct its antisense eukaryotic expression vector. Methods The ?_2-AR full length cDNA was cloned from human detrusor cells through RT-PCR and subcloned into clone vector (pUC18).The objective gene was then cut from ClaⅠ/HindⅢ sites of pUC18 with restriction endonulcease and subcloned into pLNCX vector in trans-direction.Finally the constructed ?_2-AR gene antisense expresstion vector was identified through restriction endonuclease analysis and sequencing. Results The sequence of cloned ?_2-AR full length cDNA was certified by comparison with the database of the Genebank.The constructed antisense eukaryotic expression vector was proved to be same with designed by restriction endonuclease analysis and sequencing. Conclusions ?_2-AR full length cDNA was cloned and its antisense eukaryotic expression vector was successfully constructed.This technique establishs the foundation for the further research on drug treatment of bladder dysfunction.

Oil of Eveningprimrose was extracted from the seed of Oenothera biennis by supercritical CO2 extraction.A yield of 20% oil was obtained at 50℃ and 25MPa. Its ?-linolenic acid content was 10. 6%. Acid value and saponification number of the oil was determined.

Objective To study the association between platelet distribution width (PDW) and other indexes in coronary heart disease (CHD) patients with heart failure (HF).Methods Five hundred and ninety-one CHD patients with HF were divided into control group with their NT-proBNP＜300 ng/L (n=228),mild HF group with their NT-proBNP=300-3000 ng/L (n=191),and severe HF group with their NT-proBNP＞3000 ng/L(n=172).Their blood and echocardiographic parameters were recorded.Results The PDW was significantly shorter in 3 groups after treatment than before treatment (0.129±0.031 vs 0.133±0.027,0.133±0.030 vs 0.141± 0.029,0.139±0.028 vs 0.148±0.029,P＜0.05,P＜0.01),and was significantly longer in mild HF group and severe HF group than in control group and in severe HF group than in mild HF group after tgreatment (P＜0.05,P＜0.01).Multivariate stepwise regression analysis showed that PDW was positively associated with NT-proBNP and LAD (P=0.001,P=0.000) and negatively associated with LVEF (P=0.003).Conclusion PDW increases with the severity of HF in CHD patients and can thus play an auxiliary role in assessing the severity of their HF.

Objective To preliminarily explore the clinical significance of word recognition scores in evalua_ting the curative effects of sudden hearing loss .Methods Pure tone averages (PTA) and word recognition scores (WRS) in 11 cases of sudden hearing loss were obtained on the pre-treatment day and the post -treatment day , and the results were compared .ResuIts The average PTA of 11 cases was 62 .27 ± 15 .36 dB HL on the pre-treat_ment day ,and 60 .23 ± 14 .32 dB HL on the post -treatment day .The average WRS in 11 cases was 42 .91% ± 30 .83% on the pre-treatment day ,and 72 .73% ± 26 .72% on the post-treatment day .There were no significant differences in PTA and the intensity of the sound for testing WRS before and after treatment(P> 0 .05) ,and the difference of the WRS between the pre -treatment and the post -treatment day was statistically significant(P<0 .05) .ConcIusion The WRS could be used in evaluating the curative effect of sudden hearing loss ,which is a sup_plement for PTA .

OBJECTIVE:To investigate the inhibitory effect of the total flavoniods of Ampelopsis grossedentata on human hepatoma carcinoma cells(HCC) in vitro.METHODS:The inhibitory effect of the total flavoniods of Ampelopsis grossedentata extract on human hepatoma carcinoma BEL7404 cells in vitro was analyzed by MTT,cell growth curve method and colony formation method,respectively.RESULTS:The total flavonoids of Ampelopsis grossedentata significantly inhibited the growth of BEL 7404 cells in vitro.MTT assay showed that the IC50 was 28.59 ?g?mL-1;the cell growth curve measurement showed that the total flavonoids of Ampelopsis grossedentata significantly inhibited the growth of BEL 7404 cells;the colony formation assay showed that the IC50 was 100% at a drug concentration of above 22.22 ?g?mL-1 and the IC50 was 58.05% at a drug concentration of 14.81 ?g?mL-1.CONCLUSION:The total flavonoids of Ampelopsis grossedentata have a remarkable inhibitory effects on the growth of BEL 7404 in vitro.

