1.Experimental Study on Anti-inflammatory and Analgesic Effects of Jitongning Capsule

Objective To study the anti-inflammatory and analgesic effects of Jingtongning Capsule. Methods Models of acute inflammatory reactions of croton oil-induced mouse ear edema and carrageenan-induced rat raw swelling and chronic inflammatory reaction of cotton ball-induced mouse granuloma in abdominal cavity were performed to study the anti-inflammatory effects of Jingtongning Capsule. Model of acetic acid-induced writhing in mice was used to study the analgesic effect of Jingtongning Capsule. Results Jingtongning Capsule significantly suppressed croton oil-induced mouse ear edema and carrageenan-induce rat paw swelling. Cotton ball-induced mouse granuloma in abdominal cavity was also markedly reduced by Jingtongning Capsule. The writhing times of mice induced by acetic acid were decreased by Jingtongning Capsule. Conclusions Jingtongning Capsule has obvious anti-inflammatory effect on acute and chronic inflammatory reactions. The pain reactions induced by chemical stimulus was significantly relieved by Jingtongning Capsule.
2.Effects of water improvement on skeletal fluorosis in drinking-water-borne endemic fluorosis areas in Liaoning Province

Objective To investigate the prevalence of adult skeletal fluorosis in drinking-water-borne endemic fluorosis areas in Liaoning Province and to observe the effects of preventive measures.Methods Three water-changed villages were selected from six drinking-water-borne endemic fluorosis counties,Faku County,Liaoyang County,Haicheng City,Linghai City,Fu Mongolia County,and Jianping County.Totally 18 diseased villages were selected as the investigation sites.The operating condition of the improvement projects was investigated and the fluoride level of drinking water was tested.People over the age of 25 and living in the local for more than 5 years in each survey site were selected and divided into five groups according to age groups of 25-,35-,45-,55-,and ≥65 years old.Ten people in each age group were selected,half male and half female,to examine skeletal fluorosis by X-ray.Clinical symptoms and bone change,including sclerotin,bones,and joints changes were also examined.Results The water fluorine values of two villages were 1.32 and 1.69 mg/L,more than the national standard (1.2 mg/L),while other 16 villages were between 0.5-1.0 mg/L.X-ray images were analyzed for diagnosis of skeletal fluorosis in 905 adults,and 46 cases were diagnosed as skeletal fluorosis in the 18 monitored villages.Xray detection rate was 5.08%,of which the X-ray detection rate of adult fluorosis in water fluoride qualified monitoring villages was 4.11% (33/803).The detection rate of adult skeletal fluorosis was 12.75% (13/102) in water fluoride unqualified monitoring villages.There was a significant difference of the total detection rates between the two groups of monitored villages (x2 =13.99,P < 0.05).Skeletal fluorosis was mainly happened in the age group over the age of 45,account for 89.13% (41/46).Peri-bone changes were observed in the 46 cases of patients with skeletal fluorosis.Conclusions The condition of skeletal fluorosis has been effectively alleviated after implementing the improvement measures.Prevention and treatment can effectively control the occurrence of fluorosis.
3.Minimally invasive managements for non-anastomotic biliary stricture after orthotopic liver transplantation

Objective To evaluate the clinical value of minimally invasive methods for non-anastomotic biliary stricture (NABS) after orthotopic liver transplantation.Methods The clinical data of 403 patients who underwent liver transplantation during recent 10 years in Liver Transplantation Center at General Hospital of Guangzhou Military Commanmol were analyzed retrospectively,and 13 patients with NABS were selected.The outcomes of 3 types of NABS patients treated by endoscopic retrograde cholangiopancreatography(ERCP) or percutaneous transhepatic cholangial drainage(PTCD) were compared and the indication for re-transplantation was identified.Results PTCD treatments of 4 patients were proved ineffective.The shortterm curative rate of minimally invasive treatments was 8/13.Five patients eventually required surgical treatments (re-transplantation in 4,Roux-en-Y anastomosis in 1).According to cholangiography results,NABS were divided into 3 types,namely hepatic bile duct strictures (n =4,type Ⅰ),multiple extra-hepatic and intrahepatic biliary strictures (n =7,type Ⅱ),intrahepatic biliary strictures (n =2,type Ⅲ).The success rates of minimally invasive treatment in 3 types of NABS were 3/4,4/7 and 1/2,respectively.Nearly half of type Ⅱ and type Ⅲ patients needed re-transplantation,which was more likely for those patients with hepatic artery stenosis (2/3).Conclusion NABS treated with minimally invasive methods are preferred.Based on the appearance of biliary stricture,type Ⅰ patients had the best prognosis.For those type Ⅱ and type Ⅲ patients who failed minimally invasive treatment,especially combined with hepatic arterial stenosis,surgical treatment should be timely,so as not to lose a chance for re-transplantation.
4.Herbal Textual Research on SemenLepidii seu Descurainiae

