OBJECTIVE:To prepare warfarin sodium granules(WSG)and to establish it’s quality control methods .METHO_DS:To prepare WSG with part.aeq.geometric method,to optimize the amounts of all sorts of agents in the formulation in terms of dissolution time and abilityof WSG by orthogonal experiment.To determin the content of WS by UV. RESULTS:The formulation was designed and optimized,the WSG’s size were evenly.The concentration of WS was linear within the range of 5.92～17.86?g/ml with r=0.9 998(n=7),the average recovery rate was 101.26%～101.84%(n=5)with the RSD lower than 1.0%.The solution dissolved by granules is stable within 10 days.CONCLUSIONS:The preparation method is simple and feasible,the content determination methods is accurate,realible and sensitive,suitable for the quality control of WSG.The dissolved solution can be divided in accurate doses.

Objective To collect information of patient doses of interventional radiology in Beijing Xuanwu Hospital,and investigate correlation between the peak skin dose(PSD) and dose-area product(DAP).Methods Radiation doses from 135 patients have been studied including 84 coronary angiographies(CAG) and 51 percutaneous transluminal coronary angioplasties(PCI).Dose-area product(DAP) values,cumulative dose(CD) at interventional reference points,fluoroscopy times,total number of cine frames were collected for each patient.Skin dose measurements were made with thermoluminescent dosimeters(TLD) placed as a 10 ? 9 arrays of TLDs on the body.The grid of TLDs arrays was 5 ? 4 cm.Results Mean values for dose-area product were 2690.84 ?Gym2 for CAG and 7946.91 ?Gym2 for PCI.Mean values for CD were 431.6 mGy cm2 for CAG and 1395.3 mGy for PCI.Mean fluoroscopy times were 2.9 min for CAG and 10.9 min for PCI and mean number of frames were 544 and 945 for CAG and PCI,respectively.PSD values ranged from 26.18 to 120.37 mGy for CAG and 38.91 to 184.79 mGy for PCI.The relationship between DAP and PSD was r = 0.52 for CAG and r = 0.54 for PCI.The correlation of PSD with CD was r = 0.45 for CAG and r = 0.53 for PCI.Conclusion Comparison shows that patients DAP,CD and fluoroscopy time values were comparable with other publications.Skin dose values of investigated patients are below the threshold dose for radiation skin injury(2 Gy).There is no good relationship between DAP and PSD.So calculation of individual maximum skin dose based on DAP data is not reliable and needs to find a new reference value for skin dose.(J Intervent Radiol,2007,16:222-225)

AIM: To investigate the constituents of the effective fractions from Shenshao Xinxin Prescription(Radix et rhizoma ginseng;Radix paeoniae rubra;Radix puerariae lobatae). METHODS: The compounds were isolated by column chromatography on silica gel,polyamide,sephadex gel and their structures were determined on the basis of their spectroscopic evidence including UV,FAB-MS and NMR data. RESULTS: Eight compounds had been isolated and identified as daidzein,4′-O-methyl-daidzin,genistin,PG-3,puerarin,paeoniflorin,daidzein 8-C-Apiosyl(1→6) glucoside,4′,7-diglucoside. CONCLUSION: The compounds obtained were the alcohol-soluble compounds from Shenshao Xinxin Prescription.

Objective To expand the NKT cells in vitro and to determine the cytokine releasing of NKT cells during proliferation.Methods Several kind of methods were established to expand TCRV ?24 +/TCRV ?11 + NKT cells from PBMC or purified T lymphocyte.The levels of IL 4,IFN ?and TNF ? of NKT cells were determined by flow cytometry.Results After 19 days' expansion,the largest number of NKT cells increased (23.0?16.7)?10 3 times and the largest ratio of NKT cells to total T lymphocyte was (25.5?7.2)%.The ratio of TCRV ?11 + NKT cells that secreted IL 4,IFN ?and TNF ? was higher than the TCRV?11 T lymphocytes.Conclusion TCRV ?24 +/TCRV ?11 + NKT cells can be expanded in vitro with ? Galcer presented by CD 1d molecule.The NKT cells can also be expanded from PBMC because the monocyte lineage cells within PBMC can express CD 1d molecule and present specific glycolipids like ? Galcer to them.

