1.The expressions of PTEN,Survivin in tissues of non-small cell lung cancer and its relationship with microvas-cular density

Objective To study the expressions of PTEN ,Survivin in tissues of non-small cell lung cancer and their relationship with microvascular density ( MVD) .Methods 66 cancerous tissues of non-small cell lung canc-er were selected as the observation group ,and adjacent tissues were selected as the control group .PTEN,Survivin and MVD were determined by immunohistochemical staining .Results 17 cases(25.8%) were PTEN positive in the ob-servation group,51 cases(77.3%) were PTEN positive in the control group ,there was significant difference between the two groups(χ2=6.92,P<0.05).Survivin positive expression was 48 cases(72.7%) in the observation group, which was significantly higher than 12 cases(18.2%) in the control group(χ2 =5.72,P<0.05).MVD value was (74.6 ±18.3) in the observation group,which was significantly higher than (23.5 ±6.1) in the control group(t=6.722,P<0.01).There was no significant relationship between PTEN ,Survivin expression and gender ,histological type(χ2 =0.16,0.27,0.22,0.24,all P>0.05),and PTEN,Survivin expression had close relationship with tumor differentiation,lymph node metastasis and distant metastasis (χ2 =4.39,4.92,5.76,5.29,6.26,4.25, all P <0.05).PTEN was negatively correlated with MVD (r=-0.393,P<0.05).Survivin was positively correlated with MVD(r=0.544,P<0.05).Conclusion Low expression of PTEN and overexpression of Survivin are related with the occurrence and development of non-small cell lung cancer .
2.S100B Protein and Central Nervous System Injury

S100B protein has a higher sensitivity to central nervous system(CNS),which can be used in clinical and prognostic prediction of various forms and different degrees of CNS injury,even in the judgment of brain death.The measurement of S100B protein and prediction of secondary brain injtry enable physicians to select an appropriate opportunity for therapy intervention,which will be a focus in the future studies.S100B protein is a potential protein biomarker that has clinical prospect.
3.Advances of Oct4 gene research

Oct4 gene,a member of POU transcription factor family,is expressed in embryonic stem(ES) cells and embryonic carcinoma(EC) cells,but not in cells of differentiated tissues.It plays a critical role in maintaining pluripotent and self-renewing state of embryonic stem cells.The molecular structure,function,regulation passageway and the relationship with tumors of Oct4 gene have been briefly reviewed.
4.Detection of Chlamydia trachomatis Infection by Ligase Chain Reaction in STD Patients

Objective To evaluate the performance of ligase chain reaction(LCR) in the detection of Chlamydia trachomatis(Ct) from urethral or cervical samples in STD patients. Methods Ct was detected by LCR in urethral or cervical swabs from 276 cases with urethritis or cervicitis attending our STD clinic. All 276 urethral or cervical swab specimens were pooled by 4 into 69 pools for detection. Chlamydial cell culture was performed in 56 cases with nongonococal urethritis or cervicitis. Discrepant results were analyzed by PCR with primers targeting Ct major outer membrane protein gene. Results The sensitivity and specificity of LCR assay were 96.7% and 100% respectively. Two LCR approaches, pooling or individual testing, yielded 100% of consistency in the detection of Ct in urethral or cervical specimens. Conclusions LCR provides a highly sensitive and specific assay for detection of Ct from urethral and cervical samples, and could be recommended for the diagnosis of genital chlamydial infection. Pooling LCR is suitable for screening of urethral or cervical Ct infection in population study.
5.Solid tumor cell apoptosis induced by arsenic trioxide is related to generation of reactive oxygen species

