In addition, allopurinol has protective effects on ischemical reperfusion injury,inflammation and tumor.
2.Long-term follow-up for laparoscopic anterior 180° partial fundoplication for gastroesophageal refulx disease

Objective To evaluate long-term follow-up for laparoscopic anterior 180° partial fundoplication for gastroesophageal refulx disease (GERD).Methods A total of 48 patients had undergone a laparoscopic anterior 180° partial fundoplication from July 2004 to October 2007.Patients were followed up at 3 months,12 months,3 years,5 years by using a structured questionnaire via phone or e-mail which evaluated symptoms of reflux,dysphagia,side-effects,and overall satisfaction with the clinical outcome.Results Follow-up data were collected from 43 patients,ranging from 60 to 98 months.Postoperative heartburn significantly improved in 37 patients.Normal belching was preserved in 40 patients,and 38 patients were able to eat normally.Thirty nine (90.7%) patients reported a good or excellent result (minimal or no symptoms) at the late follow-up.Two patients underwent laparoscopic anterior 180° partial fundoplication again due to acid reflux at the 12th and 38th month respectively.Conclusion At minimum 5 years followup,laparoscopic anterior 180° partial fundoplication for GERD is effective and lasting,and most patients are satisfied with the outcome.
3.Effect of median nerve electrical stimulation on expression of α1-adrenergic receptor in prefrontal cortex of coma rats induced by traumatic brain injury

Objective To investigate the wake-promoting action of median nerve electrical stimulation(MNES) in coma rats induced by traumatic brain injury(TBI) and its influence on the expression of α1-adrenergic receptor(α1 R) in the prefrontal contex (PFC).Methods Seventy-two healthy Sprague Dawley(SD) rats were randomly divided into the control group,sham-stimulated group(TBI),stimulated group (TBI+ MNES) and antagonist group(TBI+ OX1R antagonist +MNES).The control group had no any treatment.The TBI coma rat models were prepared in the other 3 groups.The sham stimulated group had no treatment.The antagonist group was injected with orexin receptor-l(OX1R) antagonist SB334867 into lateral ventricle,and both the antagonist group and stimulated group received MNES treatment.Then the behavior changes of rats in each group were observed and the α1 R expression level in PFC was detected by using the immunohistochemistry technique.Results Thirteen rats in the stimulated group and 8 rats in the antagonist group revived,while only 4 rats in the TBI group.The α1R levels from low to high were the blank control group,sham-stimulated group,antagonist group and stimulated group,showing the increasing trend,and the difference was statistically significant(P<0.05).Conclusion MNES can improve the rat consciousness level after TBI coma,and its mechanism may be related with up-regulating the α1 R expression level in PFC area,moreover Orexin-A participates in this regulation process.
4.Spectrum-Effect Relationship of Ethyl acetate Fraction in Ethanol Extract fromXiaoyaosan(XYE-E) for Antidepressant Effect Based on1H-NMR

Thearticle is aimed to find the correlation between bioactive components of XYE-E and the antidepressant efficacy, by analyzing the immovability time in tail suspension test (TST) and forced swimming test (FST). Using the method of gray relational analysis, correlation analysis and regression analysis, relating the peak area of each common peak of1H-NMR spectra with the immovability time in TST or FST, we found that there were total 14 chemical components identified in the1H-NMR spectrum of XYE-E. Among them, 8 compounds, including saikosaponin a, saikosaponin c, saikosaponin E, saikosaponin F, saikosaponin G, saikosaponin b2, atractylenolide I and atractylenolide II, had significant correlation with antidepressant efficacy.
5.Treatment of children with EB virus infection by Chinese medicine: a clinical study.

OBJECTIVETo provide evidence for Chinese medical treatment of children with EB virus infection by exploring its clinical efficacy from multiple angles.
METHODSTotally 81 children patients were randomly assigned to the treatment group (46 cases) and the control group (35 cases). Patients in the treatment group took Chinese medical decoction, while those in the control received intravenous dripping of Ganciclovir and oral administration of pidotimod. The treatment period for the two groups was 2 weeks. Patients were followed-up till the 12th week. Clinical symptoms such as fever, lymphadenopathy and hepatosplenomegaly, as well as lab indices such as abnormal lymphocyte percentage, EB virus antibody, virus DNA load, T cell subsets, immunoglobulin, and so on were observed before and after treatment, at week 4 and 12 of follow-ups.
RESULTS(1) The total effective rate at week 2 was 95.6% in the treatment group, higher than that of the control group (94.3%), but there was no statistical difference between the two groups. (2) The time for defervescence, duration of pharyngeal hyperemia, duration of swollen tonsils was shorter in the treatment group than in the control group (P<0.05). The subsidence of lymphadenopathy, hepatomegaly, and abnormal lymphocytes was better in the treatment group than in the control group (P < 0.05). (3) The positive cases of peripheral blood hetero-lymphocyte was significantly reduced after treatment, at week 4 and 12 of follow-ups both in the treatment group and the control group (P < 0.01). The expression of IgA and IgM decreased after treatment in the two groups when compared with before treatment in the same group (P < 0.05, P < 0.01). IgG in the treatment group also obviously decreased after treatment, at week 4 and 12 of follow-ups (P < 0.05, P < 0.01), while it decreased only after treatment in the control group (P < 0.05). Activities of AST and ALT in the treatment group and the AST activity in the control group were markedly improved when compared with those before treatment (P < 0.05). Compared with the control group, the abnormal lymphocyte positive case number obviously decreased in the treatment group after treatment, at week 4 and 12 of follow-ups (P < 0.05). (4) After treatment, at week 4 and 12 of follow-ups, CD3+ and CD8+ significantly decreased; CD4+, CD4/CD8, and B cells significantly increased in the two groups, when compared with before treatment (P < 0.05). NK cells significantly increased more in the treatment group after treatment, at week 4 and 12 of follow-ups, higher than before treatment as well as the control group (P < 0.05). (5) EB viral DNA and EB viral CA-IgM negative conversion case numbers significantly increased in the two groups after treatment, at week 4 and 12 of follow-ups (P < 0.05). Compared with the control group, EB viral DNA and EB viral CA-IgM negative conversion case numbers significantly increased in the treatment group after treatment and at week 4 of follow-ups (P < 0.05).
CONCLUSIONSTreatment of EB virus infection by Chinese medical treatment was effective. It could promote the recovery of EB viral infection, and reduce the risk of vicious disease after EB viral infection.

