Chinese Journal of Geriatrics 2021;40(4):464-468
Objective:To investigate the effects of Leflunomide and Azathioprine on serum chemokine cx3cl1(fractalkine), cathepsin S and vascular cell adhesion molecule-1(VCAM-1)in elderly patients with lupus nephritis. Methods:Sixty elderly patients with lupus nephritis admitted to our hospital from January 2017 to July 2019 were selected as the research objects.Patients were divided into two groups by using a random number table method: the Leflunomide group(treated with Leflunomide plus conventional treatment)and the Azathioprine group(treated with azathioprine plus conventional treatment)(n=30, each group). Serum levels of fractalkine and cathepsin S and urine VCAM-1 level before and 6, 9 and 12 months after treatment were measured in the two groups and statistically analyzed. Results:In both groups, serum fractalkine levels were decreased after versus before treatment( F=123.029 and 99.041 respectively, P=0.000). At 6 months after treatment, serum fractalkine level was lower in the leflunomide group(2.34±0.95)μg/L than in the Azathioprine group(2.97±1.01)μg/L( t=2.489, P=0.016). Serum cathepsin S levels were decreased in both groups after treatment( F=106.733 and 64.928, P=0.000). Serum cathepsin S levels were lower in the Leflunomide group that in the azathioprine group at 6 and 9 months after treatment[(24.31±3.15)μg/L vs.(26.92±4.02)μg/L, (21.72±2.67)μg/L vs.(23.89±2.75)μg/L, t=2.799 and 3.101, P=0.007 and 0.003]. After treatment, urinary VCAM-1 levels were decreased in both two treatment groups( F=907.450 and 858.114, P=0.000). At 6 months after treatment, urine VCAM-1 level was lower in the Leflunomide group than in the Azathioprine group[(74.36±9.17)μg/L Cr vs.(86.91±9.22)μg/L Cr, t=5.286, P=0.000)]. The incidences of adverse reactions were low in the two groups, and the difference was not statistically significant. Conclusions:Leflunomide and Azathioprine can effectively reduce serum levels of fractalkine and cathepsin S and urinary VCAM-1 in elderly patients with lupus nephritis, which suggests that Leflunomide and Azathioprine have similar effects in inhibiting inflammatory reaction, cell matrix degradation and remodeling and vascular cell adhesion. But at early stage of therapy, the effect of Leflunomide was better than that of Azathioprine. And two drugs can be selected according to the specific clinical situation.

Objective To observe the effect of propofol on phosphorylation of a-amino-3-hydroxy-5-methyl-4-isoxa-zolep-propionate receptors (AMPARs) GluR1 subunit and long-term potentiation (LTP) in cultured hippocampal neurons in aged rats. Methods A total of 30 18-month-old rats were decapitated, the brains were rapidly removed and hippocampal slice were prepared. The slices were randomly divided into control group (perfused with artificial cerebrospinal fluid,n=10),propofol-treated group (perfused with propofol in artificial cerebrospinal fluid,n=10)and propofol+ phorbol-12-myristate-13-acetate (PMA)-treated group (perfused with propofol and phorbol ester in artificial cerebrospinal fluid,n=10).Extracellular excitatory postsynaptic potentials (EPSP) were recorded from the CA1 region of hippocampal slices. After perfusion for 20 min,LTP was induced using higher-frequency stimulation (HFS,100Hz,400 pulse) by the Schaffer-collateral pathway. The phosphorylation of AMPA-GluR1 subunit was assayed in cultured rat neurons by Western blot. Results The value of EPSP in propofol-treated group (105.50 ± 3.77) was much lower than in control group (242.10±14.68) and in propofol+ PMA-treated group (239.40±8.98) (F=2.90,P<0.05),and there was no significant difference in the value of EPSP between control group and propofol+ PMA-treated group (P>0.05).The level of P-Glu1/Glu1in propofol-treated group (0.68±0.15) was much lower than in control group (1.67±0.20) and in propofol+PMA-treated group (1.57±0.18) (F=6.84,P<0.05),while there was no difference in the level of P-Glu1/Glu1 between control group and propofol + PMA-treated group (P > 0.05).There was no difference in the value of GluR1/β-actin among the three groups (F=0.31,P>0.05).Conclusions Propofol possesses the ability to inhibit LTP induction and attenuate AMPA receptor GluR1 subunit phosphorylation through modulation of PKC pathway.