Objective: To estalish a rat model system in which periapical lesions were induced by pulp exposure and infection from the oral environment. Methods: The pulps of mandibular molar of 16 SD rats were surgically exposed and kept open to be induced for infection from the oral environment. The rats were killed 1, 2, 3, 4 weeks after operation respectively. Radiographic image analysis was performed by means of computer linked to a video image digital analysis system. In addition, specimens were decalicified, sectioned, HE stained and examined under microscope. Results: Periapical lesions were readily detectable as early as 1 week after pulp exposure. A significant increase in lesion area was detected from 1 to 3 weeks in a time-dependent manner, the lesion declined a little bit in 4 weeks. Slight inflammatory cell infiltration and alveolar bone resorption were observed in the periapical tissue in the 1-week-samples, total pulp necrosis in the 2-week-samples, apical abscess and bone destruction in the 3-week-samples. Decreased infiltration and the matured osteoblasts were found on the surface of the bone in the 4-week-samples. Conclusion: Pulp exposure can induce periapical lesion in rat. A period of rapid lesion development occurs between day 7 and day 21 after pulp exposure (active phase), and a period of slower development follows thereafter (chronic phase).

Objective To develop a new type of gene vaccine against plague. Methods caf1 gene of caf Operon Encoding F1 Antigen of Y. pestis was inserted into pHSS6-mTn-3xHA/lacZ library plasmids, the recombinant plasmids were linearized with Not I and transformed into yeast cell by lithium acetate (LiAc) method to induce homologous recombination. Positive recombinants were then selected with uracil-lack medium. Results These recombinants were confirmed by colony PCR to have the target gene fragments. Conclusion The present study provided a platform for constructing a novel expression system for expressing Y. pestis F1 Antigen via homologous recombination in Saccharomyces cerevisiae, which may contribute to the genetic prevention of plague through digestive-tract-route (DTR).

Neural crest stem cells originated from hair follicle (epidermal neural crest stem cell, EPI-NCSC) are easy to obtain and have potentials to differentiate into various tissues, which make them eminent seed cells for tissue engineering. EPI-NCSC is now used to repair nerve injury, especially, the spinal cord injury. To investigate their effects on repairing peripheral nerve injury, EPI-NCSC from a GFP-SD rat were primarily cultured on coated dishes and on a poly lactic acid coglycolic acid copolymer (PLGA) membrane. Methyl thiazolyl tetrazolium (MTT) assay showed that the initial adhesion rate of EPI-NCSC was 89.7% on PLGA membrane, and the relative growth rates were 89.3%, 87.6%, 85.6%, and 96.6% on the 1st, 3rd, 5th, 7th day respectively. Cell cycles and DNA ploidy analysis demonstrated that cell cycles and proliferation indexes of cultured EPI-NCSC had the same variation pattern on coated dishes and PLGA membrane. Then cultured EPI-NCSC were mixed with equal amount of extracellular matrix and injected into a PLGA conduit to connect a 10 mm surgery excision gap of rat sciatic nerve, Dulbecco's Modified Eagle's medium (DMEM) was used to substitute EPI-NCSC in the control group. After four weeks of transplantation, the defected sciatic nerve achieved a histological restoration, the sensory function of rat hind limb was partly recovered and the sciatic nerve index was also improved. The above results showed that a PLGA conduit filled with EPI-NCSC has a good repair effect on the peripheral nerve injury.

Objective To investigate the infection situation of L-form bacteria in urinary calculi from the patients with urolithia-sis in a hospital to provide a scientific basis for postoperative anti-infection and prevention of urinary stone recurrence.Methods The calculi samples in 265 cases of urinary calculi from October to December 2015 were collected and performed the culture of com-mon bacteria and L-form bacteria respectively.Culture of common bacteria and bacterial L-forms.Results Among 265 cases of uri-nary calculi ,8 cases(3% ,8/265) were L-form Bacterial combined with common bacterial infection ,only 7 cases(2.6% ,7/265) were L-form bacterial infection ,80 cases (30.0% ,80/265) were common bacterial infection.15 strains of L-form bacteria were detected and 96 strains of common bacteria were detected.The drug resistance of L-form bacteria was significantly increased compared with common bacteria.Conclusion The positive rate of L-form bacteria culture of urinary calculi is lower than other domestic reports. Adding hypertonic medium for conducting L-form bacterial isolation and culture in the patients with urinary tract infection can re-duce the false negative.

Objective To generally analyze the current situations of clinical research and applications in early enteral nutrition (EEN) after abdominal surgery. Methods The published papers about the current situations of clinical research and applications in EEN after abdominal surgery were reviewed. Results EEN after abdominal surgery seems to be safe and effective, produces a positive nitrogen balance, keeps the integrality of structure and function of the apparatus, protects gut barrier, and reduces or prevents septic complications. Conclusion EEN may be of more benefits and will be one of the best methods of nutrition support after abdominal surgery.
