1.Auditory Verbal Memory Test in Chinese Elderly

Objective: To develop Auditory Verbal Memory Test (AVMT) suitable for Chinese elderly. Method:100 normal elderly and 22 patients with mild to moderate Alz heimer' s disease were evaluated by AVMT, mini mental state examination. 40 person randomly selected from the 100 sample were assessed in follow up by AVMT, Wechsler Memory Scale ( Chinese Revised Version), information and similaritias subtests of Wechsler Adult Intelligence Scale - Chinese Revised Version (WAIS- RC) . Results: The correlation between inter- raters was 0.99, test- retest reliability at 3 months interval were 0.87~0.94. Education was a prominent influence factor of memory and recognition of AVMT. There were significant correlation between third recall, short delayed recall and total scores of VMS- RC, between long delayed recall and verbal sceres of WAIS - RC. Semantic clustering strategy showed gradual strongly correlation with measures of recall ability in higher education group. Performance of Alzheimer group was poorer than that of matched normal control. Scores of long delayed re call differed most obviously in group comparison. Conclusion: AVMT had good reliability and validity
2.Application of Rey-Osterrieth Complex Figure Test in Chinese Normal Old People

Objective: To evaluate the applicability, reliability and validity of the Rey-Osterrieth Complex Figure Test (CFT)in Chinese normal elderly. Methods: A total of 111 normal elderly were assessed by the CFT (Form A) and the mini-mental state examination (MMSE). Among these subjects, 40 were also administered the CFT (Form B), the Block Design and the Visual Reproduction tests. Results: No significant difference was found between CFT (Form A) and CFT (Form B) with respect to copy, delayed recall and recognition. Recall performance was related to copying strategy and level of organization. Details of accuracy and their respective placement were evaluated in both copy and delayed recall trials for CFT (Form A). Conclusion: As a measure for visual-spatial constructional ability and visual memory, the CFT (Form A & B) was assessed to be psychometrically reliable and valid to be used in normal Chinese elderly.
3.Research for Difference between Neurodegenerative and Vascular Mild Cognitive Impairment with Neuropsychological Assessment

Objective: To analyze neuropsychological test identification of neurodegenerative (NMCI) and vascular (VMCI) subtypes of mild cognitive impairment. Methods: 94 normal elderly, 107 NMCI and 32 VMCI were evaluated by neuropsychological tests, such as mini mental state examination(MMSE), auditory verbal memory test (AVMT), Rey-Osterrich complex figure test (CFT), trailmaking test (TMT), Stroop Color Words Test (CWT), Verbal fluency test (VFT) and Clock Drawing Test (CDT). MMSE total score of 3 groups were 28.1?1.6, 26.9?1.8 and 26.2?1.7 , respectively. Results: There are similar global severity between NMCI and VMCI groups. VMCI group performed better than NMCI group in verbal delay memory rather than verbal short memory, or figure memory. Stroop interference effect and TMT interference effect of VMCI group were more powerful significantly than that of NMCI. VMCI group also generated fewer figure copy score on CFT. But there wasn't difference on VFT and CDT. Conclusion: These results suggest that decline in executive function is associated with cerebral vascular factors, and decline in episodic memory (such as verbal delay memory) is associated with neurodegenerative factors.
4.A prospective,randomized controlled clinical study to evaluate porous β-tricalcium phosphate for lacunar bone defects repair

Objective To evaluate the clinical outcomes of a hovel porous β-tricalcium phosphate (β-TCP)and control allograft for the repair of lacunar bone defects caused by solitary bone cyst curettage.Methods From January 2003 to December 2008,the patients with solitary bone cyst were randomized into an experimental(55 cases)and a control(40 cases)group.The control group received particulate allograft bone as the graft material,and the experimental group received β-TCP.At 1 week,1,2,3,6,12,24,48months after surgery,a new radiographic scoring system was employed to calculate the biodegradation of bone graft and evaluate the influence of multiple factors.Histologic characteristic of the degradation process of β-TCP were also evaluated.Results All the cases were followed up for average 28.4 months.Radiographic semi-quantitative analysis revealed that the biodegradation effieiencies were not significantly difierent between β-TCP and allografts(P=0.424).Degradation percentage of the implanted β-TCP or allograft was higher in younger patients than those in the older ones.Degradation of β-TCP was significantly higher than that of allografts over 3 years after surgery(P=0.04).In the experimental group,β-TCP degradation was greater in the loose packing treatment than that in the dense packing treatment (P=0.03).Histological observation demonstrated that the process of new bone formation accompanied the degradation of β-TCP.Conclusion The interporous β-TCP could be an advantageous alternative to allografts for repair bone defects caused by bone cyst.The clinical application of β-TCP is safe and reliable,which shows better biodegradation and osteogenesis than allografts in long-term follow-up.
5.Effects of deep brain stimulation on expression of DARPP-32 and its phosphorylated proteins in corpus striatum of rats with dyskinesia

