2.Posthepatic manipulative bleeding alleviates lung injury induced by liver ischemia-reperfusion

AIM:To determine the effects of posthepatic manipulative bleeding on the lung injury induced by hepatic ischemia-reperfusion (HIR)in rat model. METHODS:Rat 35 min total hepatic ischemia model was used in this study. We treated the experimental rats by posthepatic manipulative bleeding or IH-CV manipulative bleeding at 10 min after reperfusion. RESULTS:Two percent of body weight posthepatic manipulative bleeding with blood transfusion at 10 min after reperfusion significantly decreased circulating malondialdehyde(MDA). Tissue edema,myeloperoxidase(MPO) and MDA concentrations in lung were significantly decreased 6 h after treatments. The 7 d survival rate was remarkably improved in experimental group. CONCLUSION:Two percent of body weight posthepatic manipulative bleeding with blood transfusion at 10 min after reperfusion significantly protects the rats from lung injury induced by HIR.

4.Central neuroregulatory machanisms between early life environment and glucose metabolism in adulthood

Traditionally it has been widely accepted that our genes together with adult lifestyle factors determine our risk of developing noncommunicable diseases such as type 2 diabetes mellitus ,cardiovascular disease and obesity in lat-er life.Currently, there is substantial evidence that the early life environment plays a key role in determining our suscepti -ble to such diseases in later life .As an increased focus on the understanding of central insulin resistance , central nerve sys-tem plays an extremely important role in regulating the peripheral glucose homeostasis .Substantial studies have indicated that it may be regulated by epigenetic mechanisms .Epigenetics can be defined as the study of heritable changes in gene ex-pression that do not involve alterations in the DNA sequence .Epigenetic processes play a central role in regulating tissue specific gene expression and hence alterations in these processes can induce long -term changes in gene expression and me-tabolism which persist throughout the lifecourse .This review will focus on the epigenetic machanisms of central neuroregula-tion between early life environment and glucose metabolism in adulthood .
5.The influence factors of lower extremities deep venous thrombosis after hip replacement

Objective To analyze the influence factors of lower extremities deep venous thrombosis after hip replacement,and provide guidance for perioperative prevention.Methods 52 cases of patients underwent total hip replacement were selected,who were given color doppler ultrasound examination for lower limb venous thrombosis after hip replacement.And analyzed risk factors of lower extremity deep vein thrombosis from patient's gender,age,blood lipid,D -dimer,complications and surgical index.Results During the 52 cases,16 cases were found with postopera-tive lower limb deep vein thrombosis (30.8%);Single factor analysis showed that patients with age ≥65 years,over-weight or obese (BMI≥25kg/m2 ),high triglycerides (TG ≥ 1.7mmol/L)and high D -dimer level (≥500μg/L), diabetes or high blood pressure,large blood transfusion combined with a greater incidence of postoperative deep vein thrombosis (P <0.05).Further multiple factors regression analysis showed that age ≥65 years,overweight or obesity, high triglycerides,high D -dimer,diabetes,large blood transfusions were independent risk factors of lower extremity deep vein thrombosis (OR =15.693,1.278,9.832,3.053,1.855,1.014,all P <0.05).Conclusion Patients with total hip replacement should be given preoperative vein thrombosis risk assessment,patients with age ≥65,overweight or obese,high triglycerides,D -dimer level,diabetes,large number of blood transfusions have higher risk of deep vein thrombosis,we should focus on perioperative monitoring,application of low molecular heparin,lower limb vein pump, etc.early postoperative lower limb rehabilitation exercise,to reduce the occurrence of deep vein thrombosis.
9.Association analysis between Alzheimer's disease and the promoter polymorphism of PSEN2 gene

Objective Alzheimer's disease (AD) is the most common neurodegenerative disorder of aging.Identifying novel AD genetic risk factors is important for understanding its pathogenesis.Presenilin- 2 (PSEN2) is one of the causative genes for familial AD.Polymorphism of the promoter region of PSEN2 has recently been reported to be associated with sporadic AD in a Russian population.The purpose of this study was to determine whether AD is associated with the PSEN2 gene polymorphism in a case-control study.Methods We examined PSEN2 and ApoE genotypes of 160 patients with AD and an equal number of age- and sex-matched controls from the same community by using PCR-RFLP method.Statistical analyses included a Chi-square test for homogeneity and a Logistic regression analysis.Results Examination of PSEN2 genotypes revealed no statistically significant differences in the frequency of the+A/-A polymorphism when compared with the total sample of AD patients and control individuals (P=0.11). However,subgroup of ApoE 84 non-carriers suggested significant differences of the three genotypes of PSEN2 promoter between AD and controls (P=0.038).Logistic analyses indicated ApoE genotypes were related with AD,and after stratification by e4 allele,PSEN2 genotypes were related with AD of e4 non-carriers.In addition,there was no interaction between ApoE and PSEN2 gene.Conclusion The+A/-A polymorphism of PSEN2 promoter may be a minor genetic risk factor for sporadic AD,especially in the group of AD without ApoE ?4 allele.
