1.Effects of alendronate on articular cartilage degeneration in rats with anterior cruciate ligament transaction

[Objective]To investigate the effects of alendronate(ALN)on subchondral bone and articular cartilage in the rat osteoarthritis model induced by anterior cruciate ligament transaction(ACLT).[Method]Twenty-four three-month-old female Sprague-Dawley rats were divided randomly into three groups:Sham group(n=8),ACLT+NS group(n=8) and ACLT+ALN group(n=8)(20 ?g?kg-1?2 d-1 subcutaneous injection for 12 weeks).Bone mineral density(BMD) measurement and bone histomorphometric analysis were made for the subchondral bone of right distal femur and proximal tibia in all rats at 12 weeks post surgery.Degree of cartilage degeneration was scored by Mankin scoring system.Immunostaining for matrix metalloproteinase 13(MMP-13) was performed to investigate the effect of ALN on cartilage matrix loss.[Result]BMD of the subchondral bone in the ACLT+NS group were significantly lower than those in the Sham group and the ACLT+ALN group(P
2.Construction and Eukaryotic Expressing of GPI-Anchored Met-RANTES

Objective: To express glycosyl-phosphatidylinositol (GPI) modified Met- RANTES fusion protein on Chinese hamster ovary (CHO) cells and to develop a novel immunosuppressant GPI anchored form of Met-RANTES. Methods: The eukaryotic expression vector PEF/GPI-Met-RANTES were constructed and transfected into CHO cells by electroporation. The transfectants were selected with methotrexate (MTX). Expression of the recombinant protein was assessed by flow cytometric analysis, cell immunofluorescence staining and immunogold electron microscopy. Results: The chimeric molecules of GPI anchored form of Met-RANTES including the whole reading frame were constructed, and the sequence was identical to the designed sequence. GPI anchored form of Met-RANTES was stably expressed on CHO- DHFR- cells. Conclusion: A large amount of GPI modified Met-RANTES fusion protein was expressed on CHO cells. GPI anchored form of Met-RANTES may be used as novel immunosuppressant for suppressing reaction in graft rejection.
3.Comparison of Clinical Efficacy of Lyophilized Recombinant Human Brain Natriuretic Peptide and Dobutamine in Treatment of Acute Heart Failure and Plasma Gal-3, CysC and ET-1 Levels

Objective:To explore and compare the clinical efficacy of lyophilized recombinant human brain natriuretic peptide (Lrh-BNP) and dobutamine (Dob) in the treatment of patients with acute heart failure (AHF) and impacts on the plasma galectin (Gal)-3,Cystatin C (CysC) and endothelin (ET-)-1 levels.Methods:114 cases of patients with AHF in our hospital from February 2015 to February 2017 were selected as the research objectives and randomly divided into two groups.Dob group was treated by Dob,while Lrh-BNP group was treated by Lrh-BNP.The cardiac function parameters,plasma Gal-3,CysC,ET-1 levels before and after treatment,clinical comprehensive efficacy and incidence of adverse reactions were compared between two groups.Results:The FS,LVEF levels of both groups at 72 hours after treatment were significantly higher than those before treatment (P<0.01),but the LVEDD,plasma Gal-3,CysC,ET-1 levels were obviously decreased (P<0.01),the index mentioned above of Lrh-BNP group improved more significantly than those of the Dob group(P<0.01).The overall effective rate of Lrh-BNP group was 89.5 %,which was significantly higher than that of the Dob group (73.7%,P<0.05).No significant difference was found in the incidence of adverse reaction between two groups(P>0.05).Conclusion:Lyophilized recombinant human brain natriuretic peptide was more effective in the treatment of AHF than Dobutamine with equal safety,which might be related to the decrease of plasma Gal-3,CysC,ET-1 levels.
4.Comparison of retinopathy of prematurity incidences between single and multiple premature infants and analysis of related factors

