2.Clinical characteristics and surgical treatment of ganglioneuroma in spine.

OBJECTIVETo summarize the clinical manifestation and diagnosis of ganglioneuroma in spine and investigate the clinical effect of surgical treatment.
METHODSThe clinical data of 6 patients underwent a surgery for ganglioneuroma in spine from January 2008 to January 2015 were retrospectively analyzed. There were 4 males and 2 females, aged from 2 to 63 years old with an average of 34.6 years. The courses of disease were from 3 days to 17 years. Five patients complicated with superficial hypesthesia in correlative level of tumor, and the muscle strength under tumor plane had decreased at different levels, with the strength of grade II-IV. Two cases complicated with hypermyotonia and positive bilateral Hoffmann's and Babinski sign. Five cases were sporadic lesion in correlative spinal canal and one case complicated with the giant occupying lesion in thoracic cavity.
RESULTSSix operations had been performed including 5 en bloc and 1 subtotal resection. Postoperative pathological results showed tumor cells scattered or fasciculated inserted into Schwann cells in the stroma. In 2 patients complicated with radiculalgia before operation, 1 case was relieved and 1 was invariant after operation. All 4 patients with preoperative dyscinesia in the limbs obtained improvement after operation. All the patients were followed up from 0.3 to 6.8 years with an average of 2.5 years. At the final follow-up, according to ASIA grade, 5 cases were good and 1 case was invariant. During the follow-up, only 1 patient experienced chemoradiation because of merging ganglioneuroblastoma and receiving subtotal resection. No recurrence in other 5 cases.
CONCLUSIONGanglioneuroma is a benign and rare tumors in spine. Clinically, radicular pain and sensory-motor disorders are the main manifestations. Its diagnosis depends on pathological examination. Prognosis of surgical treatment is good.

3.P-VEP games aided combined treatment of amblyopia in children

AIM: To evaluate the efficacy of combined therapy for amblyopia in children by making use of pattern visual evoked potential ( P-VEP) game.
METHODS: This was a prospective case control study. These asthenopic children were divided into two groups. The control group ( 66 eyes of 49 patients ): occlusive therapy with glasses, cover, precision work, red light treatment and so on, later the stereo vision training was added. The experimental group (72 eyes of 52 patients):conventional methods mentioned above with P - VEP games.
RESULTS: The total effective rate and cure rate of experimental group in 6mo were higher than those of control group. The overall effective rate was 94. 4% in the experimental group and 83. 3% in the control group. There was a statistically significant difference between them (P<0. 05).
CONCLUSION: The comprehensive therapy by making use of P-VEP game is an individualized effective new way in treating amblyopia.
4.Determination of residual organic solvents and macroporous resin residues in Akebia saponin D.

According to ICH, Chinese Pharmacopoeia and supplementary requirements on the separation and purification of herbal extract with macroporous adsorption resin by SFDA, hexane, acetidine, ethanol, benzene, methyl-benzene, o-xylene, m-xylene, p-xylene, styrene, diethyl-benzene and divinyl-benzene of residual organic solvents and macroporous resin residues in Akebia saponin D were determined by headspace capillary GC. Eleven residues in Akebia saponin D were completely separated on DB-wax column, with FID detector, high purity nitrogen as the carry gases. The calibration curves were in good linearity (0.999 2-0.999 7). The reproducibility was good (RSD < 10%). The average recoveries were 80.0% -110%. The detection limit of each component was far lower than the limit concentration. The method is simple, reproducible, and can be used to determine the residual organic solvents and macroporous resin residues in Akebia saponin D.

5.Simultaneously preparation of grams of high purity tyrosol, crenulatin and salidroside from Rhodiola crenulata.

Tyrosol, crenulatin and salidroside are the main active constituents of Rhodiola crenulata, with extensive pharmacological activities. In the study, grams of high purity tyrosol, crenulatin and salidroside were simultaneously separated from R. crenulata by the first time. Firstly, R. crenulata was extracted by 70% alcohol. Then, with the yields of three compounds as the index, the macroporous resin was optimized. At last, grams of high purity tyrosol, crenulatin and salidroside were isolated by D-101 macroporousresin, purified by column chromatography. Detected by HPLC, the purity of three compounds were higher than 98%. This method has the advantages of simple process and operation, less dosage of organic solvent, highly yield and reproducibility, suitable for the simultaneously preparation of tyrosol, crenulatin and salidroside.

