Objective Study the relationship between the component of medium and spore formation of Bacillus sp WTFY. Methods Alter the carbon source、nitrogen source、inorganic concentration and kinds in turns, observe the sporulation circumstances of Bacillus sp WTFY.Results When glucose concentration was higher over 0.1%, spore did not emerge;Nitrogen source、phosphorus、magnesiumions concentration and kinds have some degree influence,but not significantly.Conclusion Carbon source concentration is a key factor that influence spore formation.
2.Effects of bone marrow mesenchymal stem cell transplantation on ethology of rats with Alzheimer's disease

BACKGROUND: Previous studies have shown that bone marrow mesenchymal stem cells (BMSCs) may be a promising candidate for cell-replacement therapies for neurodegenerative diseases including Alzheimer's disease (AD). But, it is still unclear whether BMSCs have therapeutic effects on AD.OBJECTIVE: To investigate the influence of BMSC transplantation on ethology of rats with AD, and to find a potential strategy for the development of effective therapies for the treatment of AD.DESIGN, TIME AND SETTING: The randomized controlled study was performed at the Laboratory of the Department of Physiology and Pathophysiology, Medical College of Chinese People's Armed Police Force in March 2009.MATERIALS: A total of 40 healthy male Wistar rats aged 24 months were used in this study, weighing approximately 450 g.METHODS: The natural senile AD rat model was chosen by maze test. The AD rats were randomly divided into four groups with ten animals in each group. In the control group, rats were injected bilaterally with physiological saline into the hippocampus. In the BMSC transplantation group, rats received BMSCs. In the normoxic differentiation and transplantation group, rats received injection of BMSCs induced from neuron-like cells under normoxic condition. In the hypoxic differentiation and transplantation group, rats received injection of BMSCs induced from neuron-like cells under hypoxic condition.MAIN OUTCOME MEASURES: The learning and memory ability of the AD rats were detected by Y type maze test 8 weeks later.Memory ability was tested 48 hours after learning test.RESULTS: The learning and memory scores decreased in control group and increased in BMSC transplantation group, there were all not statistics significance compared with that before transplant treatment (P > 0.05). The learning and memory scores were all higher than before in normoxic differentiation and transplantation group and hypoxic differentiation and transplantation group. There was significant difference between control group and the other groups (P < 0.01).CONCLUSION: BMSC transplantation may improve the learning and memory ability of AD rats. Our results demonstrate that BMSCs not only play an important role in improving cognitive disturbance of AD rats. Oriented differentiated BMSC transplantation was superior to non-differentiated BMSCs. Transplantation of hypoxic differentiated BMSCs has obtained optimal outcomes.

5.β-arrestin2 promoting mice colitis through impairment of epithelial barrier function

Objective To investigate the role of β-arrestin2 in intestinal inflammation and illustrate the mechanisms from the perspective of epithelial barrier function. Methods Dextran sodium sulfate(DSS)is used to induce acute intestinal colitis in mice. The experiment groups are designed as the wild type control(WT),the wild type colitis (WT+DSS) and the β-arrestin2- knockout colitis (KO+DSS). The expression of β-arrestin2 gene by mRNA and protein level is compared between the WT and WT + DSS groups. The difference of weight loss , disease activity index(DAI),spleen weight,colon length,histological score,intestinal permeability and important tight junction proteins (occludin ,claudin1 and ZO-1) were detected in the WT+DSS and KO+DSS groups. Results Compared with the WT group,the expression of β-arrestin2 was significantly higher in the colon of the WT+DSS group. Compared with the WT+DSS group,the KO+DSS group had less weight loss(P < 0.05),lower DAI(P<0.05),smaller spleen,longer colon and lower histological score(P=0.002). The KO+DSS group had a lower intestinal permeability(P = 0.009)and higher protein level of occludin and claudin1.There was no signifi-cant difference of ZO-1 in the two groups. Conclusion β-arrestin2 may promote mouse colitis through impairment of epithelial barrier function.
6.Effect of 3 isoforms of Ikaros on proliferation of human ovarian cancer SKOV3 cells

