3.Study on a Novel Hepatitis B Virus Multi-epitope DNA Vaccine with siRNA,hIL-12 Coexpressing

Objective:To construct a DNA vaccine co-expressing the HBV compound multi-epitope antigen gene, the hIL-12 and the anti-HBV siRNA genes, and to express this DNA vaccine in HepG2 cells. Methods:The HBV multi-epitope antigen gene was designed and synthesized before it was fused with enhanced green fluorescent protein(EGFP) gene, and cloned into the multi-clone site(MCS) of the eukaryotic expression vector pVAX1. The expressinig units of hIL-12 and siRNA were cloned into the BspH I and Mlu I site of pVAX1 respectively. Then the recombinant plasmid pVAX1-siHBV-HB-EGFP-hIL12 was transiently transfected HepG2 cells. The expression of HBV compound multi-epitope gene was observed through EGFP report gene. The expression of hIL-12 was analyzed by ELISA and the effects of anti-HBV siRNA was confirmed with rtPCR . Results: The analysis of enzyme digestion and sequencing both demonstrated that the trible-expressing HBV DNA vaccine has been constructed successfully. The green fluorescent image was detected in the transfected cells which could confirm the expression of the multi-epitope antigen gene. The amount of hIL-12 secretion was 1289pg/ml in supernatant at 48h after transfection and 1712pg/ml at 72h after transfection. The mRNA amount of HBV S gene, which was the siRNA target, had been obviously knockdown. Conclusion: The DNA vaccine co-expressing the HBV compound multi-epitope antigen gene, the hIL-12 and the siRNA genes was constructed and transiently expressed in HepG2 cells, and siRNA had shown us a good anti-HBV effect. It laid a foundation of further study on anti-HBV effect of the new DNA vaccine.
4.Treatment of portal vein tumor thrombosis in patients with hepatocellular carcinoma by TACE

Objective To evaluate the theraputic effect of transcatheter arterial chemoembolization (TACE)for hepatocellular carcinoma with tumor thrombosis of portal vein.Methods One hundred and six patients of hepatocellular carcinoma with tumor thrombosis of portal vein under treament of TACE were observed before and after the procedure.Results After TACE tumor size reduced>50% in 23 patients,<50% in 25, no significant change in 44.The size of tumor enlarged in 12.The disappearance of portal vein tumor thrombosis accessed in 14,with reduction in 39,and no significant change in 51.Two patients died within one week.Conclusion TACE provides good therapeutic effect on hepatocellular carcinoma with tumor thrombosis of portal vein.
5.Purification of recombinant human IE—2 by McAb affinity chromatography

For purifying recombinant human IL—2 (rhIL—2),the columns of immunoabsorptionwere prepared with 4 anti—IL—2 McAb (9B12,9F5,9B2 and 8H7) purified by caprylic acid.Although 4 McAbs differ as regards their antigen—antibody binding characteristics,all they canserve as effective immnoabsorbents,provided optimum condition was adopted.The recoveryrate of 9B12,9F5,8H7 and 9B2 columns were 49.2%,37.5%,31.5% and 18.8% respec-tively.The purity of rhIL—2 obtained was more than 95% and biological activity remainedhigher.
6.Percutaneous coronary intervention in mainland China in 2008: register results.

OBJECTIVETo collect data on percutaneous coronary intervention (PCI) performed in mainland China.
METHODSQuestionnaires on PCI were distributed to all hospitals capable of performing PCI through national society of cardiology. Data in calendar year 2008 were analyzed.
RESULTS(1) 182,312 PCI were performed in mainland China in 2008 (+26.02% vs. 2007), average PCI rate was 1.39 per 10,000 people. (2) PCI was performed in 1061 hospitals and 171.83 PCI was performed per hospital. Among them, there were 461 hospitals performed more than 100 PCI (+54.18% vs. 2007). (3) The number of PCI performed among provinces was positive correlated with local per capita gross domestic product (GDP, β = 0.06, P < 0.05) and the number of the hospitals performed more than 100 PCI per year (β = 434.23, P < 0.05), and negatively correlated with local population number (β = 0.49, P < 0.05). There were no correlation between the ratio of the hospitals performed more than 100 PCI per year (β = -2746.35, P > 0.05) and the ratio of people with medical insurance (β = -855.78, P > 0.05).
CONCLUSIONSFrom 2005, PCI rose 23.87% per year in mainland China. There are significant differences among provinces on the development of PCI.