Objective To investigate the effects of subthalamic nucleus (STN)-deep brain stimulation (DBS) on expression of dopamine and adenosine 3'5'-monophosphate-regulated phosphor-protein(DARPP-32) and its phosphorylated proteins in corpus striatum of rat models with levodopa-induced dyskinesia. Methods The rat models of levodopa-induced dyskinesia were set up and were given STN-DBS (stimulation group). The expression of DARPP-32 and its phosphorylated proteins in corpus striatum (damage side and normal side) were detected and compared with sham-stimulation group and sham-operation group. Results There was no significant difference in the expression of DARPP-32 total protein in corpus striatum of rats with dyskinesia among three groups (P>0.05). The expression of Phosphor-Thr34-DARPP-32 protein in the damage side of corpus striatum in stimulation group was significantly lower than that in sham-stimulation group and sham-operation group (P<0.05), while the expression of Phosphor-Thr75-DARPP-32 protein in the damage side of corpus striatum in stimulation group was significantly higher than that in sham-stimulation group and sham-operation group (P<0.05). Conclusion DARPP-32 and its phosphorylated proteins play an important role in the pathogenesis of levodopa-induced dyskinesia.
6.Research of Auditory Verbal Memory Impairment Following Stroke of Different Subcerebral Structures

Objective: To analyze auditory verbal memory impairment after isolated stroke lesions in subcerebral structure. Methods: Auditory verbal memory function was examined in 25 normal elderly and 83 patients at 20th day after their admission to hospital for acute cerebrovascular disease with non-aphasiaand solitary lesions as identified by cranial CT or MRI. Results: Test findings suggested patients with stroke of internal capsule-basal ganglia, cerebellum and pons lesion have processing impairments of recent memory and subjective organization which are related to concentration and arousal levels. Conclusion: Stroke of subcortical regions such as lesions of thalamus,internal capsule-basal ganglia,cerebellum and pons may induce memory impairments with unique impaired cognitive coding modalities.
7.Ability of Chinese Characters Reading in Patients with Alzheimer's Disease

Objective: To analyze the ability of Chinese characters reading and characteristics of alexia among patients with mild, moderate and severe Alzheimer's Disease(AD). Methods: Chinese characters reading test was performed in 20 normal controls(average MMSE total score is 27.7), 20 mild AD(average MMSE is 21.2),20 moderate AD(average MMSE is 15.2) and 20 severe AD(average MMSE is 6.9). Chinese characters reading test consists of 22 mark-symbol characters, 17 phonetic symbol characters and 17 meaning-symbol characters. Results: No significant difference were found in scores of Chinese characters reading test among normal controls, patients with mild, moderate AD group. Compared with moderate AD, severe AD showed manifest decline for scores of phonetic symbol characters and meaning-symbol characters reading. There wasn't significant different visual paralexia among 4 groups. Surface alexia occurred at early stage of AD and advanced at late stage of AD. These wrong reading were seen frequently in meaning-symbol characters. Deep alexia occurred only at late stage of AD. Compound words phanomenon is the major class of deep alexia. Conclusion: Ability of Chinese characters reading was helpful to estimate premorbid intelligence of the patients with dementia. There are different classes of alexia between Chinese and western languages.
8.The expression and clinical significance of tumor suppressor phosphatase -tension protein gene in differentia-ted thyroid carcinoma

Objective To analyze the expression and clinical significance of tumor suppressor phosphatase -tension protein gene(PTEN)in differentiated thyroid carcinoma and the relationship with the V600E gene site muta-tion of murine sarcoma viral oncogene homolog B1(BRAF).Methods The expression of PTEN was assayed by im-munohistochemical staining in differentiated thyroid carcinoma and adjacent tissues.The differences were compared between clinical pathological features.The BRAF V600E gene mutation was detected by fluorescence quantitative pol-ymerase chain reaction(PCR)and to investigate the correlation between the expression of PTEN and BRAF V600E mutation.Results The negative rate of PTEN was 78.75%(63 /80)in differentiated thyroid carcinoma,13.75%(11 /80)in adjacent tissues(χ2 =27.236,P =0.000).There were significant differences in the expression of PTEN between I -II and III -IV in TNMstage,grade I and II,and lymph node metastasis(χ2 =10.395,6.948,9.263,P =0.000,0.006,0.000).The differences were not significant in the expression of PTEN between different gender,≤45 years and >45 years different pathological types and tumor diameter <2cm and ≥2cm(χ2 =1.113,0.941, 2.301,1.567,P =0.185,0.213,0.087,0.181).There was correlation between the expression of PTEN and BRAF V600E mutations(r =0.301,P =0.004).Conclusion The expression of PTEN is involved in the occurrence and development of differentiated thyroid cancer and is associated with BRAF V600E.The PTEN has the predictive value of prognosis in tumors.
9.Experimental study on prevention of Parkinson disease by neutral amino acid

Objective To investigate the effect of neutral amino acid on preventing Parkinson disease.Methods Mice were injected with L-Valine,L-Pheylalanine,D-Valine or L-Lysine before or after paraquat administration,by which prakinsonian mouse model was constructed.The paraquat immunoreactivity was observed within nigral cell bodies.Then neurodegeneration and ?-synuclein aggregation were observed by immunohistochemistry and Western blot.Results Paraquat immunoreactivity was abolished by the administration of L-Valine,L-Pheylalanine before paraquat exposure.Pre-treatment with these two amino acids also protected the paraquat-induced loss of nigrostriatal dopaminergic cells and formation of thioflavine S-positive aggregates.In contrast, paraquat-induced toxicity was unaffected if animals were injected with these two amino acids after paraquat exposure or pre-treated with D-Valine or L-Lysine.Conclusions L-type neutral amino acids such as L Valine and L-Pheylalanine can prevent paraquat-induced neurodegeneration and a synuclein pathology through a competitive inhibition mechanism with stereospecificity in the central nervous system (CNS).Neutral amino acid could protect the dopaminergic neuron in substantia nigra and may prevent Parkinson disease.
