Aim To explore the inhibitory effect of li-gustrazine combined with paeonol on rat liver fibrosis induced by CCl4 and its mechanisms,so as to provide new treatment strategies for liver fibrosis in clinical. Methods Cleaning laboratory male SD rats were ran-domly divided into blank control group, model group (CCl4 ),ligustrazine group, paeonol group and combi-nation group (ligustrazine+paeonol).HE staining was used to observe the pathological change.Masson stai-ning and Sirius red staining was used to observe the collagen deposition. The levels of serum ALT,AST, ALP and hydroxyproline were detected by automatic bi-ochemistry analyzer.Western blot detected the markers of liver fibrosis.HSC-T6 cell was divided into model group, ligustrazine group, paeonol group and combina-tion group. The protein and gene expression of inflam-mation and apoptosis pathway was analyzed by Western blot and real time-PCR.Results Ligustrazine com-bined with paeonol could significantly improve liver tis-sue pathology changes caused by CCl4 .It could reduce serum ALT, AST, ALP and hydroxyproline levels. Moreover,it could also inhibit liver fibrosis marker protein expression, and thus reduce the deposition of collagen fibers. The effect was better than that in sin-gle intervention group.Combination group could inhib-it the inflammatory pathways related protein expression in HSC cells and promote the apoptosis of HSC cells. Conclusion Ligustrazine in combination with paeonol has significant anti-fibrosis effect, and the effect is bet-ter than both single intervention. The effect may be due to the interference with TNF-α/NF-κB pathway in the HSC cells, which promotes its apoptosis and inhib-its the generation of extracellular matrix.

Aim To investigate the blockade of cadmium on store operated calcium channels and the fluorescence interference of cadmium with Fura-2.Methods PC12 cells were used to determine the intracellular calcium concentration [Ca~(2+)]i indicated by change in Fura-2 fluorescence ratio(F_(340)/F_(380)).Results Introduction of cadmium induced a significant increase in Fura-2 fluorescence ratio following thapsigargin-evoked calcium entry;Besides, cadmium gave rise to a remarkable elevation in Fura-2 fluorescence ratio following breakdown of the plasma membrane by triton,a detergent. The Fura-2 fluorescence was increased in the presence of cadmium and Fura-2/AM,simultaneously in the absence of PC12 cells. Conclusion Cadmium can interfere with the Fura-2 fluorescence.

Journal of Applied Clinical Pediatrics 1992;0(05):-
Objective To investigate the efficacy of lamivudine( LAM) combined with thymosin ?1 (T?1) on young patients with chronic hepatitis B. Methods Sixty six chronic hepatitis B (CHB) patients aged from 12 to 16 years and weight greater than 33 kg were divided into the LAM + T?1 group and the LAM group; in LAM + T?1 group,T?1 was administered 1.6 mg hypodermic once daily and after 5 days it was changed to 1 6 mg hypodermic, twice in two weeks, and the drug was stopped after 26 weeks, while LAM had been administered 100 mg each time once daily for 52 weeks. In LAM group, only LAM had been administered 100 mg once daily for 52 weeks. The serum level of HBV DNA, HBV markers and the ALT were measured before and after treatment Results After the treatment, the rate of serum HBV UNA turned to negative in LAM + T?1 group was 100 % , and that in LAM group was 93.7 % ; recovery ratios at 52 weeks of LAM+ T?1 group was 87.1% ,and that of LAM group was 78. 1% .After 52 weeks, the HBeAg serum conversion rate of LAM + T?1 group was 58. 1 % , and that of the LAM group was 21.9%. Comparing the results of the two groups, the difference was remarkable. Conclusion The efficacy of T?1 combined with LAM on young patients with chronic hepatitis B is superior to that use of single LAM

AIM: To investigate killing effect of herpes simplex virus thymidine kinase/ganciclovior system(HSV-TK/GCV) on osteosarcoma cell and its mechanisms.METHODS: Recombinant retroviral vector (DORHyTK) containing hygromycin phosphotransferase-thymidine kinase fusion gene(HyTK) was constructed and introduced into human osteosarcoma cell line OS732 with DOTAP. DNA and total RNA extracted from HyTK expressing cells (OS732TK) were tested by PCR and RNA dot blot analysis. A chemosensitivity of OS732TK cells to GCV and "bystander effect" were measured by means of MTT colorimetric assay. Hoeschst 332258 staining and flow cytometric analysis were performed for mechanism of HSV-TK/GCV system gene therapy. RESULTS: DORHyTK was constructed successfully; the HyTK gene existed and expressed in OS732TK cells; All OS732TK cells were killed after 5 days of exposure to 5 mg/L GCV. The "bystander effect" was observed in both a high population and a low population, but the former was stronger than the latter. Hoeschst 332258 staining revealed the characteristic hallmarks of apoptosis, however necrosis also existed. Flow cytometric analysis of DNA content showed a G 0-G 1 phase blockade. CONCLUSION: HSV-TK/GCV gene therapy system showed its strong killing effects on osteosarcoma cells OS732; The phenomenon of "bystander effect" was also very apparent. GCV exposure induced both necrotic and apoptotic death in HSV-TK expressing cells, and perturbed the cell cycle. HSV-TK/GCV gene therapy system may provide a new therapeutic approach for treatment of osteosarcoma.