6.Role of Rho kinase in reorganization of the vascular endothelial cytoskeleton induced by rat burn serum.

OBJECTIVETo investigate the changes in endothelial cytoskeletal reorganization and the role of Rho in the signal transduction pathway.
METHODSECV304 cells were cultured and randomly divided into following groups: i.e. sham (with normal rat serum treatment), burn (with burn rat serum treatment), Y (with 30 micromol/L Rho kinase inhibitor Y-27632 treatment), burn plus Y (pretreatment of cells with burn serum before treated with 30 micromol/L Y-27632), Y plus burn (pretreatment of cells with Y-27632 for 1 hour before treated with burn serum), LPA (with normal rat serum and 13 micromol/L LPA), and LPA plus Y (pretreatment of cells with LPA before treated with Y-27632) groups. The indices were examined at 6, 7 and 8 posttreatment hours (PTH) in all groups except in Y group. The endothelial morphology was observed with HE staining. Endothelial cytoskeleton was observed by dual-fluorescence labeling of filamenta (F) with Rhodamine-phalloidin and monomer (G) with oregon green labeled DNAase. The actin content in the cells in all groups was measured with flow cytometry.
RESULTSIn sham and control group, the cells were in fusiform or polygonal shape, with satisfactory growth; filamentous actin (F-actin) was mainly distributed in the peripheral site of the cytoplasm and formed peripheral filamental band. The cells became confluent to form a single layer with reticular structure. Globular actin (G-actin) was concentrated in the nucleus and per nucleus. In burn group, after 6 hours of burn serum treatment, the ability of cells to adhere to vessel wall was weakened, and a striking reorganization of the actin cytoskeleton and the formation of the stress fibers were found. Furthermore, the fluorescent intensity of the peripheral filament bands was weakened, and dispersed actin monomers were seen in the cytoplasm. This reaction was enhanced along with elapse of stimulation time. In burn plus Y or Y plus burn group, the cells grew and adhered well to the wall of culture vessel. The distribution of the filamentous actin was the same as the sham group, while the stress fiber decreased in amount obviously. The structure of globular actin was condensed with little G-actin in the cytoplasm. The changes in actin cytoskeleton in LPA group was similar to that in burn group. The effects of LPA on actin reorganization could also be reversed by Y-27632. The content of F-actin in burn group at 6 PTH (0.63 +/- 0.07) was lower than that in sham group (0.75 +/- 0.08), while the content of G-actin in burn group (1.28 +/- 0.27) was higher than that in sham group (1.16 +/- 0.16, P > 0.05).
CONCLUSIONBurn serum induces vascular endothelial actin cytoskeleton reorganization in endothelial cells via the Rho-dependent signal pathway. Similar to the effect of LPA, this effect could be reversed by Y-27632.

7.A new surgical treatment for thumb (finger) reconstruction by the free moulded second toe transfer

Objective To investigate a more perfect method for a nice outward appearance of the thumb(finger) reconstructed.Methods An artery pedicle composite flap from fibular side of the great toe is inlaid in the tibial(ventral) side of the free second toe for thumb reconstruction and the same free second toe with a distal part of metatarsal bone with a double-wings flap for finger reconstruction before transplantation. Results The reconstructed thumb(finger) gets a nice looking and normal function while no blight to the great toe occurred.Conclusion It is an effective new procedure in ameliorating outward appearance of the recon- structed thumb(finger) by transferring the free moulded second toe.
8.The influence of liver fat deposition on liver iron overload quantitative assessment by fast-kilovolt-peak switching dual-energy CT imaging and material decomposition technique:a vitro experiment study

