1.Treatment of Seinsheimer type V subtrochanteric femoral fractures with inversive LISS plate combined with steel wire.

OBJECTIVETo study clinical effects of inversive LISS (less invasive stabilization system, LISS) plate combined with steel wire for the treatment of Seinsheimer type V subtrochanteric femoral fractures.
METHODSFrom January 2009 to Janurary 2012, 11 patients with Seinsheimer type V subtrochanteric femoral fractures were treated with inversive LISS plate combined with steel wire, including 8 males and 3 females, ranging in age from 35 to 61 years old (averaged, 51 years old). Causes of injury: 8 patients caused by accident and 3 patients caused by falling down. According to X-ray examination, all the patients were type V subtrochanteric femoral fractures and were treated with LISS plate combined with steel wire. Merle d'Aubigne scores were used to evaluate therapeutic effects on hip.
RESULTSThe mean period of follow-up was 16 months (ranged, 12 to 28 months). All the patients obtained bone union for average 3.8 months (ranged from 3 to 4.3 months). There were no complications such as deep infection,deep vein thrombosis, pulmonary embolism and bone nonunion. According to Merle d'Aubigne scores, 7 patients got an excellent result and 4 good.
CONCLUSIONInversive LISS plate combined with steel wire for the treatment of Seinsheimer type V subtrochanteric femoral fractures is satisfactory, Which can provide stable fixation for fracture end and provide an ideal internal fixation method for treatment of this kind of fractures in the future.

2.Comparison of Dissolubility of Lansoprazole Enteric- coated Tablets With That of Enteric- coated Capsules

OBJECTIVE: To study the method of measuring the in vitro dissolubility of lansoprazole enteric- coated tablets and to find out the difference between the dissolubility of lansoprazole enteric- coated tablets and that of lansoprazole enteric- coated capsules.METHODS: To establish a measuring method of the dissolubility of lansoprazole enteric- coated tablets and compare the dissolubility of lansoprazole enteric- coated tablets with that of lansoprazole enteric- coated capsules and to acquire the dissolving parameters for variance analysis.RESULTS: (1)The accumulative dissolving quantity of the both preparations exceeded 80% in 30 minutes.(2)The comparison of Weibull parameters,T50,Td and m,between each other in group A showed no significant differences(P>0.05).It was the same for those of group B.(3)The parameters,T50 and Td,of group A,in comparison with those of group B,showed significant differences(P<0.001).The times needed for dissoiving 50% and 63.2% of quantity in group B were both shorter than those in group A.The m parameters in different groups had no significant differences(P>0.05).There was no significant difference in slope of equation between two groups.CONCLUSION: This measuring method of the in vitro dissolubility of lansoprazole enteric- coated tablets is simple,convenient,accurate,stable,feasible and good in repetition.It is concluded that there is significant difference between the dissolubility of the two dosage forms of enteric- coated preparation of lansoprazole.
3.Expression of follistatin-like protein 1 in the peripheral blood and intestinal tissue of patients with ulcerative colitis and its clinical significance

Objective To examine the expression of follistatin-like protein 1 (FSTL1) in the peripheral blood and intestinal mucosa tissues of ulcerative colitis (UC) patients and analyze the correlation between its expression and the activity of UC.Methods From October 2010 to June 2012,sixty patients with UC were collected.From April 2012 to October 2012,thirty individuals without any obvious mucosa lesion under colonoscope and confirmed by pathological examination were set as control group.The serum expression level of FSTL1 of both UC group and control group were determined by enzyme linked immunosorbent assay (ELISA).t-test was performed for comparison between groups.The expression of FSTL1 in the intestinal mucosa of UC group and control group was detected by immunohistochemistry.Chi-square test was used for comparison between groups.The patients with UC were scored with ulcerative colitis disease activity index (UCDAI).Its correlation with plasma FSTL1 was analyzed by Pearson correlation coefficient.Results The serum expression level of FSTL1 of UC group ((14.37-±-1.80) μg/L) was higher than that of control group ((5.80±0.72) μg/L)and the difference was statistically significant (t=25.01,P< 0.05).The serum expression level of FSTL1 of UC group was positively correlated with UCDAI (r=0.814,P<0.05).The positive expression rate of FSTL1 in the intestinal mucosa tissues of UC group (86.7%,52/60)was higher than that of control group (46.7%,14/30) and the difference was statistically significant (x2 =52.334,P<0.05).Conclusions The expression of FSTL1 of UC patients increases and is positively correlated with disease activity.FSTL1 may play a role in the development of UC.
5.Clinical observation on carbon dioxide fractional laser combined with liquid nitrogen cryotherapy for flat warts

