1.Content Determination of Icariin in Fucongxiang Liquid by HPLC

OBJECTIVE:To establish the content determination method of Icariin in Fucongxiang liquid by HPLC.METHODS:HPLC was performed to determine Icariin on VP-ODS column.The mobile phase was acetonitrile:water(30∶70).Detecting wavelength was270nm.RESULTS:The detected concentration of Icariin was in the good linear range of2.8~28.0?g/ml(r=1.0000),and the average recovery was100.03%(RSD=0.54%,n=5).CONCLUSIONS:The estab-lished method is simple,sensitive,accurate and available for the quality control of Fucongxiang liquid.

3.Clinical investigation of rheumatoid arthritis associated lung disease

Objective To identify the clinical features and risk factors of rheumatoid arthritis-associated interstitial lung disease (RA-ILD).Methods The clinical and laboratory data of 595 patients with RA hospitalized in Chaoyang Hospital.550 cases of patients were performed pulmonary high-resolution CT examination.Risk factors were analyzed with logystic regression analysis.Results ① There were 237 (39.8%)patients with RA-ILD.13.5% of RA-ILD occurred before the dia-gnosis of RA,69.7% of RA-ILD occurred within ten years after the diagnosis of RA,16.8% of RA-ILD occurred more than ten years after the diagnosis of RA.② The most common manifestations of HRCT were reticular opacity (57.8%),pleural thickening (57%),ground-glass opacity (53.2%),followed by septal thicke-ning,nodules,bullae,cellular-like changes and bronchiectasis.③ Pleural disease,bronchiectasis,pulmonary hypertension in RA patients were associated with RA-ILD.④ Compared with the RA-N-ILD patients,RA-ILD patients had a higher proportion of male patients,higher rate of smoking,older mean age at onset,shorter duration,more swollen joints higher levels of lactate dehydrogenase,globulin,erythrocyte sedimentation,C-reactive protein and rheumatoid factor,the difference was statistically significant (P<0.05).The forced vital capacity,forced expiratory volume in one second,carbon monoxide diffusion rate was lower,the difference was statistically significant (P<0.05).Logistic regression analysis showed that age,smoking,elevated lactate dehydrogenase,RF-positive rate were relevant factors of RA-ILD.Conclusion ILD is common in RA,and most of it occurrs within ten years after the diagnosis of RA,and often coexists with other lung disease.Age,smoking,elevated lactate dehydrogenase,RF-positive rate

5.Inhibitory effect of rapamycin on proliferation of H_(22) hepatic cancer in mice

Objective To explore rapamycin's inhibitory effect on proliferation of H_(22) hepatic cancer in mice. Methods In vitro study: H_(22) hepatic cancer cell lines were cultured with rapamycin, CsA, FK506, and proliferation was determined through MTT. The influences of different agents on the H_(22) hepatic cancer cell cycle were observed by flow cytometry. The vascular endothelial growth factor (VEGF) concentration of the supernatant fluid of the cultured H_(22) hepatic cancer cell was detected by ELISA. In vivo study: C57BL/6 to Balb/c mice allogenic skin transplant was established, and the H_(22) hepatic cancer cell was implanted under skin. Rapamycin, CsA, FK506 and 5-FU were fed to the mice, respectively. The effect of different immunosuppressors on the survival of skin graft was observed while the proliferation of the transplant tumor was investigated. VEGF concentration of treated mice serum was examined by ELISA. The microvessel density of the transplanted tumor was observed through immunohistochemistry staining of CD34. Results The proliferation of the H_(22) hepatic cancer cells was inhibited by rapamycin at the concentration different dose of rapamycin, the VEGF concentration of the supernatant fluid decreased significantly (P<0.05). The number of S phase cells decreased significantly compared to that of other agents (P<0.05). When rapamycin, the lengthened survival time of the skin grafts was similar to that in CsA and FK506 groups. But the tumor volume was smaller than that in CsA and FK506 groups (P<0.05). Compared to that in the control group, the VEGF concentration of mice serum decreased in rapamycin group (P<0.05), and the microvessel density of the transplant tumor was reduced greatly (P<0.05). Conclusion Rapamycin, as an immunosuppressor, significantly resists immunologic rejection and inhibits the proliferation of H_(22) hepatic cancer, thus having its advantage in treating malignant hepatic cancer with liver transplantation.

