1. Tumorigenesis of bone marrow stromal cells transplanted into the brain of Sprague-Dawley rats: Report of three cases

Objective: To investigate the possible oncogenicity of bone marrow stromal stem cells (BMSCs) transplanted into brain and its underlying mechanism. Methods: The contralateral hemiplegia became gradually worse in three Sprague-Dawley rats among twenty rats after intracerebral transplantation with BMSCs. The intracerebral space occupying lesions were examined by MRI, and the life span of these rats was observed and recorded. The tumor tissues were obtained after the three rats were sacrificed. The telomerase activity was detected by telemeric repeat amplification protocol (TRAP)- PCR-enzyme-linked immunosorbent assay (ELISA). The expressions of proliferating cell nuclear antigen (PCNA), nestin and glial fibrillary acidic protein (GFAP) were detected by immunohistochemistry. The relative expression levels of epidermal growth factor receptor(EGFR), vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 (MMP-9) proteins in intracerebral tumor tissues and normal tissues were examined by Western blotting. Results: Of twenty rats, three rats displayed left hemiplegia which was continuously getting worse, and the life spans of the three rats were 129, 174 and 187 d, respectively. The result of TRAP-PCR-ELISA showed that the telomerase activity of intracerebral tumor tissues was higher than that of intracerebral normal tissues. The expressions of PCNA and nestin proteins in intracerebral tumor tissues were both higher but the expression of GFAP was lower than those of intracerebral normal tissues. The results of Western blotting showed that among three rats with intracerebral normal tissues, the expression level of EGFR protein was increased in one rat while the expression level of VEGF protein was increased in two rats. There was no significant difference in the expression of MMP-9 protein among the three rats. Conclusion: The telomerase activity and the ability of cell proliferation of BMSCs can be enhanced and the activation of oncogenes can be observed after long-term cell culture in vitro, and these findings suggest that the tumorigenesis potential can be induced after intracerebral transplantation with MSCs. Results This system can realize the real-time monitor of the medical equipment and improve the management of hospital. Conclusion The system resolves the problem of charge evasion effectually and increases the income of the hospital.
3.Recent development of informatics nurses displayed in investigation by US HIMSS

The status quo of American informatics nurses investigated 4 times by US HIMSS from 2004 to 2014 was analyzed.The following characteristics of American informatics nurses were summarized in terms of their educa-tion level, income, qualification authentication, occupation satisfaction, work experience and duties, including high education level and interdisciplinary background, high income and occupation satisfaction, necessary clinical background, and systematic professional development, which may provide reference for nursing staff in China to have a better understanding of informatics nurse.
5.Chitosan/PVA nerve conduits repair sciatic nerve defect in rats

ObjectiveTo investigate the effects of chitosan/PVA nerve conduits which used for repairing sciatics nerve defect in rats.MethodsTwenty-seven rats were divided into 3 groups randomly,with 9 rats in each group. Firstly, the 15mm defects in the left sciatic nerves were made in the rats and were respectively repaired with chitosan/PVA conduits graft (group A), the silicon conduits graft (group B),and autografts (group C). At 12 weeks after the operations, the left sciatic nerves were taken out, and the comparative evaluation was made on the repairing effects by wet weight of gastrocnemius and soleus muscles, histological examination,computerized imaging analysis and True Blue retrograde tracing. ResultsThe wet weight of gastrocnemius and soleus muscles showed no significant difference between the chitosan/PVA graft and autograft groups (P > 0.05). The wet weight of gastrocnemius and soleus muscles in significant difference between the chitosan/PVA graft and the silicon group at 12 weeks after the operation(P < 0.05). The nerve fiber density showed no statistically significant differences between the chitosan/PVA and autograft groups(P> 0.05).The regenerative nerve fiber in group B had normal morphological and structural characters under transmission electron microscope.True Blue-labeled neuron cell bodies were found within both anterior horn of gray matter in the spinal cord and dorsal root ganglions (DRGs) ipsilateral to the operated side of the tested rats on illumination with ultra-violet light 1 week after the injection of True Blue.Conclusion Chitosan/PVA nerve conduit can effectively promote the nerve regeneration and myelinization of rat sciatic nerve, which is expected to substitute for autograft to repair nerve defects succesfully.
6.The brain edema and Mip1 expression of perihematoma after intracerebral hemorrhage in rat model

