Histone acetylation and deacetylation is an important regulatory way of gene expression in eukaryotic cells. As a key regulatory enzyme, histone deacetylase is overexpressed in many malignant tumors, including hepa- tocellular carcinoma. In addition, it has been suggested to be a potential therapeutic target for solid tumors. In this study, we review the classification, mechanisms, as well as the expression and regulation of histone deactylases in hepatocelhlar carcinoma.

Objective To determine pathogenesis and the suitable time of operation for myelomeningocele associated with hydrocephalus in neonate.Methods 6 underwent head CT scanning, 2 lumbosacral CT scanning and 6 lumbosacral X radiography on 6 patients myelomeningocele complicated with hydrocephalus.Ventriculoperitoneal shunt and repair of the myelomeningocele were performed respectively for one patient.from 1 day to 28 day old.Operation stage 1 in 5 patients.Repair of the myelomeningocele concurred with ventriculoperitoneal shunt. Intracranial pressure was measured in shunting procedure.Results 4 patients had normal intracranial pressure,2 patients increased intracranial pressure in the 6 patients.The volume of the hernial sac had markedly diminished and status of hernial sac had greatly improved wall in the patient who wnderwent two-stage procedures after shunt procedure. Lumbosacral wound healing was good . No recurrent myelomeningocele was found, no hydrocephalus was seen using head CT scanning and clinical manifestation has improved in these patients who were followed up 6 month to 3 year.Conclusions Hydrocephalus may deteriorate the degree of lumbosacral myelomeningocele. Effecacy of vntriculoperitoneal shunt and repair of the myelomeningocele was excellent in myelomeningocele complicated with hydrocephalus in neonate.Micro-operative technique might prevent the occurrence of tethered cord.

We hypothesized that the concentrated urea and NaCl solutions may opened the blood-brain barrier to the horseradish peroxidase and trypan blue-albumin complex by shrinking barrier cells and opening up spaces between them. The experiments were carried out on 30 healthy, adult rats. Two experimental groups were used. First, intracarotid perfusion of anesthetized rats were prepared by exposing and catheterizing the left common carotid artery. A test solution of 3.4 M, 3.0 Osm urea and 0.87 M, 1.6 Osm NaCl, was perfused manually for 30 sec. in a cranial direction so as to expel the blood from the pial arterioles of the exposed brain. The pressure, which was not measured, varied between what was required which to expel blood from both arterioles and venules. Five milliliters of the test solution usually were used. Horseradish peroxidase and trypan blue was injected intravenously or through the carotid artery after perfusion. Threshold of barrier damage due to the intracarotid substance was defined as the lowest osmotality which produced obvious blue staining of the brain both on surface observation and coronal section. The effect of a substance was defined as reversible if a threshold concentration did not produce blue staining when the dye was injected 30 min following perfusion. Second, we applied a concentrated solution of 3.0 Osm urea and 1.6 Osm NaCl to the pia-arachnoid of the cerebral cortex, to study the barrier to the intravascular horseradish peroxidase and trypan blue-albumin complex. Hyperosmotic solution of 3.0 Osm urea and 1.6 Osm NaCl, either infused into one internal corotid artery or applied topically to the pia-arachnoid surface of the brain of rats, results in the opening of endo thelial tight junction through which horseradish peroxidase passes from blood to the basal menbrane and astrocytes and neurons. The evidence for this opening of the blood-brain barrier to protein is the entry of peroxidase into the neurons. It was postulated that sufficiently high concentrations of electrolytes or relatively lipid-insoluble non-electrolytes such as urea, osmotically pulled water from the cerebral endothelial cells resulting in their shrinkage. The shrinkage, in turn, was believed to open the tight junction between continuous endothelial cells so that the dyeprotein complex could pass through the junction from blood to neurons. The present study shows that these tight junctions, the sites of the barrier to neuron movement of protein, are indeed opened by the osmotic action of urea or NaCl.