6.Study on quality standard for Huanglongganzhixiao Granule

AIM: To establish the quality standard for Huanglongganzhixiao Granule(Resina Draconis, Radix Astragali, Herba Epimedii, etc.). METHODS: Radix Astragali, Resina Draconis, Herba Epimedii in Huanglongganzhixiao Granule were identified by TLC. The content of loureirin B in this Granule was determined by HPLC. RESULTS: Radix Astragali, Resina Draconis, Herba Epimedii could be identified by TLC. Loureirin B showed a good linear relationship at a range of 0.032~0.096?g, r =0.9997( n =5). The average recovery was 100.1% and RSD was 3.33%. CONCLUSION: The method is simple, accurate and with strong specificity and can be used for the quality control of Huanglongganzhixiao Granule.
7.Effect of Vagus Nerve Stimulation on Wake-promoting and Expression of γ-aminobutyric Acid b1 Receptor in Prefrontal Cortex of Coma Rats post Traumatic Brain Injury

Objective To investigate the wake-promoting effect of vagus nerve stimulation (VNS) on coma rats after traumatic brain in-jury (TBI), and the related mechanism. Methods A total of 168 healthy Sprague-Dawley rats were randomly divided into blank group, TBI group, antagonist group and VNS group, 42 rats in each group. The latter three groups were established TBI model with impact, and the rats in coma at least 30 minutes were included. VNS group accepted VNS, the antagonist group were injected intralateroventricularly Orexin A receptor 1 (OXR1) antagonist SB334867, and TBI group accepted sham VNS. Their behaviors were observed to determine the level of con-sciousness six, twelve and 24 hours after intervention, while the expression ofγ-aminobutyric acid b1 receptor (GABAb1R) in prefrontal cortex was detected with immunohistochemistry and Western blotting. Results There were 42 rats in the blank group, 11 rats in TBI group, 13 rats in the antagonist group, and 28 rats in VNS group awakened finally. The expression of GABAb1R in prefrontal cortex ranged as TBI group, antagonist group, blank group and VNS group from more to less twelve and 24 hours after intervention under Western blotting (F>60.412, P<0.001), and it ranged as TBI group, antagonist group, VNS group and blank group under immunohistochemistry (H=15.121, P=0.002), with no significant difference among time points (H=3.028, P=0.220). Conclusion VNS can promote waking from coma in rats after TBI, which may relate with the decrease of GABAb1R in prefrontal cortex that induced by Orexin A.

9.Pharmacokinetic comparison of two ozagrel polymorph forms in SD rats.

To enhance the quality and efficiency of ozagrel by investigating the differences between the ozagrel polymorphs in bioavailability. Solid ozagrel in different polymorph forms were orally administered to SD rats. An HPLC method was established to determinate plasma level of ozagrel. The bioavailabilities of two polymorph forms were calculated and compared. The pharmacokinetic parameters of ozagrel, were as follows: Cmax was 32.72 ± 17.04 and 34.01 ± 19.13 mg · L(-1), respectively; AUC0-t was 61.14 ± 14.76 and 85.56 ± 18.08 mg · L(-1) · h, respectively; t½ was 1.53 ± 0.51 and 4.73 ± 3.00 h, respectively. There was no significant difference in pharmacokinetic parameters between form I and II polymorphs of ozagrel while the t½ of form II is longer, which indicates that the use of form II polymorph as pharmaceutical product may prolong the effective action time in clinics. This would help the polymorph quality control in drug production.

10.Research on ultrafine grinding technology of improving dissolution rates of effective components in Sanjie Zhentong capsule.

The effects of ultrafine grinding on the dissolution rates of the effective components in Sanjie Zhentong capsule (SZC) were studied in this experiment. Fine and ultrafine powder of SZC intermediates were made by ordinary grinding and ultrafine grinding technology, and then granulated by wet granulation. SZC were prepared by fine powder, ultrafine powder and ultrafine granules, respectively. With resveratrol and loureirin B as investigated indexes, dissolution rates of the four intermediates in SZC were determined by cup method and HPLC. The dissolution rates of resveratrol in SZC prepared by fine powder, ultrafine powder and ultrafine granules were 26.11%, 63.27%, 67.49%, respectively; and the dissolution rates of loureirin B were 7.160%, 20.29%, 23.05%, respectively. The dissolution rate of resveratrol and loureirin B in SZC prepared by ultrafine granules was the best. D90 size of ultrafine grinding was 13.221 μm and could improve the dissolution rates of resveratrol and loureirin B in SZC.
