1.The effect and influence factor analysis ofⅠandⅡ stage DLBCL patients who receive radiotherapy after chemotherapy

Objective:To study the effect and influence factor analysis of DLBCL patient with early stage who receive radiotherapy after chemotherapy.Methods: 374 cases of patients with DLBCL was selected from January 2010 to December 2015 in our hospital. By random number table method, the patients were divided into CHOP group (n=104), R-CHOP+RT group (n=93), R-CHOP group (n=80), CHOP+RT group (n=97). CHOP chemotherapy was given to all patients, 180 patients received radiotherapy after chemotherapy, and 169 patients received rituximab. Survival rates were compared between the 4 groups.Results: The survival rate of R-CHOP group in 12 months, 24 months, 50 months and 100 months were lower than R-CHOP+RT group, but the difference was not statistically significant in twelfth months(x2=2.02,P>0.05). The differences of 24 months, 50 months and 100 months were statistically significant (x2=4.08,x2=4.03,x2=8.79;P<0.05); The survival rate of CHOP group was 12 months and 24 months was higher than CHOP+RT group which the difference was not statistically significant (x2=1.05,x2=0.22;P>0.05); The survival rate of CHOP group in 50 months and 100th months was lower than CHOP+RT, but the difference was no significant difference (x2=1.62,x2=0.03;P>0.05). Smoking index, whether the use of rituximab, the age associated with the survival of patients, the difference was statistically significant.Conclusion: Early DLBCL patients with R-CHOP and radiotherapy combined treatment can be effective in patients with survival, while the use of rituximab chemotherapy, in addition to smoking on the prognosis of patients with serious adverse effects.

5.Self-assembly tissue engineering fibrocartilage model of goat temporomandibular joint disc.

OBJECTIVETo construct self-assembly fibrocartilage model of goat temporomandibular joint disc and observe the biological characteristics of the self-assembled fibrocartilage constructs, further to provide a basis for tissue engineering of the temporomandibular joint disc and other fibrocartilage.
METHODSCells from temporomandibular joint discs of goats were harvested and cultured. 5.5 x 10(6) cells were seeded in each agarose well with diameter 5 mm x depth 10 mm, daily replace of medium, cultured for 2 weeks.
RESULTSOne day after seeding, goat temporomandibular joint disc cells in agarose wells were gathered and began to self-assemble into a disc-shaped base, then gradually turned into a round shape. When cultured for 2 weeks, hematoxylin-eosin staining was conducted and observed that cells were round and wrapped around by the matrix. Positive Safranin-O/fast green staining for glycosaminoglycans was observed throughout the entire constructs, and picro-sirius red staining was examined and distribution of numerous type I collagen was found. Immunohistochemistry staining demonstrated brown yellow particles in cytoplasm and around extracellular matrix, which showed self-assembly construct can produce type I collagen as native temporomandibular joint disc tissue.
CONCLUSIONProduction of extracellular matrix in self-assembly construct as native temporomandibular joint disc tissue indicates that the use of agarose wells to construct engineered temporomandibular joint disc will be possible and practicable.

6.Effects of Shenfu Injection on inflammatory cytokines during cardiopulmonarbypass in infants.

OBJECTIVETo evaluate the effects of Shenfu Injection (SF) on cytokines during cardiopulmonary bypass (CPB) in infants.
METHODSTwenty-four infants with congenital heart disease, aged below three years, were randomly assigned to the control group and the SF group equally. In the SF group, 1 mL/kg of SF was given through center vein pump after center vein puncture being performed, while only normal saline was given instead in the control group. Blood sample was obtained for measurement of serum necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) concentration by ELISA, at various time points in the process, i.e. right after anesthesia induction (T1), beginning of CPB (T2), aortic off-clamping (T3), 20 min after CPB (T4), the end of CPB (T5), and 6 h (T6) and 24 h (T7) after CPB.
RESULTSThe time for reverting to sinus heart rhythm and analepsia after CPB was shorter in the SF group than in the control group (P < 0.05). Serum concentration of TNF-a and IL-6 was equal in the two groups at T1; they increased significantly after CPB (P < 0.05), reached the peak value at T4 and reduced to the normal level at T7 in the control group. TNF-alpha concentration was significantly lower at T3, T4 and T5, and IL-6 concentration was lower at T4 in the SF group than that in the control group at corresponding time point (P < 0.05).
CONCLUSIONSF could shorten the time for reverting to sinus heart rhythm and analepsia after CPB in infants, and suppress the inflammatory response caused by CPB.

7.Effects of shikonin on stemness maintance of glioma stem cells

Aim To explore the effect of shikonin on stemness maintance of glioma stem cells ( GSCs ). Methods After the U87-MG cells were cultured and isolated, the sphere cells were identified by immuno-fluorescent staining. The alteration of stemness of GSCs by shikonin treatment(2 μmol·L - 1 ) for 12 h, 24 h and 48 h was valued by morphological detection using optical microscope and sub-sphere forming assay. Mo-reover, the related markers of stem cells, such as CD133, were detected in shikonin treated GSCs by western blot assay. Protein expression of PI3K, p-PI3K, Akt and p-Akt was detected by western blot af-ter shikonin treatment alone. Furthermore, by combi-nation with insulin-like growth factor-1 ( IGF-1), we observed the alteration of stemness maintance of shiko-nin-treated GSCs. Results The presence of neural stem cell related markers CD133 and nestin proved the characteristics of GSCs. Shikonin treatment significant-ly inhibited the morphology of GSCs and the sub-sphere forming. Besides, the reduced expression of CD133 was detected in shikonin treated GSCs. Though, the expression of PI3K and Akt did not change compared with the control group, the expression of p-PI3K and p-Akt was reduced. Furthermore, the combination of IGF-1 markedly attenuated the inhibitory effect of shikonin on stemness maintance of GSCs. Conclusion The stemness maintance of GSCs can be significantly inhibited by shikonin treatment, in which PI3K/ Akt pathway is involved.
8.Effects of optic disc topography and retinal nerve fiber layer thickness measurement by spectral-domain OCT on diagnosis of glaucoma