AIM: To study the effects of As_2O_3 on the growth of human cervical carcinoma cell line, HeLa cells, human pancreatic carcinoma cell line, AsPC-1 cells, and the generation of reactive oxygen species (ROS) in the cells. METHODS: HeLa cells and AsPC-1 cells were treated with various concentrations of As_2O_3. The effects of As_2O_3 on HeLa cells and AsPC-1 cells survival and apoptosis were determined by MTT assay and light microscope, and cellular ROS was also measured by fluorometer. RESULTS: After being treated with 2 ?mol/L As_2O_3 for 48 h, the survival of HeLa cells was decreased, a marked apoptosis characteristic was observed in time- and dose-dependent manner. However, the survival of AsPC-1 cells markedly decreased after being treated with even 1?mol/L As_2O_3 for 24 h. When we measured the cellular hydrogen peroxide (H_2O_2) level, we found that the H_2O_2 level in HeLa cells started to rise after being incubated with As_2O_3 for 2 h, and sustained increased, reached the peak of H_2O_2 level at 8 h, then it began to decrease. While the H_2O_2 level in AsPC-1 cells started to rise at 2 h, reached the highest point at 5 h, after which it began to descend. At 12 h, the H_2O_2 level in HeLa cells and AsPC-1 cells were similar to that of the control group. CONCLUSION: The molecular mechanisms underlying As_2O_3 -induced apoptosis in cells derived from the solid tumors may be related to the cellular ROS level.
6. Expression of connexin43 and its correlation with the expression of bcl-2 and bax protein in transitional cell carcinoma of the bladder

Objective: To study the expression of connexin43 (Cx43) gene and its correlation with the expression of apoptosis related genes bcl-2 and bax in transitional cell carcinoma of the bladder (BTCC), and to investigate the role of Cx43 in the BTCC. Methods: Reverse transcription polymerase chain reaction was used to detect the expression of Cx43 mRNA and immunocytochemistry technique was used to detect the expression of bcl-2 and bax proteins in 60 cases of BTCC tissue, and compared with that of 15 cases of pericancerous tissue and 15 cases of normal bladder tissue. Results: The positive rate of Cx43 mRNA expression in 60 cases of BTCC tissues was 43.33% which was significantly lower than that in pericancerous tissues (73.33% ) and normal tissues (100%) (x2 = 17.58, P < 0.01). The expression of Cx43 had significant negative correlation with the pathological degree and lymph node metastasis of BTCC (x2 = 9.33 and 9.74, respectively, P < 0.01). However, no correlation was found between the expression of Cx43 and the sex, age, clinical staging, and the diameter and the growth pattern of BTCC (P >0.05). Expression of Cx43 negatively correlated with bcl-2 protein (r = 0.63, P < 0.01) and positively correlated with bax protein (r = 0.52, P < 0.01). Conclusion: The down-regulated expression of Cx43 gene was closely associated with the development, invasion and metastasis of BTCC. It could be used a prognostic indicator for BTCC. Cx43 gene may have antagonistic effects with bcl-2 gene and synergistic effects with bax gene in the initiation and progression of BTCC.
7.Clinical Study on Treating Lumbar Intervertebral Disc Protrusion Accompanied with Secondary Lumbar Spinal Stenosis by Acupuncture Combined with Medication Plus Joint Mobilization Treatment

Objective To study the clinical efficacy of acupuncture combined with medication plus joint mobilization in the treatment of lumbar intervertebral disc protrusion accompanied with secondary lumbar spinal stenosis.MethodsTotally 66 cases of lumbar intervertebral disc protrusion accompanied with secondary lumbar spinal stenosis were collected randomly and divided into treatment group (34 cases) and control group (32 cases). The control group was given treatment of simple acupuncture and TCM medication, while the treatment group was given joint mobilization treatment besides acupuncture and TCM medication. Functions of lumbar vertebra were evaluated by ODI scale and the degrees of pain were evaluated by VAS. The clinical efficacy in the two groups was compared. Results ODI in both groups were significantly improved after treatment compared with that before treatment. However, the changing range of the ODI of the treatment group was more significant than that in control group (P<0.01). After treatment, VAS scores were relieved (P<0.05) in both groups, and treatment group was more significant than that in control group (P<0.05). The total clinical efficacy was 97.06% (33/34) in the treatment group, and 84.38% (27/32) in the control group, with statistical significance (P<0.05).Conclusion Acupuncture combined with medication plus joint mobilization in the treatment of lumbar intervertebral disc protrusion accompanied with secondary lumbar spinal stenosis has good efficacy.
8.Clinical analysis of 12 patients caused by long-acting anticoagulant rodenticide occult poisoning

