1.Biomechanic results of PLGA/TCP scaffold with bovine BMP in treating peri-porous-titanium bone defects in femur of adult rabbits

Objective To evaluate the feasibility of composite material of bone morphogenetic protein (BMP) and PLGA/TCP in repair of peri-porous-titanium bone defects in adult rabbits.Meth- otis The composite of PLGA/TCP scaffold with bovine BMP was made and implanted in distal bone de- fects peri-porous-titanium in femur of adult rabbits.The effect of BMP with PLGA/TCP on peri-prosthesis was evaluated by means of scanning electron microscope (SEM),energy dispersive X-ray analysis (EDX) and biomechanics.Results BMP with PLGA/TCP showed obvious peak of Ca and P of EDX in the pores of titanium in experiment group six weeks after operation and higher than that in control group (P<0.05).Push-out test demonstrated that the bonding strength between the composite of HA coating/ porous titanium and bone was increased significantly with time (P<0.05).In experiment group,at 6 and 12 weeks,peri-porous-titanium had higher shearing force compared with control group (P<0.05). Conclusion BMP loaded with PLGA/TCP is a promising bone graft for bone defects in revision arthro- plasty,as indicates that bone induction of BMP plays an important role in biological stabilizaiton.
2.Numerical Taxonomy and 16S rDNA PCR-RFLP of Rhizobial Strains Isolated from Psoralea corylifolia etc

24 strains obtained from root nodules of Psoralea corylifolia, Pueraria lobata, and Campylotropis macrocarpa of Yunnan province were studied with numerical taxonomy and 16S rDNA PCR-RFLP. Results of numerical taxonomy indicated that all strains included 10 reference strains were divided into 3 groups at 84% similarity. Group III is an unknown group with no reference strains. Group I is slow-growing kind, and group II fast and middle-slow-grower. The dendrogram derived from 16S rDNA PCR-RFLP showed that all strains divided into five phylogenetic branches at the similarity of 70%. They are branches I and V with no reference strains, Agrobacterium-Sinorhizobium-Rhizobium, Mesorhizobium and Bradyrhizobium. Not all results of numerical taxonomy are accord with 16S rDNA PCR-RFLP, and 2 strains at the same group with A. tumefaciens IAM13129T.
3.Expression and purification of spike protein of severe acute respiratory syndrome coronavirus in Saccharomyces cerevisiae

Objective To construct the recombinant plasmid pYES6-S and express and purify spike protein of severe acute respiratory syndrome(SANS)coronavirus in Saccharomyces cerevisiae. Methods DNA fragments of SANS coronavirus were obtained by reverse transeription.Four over- lapped fragments of spike protein genes were amplified by polymerase chain reaction(PCR)and ligated into an integral spike protein gene by restriction enzyme digestion.The spike protein gene recombined with pYES6 and cloned into E.coll.The recombinant plasmid pYES6-S was induced and expressed in Saccharomyces cerevisiae(INVScl)by galactose.Results The recombinant plasmid pYES6-S was confirmed that inserted fragment was right in length,direction and base matching by restriction enzyme digestion and sequencing.The purified protein encoded by the whole spike protein gene was about Mr 110?10~3 identified by electrophoresis.Conclusion The whole spike protein gene of SARS coronavirus is cloned into E.coli and the protein is expressed in Saccharomyces cerevisiae successful ly.which can be helpful in SARS vaccine research.
4.Relationship between Chinese medicine pattern with blood levels of sex hormone, lipid and glucose in climacteric women patients with hypertension.

OBJECTIVETo find the objective indices for traditional Chinese medicine (TCM) pattern identification in women patients with climacteric hypertension (CH) by exploring the relationship between TCM pattern with blood levels of sex hormone, lipid and glucose.
METHODSTCM syndrome differentiation was made in 75 women CH patients, 43 as yin-deficiency with inner dryness pattern (YDD) and 32 as non-YDD, and correlation analyses were conducted on TCM patterns and blood levels of estradiol (E2), lipids and glucose.
RESULTSBesides the level of E2 in YDD was lower than that in non-YDD [(112.26 -/+ 66.81) pmol/L vs (146.69 +/- 83.59) pmol/L, P < 0.05), no difference between the two patterns was found in the other indices measured (P > 0.05). Further subunit analysis showed that in patients with CH grade 1, blood levels of total cholesterol (TC), high density lipoprotein (HDL) and apoprotein A-I (ApoA-I) were higher in the 13 patients of YDD than those in the 9 of non-TDD (P < 0.05); in patients with CH grade 2, levels of E2, progesterone (T), HDL and ApoA-I were lower but level of follicle stimulating hormone (FSH) was higher in the 20 YDD than those in the 10 non-YDD (all P < 0.05); while in patients with CH grade, no statistically significant difference was shown between 3 patients of 10 with YDD and 13 with non-YDD in all the indices measured (P > 0.05).
CONCLUSIONBlood level of E2 shows different characteristics between CH patients of YDD and non-YDD patterns, suggesting that it may provide some help to syndrome micro-differentiation, and taking it as an objective index for syndrome typing of climacteric hypertension in female is considerable.

