2.Biocompatibility of polycaprolactone with bone marrow stromal cells

BACKGROUND: Bone defect repair is a difficulty in orthopedic field all the time, researches on the tissue engineered bone tissue have provided completely new thoughts and methods for bone defect repair, and it is an important link to detect the biocompatibility of the biomaterials.OBJECTIVE: To investigate the biocompatibility of PCL with bone marrow stromal cells (BMSCs).DESIGN: A controlled observation.SETTING: Department of Orthopaedics, the First Affiliated Hospital of Xinjiang Medical University.MATERIALS: The experiments were carried out in the orthopedic laboratory of the First Affiliated Hospital of Xinjiang Medical University. Healthy 4 to 8-week-old New Zealand rabbits of about 2 kg were used.METHODS: ① Bone marrow was extracted from bilateral femurs of the rabbits, then mixed into 10 mL RPMI1640 complete medium, and then entered the passage culture. ② The cells were inoculated and then divided into PCL group and control group, the cells were only inoculated in the control group, and the BMSCs were co-cultured with PCL in vitro in the PCL group. The morphological observed, cell proliferation, protein content and enzymological determinations were conducted.MAIN OUTCOME MEASURES: The growth and the adhesion of BMSCs on PCL biomaterials were mainly observed.RESULTS: In the control group, most of the BMSCs changed to the shape of fusiform or multi-angles. In the PCL group, the BMSCs could adhere and proliferate on PCL, and the growth and function were not affected, PCL also played a certain role in promoting the cell proliferation.CONCLUSION: PCL possess satisfactory biocompatibility, and it is possible to be used as the carrier of BMSCs in tissue engineering.

4.Application of continuous monitoring of jugular venous oxygen saturation in the course of mild hypothermia treatment of severe traumatic brain injury

Objective To discuss significance of continuous monitoring of jugular venous oxygen saturation(S_(jv)O_2)in the course of mild hypothermia treatment(MHT)for severe traumatic brain injury (sTBI).Methods Intracranial pressure(ICP),S_(jv)O_2 and brain tissue pressure(P_(bt)O_2)were contin- uously monitored in 36 cases with sTBI for analyzing the correlation between S_(jv)O_2 and P_(bt)O_2.Results (1)There was negative linear correlation between P_(bt)O_2 and ICP(r=-0.978,P<0.05),negative lin- ear correlation between S_(jv)O_2 and ICP(r=-0.947,P<0.05)and positive linear correlation between P_(bt)O_2 and S_(jv)O_2(r=0.965,P<0.05)within 24 hours and at 36 hours and 48 hours after injury.(2) The cases with decreased S_(jv)O_2 value had a worse outcome than those with normal S_(jv)O_2.meanwhile,the cases with abnormal increase of S_(jv)O_2 value had worse prognosis.Prognnsis was improved significantly with increase of S_(jv)O_2 in certain range(P<0.05).Conclusion Continuous monitoring of S_(jv)O_2 can reflect the condition of hemicerebral oxygen metabolism and guide treatment and predicting outcome.
5.Effects of shikonin on stemness maintance of glioma stem cells

Aim To explore the effect of shikonin on stemness maintance of glioma stem cells ( GSCs ). Methods After the U87-MG cells were cultured and isolated, the sphere cells were identified by immuno-fluorescent staining. The alteration of stemness of GSCs by shikonin treatment(2 μmol·L - 1 ) for 12 h, 24 h and 48 h was valued by morphological detection using optical microscope and sub-sphere forming assay. Mo-reover, the related markers of stem cells, such as CD133, were detected in shikonin treated GSCs by western blot assay. Protein expression of PI3K, p-PI3K, Akt and p-Akt was detected by western blot af-ter shikonin treatment alone. Furthermore, by combi-nation with insulin-like growth factor-1 ( IGF-1), we observed the alteration of stemness maintance of shiko-nin-treated GSCs. Results The presence of neural stem cell related markers CD133 and nestin proved the characteristics of GSCs. Shikonin treatment significant-ly inhibited the morphology of GSCs and the sub-sphere forming. Besides, the reduced expression of CD133 was detected in shikonin treated GSCs. Though, the expression of PI3K and Akt did not change compared with the control group, the expression of p-PI3K and p-Akt was reduced. Furthermore, the combination of IGF-1 markedly attenuated the inhibitory effect of shikonin on stemness maintance of GSCs. Conclusion The stemness maintance of GSCs can be significantly inhibited by shikonin treatment, in which PI3K/ Akt pathway is involved.
6.Genetic characteristics of hemagglutinin in measles viruses isolated in Henan Province, China.

This study aims to investigate the genetic characteristics of hemagglutinin in wild-type measles viruses in Henan Province, China and to provide a basis for measles control and elimination. Specimens were collected from suspected measles cases in Henan during 2008-2012. Cell culture was performed for virus isolation, and RT-PCR was used to amplify hemagglutinin gene. The PCR products were sequenced and analyzed, including construction of phylogenetic tree and analysis of the distance between the isolated virus and the reference virus; then, the variations in predicted amino acids were analyzed. The results showed that 12 measles viruses were isolated in Henan Province and identified as H1a genotype; the nucleotide and amino acid homologies were 98.0%-100% and 97.2%-99.8%, respectively. One glycosylation site changed in all the 12 sequences because of the amino acid mutation from serine to asparagine at the 240th site, as compared with Edmonston-wt. USA/54/A. Overall, the wild-type measles virus genotype circulating in Henan Province from 2008 to 2012 was H1a, with high homology between strains; there were some variations in amino acid sequences, resulting in glycosylation site deletion.

10.The impact of repeated app1ication of contrast media on rena1 function within a short period of time ;in different occasions

Objective To discuss the impact of repeated contrast media exposure on renal function in patients who received coronary angiography ( CAG) or percutaneous coronary intervention ( PCI) within 1 week after CTA of coronary ateries. Methods A total of 258 patients who received CAG or PCI after coronary CTA were divided into the study group ( n=132, patients had CAG/PCI within 1 week after CTA) and the control group ( n=126, patients had CAG/PCI 1-2 weeks after CTA). Serum creatinine, cystatin C and estimated GFR were tested before and on day 1, 2 and 3 after procedures. The occurance of contrast-induced nephropathy ( CIN ) was recorded. Resu1ts The baseline clinical characteristics of the patients between the two groups had no significant difference. Preoperative and postoperative serum creatinine, cystatin C and eGFR values on day 1, 2 and 3 had no significant difference between the two groups (all P﹥0. 05). There was no significant difference in the incidence of CIN between two groups (5. 3% in the study group vs. 4. 8% in the control group, P﹥0. 05 ) . Conc1usions It is safe and feasible for patients with eGFR≥60 ml/( min?1. 73 m2 ) to undergo CAG or PCI within 1 week after coronary CTA.
