OBJECTIVETo study the effects of continuous dry-stress and full-water treatments in different periods of spring on the water condition, permeability of plasma membrane and protective enzymes activities in leaves of Ligusticum chuanxiong.
METHODPot cultivation method was applied and physical and biochemical indexes were measured.
RESULTUnder dry-stress treatment the soil relative water content (SRWC) and the relative water content (RWC) in leaves decreased gradually with the days of treatment increased, the content of malondialdephyde (MDA) and permeability of plasma membrane increased significantly. The activities of superoxide dismutase (SOD) and catalase (CAT) increased at first and then decreased while the activity of peroxidase (POD) increased. The influence of full-water treatment to all above indexes was the same trend with that of dry-stress treatment approximately but was not significant.
CONCLUSIONIn this experiment, the suitable soil relative water content for growth of Chuanxiong is about 60%.

Objective To study the role of Th17 cells proportions and the cytokines levels in the diagnosis and treatment of acute pancreatitis (AP).Methods Patients with AP (n =82) treated in our hospital between August 2017 and August 2018 were divided into the mild AP group (MAP,n =36),the moderately severe AP group (MSAP,n =26) and the severe AP group (SAP,n =20).Twenty-five healthy subjects were chosen as the control group. The proportions of Th17 cells and the levels of cytokines including IL-17,IL-21,IL-22 and IL-23 in peripheral blood from the four groups were analyzed. The APACHE Ⅱ and Ranson scores were used to evaluate the illness severity.Spearman correlation analysis was conducted to detect the correlation between the Th17 cells, cytokines, and inflammatory factor,APACHE Ⅱ and Ranson scores. Results Compared with the control group, the proportion of Th17 cells and the levels of IL-17,IL-21,IL-22 and IL-23 in the peripheral blood of MAP,MSAP and SAP groups were significantly increased (SAP > MSAP > MAP,P < 0.05).The levels of inflammatory cytokines IL-6,IL-8 and TNF-α,and the scores of APACHE Ⅱ and Ranson in the SAP group were significantly higher than those in the MSAP group and MAP group (P < 0.05).The proportion of Th17 cells and their cytokine levels were positively correlated with IL-6,IL-8,TNF-o,APACHE Ⅱ and Ranson scores. The levels of Th17 cytokines returned to normal in AP patients after treatment. Conclusion Th17 cells and the cytokines have certain clinical significance in evaluating early inflammatory response, severity of illness, and therapeutic effect in AP patients.

OBJECTIVETo explore the effects of bone marrow mesenchymal stem cells (MSCs) on in vitro expansion potential, the adherent molecules expression of cord blood (CB) CD34(+) cells.
METHODSMSCs were obtained from human bone marrow and their differentiation function and phenotype were identified. CB CD34(+) cells were expanded in culture systems with or without MSC layer. Hematopoietic progenitor cells and adhesion molecules expression were assessed by semisolid culture assay and flow cytometry.
RESULTSThy-1, SH2, SB10, CD44, CD13, CD49e and CD29 were highly expressed on MSCs with no expressions of CD34, CD45, HLA-DR, CD14 and CD31. The MSCs could differentiate into adipocytes and osteoblasts under specific induction conditions. After culturing on MSCs layer with supplement of cytokines for 8 days, the absolute numbers of nuclear cells, CD34(+), CD34(+)CD38(-), CD34(+)CD62L(+) cells and CFU-Cs were increased by 145.57 +/- 17.89, 37.47 +/- 13.78, 69.78 +/- 50.07, 10.74 +/- 5.89 and 20.73 +/- 5.54-folds, respectively, being significantly higher than that cultured with cytokines alone. The expression of ALCAM, VLA-alpha4, VLA-alpha5, VLA-beta1, HCAM, PECAM and LFA-1 on CD34(+) cells remained unaffected. The expressions of ICAM-1 and L-selectin were downregulated during expansion, while the absolute numbers of CD34(+)CD62L(+) and CD34(+)CD54(+) cells were increased.
CONCLUSIONSMSCs layer improves expansion of CB CD34(+) cells while inhibiting their differentiation and retaining their homing ability.

OBJECTIVETo summarize and discuss the lapsus and the treatment of the lumbar intervertebral disc herniation using percutaneous endoscopic lumbar discectomy (PELD).
METHODSBetween July 2002 and October 2010, 689 patients with lumbar intervertebral disc herniation treated by PELD were analyzed, including 448 males, and 241 females. Single lumbar intervertebral disc herniation were 669 cases. double lumbar intervertebral disc herniation were 19; three lumbar intervertebral disc herniation were 1. Central type in 66, side central type in 365, lateral type in 242, extreme lateral type in 10, sequestered type in 6. These cases with complications in operation and postoperation were studied retrospectively.
RESULTSThere were nucleus pulposus omissions in 5 patients and 2 patients underwent open resection of nucleus pulposus during operation immediately and the second operation was needed in 3 cases, 1 case with transforaminal lumbar interbody fusion (TLIF) and the others with open resection of nucleus pulposus. Two patients had nerve root injury, but all completely recovered in 3 - 6 months after operation. Spinal dura mater disruption was in 2 patients, recovered after suturing of skin wound. All 689 patients were followed up for 6 - 96 months, mean follow-up time was 33 months. Postoperative spondylodiscitis was in 7 patients, recovery after expectant treatment in 1, percutaneous puncture irrigation and drainage for continued use of local antibiotics in 4, posterior infective lumbar discectomy in 2. Postoperative relapse was in 6 patients, operated secondly by PELD in 4 and by TLIF in 2, recovery after the second operation. Nerve root induced hyperalgesia and burning-like nerve root pain was seen in 19 patients, the symptom was improved by analgesic drug, neurotrophy drug and physiotherapy. The effect of single segment PELD was not good in 10 patients with spinal stenosis, who underwent multiple segment TLIF later.
CONCLUSIONSThe complications during operation usually are nucleus pulposus omissions, nerve root injury, spinal dura mater disruption. Accordingly the complications after operation include spondylodiscitis, recurrence, nerve root induced hyperalgesia or burning-like nerve root pain. Strict indication, aseptic technique, skilled operation and proper rehabilitation exercise are effective ways to reduce complications.
