1.Research progress on differentiation of bone marrow stem cell into cardiomyocytes
International Journal of Biomedical Engineering 2006;0(06):-
Bone marrow stem cells(BMSCs) can differentiate into cardiomyocytes when induced with proper reagent. The cardiomyocytes from BMSCs have cardiomyocyte phenotypes with the expression of cardiac-specific antigen and receptor. BMSCs differentiate into cardiomyocytes in host myocardial microenvironments. Mechanism of the differentiation is unclear and need to be studied further.

Objective To investigate the feasibility of pig lung cancer model induced by bronchial perfusion of 3,4-benzopyrene. Methods 24 experimental pig were randomly divided into model group and control group, each containing 12 cases. Experimental pigs were under the anaesthetic state, pigs in the model group were given endobronchial infusion of 3,4 - benzopyrene - corn oil mixture, pigs in control group were injected with equal capacity of corn oil.Perfusion 1 times a week for 16 weeks. In week 16,32 and 48,all experimental pig were given lung CT scans, then the lung lesions were observed. After 48 weeks, the pig lung, esophagus, and gastrointestinal tract, liver and brain and other organs were dissected, the presence of tumor formation was observed, and the mass, and experimental pig lung biopsy were given hematoxylin and eosin staining analysis. Results Lung cancers were not found in control group by both CT lung cancers and anatomy. In the model group, pulmonary CT showed space-occupying lesions with different location and size in lungs of 8 pigs, and the space-occupying lesions were confirmed as malignant tumors by pathology, including 3 cases of moderately differentiated adenocarcinoma, 2 cases of highly differentiated squamous cell carcinoma, 1 case of alveolar cell carcinoma,2 cases of adenosquamous carcinoma. Three other pigs and pigs in the control group were not found with tumor by both lung CT and anatomy. Pigs in model group were induced successfully to malignant tumor in 1 year , the total tumor formation rate was 75%,lung tumor formation rate was 66.66%. Conclusion The trachea bronchial perfusion of 3,4- benzopyrene is a simple, safe and reliable way to construct animal models of lung cancer.

Objective To construct the plasmid that can produce long,double-stranded RNA and identify nonspecific apoptosis induced by long, double-stranded RNA in human osteosarcoma cells. Methods Plasmid pCI-neo-dsRNA was constructed by two promoters driven DNA sequence based on nontoxic, completely exoteric gene EGFP from the head and the end simultaneously to synthesize sense and antisense RNAs. Human osteosarcoma cells MG63 were transfected with pCI-neo-dsRNA and 48 h later the transfected cells were assessed by Apoptosis Detection System, FCMand MTT method. Results Plasmid pCI-neo-dsRNA was constructed successfully, MTT measurement showed that the absorbance of cells decreased gradually with the increase of plasmid pCI-neo-dsRNA dose(0.05 ?g, 0.10 ?g, 0.15 ?g, 0.20 ?g,0.25 ?g,0.30 ?g). MG3 transfected with pCI-neo-dsEGFP presented a great amount of irregular green fluorescence that indicated cell apoptosis. As the quantity of dsRNA increased, the percent of apoptosis cells also raised. While the dose of pCI-neo-dsEGF was 3.0 ?g, the apoptotic results of pCI-neo-dsEGF in MG3 were similar to that of 5-FU. Conclusion Long, double- stranded RNA could trigger human osteosarcoma cell apoptosis and the further research on using long, double-stranded RNA in osteosarcoma gene therapy can be done based on this theory.

[Objective]To observe the bone repairing efficacy of large segmental femur defects in rabbits with calcium phosphate cement(CPC)combined with bone morphogenetic protein(BMP)and massive bone allograft, which may benefit the clinical application of large segmental bone allograft transplantation.[Method]Fifty-four New Zealand white rabbits were divided into three groups and a 2 cm femur defect was created on one side of each rabbit, followed by implantation with:CPC combined with BMP and massive bone allograft(Group A),bone allograft only(Group B)and autograft transplantation(Group C).Intramedullary nails with a 3 mm diameter fixed all the grafts transplanted. The bone defect repair efficacy was evaluated by radiology and histology exam at 4,8 and 12 weeks after operation.[Result]The bone reparation capacities of allograft with CPC/BMP complex was better than that of the allograft alone after 4 ~8 weeks of transplantation, which were similar to the result of autograft transplantation.Complete bone union was achieved for all the groups after 12 weeks of operation, with better bone remodeling for group A and group C transplantation. The healing process of CPC/BMP combined with allograft transplantation was featured with large amount of bone callus forming surrounding the graft-host bone union area and the surface of allograft, which composd the extra cortical bone bridge and ingrowth(EBBI).Bone invasion, resorption as well as new bone genesis were seen in the surface of bone allograft at early stage, companied by expansion of Haversians canal, with more lanner cells, osteoblasts, osteoclasts and blood cells inside the allograft.CPC was slowly biodegraded with the bone graft resorption and new bone regeneration.[Conclusion]CPC combined with BMP can improve the bone reparation and substitution process in massive bone allograft transplantation for the treatment of large segmental bone defects.