People wearing positive pressure biological protective suits might face some potential safety risks when work -ing in a high-risk biological contamination environment .These risks could be effectively prevented by simulation experi-ments.This paper elaborates on the progress in risk studys on positive pressure biological protective suits in terms of dynam -ic detection methods and risk assessment , and points out the problems to be solved and the direction of future development .
7.ERCP in patients with prior BillrothⅡgastrectomy

Objective To evaluate the efficacy and safety of endoscopic retrograde cholangiopancre-atography (ERCP)by using a duodenoscope in patients with prior Billroth Ⅱ gastrectomy.Methods Data of 98 patients(including 7 who underwent Billroth Ⅱand Braun)with prior Billroth Ⅱ gastrectomy who un-derwent ERCP from January 2008 to May 2014 at Endoscopy Center of Ruijin Hospital were retrospectively studied.Results A total of 98 patients accepted 111 ERCP procedures,the overall success rate was 75.7%(84 /111).The success rate of accessing Vater′s papilla was 82.0%(91 /111)and the selective can-nulations success rate was 92.3%(84 /91).In the cases of successful access,therapeutic procedures were achieved in 100.0%(84 /84)of all patients.The overall rate of complications was 4.5%(5 /111),with mild post-procedure pancreatitis occurring in 3 cases,cholangitis in 2 cases,respectively.Conclusion ERCP u-sing a duodenoscope can be safely and effectively performed in patients with prior Billroth Ⅱ gastrectomy with high cannulation and treatment success rates.
8.Determination Optimization of Total Amino Acids in Runing Pills by Orthogonal Design

Objective:To optimize the determination technology for the total amino acids in Runing pills. Methods:The process was optimized by L9 (34 ) orthogonal design using the hydrolysis temperature, hydrolysis time and hydrochloric acid concentration as the evaluation factors,and the content and yield of total amino acids as the evaluation indices. Results: The optimal determination condi-tions were as follows:the hydrolysis temperature was 110℃, the hydrolysis time was 22 h and the hydrochloric acid concentration was 6 mol·L-1 . Conclusion:The optimal determination technology can be applied in the total amino acids from Runing pills,which pro-vides the basis for the further research of Runing pills.
9.Isolation, culture and identification of neural stem cells in new-born rats.

The cortexes were obtained from new-born rats and dissociated to single cells by triturating. The cells were cultured in neural stem cell (NSC) culture medium (DMEM supplemented with bFGF, EGF and B27) and formed primary neurospheres after 7 days. Single cells dissociated from neurosphere were cultured in 96-well plates and formed single-cell cloning neurosphere 7 days later. The primary and single-cell cloning neurospheres were both positive for the immunofluorescent staining of nestin and were identified as NSC. It was proved that NSC can be expanded in vitro and provide seed cells for neural tissue engineering.

10.Surgical Treatment of Infected Femoral Artery Pseudoaneurysm

Objective To evaluate surgical treatment of infected femoral artery pseudoaneurysm.Methods The data on surgical treatment of 45 patients with infected femoral artery pseudoaneurysm admitted from January 2003 to June 2008 were analyzed retrospectively.Fourty-three patients underwent operative treatment including excision of infected femoral artery pseudoaneurysm,exhaustive debridement and bypass graft with vascular prosthesis.Two patients were unavoidable to undergo removing of infected femoral artery pseudoaneurysm and ligating the proximal and distal artery of pseudoaneurysm because of severe infection and large volume.Results The patients were followed up from 3 to 12 months(mean 7.82 months).The limbs of all the patients underwent bypass graft with vascular prosthesis were salvaged successfully,patients of which had secondary wound healing and had not intermittent lameness.One of two patients performed ligation of artery was salvaged successfully but had severe intermittent lameness,another patient underwent high amputation above knee because of ischemic gangrene.ConclusionFor infected femoral artery pseudoaneurysm,the operative treatment including excision of infected femoral artery pseudoaneurysm,exhaustive debridement and bypass graft with vascular prosthesis is effective and safe.