Objective To investigate the influence of liver fat deposition on the quantification of the liver iron overload using fast-kilovolt-peak switching dual-energy CT imaging and material decomposition technique. Methods A total of 20 healthy SD rats were taken to make 18 PVC tube of homogenate of fresh liver tissue. The dextran with concentration of 50, 40, 30, 20, 10 and 0 mg/ml were mixed with rat liver homogenate and triglyceride with three different concentrations (add fat with volume percentage of 60%, 30%, 10% and to simulate severe, moderate and mild fatty liver respectively). All samples were placed in standard phantom according to the order of iron concentration from high to low and scanned by GE Revolution CT 256 slices scanner in GSI mode with rapid tube voltage switching between 80 and 140 kVp and with tube current 200 mA, 320 mA, 485 mA respectively. The images of iron (fat)-based substance pair were reconstructed and the virtual iron concentration (VIC) value were recorded. The correlation between VIC and the actual liver concentration (LIC) of the three sets of tube currents (200, 320, 485 mA) was analyzed by Spearman correlation analysis and linear regression. Results LIC and VIC were highly positively correlated with the liver iron deposition model under different tube currents (r value was 0.900 to 1.000, P<0.05). The presence of fat will result in the decrease of VIC value. In a certain X-ray energy range and certain liver iron concentration, the higher the fat content, the more liver iron concentration underestimation were happened in VIC. At 200, 320 and 485 mA, the crossing points of linear equations for 30%and 10%fatty liver iron deposition models were located at VIC=12.682, 12.470 and 13.447 mg/cm3, respectively. Conclusions The fast-kilovolt-peak switching dual-energy CT imaging and material decomposition techniques can be used for quantitative evaluation of liver iron with hepatic steatosis. The presence of fat will lead to a decrease in VIC measurement.
9.Expressions of ATM, ATR, Chk1 and Chk2 genes in human glioma

Objective To detect the expressions of DNA damage checkpoint genes including A TR, A TM, Chk1 and Chk2 in human primary gliomns and explore their relations with tumor progression. Methods SYBRTM Green real-time quantitative PCR was performed to detect the expressions of ATR, A TM, Chk1 and Chk2 genes in 35 cases of primary gliomas and 10 of normal brain tissues. Results In glioma tissues of various pathological grades, the expressions of the target genes, with the exception of A TM gene, were significantly increased as compared to those in normal brain tissues (P<0.05). Chk1 gene expression was significantly higher in grade Ⅳ than in grade Ⅱ and Ⅲ gliomas (P<0.05), but no significant differences were found in A TR or Chk2 gene expression between grade Ⅱ, Ⅲ and Ⅳ gliomas (P>0.05). Conclusion The up-regulation of ATR, Chk1 and Chk2 genes in primary glioma suggests their association with the pathogenesis of glioma. Chk1 expression may indicate the malignancy of glioma and help evaluate the pathological grade of glioma.
10.A case-control study on the relationship of drinking water from farmland edge and digestive tract cancers

Objective To explore the relationship between drinking water from farmland edge and digestive tract cancers in rural areas.Methods A total of 180 cases of digestive tract cancers from Longquan city,Jinyun county and Qingtian county were selected and 360 controls with the same age group and sex were enrolled.1∶2 matched design of case-control study was used.The data was analyzed by Wilcoxon matched -pairs signed -ranks test.Results Overall,individuals who drank water from farmland edge were more likely to have digestive tract cancer among 3 areas (OR=4.005 ,95%CI:2.228-7.199).In Qingtian county the risk of digestive tract cancers in individuals who drank water from farmland edge was 5.375 times of control group population (95%CI:2.287 -12.635 ),But there was not statistically significant in Longquan city and Jinyun county (P >0.05 ).For gastric cancer,the proportion of drinking farmland edge water was significantly higher in cases than that in control group (OR=4.875,95%CI:2.007-11.839),while the difference was not statistically significant for liver cancer,colorectal cancer and esophageal cancer (P>0.05 ).In Qingtian county ,for gastric cancer,the proportion of drinking water from farmland edge was 5.750 times of control group population (95%CI:1.683-19.649).But there was not statistically significant in other cancers(P>0.05).Conclusion Drinking water near farmland edge may be one of the risk factors of gastric cancer in rural areas.
