1.Effect of tumor-stroma ratio in prognosis evaluation of tumor

Tumor-stroma ratio is the ratio of tumor cells and stromal tissue. It can be evaluated through HE-stained sections. Usually a 50% cut-off value is taken. TSR is stratified as stroma rich(TSR < 50% )and stroma poor(TSR≥50% ). The different prognosis between the two groups can be assessed. Several previous studies show that TSR is an independent prognostic factor for colon carcinoma,breast cancer,esophageal squa-mous cell carcinoma,cervical carcinoma and non-small cell lung cancer;patients with stroma-rich tumor suffer worse prognosis. The cause might be that tumor cells could activate stroma cells while activated stroma cells could promote the growth,infiltration and metastasis of tumors. And tumor stroma should be recognized as an indispensible participant in progression and invasion of carcinoma.

3. A new triterpene saponin from roots of Dolichos falcate

Objective: To study the chemical constituents from the roots of Dolichos falcata. Methods: The chemical constituents were isolated by column chromatography with silica gel and Sephadex LH-20. Their structures were identified on the basis of spectroscopic analysis. Results: Three compounds were isolated from the roots of D. falcata and identified as 3β, 23-dihydroxy-2-oxoolean-12-en-28-dioic acid 3-O-β-D-glucopyranoside (1), 3β-hydroxyolean-12-en-23, 28-dioic acid 3-O-β-D-glucopyranoside (2), and 2β, 3β-dihydroxyolean-12-en-23, 28-dioic acid 3-O-β-D-glucopyranoside (3). Conclusion: Compound 1 is a new triterpene glycoside named doliroside C and compound 2 is isolated from this plant for the first time.

5.Mechanism by which dihydroartemisinin inhibits invasion and migration of glioma stem cells

BACKGROUND:Dihydroartmisinin can promote apoptosis of glioma cels GL261, but its effect on glioma stem cels is stil unknown. OBJECTIVE:To investigate the preliminary mechanism that dihydroartemisinin inhibits migration and invasion of glioma stem cels. METHODS: Glioma stem cels were isolated from mouse malignant glioma cel lines GL261. Immunofluorescence analysis was conducted to identify the characteristics of glioma stem cels. Migration and invasion abilities of glioma stem cels were analyzed by Transwel assay. The mRNA expressions of Tol-like receptor 2, matrix metaloproteinase-2 and matrix metaloproteinase-9 were examined by real-time fluorescence quantitative PCR. RESULTS AND CONCLUSION:The characteristics of glioma stem cels were identified by CD133 and Nestin staining. The migration and invasion of glioma stem cels were attenuated by dihydroartemisinin dose-dependently. Moreover, the mRNA expression of Tol-like receptor 2, matrix metaloproteinase-2 and matrix metaloproteinase-9 was also decreased by dihydroartemisinin in a dose dependent manner. These results suggest that dihydroartemisinin inhibits the migration and invasion of glioma stem cels probably through attenuation of Tol-like receptor signaling pathway.
6.Construction of Eukaryotic Expression Vector for Tumor-Associated Gene SNC90 and Transfection into Cancer Cells

Objective: To construct eukaryotic expression vector of tumor-associated gene SNC90 and transfect it into human colorectal cancer cell lines. Methods: A 1.5 kb tumor-associate gene SNC90 full length cDNA was inserted into a mammalian expression vector pREP9 to make recombinant vector pREP9-SNC90, which was then introduced into three kinds of colorectal cancer cell lines, SW1116, COL0205 and SW620, by lipofection or electroporation. The cells resistant to G418 drug were selected. Results: Cells transfected with pREP9-SNC90 showed fewer G418-resistant colonies than those transfected with void vector, the inhibitory rates are 72.2%, 74.2% and 59.7%, respectively. Conclusion : SNC90 may play a negative role in regulating growth of colorectal cancer cell.