5.Effect of Erigeron breviscapus injection on ventricular and vascular remodeling in spontaneous hypertension rats.

OBJECTIVETo observe the ventricular and vascular remodeling reversal effect of Erigeron breviscapus injection (EBI), a protein kinase C inhibitor, in spontaneous hypertension rats (SHR).
METHODSTwenty-four SHR were divided into 4 groups, they were treated respectively with EBI, Fosinopril, Enalapril and normal saline 10 mg/kg per day by intraperitoneal injection for 8 weeks. Systolic blood pressure (SBP), ventricular weight index (VWI) and protein kinase C (PKC) activity in myocardial cells were determined, ultrastructural changes of heart and vessel were observed by polaroscope and transmission electron microscope, and the area and content of myocardial interstitial collagen (MIC) were determined by image analyzer system.
RESULTSThe left ventricular hypertrophy was regressed to certain degree after EBI, Fosinopril and Enalapril treatment, but no significant change in heart rate and right VWI was found. Fosinopril and Enalapril were superior to EBI in lowering SBP and left VWI, and EBI was more obvious in improving myocardial ultrastructure such as hypertrophy and degeneration. All the 3 drugs could improve the MIC and vascular remodeling, the MIC area, content and collagen volume fraction in the EBI group were lowered after treatment, as compared with those in the control group, but comparison between the three groups showed no significant difference. The 3 drugs could reduce the PKC activity in myocardial cell membrane, and EBI showed the effect more significant than that of the other two (P < 0.05).
CONCLUSIONEBI could reverse the myocardial, interstitial and vascular remodeling, improve the rigidness of cardiac muscle, thus, has protective effect on heart.

6.Study on Interaction Between Boronic Acid and Hydroxyethylamines

The reversible covalent interaction between boronic acids and cis-diol-containing compounds provides unique affinity for recognition and separation of cis-diol-containing biomolecules such as glycoproteins and sugars. Herein, by using β-blockers and β-agonists as representative hydroxyethylamines, the interaction between phenylboronic acid and hydroxyethylamines was investigated through nuclear magnetic resonance ( NMR) and high performance liquid chromatography ( HPLC ) . The results showed that strong interaction between hydroxyethylamines and phenylboronic acid occurred at high pH value, while the interaction became much weaker and even disappeared at low pH value. This interaction was similar to boronate affinity interaction between boronic acids and cis-diol-containing compounds. However, unlike boronate affinity, the presence of an aprotic solvent disrupted the interaction. The above findings not only provided new insights for in-depth understanding boronate affinity interaction, but also paved the basis for the application of the interaction between boronic acid and hydroxyethylamines.
7.A study on teratogenic effects of Chimonanthus salicifolius S. Y. Hu extract on SD rats

Objective To study maternal toxicity, embryotoxicity and teratogenecity of Chimonanthus salicifolius S. Y. Hu in SD rats.Methods A total of 64 successfully mated female SD rats were randomly divided in to 4 groups (16 per group), in which 3 experimental groups were daily treated with 3.75, 7.5 and 15.0 g/kg. bw test substance by lavage from 7th to 16th day during gestation respectively. Body weight and general conditions of the pregnant rats were recorded during the study. On the 20th day in pregnancy, the rats were anatomized and examined grossly, the fetuses were removed and counted, weight, length, visceral and skeletal changes were then examined. Results There was no significant difference in the conception rate, total weight gain during the pregnancy and the number of living, dead and resorbed fetuses between each dosage groups and the control group (P>0.05) . The number of the rib, sternum, the fifth sternum punctated and the parietal bone which were ossified defectively all showed no difference among the four groups (P>0.05) . Conclusion Chimonanthus salicifolius S. Y. Hu extract had no obvious maternal toxicity, embryotoxicity and teratogenecity in SD rats under this experiment condition.
8.Sub-health status of the young and middle-aged populations and the relationships among sub-health, sleep ness and personality in Hubei province, China

Objective To investigate the epidemiological condition of the sub-health status of the young and middle-aged persons in urban and rural areas of Hubei province, and to explore the relationships among personality, quality of sleep and sub-health status. Methods Purposive sampling method and Personal Health Status Questionnaire developed by Chinese Academy of Medical Sciences were used to investigate the sub-healthy status of 3187 subjects aged 18-55 and evaluate the relationships among sub-health status, personality and the quality of sleep with structural equation model. Results Gender, education, marriage, age and occupation were factors influencing the status of sub-health. Among all of the subjects, sub-health status of females were more serious than that of males (P<0.0001 of all). The divorced, separated and widowed ones were more likely to experience sub-health than other status of marriage (P= 0.0006, P=0.0102). The sub-health status of the subjects aged 30-55 in the urban (P=0.0197)and low-literacy residents in rural (P=0.0003)were among the worst. The condition of the sub-health in migrant workers and professional were worse compared to other occupational groups (P=0.0043). Structural equation model analysis revealed that sub-health status associated with the quality of sleep and the type of characteristics (P<0.01 ). Conclusion Sub-health status did exist among the young and middle-aged persons and was influenced by the quality of sleep and personality. However, it could be adjusted by improving the quality of sleep and personality to prevent sub-health.
9.Effects of tea polyphennols on hepatic lipase activity in rabbits with fatty liver.