Objective To explore the protective effect of Mip1 on cerebral edema by observing the Mip1 expression of perihematoma in intracerebral hemorrhage (ICH) rat model. Methods Ninety male SD rats were randomly divided into sham-operated group (n=15) and ICH group (n=75). ICH group was equally subdivided into 6 h, 12 h, 1 d, 3 d, and 7 d groups according to cerebral hemorrhage time, 15 in each group. Hematoma was formed by infusing autologous femoral artery blood into the right caudate nucleus. Rats were killed at the above time points, brain tissues around hematoma were taked. The pathological morphological changes in brain tissue around hematoma were observed using HE staining. The water content of brain (BWC) was measured with dry/wet weight method, and immunohistochemistry was used to analyze the average optical density value of Mip1 protein in perihematoma. Results Neuronal cell body of perihematoma was significantly decreased after ICH, the cytoplasm light staining, Nissl body reduced obviously in cytoplasm, and edema fissure appeared around cells, however, no obvious changes of nerve cells were detected in sham-operated group. Brain edema gradually increased at 6 h, and reached peak at 3 d (all P<0.05), then up to normal at 7 d after intracerebral hemorrhage (P>0.05). A large amount of brown Mip1 positive cells were found to distribute in cytoplasm and nucleus of perihematoma after intracerebral hemorrhage. The Mip1 protein expression level was higher in 6 h after intracerebral hemorrhage than that in sham-operated group (all P<0.05), and returned to normal level at 7 d (P>0.05). Conclusion The high expression of Mip1 after ICH is in line with the trend of the brain edema development, and shows that the activation of Mip1 may participate in the pathological process of hemorrhagic cerebral edema.
7.Expression of NogoA in 21a area of visual cortex in visual developing normal cat and strabismic amblyopia cat

BackgroundThe study of the molecular mechanism of visual plasticity is helpful for the explaining and prevention of strabismus and amblyopia.The effect and significance of NogoA in the strabismus and amblyopia formation are attracting more attention.ObjectiveThe present study was to investigate the expression of NogoA in 21a area of visual cortex in strabismic-induced amblyopia cats and explore the possible molecular mechanism of strabismic-induced amblyopia.MethodsSixteen 4-week old clean cats were randomizedly divided into normal group and strabismic-induced amblyopia group and eight for each group.The strabismic-induced amblyopia models were created by cutting off the external rectus in 8 cats.Pattern visual evoked potentials ( P-VEP ) were recorded 1 week after operation and compared with normal cats,and depression of amplitude and prolongation of implied time of P100 wave were as the successful criterion of model.The 200 ml paraformaldehyde was infused via heart to fax the brain under the deep anesthesia and then the cats were sacrificed and the brain cortex sections were prepared.The morphology of 21a zone of cat visual cortex was examined by haematoxylin and eosin staining,and the expressions of NogoA in 21a area of visual cortex were detected by immunohistochemistry with a monoclonal antiNogoA antibody.The use of the animals complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission.ResultsThe implied time and amplitude ratio of VEP P100 wave were (98.10±7.07)ms and (0.83±0.14) in normal group,and those in strabismic-induced amblyopia group were (108.50±6.95 )ms and (0.35 ±0.09 ),showing significant differences between two groups (t=4.450,P=0.005 ; t =5.970,P =0.005 ).The numbers of neurons were similar in 21a area of visual cortex between the two groups,but the volume of the neurons was lessen in strabismic-induced amblyopia group.The positive cell densities for NogoA were ( 387.37±2.01 ) cells/mm2,( 354.58± 1.85 ) cells/mm2 and ( 289.68± 1.81 ) cells/mm2 in layer Ⅱ/Ⅲ,Ⅳ and V/V[ respectively in strabismic-induced amblyopia group,and those in normal group were ( 161.39± 1.98 ) cells/mm2,( 128.93 ± 1.26 ) cells/mm2 and ( 96.25 ± 1.49 ) cells/mm2,indicating a significant increase in model cats ( t =- 160.400,- 201.890,- 164.740,P =0.000 ).Conclusions NogoA may play an important role in the regulation of the sensitive developmental period of visual sense and its plasticity.
8.Study on mechanism of sleep disorders associated with Parkinson's disease