8.Changes of atrial endothelial ultrastructure during acute atrial fibrillation in canines

Objective: To study changes of ultrastructure of atrial endothelial cells during acute atrial fibrillation (AF) in canines, and explore the possible mechanism of AF left atrial thrombosis. Methods: A total of 16 healthy adult mongrel canines were randomly and equally divided into blank control group (only received thoracotomy without pacing) and rapid atrial pacing (RAP) group (established acute AF model). Myocardial tissue of left and right appendage were taken from two groups and received hematoxylin eosin (HE) staining, then myocardial cell morphological changes was observed under ordinary light microscope; morphological changes of appendage endothelial cells was observed under electron microscope. Results: (1) Paroxysmal AF was successfully induced in all canines of RAP group; (2) There were no significant difference in morphology of appendage and endocardial tissue under ordinary light microscope between two groups; 3. Under transmission electron microscope, endothelium cell of appendage tissue presented defect of different extent, and some shedding in RAP group; while endothelial cell layer was complete with absence of necrosis and shedding in blank control group. Compared with blank control group, there was significant rise in endothelial cell incompleteness (12.5% vs. 75.0%) in RAP group, P=0.041. Conclusion: When acute atrial fibrillation occurs, endothelial cell ultrastructure has already changed, which may be related to thrombosis adhered to wall during atrial fibrillation.
9.Study on biologic characteristics of cultured dermal papilla cells and hair follicle regeneration in vitro and in vivo

Objective To observe the ability of cultured dermal papilla cells(DPCs) to induce hair follicle regeneration and to sustain hair growth in vivo and in vitro. Methods The expression of basic fibroblast growth factor (bFGF), endothelin-1 (ET-1) and stem cell factor (SCF) in different passages of DPCs, and their possible effects on biologic behaviour of DPCs were measured by in situ hybridization and immunohistochemistry. Hair follicle regeneration induced by DPCs in hair follicle organotypic culture model and the model implantated into nude mice were studied. Results The expression of ET-1 and SCF in the early passages of cultured DPCs was strong, but became weak and even negative after 6 passages. Hair follicle-like structures were formed in the hair follicle organotypic cultures, composed of DPCs and hair follicle epithelium cells. When the hair follicle organotypic cultures were implanted into the subcutis of nude mice, relatively intact hair follicles were formed. Injection of the early passage DPCs mixed with hair follicle epithelial cells into the subcutis of nude mice resulted in the formation of hair follicle-like structures, while the structures were not formed after the injection of the mixture of hair follicle epithelial cells with dermal sheath fibroblasts or with scalp fibroblasts. There was a positive correlation between the expression levels of ET-1 and SCF in DPCs and the ability of DPCs to induce hair follicle regeneration . Conclusions Cultured DPCs can induce hair follicle regeneration and sustain hair growth in vivo and in vitro. Moreover, the expression levels of ET-1 and SCF are positively correlated with the ability of DPCs to induce hair follicle regeneration.
10.Epidemiological characteristics of viral hepatitis E in Fujian Province from 2012 to 2021

Abstract: Objective To analyze the epidemiological characteristics of viral hepatitis E in Fujian Province from 2012-2021, and to provide scientific evidence for the prevention and control of hepatitis E in the future. Methods Descriptive epidemiological method was used to analyze hepatitis E cases in Fujian Province from 2012 to 2021. Results From 2012 to 2021, a total of 8 877 cases of hepatitis E were reported in Fujian Province from 2012-2021. The overall incidence rate showed a decreasing trend (χ2trend =458.14, P<0.001), with the lowest incidence rate of 1.32/100 000 in 2020 and an annual average incidence rate of 2.29/100 000 per year. The incidence was higher in winter and spring, with the months of March and April having the highest number of reported cases (2 146, 24.17%) and the fewest cases were reported in September (571, 6.43%). The difference in reported incidence rates between cities was statistically significant (χ2=1 877.75, P<0.01). The comprehensive experimental zone of Pingtan had the highest average reported incidence rate of 6.03/100 000, while Zhangzhou had the lowest at 0.94/100 000. The number of male cases was higher than the number of female cases, with a male to female ratio of 2.04∶1. The disease was most prevalent among middle-aged and elderly individuals, with the age group of 40-<65 years having the highest number of reported cases, accounting for 57.44% (5 099/8 877) of all cases. The age group of 50-<55 years had the highest reported incidence, with the number of reported cases increasing with age below 50 years, but decreasing with age over 50 years. As for occupational distribution, peasants had the highest proportion of the disease, accounting for 34.49% (3 062 cases) of the total cases. Conclusions The reported incidence rate of hepatitis E in Fujian showed a downward from 2012 to 2021. Due to the impact of COVID-19, incidence of the lowest was 2020, but it did not reflect the true situation of the disease, which may have affected trend of hepatitis E. In order to control and reduce the incidence of hepatitis E, efforts should be made to increase publicity and education on health knowledge and vaccination among key areas and populations, strengthen monitoring and diagnostic capability, and implement comprehensive prevention and control measures.