OBJECTIVETo investigate the relationship between hepatic lipase activity and fatty degeneration of hepatocytes and explore the effects of tea polyphennols (TP) on the changes of hepatic lipase (HL) activity in rabbits with fatty liver.
METHODSAccording to serum cholesterol and triglyceride (TC) levels, 19 rabbits were divided into fatty liver group (FL, n=6) fed with high cholesterol diet, TP group (n=7) fed with high cholesterol diet and 20mug/g/d tea polyphennols everyday orally, control group (n=6) fed with normal diet. After 8 weeks, the levels of serum TC, HL activity, HL activity and malondildehyde (MDA) in hepatic tissue were detected, and the pathomorphology of hepatic tissue were determined in all rabbits.
RESULTSThe fatty degeneration of hepatocyts in FL group was more severe than that in TP and control group. The serum TC level in TP group (16.87 6.58) mmol/L was higher than that (1.11 0.82) mmol/L in control group (t=5.786, p<0.05), but lower than that (28.49 5.99) mmol/L in FL group (t=3.968, p<0.05). The serum low density lipoprotein-cholesterol level in Tp group (5.10 4.19) mmol/L also higher than that (0.71 1.14) mmol/L in control group (t=3.763, p<0.05), but lower than that (12.15 1.95) mmol/L in FL group (t=2.478, p<0.05). The number of positive dots presenting HL activity level in 100 square micron, hepatic tissue in TP group (3.24 0.17) was higher than that (1.76 0.10) in FL group (t=-3.153, p<0.05), but lower than that (4.14 0.05) in control group (t=-2.902, p<0.05). The levels of MDA in hepatic tissue in TP group (44.66 26.18) nmol/mg was significantly lower than that (75.58 29.88) nmol/mg in FL group (t=2.261, p<0.05), but no evidently different from that (43.64 16.95) nmol/mg in control group. The plasma HL activity was no difference among the three groups.
CONCLUSIONThe HL activity in hepatic tissue with fatty degeneration of hepatocytes was lower than that in normal liver. Tea polyphennols can increase HL activity in hepatic tissue and protect hepatocytes from fatty degeneration.

10.A study on the influence of dichloromethane exposure on liver function among furniture manufacturing workers

Objective To explore the effects of dichloromethane(DCM)exposure on serum biochemical indexes among furniture manufacturing workers. Methods A total of 65 workers who engaged in adhesive operations in furniture manufactory and 56 workers who would be recruited into another factory as new employees were investigated. Twenty two pairs of workers were selected as occupationally exposure group and control group,respectively. The concentration of DCM in workplace of adhesive operations in furniture manufactory was determined. Serum levels of TP,ALB,GloB,A/ G, ALT,AST,GGT,AFU,GPDA,ADA,ALP,CHE,CHOL,TG,TBA,TBIL,DBIL and IBIL were tested by automatic biochemical analyzer in all groups. Results The mean levels of serum TP,ALB,GloB in exposure group( the concentrations of DCM in workplace were 1 300 - 7 760 mg/ m3 )were 73. 7 ± 3. 6,47. 5 ± 2. 3 and 26. 2 ± 2. 8 g/ L,which were significantly lower than that of control group(P < 0. 05). Compared to the control group,the mean levels of CHE, ADA,AST and ALT activity in exposure group(7 477. 8 ± 1 240. 8,7. 3 ± 1. 7,21. 6 ± 5. 6 and 13. 0 ± 5. 6 U/ L)were significantly lower(P < 0. 05). The mean levels of TBIL,DBIL and IBIL were 13. 9 ± 7. 6,6. 4 ± 4. 3 and 7. 5 ± 3. 4μmol/ L,respectively,which were significantly higher than that of control group(P < 0. 05). No significant difference in the mean levels of the other biochemical indexes in serum was observed between exposure and control group. However,no correlation was found between the levels of ALB and TBIL,ALB and DBIL,or ALB and IBIL. Conclusion Occupational exposure to DCM is associated with the decrease of TP,ALB,GloB and CHE levels in serum. Meanwhile,DCM occupational exposure is correlated with the increase of TBIL,DBIL and IBIL levels in serum. Our data suggest that occupational exposure to DCM may be related to inhibition of synthetic function in live,damage of blood cells and declined metabolism of bilirubin.