OBJECTIVE:To study inhibitory effects of propofol on PC12 cells injury induced by glutamic acid via mitogen-acti-vated protein kinase/extra-cellular regulated kinase (MAPK/ERK) signal pathway. METHODS:PC12 cells were randomized into normal control group,model group(10 mmol/L glutamic acid),propofol low-concentrations,medium-concentrations and high-con-centrations groups(12.5,25,50 μmol/L+10 mmol/L glutamic acid). The optical density of cells,cell apoptosis,the phosphoryla-tion of ERK1/2 and the expression of c-fos,Bax,Bcl-2 were detected after treated with relevant medicine for 48 h. RESULTS:Compared with normal control group,optical density of cells,the phosphorylation of ERK1/2 and Bcl-2 decreased in model group (P<0.01);apoptotic rate,the expression of c-fos and Bax increased (P<0.01). Compared with model group,optical density of cells,the expression of Bcl-2 and the phosphorylation of ERK1/2 increased in propofol group (P<0.01);apoptosis rate,the ex-pression of c-fos and Bax decreased (P<0.05 or P<0.01). There were statistical significant between the different concentrations (P<0.01). CONCLUSIONS:Propofol can inhibit the apoptosis of PC12 cells induced by glutamic acid,which is associated with the up-regulation of ERK1/2 phosphorylation.

Objective: To explore the effect of low-level laser irradiation (LLLI) preconditioning on milieu of infarcted myocardium in experimental rats.
Methods: The myocardial infarction (MI) model was established by left anterior descending (LAD) artery ligation in female rats. 3 weeks later, the qualified MI rats were randomly divided for 3 groups: ① LLLI preconditioning group, the rats received thoracotomy for LLLI by a 635nm, 5mW diode laser with the energy density of 0.96 J/cm2 for 150 seconds, n=26. ② Control group, the rats received thoracotomy for daylight irradiation, n=27. ③ Sham operation group, the rats received thoracotomy without LAD ligation, n=24. The Expressions of myocardial vascular endothelial growth factor (VEGF), glucose-regulated protein 78 (GRP78), superoxide dismutase (SOD) and malondialdehyde (MDA) were evaluate by real time-PCR, Western blot analysis and other relevant laboratory test at 1 hour, 1 day and 1 week after treatment. The myocardial cell apoptosis was examined by TUNEL staining, and left ventricular function was detected by echocardiography.
Results: LLLI preconditioning obviously increased the myocardial VEGF, GRP78 expression and SOD activity, decreased MDA production; while it could not really improve the myocardial cell apoptosis at peri-infarcted area and left ventricular function in experimental rats.
Conclusion: LLLI preconditioning may improve the milieu of infarcted myocardium via decreasing the oxidative stress in experimental rats.

Objective: To explore the effect of omentopexy combining autologous atrial tissue patch cardiomyoplasty for treating the chronic myocardial infarction (MI) in experimental rats.
Methods:The MI model of SD rats was established by left coronary artery ligation. 3 weeks later, the qualiifed animals were randomized into 4 groups for the 2nd operation. ① Control group, the rats only received re-thoracotomy, ② Atrial appendage group, the autologous atrial tissue patch was harvested from left atrial appendage of rats and transplanted to infarcted zone, ③ Omentum group, the omentum of rats was transplanted to infarcted zone through diaphragm and ④ Combination group, the left atrial appendage tissue and omentum were transplanted to infarcted zone together. 4 weeks after the 2nd operation, the cardiac function was evaluated by echocardiography, the size and scar thickness of the infarction were examined by Masson staining, the survival of transplanted atrial tissue and angiogenesis were measured by immunohistochemistry, the protein expressions of VEGF, MMPs and TIMP-3 were examined by Western blot analysis, and the activities of MMPs were
identiifed by gelatin zymography.
Results:4 week after the 2nd operation, transplanted left atrial appendage tissue only survived in Combination group. The areas of infarction were similar among different groups, P>0.05. Compared with Control group, Combination group had the increased scar thickness (329 ± 33)μm vs (391±31)μm, improved LVEF (47.5 ± 4.5)%vs (57.9 ± 5.8)%, improved LVFS (20.7 ± 2.0)%vs (25.2 ± 3.6)%, all P<0.05. Compared with Control group, both Omentum group and Combination group demonstrated higher density of angiogenesis at infracted area (33/0.2) mm2 vs (49/0.2) mm2 and (33/0.2) mm2 vs (48/0.2) mm2, all P<0.01. Combination group had decreased protein expressions of MMPs, while the expressions of TIMP-3 were similar among different groups, the activities of MMP-2 and MMP-9 were decreased by 68%, P=0.002 and 64%, P=0.016 respectively.
Conclusion:Omentopexy could improve the angiogenesis and support the survival of transplanted autologous atrial tissue patch, therefore improve the cardiac function in experimental rats with chronic MI.

