1.Construction of a middle fragment-deleted class Ⅱ molecule transactivator mutant by modified OE-PCR technique

Objective:To develop a simple and efficient method for constructing a middle fragment-deleted mutant of MHC class Ⅱ molecule transactivator(CⅡTA)mutant with the 109~(th)to 226~(th) amino acid codons deleted.Methods: Two gene fragments at each end of the deleted CⅡTA gene were obtained by OE-PCR method and were mixed together for 8 PCR cycles without primers to achieve effective overlapping,then 2 primers was added for amplification of the desired fragments.The amplification products were subsequently cloned into eukaryotic vector pIRES for identification.Results: A mutant of CⅡTA with the 109~(th)to 226~(th) amino acids deleted was successfully constructed.Conclusion: This modified OE-PCR technique overcomes some shortcomings of traditional method and is very suitable for constructing mutants with middle fragment deletion,making it worth to be popularized.

3.Survey on human resources and the cost of essential public health services based on community health service centers in Beijing’s rural areas

Objectives:To understand the human resources and costs of essential public health services based on community health service centers in Beijing’s rural areas and to offer policy recommendations for advancing the progress of essential public health services. Methods:With the self-designed questionnaire, the paper collected data from the districts of Changping, Tongzhou, Huairou and Miyun. By cluster random sampling, 12 community health service centers and 108 personnel were surveyed based on the level of economic development. Results: The number of young women, preventive personnel and staff with low ranking positions represents a significant share of all essen-tial public health service providers. Beijing’s rural areas deploy a great deal of human resources for vaccination and health education with little set aside for women’s health and reproductive health. The unit cost of setting special col-umns for health education and field disease treatment is higher than other service items. The suburbs deploy a great deal of human resources for service items than the outskirts while the unit costs are lower. Conclusions: Human re-source for essential public health services in Beijing’s rural areas should be strengthened. The costs of service items differ greatly so that policy adjustments and finances should be implemented based on actual demand and resources.

7. Effects of puerarin combined with edaravone on inhalation lung injury induced by black gunpowder smog in rats

Objective To explore the protective effects of puerarin combined with edaravone on inhalation lung injury induced by black gunpowder smog in rats. Methods Forty healthy male Wistar rats were randomly divided into normal control group (group N), inhalation group (group X), puerarin group (group P), edaravone group (group E) and edaravone combined with puerarin group (group L), with 8 rats in each group. Rat model of inhalation lung injury was reproduced by a self-made smoke generator. Rats in group E were given intraperitoneal injections of edaravone (9 mg/kg) at 30 minutes and 1 day after modeling (twice totally). Rats in group P were given intraperitoneal injections of puerarin (100 mg/kg) at 30 minutes and 1, 2, 3, 4, 5 days after modeling (6 times totally). Rats in group L were treated the way of both group E and P. The rats in group N and group X were given intraperitoneal injections of normal saline (12 ml/kg) at the time-points above. The animals were sacrificed 6 days after modeling, and the blood samples were collected from abdominal aorta to assess arterial blood gas values, meanwhile the serum levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10) were determined by ELISA. Lung tissue homogenates were prepared to determine the protein content and myeloperoxidase (MPO) activity. The pathological changes in the lung tissue with HE staining were observed under light microscope. Results Arterial blood gas analysis revealed that the PaO2 levels in groups P, E and L were higher than that in group X (P<0.05), and the PaO2 levels in groups E and L were higher than that in group P (P<0.05), while the PaCO2 level in group L was lower than that in groups X and E (P<0.05). The TNF-α, IL-6 and IL-10 levels in serum, the protein content and MPO activity in lung tissue homogenate in groups P, E and L were lower than those of group X (P<0.05). The TNF-α and IL-6 levels in serum and protein content and MPO activity in lung tissue homogenate in groups E and L were lower than those of group P (P<0.05), while IL-10 level in serum was higher than that of group P (P<0.05). The TNF-α and IL-6 levels in serum and protein content in lung tissue homogenate in group L were lower than those of group E (P<0.05). HE staining revealed that lung edema was alleviated and inflammatory exudate and infiltration in groups P, E and L were less marked when compared with group X, while they were more significant in group L. Conclusions Edaravone combined with puerarin can alleviate the inflammatory reaction of inhalation lung injury induced by black gunpowder smog via reducing the production and release of pro-inflammatory mediators, thus playing a protective role against lung injury. The effect of a combination of the drugs is better than that of either drug alone.
8.Serum proteomics studies on the acute myocardial infarction patients

Objective To approach the significance of serum protein fingerprinting on the identification of different courses of acute myocardial infarction. Methods For 153 patients of acute myocardial infarction, including 45 cases of just admitted patients, 12 cases of 3h admitted patients, 22 cases of 6h admitted patients, 24 cases of 9h admitted patients, 24 cases of 12h admitted patients, 16 cases of 24h admitted patients and 10 cases of 48h admitted patients. The relative contents of serum proteins were detected by IMAC3 chip and proteinchip reader (CipherGen Inc., VS). Results On the M/Z values ranged from 4KDa to 12KDa, ten protein contents were obviously different between the different courses of acute myocardial infarction. All the patients in different courses were diagnosed correctly, the accuracy was 100%(153/153). Both sensitivity and specificity were also 100% in learning mode. Conclusion Different courses of acute myocardial infarction patients can be quickly and correctly diagnosed by this method with high sensitivity and specificity. That will be widely used in clinical application
9.Growing insights into acid-sensing ion channels

Acid-sensing ion channels( ASICs )are ubiquitously expressed both in periphery nervous system, where they are involved in nociception,mechanosensation,inflammation ,cardiac angina, and in central nervous system,where they are essential to a variety of physiologic and pathophysiologic processes,such as synaptic plasticity ,learning ,memory and ischemic brain injury. Here in the article, we present a collection of key points about ASICs, ranging from molecular identity and expression, regulatory mechanisms responsible for channel gating to their multiple functions. Finally, a future prospect for the investigation of ASICs is outlined.
10.Clinical and molecular biological characteristics of spinocerebellar ataxia type 3

Objective To explore the clinical and molecular biological characteristics of spinocerebellar ataxia type 3(SCA3).Methods Clinical manifestation and brain MRI data of 12 patients with SCA in two families were analyged.The polymorphic CAG repeated time in the encode region of SCA3,SCA1 and SCA7 genes were compared in 15 family numbers without abnormal presentations,and 12 healthy persons of controls.Results Among 27 numbers of 4 generations in the two families had 12 patients,male and female were affected,average onset was 32 years old.The main clinic features included gait ataxia,ambiguity in speech and action clumsiness.Brain MRI showed remarkable atrophy on cerebellum and brain stem.In the two families,the CAG lengths of SCA1 and SCA7 were normal in all numbers.The repeated times of CAG of SCA3 were 11~39 in two control groups,65 ~87 in 10 cases,diagnosed as SCA3 patients.The child Ⅳ2 of family 1 was 8 years old,the repeated times of CAG of SCA3 were repeats 21 and 64 times,repectively.He might be a asymptomatic patient,because he was too young to onset the disease.Conclusions SCA3 is an autosomal dominant genetic disease.The clinical manifestations are ataxia and dysarthria.The detection of repeated times CAG can provide an effective way for the genetic and asymptomatic diagnosis.
