China Oncology 2000;0(06):-
Purpose:To investigate the status of p16 gene and its influence in p16 mRNA expression in primary hepatocellular carcinoma (HCC). Methods:The methylation status of p16 gene and expression of p16 mRNA were examined by methylation-specific PCR (MSP) and RT-PCR in twenty-five primary HCC and corresponding paracancerous tissue specimens. Results:Methylation was detected in 12 of 25 (52.0%) tumor tissue specinmens, while 6 of 25 (24.0%) in corresponding paracancerous tissue specimens. Loss of p16 mRNA expression was found in 19 specimens with methylated p16 gene, while expression of p16 mRNA was detected in 27 of 31 specimens with unmethylated p16 gene. There was significant difference in mRNA expression between specimens with methylated p16 and unmethylated p16.Conclusions:p16 gene methylation is a frequent event in HCC. The methylation of p16 gene might lead to loss of p16 mRNA expression. Detection of p16 gene methylation may be a useful marker in the molecular diagnosis of hepatocellular carcinomas.

It is a trend that clinical researchers overemphasize on hypothesis testing and the P value. However, P value has some limitations in the presentation of study results. Power is a tool for study design and results explanation, especially for negative results. Confidence interval contains more information for clinical results than P value. P value, power, and 95% confidence interval should be provided in the clinical reports to facilitate the interpretation of the results.

Object To study the relationship between the essential oil constituents and the chromosome numbers of Houttuynia cordata and compare the difference of the essential oil constituents from the fresh and the dried ones, aerial and underground parts of the fresh plant. Methods The essential oil constituents of H. cordata with different chromosome numbers from the aerial and underground parts of the fresh, and the aerial parts dried in shade as well were analyzed by TLC. Results According to the difference of the spot number and definition of the essential oil TLC pattern, the essential oil constituents in the aerial parts of 20 fresh plants were classified into five types, those in their underground parts into three types, and those in 19 dried plants into four types. Conclusion There is obvious difference in the essential oil constituents between the fresh aerial and underground parts, and between the fresh and dried plants. The difference among the fresh plants is more significant than that of the dried ones. And the difference among the aerial parts of the fresh plants is more significant than that of the underground parts. There is a tendency that the spot numbers increase with the chromosome numbers. The difference of essential oil constituents in H. emeiensis and H. cordata is insignificant.

Acta Nutrimenta Sinica 1956;0(01):-
Objective To establish a modified method to evaluate the protection of the antioxidant of DNA damage induced by hydroxyl radical. Method Reaction mixture at a final volume of 1.0 ml contained 0.8mg/ml DNA,0.2 mmol/L H2O2,1.0 mmol/L ascorbic acid,in 50 mmol/L NaH2PO4-Na2HPO4 buffer (pH 7.4). EDTA(1.0 mmol/L) and FeCl2 (1.0 mmol/L)were premixed and then dispensed into the reaction mixture to trigger the Fenton reaction, and the mixture was incubated at 37℃ for 30 min. The reaction was stopped by dispensing 1ml of 10% (w/v) trichloroacetic acid ,and reaction mixture was mixed with 1ml of 1% (w/v) 2-thiobarbituric acid, then further heated at 80 ℃ for 30 min. The chromogen formed was extracted into n-1-butanol and absorbance was measured at 532 nm. Results Addition of hydroxyl radical scavenger can compete with DNA for the hydroxyl radicals and diminished TBARS formation at a defined time of reaction with dose-dependent effect. The result showed that hydroxyl radical scavenger exerted inhibition effects on hydroxyl radical-mediated DNA damage. Conclusion This method can be used to evaluate the protection of the antioxidant of DNA damage induced by hydroxyl radical.