Chinese Journal of Tissue Engineering Research 2014;(29):4724-4729
BACKGROUND:Autologous cartilage has a poor self-repair effect due to low chondrocyte density, low metabolism rate and no blood supply.
OBJECTIVE:To summarize the recent study about tissue engineering techniques and surgical treatment for cartilage injury.
METHODS:A computer-based online retrieval of PubMed database was performed by the first author for articles published between January 1992 and December 2013. The key words are“articular cartilage, injury, tissue engineering, repair”in English. According to the inclusion and exclusion criteria, 61 literatures were included into the final analysis.
RESULTS and CONCLUSION:The current clinical treatment of articular cartilage injury includes joint debridement, mosaicplasty, perichondrium transplantation and autologous chondrocyte implantation. However, their long-term result is unsatisfactory. One reason for limited clinical success is that new cartilage can be formed at the site of a defect, and the repaired tissue canot compare with the autologous cartilage in mechanical property. Tissue engineering technique is stil a hot topic in recent years, because it can potential y induce autologous cartilage formation. Through endogenous or ectogeneous seed cells and inducting factor and nutrient factors, tissue engineering technique can be applied to induce the self-repair of articular cartilage, thus regenerating into hyalinc cartilage with the similar even same biological property. How to simplify the treatment protocols and reduce treatment cost is the key to promote cartilage repair.

Acta Anatomica Sinica 1955;0(03):-
Osteoclasts were isolated mechanically from neonatal rat long bones and cultured on glass or devitalized human cortical bone slices. Both morphological and functional studies showed that the isolated cells were of typical osteoclasts, because; 1. they attached rapidly on the matrix and had pseudopodial activity when cultured on glass;2. they had multinuclei, most of them contained 3-10 nuclei;3. calci-tonin and clacitonin gene related peptide directly inhibited the cellular activity; 4. they showed high acid phosphatase activity; 5. resorption lacunae could be found when the cells cocultured with devitalized bone slices.

Chinese Journal of Tissue Engineering Research 2014;(25):3987-3992
BACKGROUND:Many studies have shown that different nanostructures produce different influences on cellbioactivity, but the nanonetwork structure is not reported yet. OBJECTIVE:To study the influence of the nanonetwork topography on the bioactivity of bone marrow mesenchymal stem cels. METHODS:The nanonetwork topography was fabricated on biomedical titanium surface by alkali-heat treatment, and pure titanium served as control group. Bone marrow mesenchymal stem cels were co-cultured with the above two types of samples. cellmorphology and cytoskeleton were observed using scanning electron microscope and immunofluorescence method. The celladhesion, proliferation and osteogenic differentiation were detected by measurement of absorbance values at different culture time. RESULTS AND CONCLUSION: The nanonetwork topography had significant advantage on the number of adherent cels at 30, 60 and 120 minutes of co-culture. The cellproliferation was significantly accelerated by the nanonetwork topography at days 1, 3, 5 of co-culture, and the absorbance values in the nanonetwork group were significantly higher than those in the pure titanium group (P < 0.05). The alkaline phosphatase activity in the nanonetwork group was also significantly higher than that in the pure titanium group at 14 days of osteogenic induction (P < 0.05). The cellshape and cytoskeleton on the nanonetwork surface were better than those on the titanium surface. These findings indicate that the nanonetwork topography has better effects on cellbioactivity compared with pure titanium.
