Autoimmune encephalitis is a group of inflammatory diseases related to autoantibodies that affect the central nervous system. Early diagnosis of patients with autoimmune encephalitis has certain difficulties, because the clinical manifestations caused by different types of autoantibodies can be non-specific, and the presence of multiple autoantibodies can cause variation and superposition of clinical manifestations. The article reported a case of autoimmune encephalitis patients with double positive anti-N-methyl-D-aspartate receptor and dipeptidyl-peptidase-like protein-6 antibodies, and reviewed relevant literature for clinical reference.

Objective To investigate the temporal expression patterns of the related transcription factors and target genes in cardiomyocyte hypertrophy, and provide valuable clues for further researches on cardiomyocyte hypertrophy. Methods Isoproterenol (ISO) was used to induce ventricular myocytes hypertrophy in neonatal mice. The survival rate of cardiomyocyte was detected by CCK-8,and the average diameters and surface areas of cells were measured by computer photograph analysis system. The mRNA and protein expression levels of related genes were respectively measured by RT-PCR and Western blotting. Results The model of cardiomyocyte hypertrophy stimulated by ISO was constructed successfully. The expression levels of GATA4,MEF2C,GATA5,BNP and ANP increased 24 hours after ISO treatment, the expression levels of P300,α-MHC and TBX5 increased 12 hours after ISO treatment, and of SRF and β-MHC mRNA increased 6 hours after ISO treatment (P<0.05).The expression levels of GATA4,α-MHC,β-MHC,SRF and P300 mRNA increased firstly, and then decreased in cardiomyocytes induced by ISO.The expression levels of GATA4,SRF,α-MHC,β-MHC and P300 mRNA were still higher than normal (P<0.05),but of MEF2C decreased to normal (P>0.0S) 72 hours after ISO treatment. The expression levels continuously elevated of GATA5,TBX5,ANP and BNP mRNA than that of controls (P<0.05),while no fluctuation was found in NKX2.5 mRNA expression (P>0.05).The expression of GATA4 protein increased, while of HEY2 protein decreased 48 hours after ISO treatment (P<0.05).Conclusions In hypertrophic cardiomyocytes, the expression pattern of MEF2C is similar to,but the patterns of GATA5,GATA4,TBX5 and SRF are different with that in the development of heart, implying these genes are important during the process from compensatory stage to decompensation stage. The expression patterns of GATA4,MEF2C and SRF are similar to that of acetylase P300,implying the temporal expressions could be regulated by P300 in cardiomyocyte hypertrophy.

BACKGROUND:The chemical compositions and structure of calcium phosphate bone cement are similar to those of human bone, which can fil the bone colapse caused by fracture and induce osteogenesis, but its degradation rate is slow. OBJECTIVE:To evaluate the biocompatibility of the calcium phosphate cement/fibrin glue and the feasibility of repairing radius defects. METHODS: In vitro cytotoxicity experiment: Mouse fibroblasts were cultured in the calcium phosphate bone cement/fibrin glue extracts, phenol solution, and RPMI 1640 culture medium containing 10% fetal bovine serum, respectively, to detect the cytotoxicity grade. Hemocompatibility experiment: Calcium phosphate bone cement/fibrin glue extracts, normal saline and distiled water were respectively added into the rabbit anticoagulation, to detect the hemolytic rate. Forty-five New Zealand white rabbits were enroled and modeled into bilateral radius defects, folowed by randomly alotted into three groups: blank control group without any intervention, experimental and control groups were given the implantation with calcium phosphate bone cement/fibrin glue and autologous radius, respectively. X-ray, histology, bone mineral density and biomechanical test were performed at postoperative 4, 8 and 16 weeks. RESULTS AND CONCLUSION:The toxicity grade of the calcium phosphate cement/fibrin glue was 0 to 1. The hemolytic rate of the calcium phosphate cement/fibrin glue was 3.15%. At 16 weeks postoperatively, X-ray showed that in the experimental and control groups, the fracture line disappeared completely, pulp cavity was recanalized, and in plastic completely. Histology showed that the reconstructed bone trabecular was obvious, plate layer of bone was mature, and medulary cavity recanalization appeared in the control group; there were a large number of new grid-shaped woven bone tissues growing into the material in the experimental group, with overt degradation, and degradation rate was in paralel to bone ingrowth. The bone density, the maximum load, maximum stress and failure energy in the experimental and control groups were significantly higher than those in the blank control group (P < 0.05), and al above indicators showed no significant differences between the experimental and control groups. These results manifest that the calcium phosphate bone cement/fiber protein glue composite material holding a good biocompatibility can promote bone tissue regeneration for bone defect repair, achieving similar curative effect with autologous bone transplantation.

E2 is an envelope glycoprotein of Classical swine fever virus (CSFV) and contains sequential neutralizing epitopes to induce virus-neutralizing antibodies and mount protective immunity in the natural host. In this study, four antigen domains (ABCD) of the E2 gene was cloned from CSFV Shimen strain into the retroviral vector pBABE puro and expressed in eukaryotic cell (PK15) by an retroviral gene expression system, and the activity of recombinant E2 protein to induce immune responses was evaluated in rabbits. The results indicated that recombinant E2 protein can be recognized by fluorescence antibodies of CSFV and CSFV positive serum (Lanzhou Veterinary Research Institute, Chinese Academy of Agricultural Sciences, Lanzhou, China) using Western blot, indirect immunofluorescence antibody test (IFAT) and ELISA, Furthermore, anti-CSFV specific antibodies and lymphocyte proliferation were elicited and increased by recombinant protein after vaccination. In the challenge test, all of rabbits vaccinated with recombinant protein and Chinese vaccine strain (C-strain) were fully protected from a rabbit spleen virus challenge. These results indicated that a retroviral-based epitope-vaccine carrying the major antigen domains of E2 is able to induce high level of epitope-specific antibodies and exhibits similar protective capability with that induced by the C-strain, and encourages further work towards the development of a vaccine against CSFV infection.