7.Effects of Electroacupuncture on Activation of Microglia in Peri-infarct Cortex of Cerebral Ischemia-reperfusion Injury Rats

Objective To explore the effect of electroacupuncture on activation of microglia in peri-infarct cortex after cerebral isch-emia-reperfusion in rats. Methods 36 male Sprague-Dawley rats were randomly divided into sham group (n=12), model group (n=12) and electroacupuncture group (n=12). The latter two groups were occluded the left middle cerebral arteries with modified Longa's method for 2 hours and reperfused. The electroacupuncture group received electroacupuncture at Quchi (LI11) and Zusanli (ST36) acupoints for 3 days. The nerve cell damage in peri-infarct cortex was observed with HE staining, while the expression of ED1 was determined with immunohisto-chemical staining, and the expression of tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β) and IL-6 were determined with Western blotting. Results The neurological deficits score improved significantly in the electroacupuncture group (P<0.05), with less nerve cell dam-age, less number of ED1 positive microglia (P<0.05) and less levels of TNF-α, IL-1βand IL-6 (P<0.05), compared with the model group. Conclusion The electroacupuncture at Quchi (LI11) and Zusanli (ST36) acupoints can protect brain from ischemia-reperfusion injury, which might be associated with inhibiting the microglial activation and proinflammatory response in peri-infarct cortex.
8.Effect of Feiyiliu Mixture on Prognosis of Patients with Middle and Advanced Squamous Cell Carcinoma of Lung

Objective::To discuss the effect of Feiyiliu mixture as an adjuvant method for chemotherapy and immune support on progression-free survival (PFS) and survival rate of patients with Ⅲb and Ⅳ stage squamous cell carcinoma of lung (SQCC), and to investigate its intervention effect on tumor markers and cytokines in peripheral blood. Method::One hundred and thirty-two patients were randomly divided into control group (66 cases) and observation group (66 cases) by random number table. Patients in control group received programme of gemcitabine hydrochloride combined with cisplatin (alternative paclitaxel combined with cisplatin) and they also got thymopolypeptides for injection, while the patients in observation group received Feiyiliu mixture, with a treatment course of 4 cycles in both groups. PFS and 12 months' survival rate were recorded during follow-up. Scores of European quality of life collaboration cancer core scale (EORTC QLQ-C30) were graded before and after treatment. Levels of CD3+, CD4+, CD8+, CD4+ /CD8+, cytokeratin 19 fragment 21-1 (CYFRA21-1), squamous cell carcinoma antigen (SCC), carbohydrate antigen 125 (CA125), vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 (MMP-2) and MMP-9 were detected, and size of solid tumor was evaluated before and after treatment. Result::Clinical remission rate of solid tumor was (35/66)53.03% in observation group, higher than(23/66) 34.85% in control group (

·AIM: To investigate the preparation of endostatin protein and its biologic activity on vascular endothelial cell.· METHODS: pBlast-hEndostatin and pBlast-Mcs were identified by digesting with Nhe Ⅰ and Sal Ⅰ, by PCR reaction, by sequencing, and by Alignments of PCR products with gene bank using NCBIBLAST software. The identified pBlast-hEndostatin as well as pBlast-Mcs were then purified with QIAGEN Endofree plasmid maxi kit.The purified plasmids transfected human fibroblasts. The expression of endostatin was detected by RT-PCR, Westem-Blot and immunohistochemistry. The endostatin prorein produced by transfected fibroblasts was purified by ultrafiltration and affinity chromatography. The inhibitory action of endostatin on human umbilical vein endothelium was measured by MTT assay.· RESULTS: pBlast-hEndostatin was found to contain human endostatin gene. Endostatin protein was produced by transfected fibroblasts. The inhibitory ratio of 2.5,5,10,20,40,80mg/L endostatin on human umbilical vein endothelium for 48h were 8.5%,13.1%,27.7%,38.1%,56.7%,63.8% respectively. IC50 value was 34.5mg/L.No inhibition action was found on fibroblasts.·CONCLUSIONS: Endostatin protein can be produced by the transfected fibroblasts. The produced endostatin has inhibitory action on human umbilical vein endothelium and has no inhibition action on fibroblasts.
10.The conventional and dynamic contrast-enhanced MR imaging findings of lymphoma of the orbit

Objective To evaluate the diagnostic value of conventional and dynamic contrast- enhanced MRI in lymphoma of the orbit.Methods Thirteen cases of lymphoma of the orbit,including B-C (10 cases),T-C (1 case),T-NK-C (1 case)lymphoma,and multiple myeloma (1 case),were studied using conventional MR and dynamic contrast-enhanced MR imaging with 3D fast spoiled gradient echo sequence.Calculated values included time to peak (Tpeak),washout ratio (WR),slope and enhancement ratio (ER),and time-intensity curves (TICs),and Tpeak,WR,slope and ER were calculated preoperatively.Results Eleven of the 13 lymphomas of the orbit was seen in the anterior portion of the orbit including eyelid and lacrimal gland.On conventional MRI,10 cases showed iso-intensity on T_1WI and T_2WI and all 13 cases showed moderate enhancement after contrast administration.TIC of all 13 cases showed rapid enhancing and wash-out,Tpeak was (58.7?8.5)s,and WR was (30.9?9.4)%.The accurate diagnosis with only conventional MRI was achieved in 6 out of 13 cases,while the accurate diagnosis was achieved in all 13 cases by using combined conventional MRI and dynamic enhanment MRI.Conclusion onventional MRI combined with dynamic contrast-enhanced MRI is useful for the diagnosis of lymphoma of the orbit.