Objective To evaluate the clinical efficacy and safety about carbon dioxide fractional laser combined with liquid nitrogen cryotherapy for flat warts.Methods Eighty-three patients with flat warts were divided into 2 groups according to the treatment method,42 patients in treatment group were treated by carbon dioxide fractional laser combined with liquid nitrogen cryotherapy,41 patients in control group were treated by liquid nitrogen cryotherapy alone.As a period for 10 days,2 groups were given 2 periods of treatment.The therapeutic effect,recurrence rate and adverse reaction were compared between 2 groups.Results The cure rates of 1-period and 2-period in treatment group were significantly higher than those in control group [73.81%(31/42) vs.46.34%(19/41) and 97.62%(41/42) vs.75.61%(31/41)],the recurrence rate and incidence of adverse reaction in treatment group were significantly lower than those in control group [2.44%(1/41) vs.19.35%(6/31) and 9.52%(4/42) vs.31.71%(13/41)],there were statistical differences (P < 0.05 or < 0.01).Conclusion Carbon dioxide fractional laser combined with liquid nitrogen cryotherapy for fiat warts is a safe and effective treatment with low recurrence rate.
6.Follow-up and retreatment strategies in the anti-vascular endothelial growth factor-A therapy for neovascularization of age-related macular degeneration

Wet age-related macular degeneration (AMD) is one of major causes of blindness in elder people.Intraocular injection of anti-vascular endothelial growth factor (VEGF) -A regimen has made big breakthrough for the treatment on choroidal neovascularization of wet AM D,while long-term follow-up and necessary retreatments are the key issues to remain obtained visual acuity.Multiple strategies of wet AMD have been used in following-up and retreating based on the visual acuity,optical coherence tomography (OCT),ophthalmoscope and fluorescein fundus angiography (FFA) in abroad.However,there also are major differences in the patient' s composition,treatment habits and distribution of medical sources in China from Western.So we suggest to standardize the follow-up and retreatment strategies about intravitreal injection of VEGF-A for wet AMD as to achieve a better effectiveness.OCT-guided individual follow-up and retreatment strategies should be very helpful for maintaining a long-term efficacy,minimizing the treatment time and reducing medical cost.
7.IKVAV-containing peptide self-assembly into hydrogel

BACKGROUND:IKVAV-containing peptide sequence is an active region that promotes the adhesion, growth, and differentiation of neural cells in the laminin. It can be fabricated into a novel tissue-engineered scaffold material by self-assembly into hydrogel.OBJECTIVE: This study was designed to synthesize IKVAV-containing (C16H31O-AAAA GGGEIKVAV) peptide. The peptide was observed self-assembly into three-dimensional and porous hydrogel after triggered with phosphate buffered saline (PBS).DESIGN, TIME AND SETTING: Single-sample experiment, performed in the Laboratory of Department of Orthopedics,Union Hospital Affiliated to Tongji Medical College, Huazhong University of Science and Technology between September 2004 and January 2005.MATERIALS: IKVAV-containing peptide was synthesized by solid phase method.METHODS: Peptide purity and relative molecular mass were detected by high performance liquid chromatogram and mass-spectrometry, respectively. 1% peptide, whose pH value was equal to 9.5, was self-assembled into hydrogel with the addition of PBS. The ultramicrostructure of the hydrogei was observed through the use of transmission electron microscope (TEM).MAIN OUTCOME MEASURES: Peptide purity and relative molecular mass were detected. Moreover, gross observation of the peptide after self-assembly and TEM observation of peptide ultramicrostructure were performed.RESULTS: Peptide relative molecular mass was 1351.6, which was in accordance with its theoretical value. Peptide purity was 95%. After triggered with PBS, 1% peptide was self-supported into hydrogel in a few seconds. TEM results showed that self-assembled hydrogel consisted of the interconnected nanofibers which varied from 3 to 6 nm in diameter and 100 to 1 500 nm m in length.CONCLUSION: The IKVAV-containing peptide was synthesized and self-organized successfully into porous hydrogel,which was triggered with PBS solution.
8.Therapeutic effect of nitrate on dextran sulfate sodium induced acute experimental colitis in mice