By reviewing name, type, function and indication of Chinese herbTing-Li-Zi (Lepidii seu Descurainiae) in literatures, there were northernTing-Li-Zi and southernTing-Li-Zi from Song dynasty. It was also known that northernTing-Li-Zi has strong effect and southernTing-Li-Zi had moderate effect. However, there was no strict classification between two types in current clinical practice. The disease prevention and treatment effect of Chinese herbs were depended on their internal features. Disease treatment must be based on syndrome differentiation with corresponding selection of herbs to achieve desired results. Therefore, it is necessary to combine properties with actions in the study of Chinese herbs to provide guidance in clinical practice.
5.Detection of TCRγ gene rearrangements in skin lesions and peripheral blood of patients with parapsoriasis ;and their clinical significance

Objective To detect TCRγ gene rearrangements in skin lesions and peripheral blood of patients with parapsoriasis, and to study their clinical significance. Methods Totally, 20 patients with parapsoriasis were included in this study. BIOMED?2 multiplex PCR was performed to detect TCRγgene rearrangements in lesional skin (n=20)and peripheral blood(n=11)samples from the patients with parapsoriasis. Statistical analysis was performed to assess the relationship of TCRγ gene rearrangements with clinical types of parapsoriasis as well as general information and histopathological manifestations(including non?specific manifestations and atypical manifestations)of patients. Results TCRγ gene rearrangements were positive in lesional skin from 7 of the 20 patients, in peripheral blood from 3 of 11 patients, and in both lesional skin and peripheral blood from 2 patients. Positive TCRγ gene rearrangements in skin lesions were significantly correlated with mycosis fungoides(MF)?related atypical histopatho?logical manifestations(P<0.05), but those in neither skin lesions nor peripheral blood were correlated with gender and age of patients or clinical course and types of parapsoriasis(all P>0.05). During an average follow?up time of 44.85 ± 18.48 months, 1 case progressed into MF, and 2 were cured. Conclusions Positive TCRγgene rearrangements in skin lesions of patients with parapsoriasis may be correlated with MF?related atypical manifestations. The presence of TCRγgene rearrangements and atypical histopathological manifestations may suggest the possibility of progression from parapsoriasis into MF.
6.DISCRETE RADIOFREQUENCY CATHETER ABLATION FOR PAROXYSMAL ATRIAL FIBRILLATION IN A DOG MODEL

To investigate an approach of treatment for atrial fibrillation with discrete ablation lesion, 12 dogs with experimental atrial fibrillation underwent radiofrequency catheter ablation. Atrial lesion was made at the most vulnerable site. The results showed that in 10 of 12 dogs persistent fibrillation could be ablated. There was no significant change in sinus heart rate, P wave duration, and corrected sinoatrial node recovery time (cSNRT). The results suggested that paroxysmal atrial fibrillation could be eliminated by point ablation of the lesion, and that trigger activity might be involved in the mechanism of atrial fibrillation.
7.EFFECT OF FUFANG GANNING ON PROTECTING LIVER AND ENHANCING IMMUNITY

Fufang Ganning(FGN)is an oral liquid,consisting of 8 kinds of chinese herbs including Radix Astragali,Radix Salviae Multiorrhizae,Radix Curcumae,Rhizoma Polygonati,etc.It was found to be effective in protecting against experimental acute hepatic damage caused by CC14,TAA and D-Glan in mice,FGN could reduce ALT and retention of BSP,and histopathological chan- ges revealed that the hepatic injury induced by CC14 in animals received FGN was much less severe than those of the control ones.It could also lower ALT and serum bilirubin in mouse and rat models with hepatic injury caused by ?- isothioeyanate.On the other hand,in the animal models with cirrhosis induced by CC14,FGN could not only cut down ALT but also lessen the degeneration and necrosis of hepatic cells and cirrhotic changes.Meanwhile,FGN could significantly increase the clearance index of carbon particles(phagocytic func- tion)and serum hemolysin in normal mice.It is concluded that FGN has the effects of protecting liver from damage and enhancing the immune function.
8.Analgesic activity of ethanol extracts from root of Daphne genkwa

Objective To elucidate the analgesic activity of the ethanol extracts from the root of Daphne genkwa (EERD). Methods The analgesic activity of EERD was evaluated by the effects on adjuvant-induced nociceptive response and paw swelling, the formation of PGE_2 and IL-1? in adjuvant arthritis (AA) rats, the activities of SOD and CAT, the levels of NO/iNOS in serum and brain tissue as well as by the effects on c-Fos protein expression in spinal cord of AA rats. Results EERD at used doses significantly delayed the adjuvant-induced nociceptive response and eased the paw swelling in AA rats. EERD also evidently inhibited the production of PGE_2 and IL-1?, and enhanced the activities of SOD and CAT in the tissue of paws being injected by adjuvant. Furthermore, it remarkably reduced the content of NO and inactivated the activity of iNOS in brain tissue of AA rats. In addition, EERD at used doses exhibited prominent inhibition on adjuvant-induced expression of c-Fos protein in the spinal cord of AA rats. Conclusion EERD is an effective agent for analgesia. The possible mechanisms for its analgesia might be the actions of inhibiting the production of PGE_2 and the release of IL-1?, reducing the activity of iNOS and hence the generation of NO in brain tissue, and blocking superoxidation through enhancing the activity of SOD and CAT.
9.Effects of straw direct vapor nitrogen method on the motility of post-thaw human sperm