Objective: To conifrm the omentopexy decreasing the susceptibility of ventricular arrhythmia in experimental rats with chronic myocardial infarction (MI) by nerve remodeling.
Methods: The MI model of SD rats was established by left coronary artery ligation. 3 weeks later, the qualiifed animals were randomized into 3 groups for the 2nd operation. ① Sham operation group, the rats received the 2nd operation without ligation, ② Isolated MI group, the rats received the 2nd operation without omentopexy, ③ Omentopexy group, the rats received the 2nd operation with omentopexy. n=20 in each group. 4 weeks after the 2nd operation, the electrophysiological characteristics were assessed by relevant techniques, the new and sympathetic nerves in MI border zone were examined by immunohistochemistry, the protein expressions of connexin43 and nerve growth factor (NGF) were measured by western blot analysis and the cardiac endothelin-1 (ET-1) level was evaluated by ELISA.
Results: Compared with Isolated MI group, Omentopexy group showed decreased susceptibility of arrhythmia (3.5 ± 1.2) vs (0.9 ± 0.2), improved electrical transduction (1.5 ± 0.2) mV vs (3.4 ± 0.3) mV and decreased capture threshold (5.5 ± 0.3) mV vs (2.2 ± 0.2) mV, all P<0.05. Compared with Isolated MI group, immunihistochemisty indicated that Omentopexy group had decreased new and sympathetic nerves in MI border zone, as for GAP43:(1388.4 ± 244.9)μm2/mm2 vs (768.6 ± 144.1)μm2/mm2, for TH:(1552.4 ± 270.3)μm2/mm2 vs (1018.5 ± 124.7)μm2/mm2, all P<0.05. Western blot analysis showed that Omentopexy group had the lower NGF expression and higher connexin43 expression;ELISA demonstrated that Omentopexy group had the lower ET-1 expression, P<0.05.
Conclusion: Omentopexy may decrease the susceptibility of ventricular arrhythmia after MI in experimental rats, which might be related to the cardiac nerve remodeling.

Objective To evaluate the left ventricular diastolic dysfunction by tissue Doppler imaging (TDI)in a suprarenal abdominal aortic coarctation model of Lewis rats. Methods Thirty male Lewis rats at age of 8 weeks were used. Myocardial hypertrophy model was obtained by suprarenal abdominal aorta banding with ligation. Mitral annular velocity measurement by TDI and regular two-dimensional echocardiography with M mode were examined at baseline and at 10-,20-,30-and 40-day post banding.Results Compared with baseline,the left ventricular end diastolic and end systolic wall thickness and ventricular dimension were progressively increased,while ejection fraction(EF)and fraction shortening(FS)were decreased(P＜0.0 1).The left ventricle demonstrated centrifugal myocardial hypertrophy.Peak myocardial systolic velocity(Sa)at the mitral annulus did not change significantly throughout this study and there was no significant correlation with EF(r=0.216,P=0.071),and peak myocardial early diastolic (Ea),late diastolic(Aa)at the mitral annulus and Ea/Aa indices also were not stable after aortic banding and there were no significant difference in repeated measures at different time(P＞0.05).But the E/Ea wasincreased and the index had a statistically significant difference(P＜0.001).Conclusions The left ventricleshowed centrifugal myocardial hypertrophy in Lewis rats with abdominal aortic banding.E/Ea radio was a sensitive and stable index to evaluate left ventricular diastolic dysfunction.