Sleep disorders are commonly occurred among patients with Parkinson's disease,such as difficulties in the initiation of sleep,fragmented sleep,sleep behavior disorder and excessive daytime sleepiness.The mechanism of sleep disorders associated with Parkinson's disease is not clear,which may be associated with the injury of brain stem,nuclei of median raphe,nuclei fasciculi solitarii,thalamencephalon and the changes of neurotransmitters as dopamine,hypocretin(orexin) and melatonin.This article gives an overview of the mechanism of sleep disorders associated with Parkinson's disease.
9.Combining Uniplate anterior cervical plate system with titanium cage to treat single level cervical spondylotic myelopathy

Objective To explore the value of combining Uniplate anterior cervical plate system with titanium cage to treat single level cervical spondylotic myelopathy. Method Observing the clinical outcome and X-ray results of 32 patients with cervical spondylotic myelopathy undergone anterior decompression, autograft fusion and internal fixation with Uniplate anterior cervical plate system and titanium cage. Results All patients were followed up for 3 to 12 months. According to bony fusion criteria,no implants breakage, dislocation or loosening was found in all cases in 3 months' postoperatively radiological review. According to Odom classification,excellent outcome was 27 patients, good was 5 patients, the excellent and good rate was 100%(32/32). Conclusions Combining Uniplate anterior cervical plate system with titanium cage to treat single level cervical spondylotic myelopathy has advantages of easy manipulation,safe, less complication and higher fusion rate. It is an ideal way for cervical anterior fixation.
10.Role of toll receptor-4 in the increase of blood brain barrier permeability caused by acute pancreatitis

Objective To investigate the relation between the expression of toll receptor-4 (TLR-4) and the increase of blood brain barrier (BBB) permeability in the rats with acute pancreatitis.Methods Male Sprague-Dawley rats were randomly divided into 8 groups:control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;acute necrotizing pancreatitis (ANP) 2 h,6 h,12 h,24 h,48 h groups with 8 rats in each group.The BBB permeability,pathological score of pancreas,TLR-4 expression were determined and the relationships between them were analyzed.Results The BBB permeability in control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;ANP 2 h,6 h,12 h,24 h,48 h groups were 1.55±0.29,1.64±0.17,1.69±0.24,1.89±0.12,2.66±0.32,2.91±0.29,2.89±0.69 and 1.84±0.07,respectively;the pathological scores of pancreas were 0,2.38±0.92,3.13±0.64,8.50±1.07,9.75±0.71,10.25±1.28,11.13±1.25 and 10.13±1.13,respectively;there was no significant difference between AEP groups and control group,while there was significant difference between AEP groups and ANP groups (P<0.05 orP <0.01).BBB permeability was correlated with pancreatic injury ( r = 0.626,P <0.01).There was no TLR-4 mRNA and protein expression in the control and AEP group,while they were significantly expressed in ANP groups,and the expression were positively related with BBB permeability ( r =0.208,P = 0.027 ).Conclusions BBB permeability was present in the course of ANP.Activation of TLR-4 signal pathway may be involved in the BBB permeability increase caused by ANP.