BackgroundGlaucoma is an optic neuropathy caused by structural damage of the optic nerve,and its early diagnosis is critical for arresting the irreversible damage of visual function. Optical coherence tomography (OCT) allows an early diagnosis of glaucoma by the measurements of the optic disc and retinal nerve fiber parameters. Objective This study was carried out to evaluate the effects of optic disc tomography and the measurement of the retinal nerve fiber layer (RNFL)thickness by spectral-domain OCT on the diagnosis of glaucomatous eye. MethodsIt was a noninterventional, cross-sectionalstudy. The optic disctopographic parameters and total and regional RNFL thickness were measured by RTVue OCT in 62 normal eyes and 67 glaucomatous eyes. The area under the receiver operating characteristic curve( ROC ) was used to assess the ability to differentiate glaucoma eyes from normal eyes of each testing parameter. This trial complied with the Helsinki Declaration and was approved by the Clinical Trial Ethic Committee of Beijing TongrenHospital. All of the participants signed the written informed consent before any medical examination. Results In the comparison of demography ,the ages of patients, the mean deficiency( MD ) and pattern standard difference( PSD ) of perimetry were obviously larger in the glaucoma group, primary open angle glaucoma ( POAG ) group and primary closure-angle glaucoma(PACG) group than those of normal controls( P<0. 01 ). No significant differences were found in the disc area between a total glaucoma group, POAG group or PACG group and normal group ( P =0. 101,0. 741 and 0. 652, respectively) ;however, the average RNFL thickness between normal eyes and glaucomatous eyes were significantly different( 109. 758 μm versus 79. 539 μm, P<0. 01 ). Among the eight regions around the optic disc, the thickest RNFL located at the inferotemporal( 150. 109 μm) and superotemporal( 146. 105 μm) regions in normal eyes,and at the superotemporal( 104. 354 μm) and inferotemporal( 102. 436 μm) regions in glaucomatous eyes. Both in normal and glaucomatous eyes,the thinnest RNFL located at the nasal(NU+NL) and temporal(TU + TL) regions. For optic disc topographic parameters,the highest ROC were observed in rim volume( ROC--0. 850,0. 841 and 0. 862 in total glaucoma,POAG and PACG, respectively) and vertical cup/disc ratio( ROC =0. 840,0. 849 and 0. 830 in total glaucoma,POAG and PACG,respectively), and the sensitivities for specificity cutoff set at 80% were 73.1% and 76. 1% in total glaucoma,73.0% and 81.1% in POAG and 73.3% and 70.0% in PACG, respectively. For RNFL thickness ,the highest ROC was observed in average RNFL( ROC =0. 925,0. 910 and 0. 942 in total glaucoma, POAG and PACG,respectively) ,and the sensitivities for specificity cutoff set at 80% were 89. 6% ,89.2% and 90. 0% in total glaucoma,POAG and PACG, respectively. Among the eight regions around the optic disc, RNFL thickness of region IT achieved the highest ROC, RNFL thickness of region TU and TL had the lowest ROC. Conclusions RTVue OCT appears to be of fair discriminating ability in distinguishing normal from glaucomatous eyes. RTVue OCT shows promise for the diagnosis of glaucoma.
9.Effects of guanxin II on cardiac protein expression after acute myocardial ischemia on rats.

OBJECTIVEGuanxin II is a famous modern formula of traditional Chinese medicine. Guanxin II after myocardial infarction (MI) has been shown to have beneficial effects on cardiac anatomy and function. The purpose of this study was to examine the effects of Guanxin 1I on cardiac protein expression after MI.
METHODRats were randomized into 3 groups, sham, model and treating groups. Model and treating groups were fed with Guanxin II and sham group was fed with water for 10 days before MI. MI operation is to ligate left coronary artery. 24 hours after MI, myocardial protein expression of junctional zone was assessed with 2D gel electrophoresis and mass spectra analysis.
RESULTGuanxin II was found to be able to improve myocardial protein expression, especially 11 proteins. These proteins are mainly involved in suppressing changes of cell shape and structure and energy metabolism.
CONCLUSIONGuanxin II after MI affected myocardial protein expression. Further experiments of larger research extent should be done to receive more results.

Mongolian folk medicine resource is the origin of Mongolian medicine development, even more important of which is the specialized Mongolian folk medicine resources with regional and high medicine quality, it processes distinctive national characteristics with irreplaceable important position in traditional Mongolian medicine. Nevertheless, due to the serious destroy of ecological environment and sharp increase of demands, etc. A lot of specialized Mongolian folk medicine resources were endangered, and there still existed some problems in the protection and exploitation and utilization. This paper intends to provide comprehensive insight into the species protection and exploitation and utilization states of specialized Mongolian folk medicine resources. The application and protection status and the existing problems were reviewed, and the development strategies of Mongolian folk medicine resource were analyzed.
