1.Comparison of two experimental ulcerative colitis models in rat

Objective:To compare the rat models of ulcerative colitis induced by 2,4,6-trinitro-benzene-sulfonic acid(TNBS) and immune-comb method.Methods:SD rats were randomly distributed into groupⅠ(immune-comb method),groupⅡ(TNBS/ethanol),groupⅢ(50% ethanol) and groupⅣ(control).The rats in groupⅠwere sensitized 2 times by antigen made of rabbit colon mucosa in 2-week intervals.After abrosia for 24 hours,each rat in four groups received enema of 0.65 ml different fluids,which were:TNBS 100mg/kg +50%ethanol for groupⅠand Ⅱ,50%ethanol for group Ⅲ,and normal sodium for control group.Rats were sacrificed on day 1,21 and week 8,12 after enema to observe the macroscopical focus of infection in bowels and the microscopical changes.Results:The typical erosion and ulcer changes appeared in groupⅠandⅡ,and there was accompanying pathological changes in the end piece of ileum in groupⅠ.The pathological changes for groupⅠcould keep for 8 weeks,while those for group Ⅱtrended to heal 3 weeks later.The pathological changes in groupⅢonly were hyperemia,oedema and a few anabrosis or superficial ulcer,keeped for 21days.There was no tissue damage in group Ⅳ.Conclusion:Immune-comb method is comparatively the ideal method to establish the model,,because it has a good reproducibility,a long persistence time,and the pathological changes resemble more to those of man..
2.Endosomatic transference of transplanted bone marrow mesenchymal stem cells in rat of ulcerative colitis

Objective:To investigate the transference of transplanted bone marrow mesenchymal stem cells (BMSCs)in rat of ulcerative colitis. Methods:SD rats were randomly distributed into group A colitis induced by immune-combined TNBS/ethanol and group B as control. All rats received caudal vein injection with 1 ml of fluorescence marked BMSCs,then 5 rats were sacrificed at day 3,7 and 14 respectively in each group. Cryostat sections of bone marrow,lung,liver,spleen and gut were made to observe the distribution of BMSCs in these organs,and the fluorimetric integral optical density(IOD)were analyzed. Results:BMSCs were mainly distributed in organs which richs vessels. The highest IOD level of BMSCs in lung was on the 7th day,and was on the 14th day in bone marrow. On the 14th day,the IOD level of BMSCs in colon of group A were higher than those of other segments of digestive tube and also higher than those in group B (P
3.Effect of survivin on the apoptosis of non-small lung cancer induced by celecoxib

Objective To investigate the inhibitory effect of celecoxib on non-small cell lung cancer in vitro and the role of survivin.Methods The inhibitory effect of celecoxib was detected by MTT.Flow cytometry and electron microscope were used for evaluation of apoptosis.The expression of survivin was detected by RT-PCR and Western blot.Results ①Celecoxib inhibited cell survives in a time-dependent and concentration-depended manner,and the effect of celecoxib was more significant in NCI-H520 than in A549.②Celecoxib caused apoptosis in concentration-depended manner.3.Celecoxib decreased the expression of survivin.Conclusion The decrease of the expression of survivin may play a critical role in the apoptosis of NSLLC in vitro induced by celecoxib.
5.Assessment of left ventricular function in patients with type 2 diabetes mellitus by quantitative tissue velocity imaging

ObjectiveTo assess the left ventricular systolic and diastolic function in patients with type 2 diabetes mellitus(DM) using quantitative tissue velocity imaging(QTVI) ,and to provide reliable evidence for early diagnosis and prevention in diabetic cardiomyopathy.MethodsOne hundred and twenty one type 2 DM patients were divided into two DM 1 groups (61 patients without microangiopathy) and DM2 group (60 patientswith microangiopathy).Fifty normal subjects were enrolled as control group.The index of echocardiography,including the LVEF, FS and mitral peak flow velocity during early and late diastole (E/A) were measured by conventional echocardiography,and the ratio of E/A was calculated.The average peak velocities of six LV wall sites at mitral annuluses during systole, early and late diastole(MEm, MEm, MAm) were measured by QTVI, and the ratio of MEm/MAm was calculated.Results Compared with the normal group, MSm ([7.13 ± 1.42])cm/s vs.([6.49 ± 1.29]cm/s), MEm ([6.22 ± 1.39]cm/s) vs.([4.53 ± 0.94]cm/s) and MEm/MAm ([0.79 ±0.17]vs.[0.59 ± 0.19]) in DM patients were significantly decreased(F = 5.32,8.01 ,4.89 ; Ps <0.05).There was no significant differences among three groups in the comparisons of LVEF ([67.45 ±5.47]%),([65.91 ±4.83]%),([68.01 ±6.16]%) and FS([38.84±4.23]%,[37.82±5.43]%),([40.17 ± 4.53]%)(F = 1.89 and 2.46 respectively, P > 0.05) .In addition, E/A of DM2 group (0.71 ±0.21)decreased more dramatically than DM1 and normal (0.91 ± 0.18,1.02 ± 0.24)(F = 4.71, P < 0.05)ConclusionCompared with EF,FS and E/A obtained by conventional echocardiography,QTVI-derived MSm,MEm and MEm/MAm are more sensitive indexes to defect early LV dysfunction.The functional disorder appears early than microangiopathy, and the left ventricular systolic and diastolic function gets worse along with the microangiopathy.
6.Detection of Hb constant spring mutation by four of primers allele-specific PCR