Objective:To explore the clinical characteristics and diagnosis and treatment of occult poisoning caused by long-acting anticoagulant rodenticides.Method:Records of 12 patients from July 2008 to April 2011 diagnosed as anticoagulant rodenticide occult poisoning,who had been misdiagnosed initially at other hospitals were analyzed retrospectively.Elements from the records included clinical symptoms and signs,laboratory findings for prothrombin time (PT) and activated partial prothrombin time (APTT),and initial misdiagnosis and treatment outcome at our hospital.Results:The clinical presentations of patients were insidious and serious,often presented as skin ecchymose,hematuria,menorrhagia and gastrointestine bleeding.Laboratory examinations showed prolonged PT and APTT; bleeding was controlled effectively by administoring vitamin K1 daily.There were statistical difference between PT and APTT before and after the treatment (P<0.01).Conclusion:Coagulation disorders might be caused by the reduced acquisition in vitamin K dependent coagulation factors,which tends to be neglected due to a hidden medical history,delayed signs of poisoning,and various organs involved.A detailed patient history and systematic review may improve the diagnostic accuracy.Once diagnosed is made,vitamin K1 should be given as soon as possible.
9.Effect of low dose fentanyl on target plasma concentration of propofol that prevents response to proseal laryngeal mask insertion in 50% of patients

Objective To evaluate the effect of low dose fentanyl on target plasma concentration (CP) of propofol (P) given by target controlled infusion (TCI) required to prevent response to proseal laryngeal mask insertion in 50% of patients (EC50). Methods Forty-six ASA Ⅰor Ⅱ patients aged 20-50 yr with body mass index ≤ 30 kg/m2 undergoing elective gynecological laparoscopy were randomly divided into 2 groups (n=23each):propofol group (P) and propofol-fentanyl group (PF). Anesthesia was induced with TCI of propofol. The initial Cp of propofol TCI was set at 2.5 μg/ml in group PF and 4 μg/ml in group P. Following equilibration between plasma and effect site concentration of propofol, fentanyl 1 μg/kg (in group PF) or normal saline (in group P) was injected iv. Laryngeal mask airway (LMA) was inserted at 3.5 min after fentanyl administration. EC50 was determined by up-and-down sequential trial. The ratio of Cp between the two consecutive patients was 1.2 in group P and 1.1 in group PF. LMA insertion response was defined as nausea, bucking and/or body movement during insertion. "Probit method was used to calculate the Echo and 95 % confidence interval required to prevent LMA insertion response. Results The EC50 and 95 % confidence interval of propofol given by TCI required to prevent LMA insertion response was 4.68 (4.20-5.21)μg/ml in group P and 2.63 (2.45-2.83) μg/ml in group PF. There was significant difference between the 2 groups. Conclusion Fentanyl 1 μg/kg iv can significantly reduce the Cp of propofol TCI required to prevent LMA insertion response.
10.Expression of Oct4 gene in cervical cancer

Objective To study the expression of Oct4 gene, an embryonic stem cell marker, in cervical carcinoma cell lines and tissues for the purpose of finding the relationship between this gene and cervical cancer. Methods The expression of Oct4 in normal cervical tissues and cervical carcinoma tissue and cell lines was detected by the immunohistochemistry and RT-PCR. Results ① The expression of Oct4 mRNA in cervical carcinomas (73.3%, 22/30) was significantly higher (P<0.01) than that in normal cervical tissues (25%, 3/12); ② The rate of Oct4 protein expression in cervical cancer (53.7%, 22/41) was higher (P<0.01) than that in normal cervical tissues (7.7%, 1/13); ③ Oct4 protein was expressed in cervical carcinoma cell lines and primary tumorspheres. ④ The expression of Oct4 in tumorspheres was down-regulated during cell differentiation. Conclusion Our results imply that Oct4 may be one of the cervical cancer stem cell markers, but this still needs further research data to support.