AIM:ToinvestigatetheeffectofIkarosisoformsontheproliferationofhumanovariancancerSK-OV3 cells.METHODS:Three isoforms of Ikaros, IK1, IK2 and IK6, were transfected into ovarian cancer SKOV3 cells. CCK-8 assay and cell counting were used to detect the effects of Ikaros isoforms on the proliferation of SKOV 3 cells.The cell cycle was analyzed by flow cytometry .The cell cycle-related proteins were detected by Western blot .RESULTS:IK1 and IK2 expression inhibited SKOV 3 cells proliferation .Flow cytometry analysis indicated that IK 1 and IK2 induced SK-OV3 cell cycle arrest at the G 1 phase.IK6 isoform exerted no obvious effect on the proliferation or cell cycle of SKOV 3 cells.Compared with control EV group , IK1 group and IK2 group showed a dramatic elevation in the expression of the cell cycle inhibitor p21, along with a substantial decrease in the expression of the cell cycle inducers cyclin D 1 and cyclin D2, which did not change in IK 6 group.CONCLUSION:IK1 and IK2 significantly inhibit the proliferation of ovarian cancer SKOV3 cells and induce cell cycle arrest at G 1 phase by regulation of cell cycle-related proteins cyclin D1, cyclin D2 and p21, while IK6 isoform exerts no obvious effect on the proliferation and cell cycle of SKOV 3 cells.

8.Research on developmental characteristics of autistic children with different functional levels

Objective To explore the developmental characteristics of different functional levels of autism and the diffe-rence in children. Methods A total of 159 autistic children aged 13-71 months including 85 high-functioning autistic (HFA) chil-dren and 74 low-functioning autistic (LFA) children were evaluated for their developmental characteristics by the development scales from the psycho-educational profile-revised (PEP-R). Results Within the same age group, both HFA and LFA children had obvious unbalance in individual ability development (H=41.68~113.51, P<0.01). Impaired cognitive performance is most common in HFA children. The development of cognitive performance was inconsistent with cognitive expression while the gross motor development was normal. The ability development in LFA children were all impaired, especially in imitation, cogni-tive performance and cognitive expression. With increasing age, the scores of imitation, perception, cognitive performance, cog-nitive expression and overall development quotient were increased gradually in HFA children (P<0.05) while the scores of cogni-tive performance and overall development quotient were decreased in LFA children. Conclusions Autistic children tend to have abnormal developmental progress and order, and individual ability development varies by functional level in autistic children with increasing age.
9.MicroRNA expression profiling in male and female model mice after renal ischemia-reperfusion injury

BACKGROUND:Renal ischemia-reperfusion injury has been shown to exhibit gender difference, but its precise mechanisms deserve further investigations. OBJECTIVE:To investigate the differential expression of microRNAs in the kidney between female and male mice in order to study the effects and mechanisms of microRNA in pathogenesis of ischemia-reperfusion injury between different genders. METHODS:Male and female mice received kidney ischemia for 45 minutes and reperfusion injury for 24 hours. Simultaneously, male and female sham surgery groups served as controls. The microRNA gene chip technology was used to detect the differences of microRNA expression in the kidney of male and female mice at 45 minutes after ischemia and 24 hours of reperfusion as wel as after sham surgery. The threshold of difference in expression among samples was double. RESULTS AND CONCLUSION:Five microRNAs were up-regulated between female and male ischemia-reperfusion injury groups. Twenty-nine microRNAs differential y expressed in the female ischemia-reperfusion group and female sham surgery group, including 25 up-regulated microRNAs and 4 down-regulated microRNAs. Thirty-eight microRNAs differential y expressed in male ischemia-reperfusion injury group and male sham surgery group, including 9 up-regulated microRNAs and 29 down-regulated microRNAs. 102 microRNAs differential y expressed in the female sham surgery group and male sham surgery group, including 22 up-regulated microRNAs and 80 down-regulated microRNAs. Results suggested that there was differential expression in microRNAs in the kidney before and after renal ischemia-reperfusion in male and female mice. These differential y expressed microRNAs may be lead to different sensitivity and tolerance to the ischemia-reperfusion injury in the kidney of male and female mice.
10.Apoptosis-inducing effects of AgLA2 on SPC-A-1 cells and its mechanism in vitro

Aim To investigate the apoptosis-inducing effects of AgLA2 on lung carcinoma cells SPC-A-1 and its mechanism in vitro.Methods The MTT assay was used to assess the proliferation of SPC-A-1 cells treated with AgLA2 in vitro.Apoptosis-inducing effects was investigated by DNA agarose gel electrophoresis,cell morphology and Elisa.RT-PCR was used to measure the expression of bcl-2 and bax mRNA,and immunocytochemistry was used to measure the expression of bcl-2 and bax protein.Results The IC50 of AgLA2 to SPC-A-1 cells was(3.447?0.436)mg?L-1.Treated with AgLA2,typical nuclear chromatine condensation and fragmentation were observed.The concentration of Caspase-3 in the group treated with AgLA2 was higher than that of the control group.Treated with AgLA2,bcl-2 mRNA,protein expression decreased while bax mRNA,protein expression increased.Conclusions AgLA2 can inhibit proliferation and induce apoptosis of SPC-A-1 cells.Its mechanism of action may be related to changing the ratio of bax/bcl-2 and the set-point of apoptosis,making the apoptosis power hold dominance.