Objective To develop a rapid method based on ARMS, named four primers allele-specific PCR(4p-AS PCR), to detect the most common non-deletional ?-thalassemia mutation Hb Constant Spring(Hb CS) by PCR technique. Methods The 4p-ASPCR was used to detect the Hb CS mutation of ?-thalassemia in 38 DNA samples of Hb H disease patients and using PCR-RE and DNA sequencing to confirm the results. Results Among the 38 Hb H disease patients 15 cases was revealed to carry Hb CS, 16 cases were deletional Hb H, and 7 cases need to be defined.Conclusion A simple, rapid and reliable method, named 4P-ASPCR, to detect Hb CS mutation have been developed. It is also may be useful in screen other point mutations such as Hb Quong Sze.
7.Assessment of left ventricular hypertrophy and impaired diastolic function in elderly hypertensive patients by brain natriuretic peptide level in combination with tissue doppler imaging

Objective To investigate the clinical values of brain natriuretic peptide (BNP) in combination with TDI in diagnosing left ventricular hypertrophy (LVH) and impaired diastolic function in elderly hypertensive patients. Methods The 140 elderly hypertensive patients were divided into LVH group (n=69) and NLVH group (n=71). Control group consisted of 50 normal subjects. Plasma BNP level and index of echocardiography, including mitral peak flow velocity during early and late diastole (E, A), ratio of E/A, average peak velocities of six LV wall sites at mitral annuluses during early and late diastole (MEm, MAm), ratios of MEm/MAm and E/MEm were measured in all patients. The correlation of plasma BNP level with cardiac ultrasonographic findings was also examined. Results The level of BNP [(61.64±37.18)ng/L, (138. 65±30. 23)ng/L] and the ratio of E/MEm (11.3±1.83, 15.7±1.45) were significantly higher in NLVH group and LVH group than in normal group (P<0. 05 or P<0. 01). MEm [(6.32±0. 94)cm/s, (4.29±0. 91)cm/s]and MEm/MAm (0.76±0.19, 0.51±0. 11) were significantly lower in NLVH and LVH group than in normal group (P<0.05 or P<0. 01). The BNP level was negatively correlated with E/A, MEm and MEm/MAm (r=- 0. 294, r= 0. 387 and r= 0. 422, all P<0. 01), and was positively correlated with LVMI and E/MEm (r=0.342, r=0.501, all P<0.01). Conclusions Left ventricular diastolic function is impaired in elderly hypertension patients regardless of LVH or NLVH.Plasma BNP level in combination with echocardiography parameter is accurate to evaluate the LVHand impaired diastolic function in elderly hypertensive patients.
8.Total laparoscopic splenectomy and pericardial devascularization for the treatment of portal hypertension

Objective To evaluate total laparoscopic splenectomy combined with pericardial devascularization. Methods In this study,40 patients with portal hypertension and hypersplenism underwent total laparoscopic splenectomy combined with pericardial devascularization,in which 26 cases had a history of up-GI bleeding. Results Procedures were successful in 36 patients.There was four conversions to open surgery.The operations were completed in a mean time of (224 ± 54) min,with a mean estimated blood loss of (296 ± 209) ml and with no serious complication. Conclusion Total laparoscopic splenectomy combined with pericardial devascularization is a safe and effective available minimally invasive procedure in patients with portal hypertension and hypersplenism.
9.Application of CRISPR/Cas9 system in virus research

To date,the clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 system has been widely used to edit the genome in many species and cells.The system is the third generation of artificial endonuclease,which can edit DNA by recognizing short DNA sequences.This paper reviews the structural features of the system and its application in virus research,such as the functional studies of virus-related genes and the exploration of antiviral therapies (including HIV,HBV,and EB virus),looking forward to the future direc-tion of virus research.
10.Progress of research on mTOR inhibitor in endocrine and HER-2 targeted therapy resistance in breast cancer

Mammalian target of rapamycin(mTOR)locates at the downstream of phosphatidylinositol 3 kinase(PI3K)-protein kinase B(Akt)cell signal transduction pathway. Studies find that the abnormal activa-tion of this pathway is correlated with the endocrine and drug resistance of anti human epidermal growth factor receptor-2(HER-2)target therapy in breast cancer. The combination with mTOR inhibitors based on the past traditional drugs can block the pathway and reflect a favourable application prospect in preventing the develop-ment of resistance and restoring the initial sensitivity on tumor cells. mTOR inhibitors are expected to be the new hope for the treatment of breast cancer.