OBJECTIVE: To investigate the current situation of early diagnosis and intervention in 0-9 months old infants with hearing loss and analysis factors that will affect early diagnosis and intervention. METHOD: One hundred and eighty-six infants referred to the West China hospital from February 2014 to September 2014 were included. All 186 children were referred due to the fact that either they failed infant hearing screening or outer ear malformation. Early diagnosis and/or intervention were performed on those 186 children and their records of early diagnosis and intervention were analyzed. RESULT: Among the 186 infants, 167 (89.8%) were diagnosed with an average age at (4.0 ± 1.4) months. Among the 167 infants with final diagnosis, there were 31 (18.6%) infants diagnosed as conductive hearing loss (CHL), and 99 cases (59.3%) diagnosed as sensorineural hearing loss (SNHL), among whom, there were 75 (44.9%) bilateral SNHL and 24 (14.4%) unilateral SNHL. There were 2 cases (1.20%) with SNHL on one side and atresia on the other side. 5 (2.99%) of all conductive hearing loss cases with unilateral atresia and 2 cases with auditory neuropathy (AN) were found. 33 infants (19.8%) were found to have normal hearing. 30.7% (23/75) infants diagnosed as bilateral SNHL and 8.3% (2/24) infants diagnosed as unilateral SNHL were fitted with hearing aids. The fitting rate in infants with bilateral SNHL with mild, moderate, severe to profound degrees were 0 (0/23), 24.0% (6/25), 66.7% (6/9), 61.1% (11/18) respectively. The average intervention age was (5.0 ± 2.1) months. CONCLUSION Although the early diagnosis and intervention situation in this study are very close to international standard, there are still infants without final diagnosis and infants with hearing loss without hearing aid fitting. Further studies and efforts to promote early diagnosis and intervention in infants with hearing loss are needed.

Objective To explore the efficiency,safety and suitable dosage of the non-invasive treatment of malignant gliomas with high intensity focused ultrasound(HIFU).Methods An extracorporeal HIFU with 9.7 MHz transducer(focal length: 4.5 mm,focal intensity: 2 500 W/cm~(2)) was used.Fifteen resected human malignant gliomas were randomly assigned to receive HIFU ablation for 10,20,30 s.After the treatment,the exposure coverage and marginal zone were collected and examined by HE staining and electron microscope.Results Exposure coverage and margin was safe in 10 s group.Typical coagulation necrosis was detected at exposure coverage with no detectable change at margin in 20 s group.Typical coagulation necrosis was seen at both exposure coverage and margin in 30 s group.Conclusion Under the parameters used in this study,20 s is a beneficial therapeutic dosage.HIFU is an accurate method for malignant gliomas ablation.

Objective: To study the effects of propafenone on action potential (AP) of rabbit ventricular myocytes with the tonic block and use-dependent block of transient sodium current (INa-T). Methods: A total of 10 adult New Zealand white rabbits were sacriifced and 10 individual ventricular myocytes were isolated by enzyme digestion method. Microelectrode technologies were used to record AP-related parameters: maximum diastolic potential (MDP), maximum rate of rise of the action potential upstroke (Vmax), action potential amplitude (APA) and action potential duration at 20%, 50% and 90% (APD20, APD50 and APD90).INa-T was measured, I-V curves and peak currents at different frequencies were detected by whole cell patch clamp before and after propafenone perfusion at 10 μmol/L. Results: There was no statistical difference in MDP at before and after propafenone perfusion as (-80 ± 6) mV vs (-82 ± 5) mV,P>0.05. After perfusion, APA was signiifcantly decreased as (95 ± 12) mV vs ( 125 ± 10) mV,P<0.05, the Vmax slowed down as (330 ± 43) V/s vs (420 ± 54) V/s,P<0.05, while APD20, APD50 and APD90 were unchanged as (8 ± 2) ms vs (6 ± 2) ms,P>0.05, (16 ± 3) ms vs (12 ± 3) ms,P>0.05 and (86 ± 14) ms vs (85 ± 12) ms,P>0.05. After propafenone perfusion, I-V curve ofINa-T was shifted upward and the peak current was decreased as (3001 ± 383) pA vs (4193 ± 378) pA, P<0.05. Before perfusion, when stimulated at 0.06 Hz, 1 Hz, 2 Hz, 5 Hz and 10 Hz, there were no signiifcant use-dependent block inINa-T , and no real difference inINa-T between the 10th and 1st pulse,P>0.05. After perfusion, no significant use-dependent block was observed when stimulated at 0.06 Hz and 1 Hz,P>0.05, while at 2 Hz, 5 Hz and 10 Hz, propafenone perfusion demonstrated signiifcant use-dependent block uponINa-T with the inhibition fractions of (22 ± 11)%, (38 ± 14)% and (52 ± 17)% respectively, those were signiifcantly different from the inhibition fractions at either 0.06 Hz or 1Hz,P<0.05. When the inhibition fractions were compared by each 2 conditions, allP<0.05. Conclusion: Propafenone may slow down the Vmax of AP, reduce APA and without the impact on APD; the effects onINa-T is not only in tonic block, but also more obviously in use-dependent block in isolated ventricular myocytes of New Zealand rabbit. Such inlfuences minimized the impact on QT interval and meanwhile, decreased the incidence of brad arrhythmia.
