1.Longitudinal investigation of chronic periodontitis during maintenance therapy

Objectives:To investigate the characteristics of clinical changes during maintenance therapy of chronic periodontitis.Methods:22 patients with chronic periodontitis were enrolled in a 9-month maintenance care program after non-surgical periodontal therapy.Oral hygiene instructions together with supra-and sub-gingival scaling and root planing were carried out every 3 months.Clinical parameters including probing pocket depth(PPD),probing attachment level(PAL),and bleeding on probing(BOP) were recorded at baseline and each re-examination point.Results:The mean PPD decreased by 0.44 mm,PAL increased by 0.38 mm and BOP point number decreased by 18.94%.The recoveries were mainly observed during the first 3 months.Sites with ≥4 mm pocket at baseline exhibited a greater percentage of pocket deepening than those with initial shallow pocket.The incidence of the sites with PPD deeping of posterior teeth increased more than that of anterior teeth(P

Objective To investigate the effect of interferon-alpha (IFN-α) on preventing pancreatic fibrosis and expression of pancreatic α-smooth muscle actin (α-SMA) and collagen type m in experimental rat model induced by diethyldithiocarbamate (DDC). Methods Forty male Wistar rats were randomly divided into three groups: normal control group, fibrotic group(PF group) and IFN-α prevention group(IFN-α group). Pancreatic fibrosis were induced by repeated intraperitoneal DDC injection. IFN-α group were treated with subcutaneous injection of IFN-α in saline, administered daily at the dosage of 105 U. Six weeks later, all the rats were sacrificed, the samples of the pancreatic tissue obtained by biopsy were submitted to histological studies. The expression of α-SMA and collagen type Ⅲ in pancreatic tissues were detected by immunohistochemistry. Results From the 4th week on, the weight of rats in PF group grew slowly, some rats 'weight even decreased, while the weight of rats in IFN-α group kept growing up slowiy, and the difference became significant at the 5th week (306.17±20.89 g vs 279.17±23.58 g, P<0.05 ). The fibrosis score, Masson stain score, α-SMA and pancreatic collagen type Ⅲ in PF group were 2.679±0.899, 218. 713± 36. 102, 148.971±30. 686 and 88. 142±42.581, respectively, and the corresponding values in PF group were 1.952±0.219, 114.732±24.912, 77.237±9.275 and 59.952±25.498, respectively. The extent of fibrosis in IFN-α group was significantly better than that in PF group (P<0.05). Conclusions IFN-α may inhibit the extent of fibrosis, decrease the expression of α-SMA and pancreatic collagen type Ⅲ, therefore, it may partially prevent pancreatic fibrosis induced by DDC.
3.The Development of Computer Game Addiction Inventory for Chinese College Students

Objective: To develop a computer game addiction inventory (CGAI) for Chinese college students. Methods:A primary inventory made by the authors was applied to 477 college students from 8 universities in Beijing for exploratory factor analysis, and then to 405 college students for confirmatory factor analysis. Results: After exploratory factor analysis, a questionnaire consisted of 33 items were established, which had four factors: dependence and addict behavior, emotion arousal, functional impairment, and shame and un-satisfaction. These four factors could explain 55.1 of the total variance, were confirmed by confirmatory factor analysis (?~2/df=2.304; IFI=0.885; CFI=0.884; NFI=0.813; RSMEA=0.057). The internal consistency of the questionnaire was good (r=0.77~0.94), the test-retest reliability was 0.907. Criteria validity shown by higher scores in addiction group than normal control was also satisfying. Conclusion: The CGAI has good psychometric features and could be applied to college students.
4.Biological characteristics of cleft palate relevant gene thyroid transcription factor-2 transgenic mice.

OBJECTIVEThe aim of this study is to establish a transgenic mouse model for cleft palate relevant gene thyroid transcription factor-2 (TTF-2), which can be used to study palatal shelf development when the expression pattern and regular activation of TTF-2 is altered.
METHODSThe C57BL/6J mouse TTF-2 gene was cloned through polymerase chain reaction (PCR) from the mouse genomic DNA. The TTF-2 gene was inserted into the expression vector pBROAD3-mcs to construct the recombinant expression vector pBROAD3-TTF-2. This expression vector was then microinjected into the male pronuclei of the fertilized mouse ovum. Thus, the TTF-2 transgenic mice model was established. The genotype of the transgenic mice was identified by PCR and Southern blot analysis. Immunohistochemistry identified the consistent expression of TTF-2 gene during its palatal shelf development.
RESULTSTTF-2 genes were microinjected into 982 fertilized ova. A total of 580 two-cell-stage embryos cultured and transplanted into the oviducts of 48 pseudopregnant female mice. Overall, 68 embryos were obtained for analysis. The genotype of the mice was determined through PCR and Southern blot analysis using genomic DNA extracted from tail biopsies of the transgenic fetus. A total of 13 TTF-2 transgenic mice were detected. The expression of TTF-2 gene during the palatal shelf development of the transgenic mice was consistently detected by immunohistochemistry.
CONCLUSIONThe recombinant expression vector pBROAD3-TTF-2 was integrated into mouse genome through microinjection. The transgenic mouse in the palatal shelf that consistently expressed TTF-2 was successfully established and displayed a cleft palate phenotype.

