1.Moxibustion Improved Transcutaneous Oxygen Tension and Exercise Capacity in Lower Limbs of Peripheral Arterial Disease.

OBJECTIVETo observe the effects of moxibustion and treadmill exercise on transcutaneous oxygen tension and exercise capacity in lower limbs of peripheral arterial disease (PAD).
METHODSTotally 58 mild-to-moderate PAD patients were assigned to the control group (18 cases), the moxibustion group (20 cases), and the treadmill exercise group (20 cases) by random digit table. Patients in the control group received conventional drug therapy for 12 weeks. Patients in the moxibustion group and the treadmill exercise group additionally received moxibustion [at Zusanli (ST36), Sanyinjiao (SP6), Yongquan (KI1)] and treadmill exercise respectively, once per day, 5 times per week for 12 weeks in total. Ankle-Brachial Index (ABI) , transcutaneous oxygen tension (TcPO₂), 6-min walking test (6MWT), and walking impairment questionnaire (WIQ) were assessed before and after treatment.
RESULTSCompared with the control group and the same group before treatment, there was no statistical difference in ABI in the moxibustion group and the treadmill exercise group (P > 0.05). But TcPO₂, 6MWT, and WIQ were obviously elevated (P < 0.01). Besides, 6MWT and WIQ assessment of the treadmill exercise group were better than that of the moxibustion group (P < 0.01) after intervention.
CONCLUSIONMoxibustion and treadmill exercise could improve the exercise capacity and TcPO₂of lower limbers in PAD patients.

3.Effect of hyperbaric oxygenation on expression of platelet membrane glycoprotein in rats of traumatic brain injury

Objective To investigate the effect and significance of hyperbaric oxygenation in down-regulation of platelet membrane glycoproteins CD31 and CD62p in rats of traumatic brain injury (TBI).Methods Fifty-six SD rats were randomly distributed into TBI group,hyperbaric oxygenation group,and sham group by the lottery method.Furthermore,TBI group and hyperbaric oxygenation group were subgrouped at 6,48,and 96 hours.There were 8 rats per group.The rat models of severe TBI were induced by lateral fluid percussion.Levels of CD31 and CD62p were measured in all groups by flow cytometry.Results At 6,48 and 96 hours,expressions of CD31 (30.8 ± 8.9,32.5 ± 9.2 and 29.0 ±5.0) and CD62p (34.5 ±9.1,33.9 ±7.5 and 30.4 ±6.4) in TBI group were significantly higher than those (18.9-± 5.5,19.5 ± 6.1) in sham group (P < 0.05).At 96 hours,expression of CD31 (22.7 ±5.5) in hyperbaric oxygenation group was significantly lower than 29.0 ± 5.0 in the TBI group (P <0.05).At 48 and 96 hours,expressions of CD62p (26.1 ± 5.8,23.6 ± 5.7) in hyperbaric oxygenation group were significantly lower than 33.9 ± 7.5 and 30.4 ± 6.4 in TBI group (P < 0.05).Conclusions Platelet activation is enhanced in the acute phase after TBI.But platelet activation may be relieved with hyperbaric oxygenation,which is conducive to inhibiting microthrombosis and mitigating secondary brain injury after TBI.
4.Preparation of new porous composite artificial bone and properties

[Objective]To prepare a new porous composite artificial bone substitute and evaluate its properties.[Method]Thermally induced phase separation was adopted to prepare the artificial bone made from oyster shell powder and PLLA which were proportionally mixed and its properties as porosity rate,pore size and mechanical strength were assessed.Meanwhile the related variation parameters were examined every 2 weeks in a course of 14 weeks after the slices of CAB and pure PLLA were immersed in NS of 37℃ in vitro,the results of which were compared in statistics.[Result]The average porosity rate of artifical bone with TIPS method was 85.1% and pore sizes ranged 100-300 ?m under the SEM,with better pore connectivity and regulation shape.The average compressive strength was 2.12 MPa.As the immersion prolonged,the regular variation was observed about the mass loss of CAB and the pH alteration of solution,there was statistically difference in the indexes between the two groups(P﹤0.05).[Conclusion]The porosity rate,pore size,mechanical strength and degradation performance in vitro of the artifical bone made by TIPS method can satisfy the requirement of bone substitute.
5.THE ESTABLISHMENT OF AN OSTEOSARCOMA CELL LINE PL-1 AND ITS BIOLOGIC CHARACTERS

