1.Expression of Inducible Co-Stimulator on T lymphocytes in Peripheral Blood from the Patients with Systemic Lupus Erythematosus

Objective To investigate the expression of inducible co-stimulator (ICOS) on T lymphocytes in peripheral blood from the patients with systemic lupus erythematosus (SLE). Methods Two-color immunofluorescent staining and flow cytometric assay were used to analyze the level of ICOS expression on T lymphocytes in peripheral blood from 33 SLE patients and 16 healthy volunteers. The data of systemic lupus erythematosus diseases activity index (SLEDAI) in SLE patients were also collected. Results The level of ICOS expression on T cells from active SLE group was markedly higher than that in the control group and the inactive SLE group (P 0.05). However, a positive correlation between the expression level of ICOS and the SLEDAI score was found in both active SLE group and inactive SLE group(r = 0.711,P = 0.001?r = 0.561,P = 0.03). Conclusions The expression of ICOS on T cell in peripheral blood from active SLE is increased, which might play a role in the pathogenesis of SLE.
2.Auditory Center Affected by Chronic Noise Exposure

In order to study the effect of chronic noise exposure on auditory center, seven guinea pigs were .exposured to 105 dB A noise 8h daily for 45 d. Then the animals were killed by decapitation. The superior olive and inferior colliculus were taken immediately to make the samples of electron microscope. The mitochondria swelling, membrane rupture, cristae breaking, ballooning degeneration were found. The rough surfaced endoplasmic reticulum was swollen and expanded. The lysosome and secondary lysosome were increased. The cell interstitial edema and synaptic besicle decrease were observed. The nuclear chromatid was decreased and nucleolus was kept to the side. The laminae of neurilemma of nerve fibers were dissociated, swollen and broken. The results indicate that the ultrastructure, neural cells and fibers in the auditory center are damaged by chronic noise exposure and most of these damages are irreversible.

Objective: To compare nephrotoxicity and ototoxicity of gentamycin administered in single dose or multiple dose daily in guinea pigs. Methods: Thirty two male guinea pigs were divided into physiological saline control, single dose group daily (gentamycin, 120 mg/kg, 1/d) and multiple dose group daily (gentamycin, 60 mg/kg, 2/d). The physiopathology of renal and cochlea in guinea pigs were examined using auditory brainstem response (ABR), SC sound irritation and electron microscope. The gentamycin concentrations in serum and in perilymph were monitored by fluorescene polarization immunoassay (FPIA).