Objective To investigate the effect of nitrate on acute experimental colitis in mice.Methods A total of 40 BALB/c mice were evenly divided into model group and treatment group.Model group were fed with 4% dextran sulfate sodium (DSS) solution and treatment group were given 4% DSS solution and nitrate (1.5 g/L) for seven days.The disease activity index (DAI) of mice was scored.The colon tissue of mice was taken for hematoxylin-eosin staining and myeloperoxidase (MPO)immunohistochemical staining observation.The MPO and activity of nitric oxide in colon tissue were measured by MPO and nitric oxide detecting kit.The data were analyzed by t test.Results At the 6th day and 7th day,the difference of DAI between treatment group and model group was statistically significant (t=5.12 and 6.72,P=0.012 and 0.008).At the 7th day,the tissue score of model group (2.5±0.5) was higher than that of treatment group (1.9±0.4) and the difference was statistically significant (t=3.82,P< 0.01).Compared with model group,the histopathological injury of colon tissue in treatment group mice significantly reduced and neutrophil infiltration also decreased.At the 7th day,the concentration of MPO,NO2-and NO3-of model group was (2.8±0.6) U/g,(10.4±4.3) mmol/g and (100.3±50.1) mmol/g respectively,treatment group was (1.5±0.3) U/g,(17.5±7.0) mmol/g and (190.7 ±85.3) mmol/g respectively.The differences were statistically significant (t=11.23,3.81 and 4.50,all P<0.01).Conclusion Nitrate can reduce DSS-induced acute experimental colitis in mice.
9.Quantitative Determination of Selenium Species in Se-enriched Yeast Using Liquid Chromatography-Hydride Generation Atomic Fluorescence Spectrometry

A rapid and sensitive method has been developed for the simultaneous determination of four selenium species Se(Ⅵ), Se(Ⅵ), selenomethionine, and Se-methylselenocysteine in Se-enriched yeast by liquid chromatography-hydride generation atomic fluorescence spectrometry (HPLC-HG-AFS). The isolation of the analytes from yeast samples was accomplished by proteaseⅩⅣ and trypsin enzymatic digestion. The target compounds were separated on a PRP-X100 anion exchange column and analyzed by HG-AFS. The mobile phase was 20 mmol/L (NH4)2HPO4. Good linearity was obtained for all the selenium species, with linear correlation coefficients higher than 0. 9996. The LODs of the four species were between 0. 5 and 5. 0 μg/kg. Average recoveries for the four analytes were in the ranges of 82 . 5%-101 . 2%, with intra-and inter-day RSD lower than 8. 6% and 14. 5, respectively. The proposed analytical method is simple, sensitive, with low operation cost, making it applicable for the determination of the selenium species in Se-enriched feeds.
10.Identification and early diagnosis for traditional Chinese medicine-induced liver injury based on translational toxicology.

Recently traditional Chinese medicine (TCM)-induced liver injury has been an unresolved critical issue which impacts TCM clinical safety. The premise and key step to reduce or avoid drug-induced liver injury (DILI) is to identify the drug source of liver injury in early stage. Then the timely withdrawal of drug and treatment can be done. However, the current diagnosis of DILI is primarily governed by exclusive method relying on administering history supplied by patients and experience judgment from doctors, which lacks objective and reliable diagnostic indices. It is obvious that diagnosis of TCM-induced liver injury is especially difficult due to the complicated composition of TCM medication, as well the frequent combination of Chinese and Western drugs in clinic. In this paper, we proposed construction of research pattern and method for objective identification of TCM-related DILI based on translational toxicology, which utilizes clinical specimen to find specific biomarkers and characteristic blood-entering constituents, as well the clinical biochemistry and liver biopsy. With integration of diagnosis marker database, bibliographic database, medical record database and clinical specimen database, an integrative diagnosis database for TCM-related DILI can be established, which would make a transformation of clinical identification pattern for TCM-induced liver injury from subjective and exclusive to objective and index-supporting mode. This would be helpful to improve rational uses of TCM and promote sustainable development of TCM industry.