To establish a xenografted tumor strain of human osteosarcoma in nude mice and a homologous cell line, the osteosarcoma speciments from patients were inoculated into the subcutaneous tissue of nude mice. After the transplanted tumor had grown to about 2cm in diameter, the tumor tissue was cultured in vitro and the continuously growing cells were analyzed by morphology, histochemistry, immunohistochemical straining, cell cycling analysis ,chromosome analysis and heterotransplantation. A newly established cell line designated PL 1 was thus obtained in this laboratory in continuous culture for over 100 passages. Under light and electron microscopes, the cells assumed the main characters of malignancy.Morphological observation, histochemistry analysis and BMP immunohisto chemical straining showed that they had the features of osteosarcoma.The cell cycle analysis showed 53 2% of the cells were in G 1. This cell line could produce ALP and BMP.Chromosome analysis confirmed that they had retained a karyotype of human cancer cells and a hypotriploid feature with a mode number of around 64~66.The tumor formation rate after heterotransplantation was 100%. It had lung metastasis characters. Therefore, PL 1 cell line derived from the xenograft in nude mice has been established and maintained for over 9 months trough 100 passages, and this cell line can provide material and model for further investigation of human osteosarcoma.
6.Cell hybridization of a murine osteosarcoma LM9 with activated B lymphocytes for the preparation of tumor vaccines

Objective To establish osteosarcoma vaccine by the LM9 osteosarcoma derived from C3h mice hybridized with activated B lymphocytes and study its biological behavior and the antitumor efficacy. Methods The LM9 osteosarcoma derived from C3h mice was fused with LPS activated B lymphocytes by using 50%PEG. The fused cells was selected by HAT medium and cultured in vitro, and the biocharacter and efficacy of the fusion vaccine were investigated. Results In contrast to LM8, the fused cells grew significantly slowly in vitro. All the mice under the protection of fused vaccine survived without tumor (8/8), while all the mice in the control group succumbed to the tumor with no survival (8/8), 75%of the mice inoculated subcutaneously with cell fusion vaccine survived without tumor burden after implantation of LM8 cells subcutaneously. The mice in the control group developed tumors and died within 45 days without any exception. Conclusion It is possible that the LM9 osteosarcoma biology characteristics change after fused with LPS activated B lymphocytes. Cell fusion osteosarcoma vaccine could produce prophylactic and therapeutic effects in mice.
7.Histocompoatibility of deantigenic bovine cancellous bone with rabbit osteoblasts

Objective: To prepare surface with collagen, so as to increase the histocompatibility of deantigenic bovine cancellous bone. Methods: Bovine cancellous bone was degreased and macerated by H 2O 2 and decalcified with 0.3 mol/L HCl to form scaffold, surface of which was treaed with collagen, then cultured rabbit osteoblasts were seeded into the scaffold and cultured in vitro . 4, 10, and 21 days later, the cells on the composite materials were observed under sanning election microscope. Results: On day 4, the cells grew in the surface of the scaffold; on day 21, the cells grow into the pores and on all of the surace of the scaffold.Conclusion: Deantigenic bovine cancellous bone with surfaces treated by collagen is histocompatable with rabbit osteoblasts.
10.The combinative biological reconstruction of bony defect following limb bone tumor resections

Objective To analyze the image and histological results of the combined use of allograft/extracorporeally frozen tumor-bearing bone and vascularized fibular flap for the reconstruction of bony defects following tumor resection,guiding clinical practice.Methods From March 2007 to June 2013,we enrolled 63 patients who had combinative biological reconstruction after bone tumor resection (11 in humerus,22 in femur,21 in tibia,4 in calcaneus).There were 36 male and 27 female in this series.The average age at time of operation was 20 years,ranging from 9 to 48 years.The follow-up ranged from 16 to 102 months with average of 48 months.We investigated the X-ray and CT images for all patients and histological findings of two patients.Patients were assessed functionally with the Musculoskeletal Tumor Society 93 score.Results Three patients with local soft tissue recurrence and one patient with infection underwent amputation.The survival of construct was 93.6%.Bone union achieved in all cases with the average MSTS score of 92.8%.Bone union ranged from 11 to 28 months in allograft group and 9 to 14 months in devitalized tumor bearing bone group.Significant difference of bone union time was found between two groups (Z=-3.638,P=0.000).Viability of the fibular grafts was verified in 58 of 63 patients (92%).Three types of images were observed in complex.Osteopenia and spongy change in fibula were found in 51 patients (81%) with stable fixation of the complex.Five complexes with failed blood supply of fibula and stable fixation revealed no density change of fibula,small amount of callous formation and relative delayed union.In seven complexes (11%) with unstable complex due to patients' incompliance,fibula reacted with dense hypertrophy and microfracture.Fusion of grafts with amount of callus was obviously observed.Union at allograft-host bone junctions occurred by residual host bone-derived external callus and fibular-derived internal callus that bridged the junction and filled the gap between abutting cortices.Callus from fibular graft was mature than that from periosteum of residual host bone.Internal repair was observed at the internal surface of the allografts.Fibula showed significant spongy changes.Conclusion Recycled tumor-bearing bone in combined with fibular flap is a reliable reconstruction as an alternative to traditional Capanna technique.The survival of the fibula is a cornerstone in success of complex reconstruction.Sponginess of fibula and internal repair of allograft compromise the intensity of complex,necessitating the strong instrumentation during reconstruction.