ObjectiveTo study the effects of two cryopreservation methods ( direct vapor nitrogen method and program freezing method) on the motility of post-thaw human sperm.MethodsThe 25 semen samples were taken from 25 donors from Shanghai Human Sperm Bank during May to July 2011,using straw as a frozen carrier, the semen samples contained modified glycerol-yolk-citrate ( GYC ) ( 1 ∶1 ) as cryoprotectant,after the liquefaction, each semen sample was divided into 2 groups,and separately cryopreserved by direct vapor nitrogen method and program freezing method.According to the recommended standardoftheWorldHealthOrganization, semensampleswereanalyzedbeforeandafter cryopreserving.Density,total motility,PR,motility parameters ( including VAL,VSL,VCL,BCF) were determinated by the IVOS sperm analyzer.Semen samples were stained by modified papanicolaou stain and 200 sperms of each sample were counted under microscope by manual analysis to calculate the rate of normal morphology sperm.The paired t test was used to compare the above indictors before and after cryopreserving in one method and after thawing between two methods.Results Before cryopreserving,the total motility,PR,VAL,VSL,VCL,BCF were(91.36±4.96)%,(81.28 ±6.41)%,(59.29 ±9.30) μm/s,(49.80±8.40) μm/s, (83.39 ± 15.70)μm/s, (16.68 ± 3.36)Hz respectively, and after cryopreserving by direct vapor nitrogen method the results were (46.04 ± 20.14)%,(40.22 ± 18.42 )%,(48.00± 11.60)μm/s,(41.11 ±8.50) μm/s,(71.55 ±17.19) μm/s,(15.94 ± 14.30) Hz respectively,the differences of the indictors above were observed with statistic significance except BCF ( t =11.324,11.958,4.266,3.964,3.620,0.258,P of BCF >0.05,P of others <0.05); and after cryopreserving by program freezing method,the results were(48.05 ± 18.27)%,(42.47 ± 15.62)%,(45.30 ± 6.51) μm/s,(38.08 ± 4.99) μm/s,(64.23 ± 11.75) μm/s,(14.00 ± 2.99) Hz respectively,the differences of the indictors above showed statistic significance (t =10.337,10.874,7.154,5.510,6.494,3.244,P <0.05).No significant differences were observed in the revival rate,total motility,PR and normal morphology sperm rate by using different methods (t =0.425,0.364,0.622,1.106,P >0.05).The results of VAL,VSL,VCL,BCF in direct vapor nitrogen method were higher than that in program freezing method,when the differences of VSL and VCL were observed with statistic significance(t =2.489,2.822,P < 0.05 ).Conclusions Using straw as carrier is practicable.Direct vapor nitrogen method is easier to operate than program freezing method.Post-thaw sperm cryopreseved by direct vapor nitrogen method has higher motility parameters,thus making this method an ideal one for fast freezing.
10.Effect of γ-rays combined with blocking VEGF expression on esophageal cancer xenografts

Objective To investigate the effect of blocking VEGF expression on the radiation sensitivity of esophageal cancer cell line TE-1 in vivo. Methods 32 male Balb/c/nu nude mice were randomly divided into four groups, including control group, radiation group, anti-VEGF group, and anti-VEGF + radiation group. The anti-VEGFcDNA cells were subcutaneously injected into the paw pats of mice (2 × 106/100 μl). The subcutaneous tumors were irradiated with 18 Gy of 60Co y-rays when the diameter of tumors varied from 0. 8 to 1.0 cm. The volume of the tumors was measured before and after irradiation, respectively. The expression level of VEGF mRNA and protein were examined by RT-PCR and Western blotting, respectively. Apoptotic cells were detected by electron micrographs. Results Latent period of the tumor formation of anti-VEGF group was lengthened compared with other groups(t = 13. 898,P <0.01 ). The volumes of tumor in anti-VEGF group [ ( 1207. 50 ± 97.07 ) mm3 ] and anti-VEGF +radiation group [ ( 1057. 5 ± 91.50 ) mm3 ] were not statistically different post-irradiation ( t = 1. 124, P >0.05 ) , but smaller than those in control group [ ( 5442. 50 ± 185.08 ) mm3 ] and radiation group [ (2922. 50 ± 152. 773)mm3 ] with statistical differences( t = 9. 475-21. 238, P < 0. 01 ). The expression level of endogenous VEGFmRNA and protein in anti-VEGF group and anti-VEGF + radiation group were statistically different from control group and radiation group (F = 387.394, 13.519, P < 0.01 ).Conclusions Antisense VEGF could inhibit the proliferation of esophageal cancer cell in the nude mice.Effect of blocking VEGF expression before irradiation on esophageal cancer xenografts might be limited.