5.Effects of interpersonal group counseling on college students with computer gaming addiction

Objective:To study the effects of group counseling which based on interpersonal activities and aimed at the college students with computer gaming addiction in China.Methods:Seventeen college students with computer game addiction were chosen as the experimental group,and 10 college students from another college were chosen as the control group.The two groups had no difference in the scores of Computer Gaming Addiction Invention(CGAI)and several other scales before group counseling.There were a pre-test and a post-test separately set when the group counseling was started and finished,focused on CGAI and negative emotions and cognitions.Results:In post-test,the score of CGAI in experimental group was significantly lower than that in control group[(92.6±16.7)vs.(105.9±13.0)] and lower than that in pre-test(98.9±16.4)by analysis of covariance(Ps<0.01).The scores of Beck Depression Inventory(BDI),Self-Rating Anxiety Scale(SAS),and the negative cognition of oneself in socical settings in experimental group were also lower than those in control group [(6.4±7.9)vs.(13.5±5.6),(39.7±6.4)vs.(49.9±10.3),(15.1±4.3)vs.(19.9±3.0),Ps<0.01],while the score of Rosenberg Self-Esteem Scale(SES)was higher than that of the control group [(29.3±2.9)vs.(24.6±4.8),P<0.01].Conclusion:Group counseling based on interpersonal activities has obvious effects on the college students with computer gaming addiction.
6.An experimental study of folic acid rivaling methylenetetrahydrofolate reductase (MTHFR) gene

Objective: To further understand the role of folic acid supplements rivaling MTHFR gene silencing in pathogenesis of NCLP, RNA interference (RNAi) was applied to knock down MTHFR in mouse embryonic palatal mesenchymal (EPM) cells. Methods: MTHFR ShRNA expression vector were transfected into the primary cultured EPM cells. MTT was used to observe cell proliferation after MTHFR gene silencing. FCM was used to observe cell cycle after MTHFR gene silencing. Results: The results showed the cells proliferation had an inequality amelioration after using folic acid supplements in MEPM cells with MTHFR gene silencing. Using folic acid supplements rivaled the effect of MTHFR gene silencing had a dose-dependent manner. Using 20 μg/ml folic acid supplements could improve the cell proliferation to achieve normal level of cell proliferation. Conclusion: MTHFR gene is an important candidate gene of NCL/P. Using folic acid supplements could prevent teratogenic MTHFR gene silencing for embryonic palate development.
7.Clinical observation of low molecular weight heparin combined with astragalus injection in the treatment of diabetic nephropathy

Objective To observe the effect of low molecular weight heparin calcium combined with astraga-lus injection in the treatment of diabetic nephropathy.Methods From January 2013 to December 2014,78 patients with stage III diabetic nephropathy in our hospital were enrolled into the study.They were randomly divided into con-trol group(41 cases) and treatment group(37 cases).The control group received general therapy,such as low protein diets,compound alpha keto acid,insulin,irbesartan,simvastatin.The treatment group was treated with low molecular weight heparin calcium and astragalus injection on the basis of general therapy.The changes of urinary albumin and 24 hours urinary protein after treatment in the two groups were observed.Results After treatment,the negative rate of urinary micro albumin in the treatment group ( 75.68%) was significantly higher than that of the control group (29.27%)(χ2 =16.765,P<0.01).24 hour urinary protein of the treatment group[(176.2 ±82.3)mg/24h]was significantly lower than that of the control group[(223.8 ±87.8)mg/24h](t=2.471,P<0.05).Conclusion Low molecular weight heparin calcium combined with astragalus injection can reduce the proteinuria of diabetic nephropa-thy,it is an effective intervention therapy for early diabetic nephropathy.
8.Effect of interferon-? on the fibroblasts from rat palatal scar

Objective:To explore the biological activities of interferon-? on the fibroblasts from rat palatal scar.Methods:Fibroblasts were cultured from rat palatal scar.The cells of pasage 4-6 were suspended into culture medium at (2.5)?105 cells/ml.Then the cells were cultured as fibroblasts-populated collagen latice(FPCL) with the final cell density of 5?104/ml.The cultures were exposed to IFN-?(U/ml) at 0,40, 400 and 4 000 for 24 h respectively.The cell proliferation was studied by MTT assay and FPCL contraction was studied by diameter measuring.Results:The absorbance of the cells treated with IFN-?(U/ml) at 0,40,400 and 4 000 was 0.247?0.014,0.235?0.014,0.190?(0.024) and 0.184?0.021 respectively,the contraction idex(%) of FPCL treated with above concentrations of IFN-? was 88.53,64.47,46.00 and 23.63 respectively.Conclusion:IFN-? may inhibit the proliferation of fibroblasts and the contraction of FPCL.
9.Clinical research on corneal perforation

AIM: To study the clinical characteristics of corneal perforation ( CP) .
METHODS:A retrospective analysis in July 1995 to July 2010 the First Affiliated Hospital of Fujian Medical University diagnosed CP 72 patients ( 72 eyes ) , clinical characteristics of all the patients were analyzed.
RESULTS: The incidence of corneal ulcer perforation rised year by year, the morbidity of male and female was 17:7, the onset age focused on 48 years old. Of 23 industrial workers ( 32%) with clear history of trauma, pathogeny identification results:top two:fungal infection and Acanthamoeba keratitis. A using history of glucocorticoid was found in 10 cases.
CONCLUSION:There are plenty of primary causes of CP such as traumas, fungal infection, Acanthamoeba keratitis, eroded keratitis, etc. CP happens in middle-aged males in Fujian province, most traumas are the causes, the main pathogenic bacteria is fungal infection.