Background Retinopathy of prematurity (ROP) is one of major causes of childhood blindness.Early screening and treatment of ROP is necessary.Objective This study was to analyze the incidence of ROP in single and multiple premature infants and related factors.Methods This study was approved by Ethic Committee of Suzhou Municipal Hospital,and oral informed consent was obtained from the parents of the infants before any medical examinations.Five hundred and four fetus with the gestational age of ≤34 weeks or birth weight of ≤ 2 000 g were collected in Suzhou Municipal Hospital,of whom 357 infants were single fetus birth and 147 infants were multiple fetus,including 26 twins and 21 three fetus birth.The fundus were examined by RetCam Ⅱ equipment and ROP was determined based on the fundus findings and history of birth.ROP incidences between the single fetus and multiple fetus were compared,and the inducing factors were analyzed.Results Sixty-six infants were ROP in all 504 fetus,with the incidence of 13.10%.The incidence of ROP in the single fetal group was 10.92% (39/357),which was significantly higher than 18.37% (27/147) in the multifetal group (x2=5.069,P=0.024),and risk of ROP in the multifetal group was higher than that in the single fetal group (OR =1.835).The oxygen inhalation rate was 42.02% (150/357) and the oxygen inhalation duration was 5.71 days in the single fetal group,and that in the multifetal group was 58.50% (86/147) and 9.42 days,respectively.Conclusions Incidence and risk of ROP in the multifetal infants are higher than those in the single fetal infants,and low-birth-weight,less gestational weeks and excess oxygen inhalation are influence factors.
5.Projections from respiratory neurons of medulla oblongata to the motor neuron pool of phrenic nerve in rat:An HRP retrograde tracing study

ObjectiveTo observe the projections from respiratory neurons of medulla oblongata to the motor neuron pool of phrenic nerve in rat. MethodsBy using horseradish peroxidase (HRP) retrograde tracing technique after injecting HRP into phrenic nerve, retrograde labelled neurons were found in C3-C5 segment. Then, HRP was injected into the area where the phrenic motor neurons occupied, retrograde labelled neurons were found in medulla oblongata. ResultsPhrenic motor neurons locate in the C3-C5 segment ipsilaterally, occuping the intermediate portion of anterior horn and appearing typical motor neurons. Retrograde labelled neurons were found bilaterally in medulla oblongata, the neurons were located in nucleus retroambigualis (RNA), ventramedial area to nucleus retrofacialis (NRF). ConclusionThe phrenic motor neurons may receive direct projection of respiratory neuron in medulla oblongata, the projecting neurons are distributed in RNA and NRF area.
6.Detection of Low-abundance Point Mutations by Competitive Strand Assisted Endonuclease Ⅳ Signal Amplification System

Genetic mutations are important molecular biomarkers for cancer diagnosis and surveillance.Therefore,the development of methods for mutation detection characterized with straightforward,highly specific and sensitive to low-level mutations within various sequence contexts is extremely needed.Although some of the currently available methods have shown very encouraging results,their discrimination efficiency is still very low.Herein,we demonstrate a fluorescent probe coupled with blocker and property of melting temperature discrimination,which is able to identify the presence of known or unknown single-base variations at abundances down to 0.1% within 20 min.The discrimination factors between the perfect-match target and single-base mismatched target are determined to be 10.15-38.48.The method is sequence independent,which assures a wide range of application.The new method would be an ideal choice for high-throughput in vitro diagnosis and precise clinical treatment.
7.Relationship among health literacy and rehabilitation compliance and prognosis in patients with diabetes combined tuberculosis

Objective To investigate the relationship among health literacy and treatment compliance and prognosis in patients with diabetes combined tuberculosis.Methods One hundred and eighty-one cases patients with diabetes combined tuberculosis were selected as the objectives and evaluated literacy level,compliance scale by the health literacy scale and compliance questionnaires.The control of tuberculosis,blood glucose control,treatment compliance and prognosis were analyzed in the patients with different health literacy level.Results Of 181 patients,there were 89 cases patients'(basic health literacy group) the total scores of health literacy and compliance were>80 points,the efficacy of pulmonary tuberculosis control and blood sugar control were respectively 84.2% (75/89) and 93.2% (83/89);92 cases patients' the total scores of health literacy and compliance were ≤ 80 points,the efficacy of pulmonary tuberculosis control and blood sugar control were respectively 59.7% (55/92) and 67.4% (62/92),the differences were significant (x2 =12.78,18.11;P <0.05).Pearson correlation analysis showed that the total scores of health literacy were positive correlated with the scores of treatment compliance (r =0.688,P < 0.001).Logistic regression analysis showed that three dimension of health literacy (health knowledge and concept,healthy lifestyle and behavior,health skills) that can combined predict the variance of rehabilitation compliance(P<0.01).Conclusion The health literacy level of diabetes combined tuberculosis patients can effectively predict the rehabilitation compliance.Measures should be taken to improve the patient's level of health literacy,so as improve the compliance of rehabilitation and prognosis.
8.Roux-en-Y gastric bypass improves carbohydrate and lipid metabolism in nonfat type 2 diabetes mellitus patients

Objective To evaluate Roux-en-Y gastric bypass operation on carbohydrate and lipid metabolism in type 2 diabetes mellitus patients with BMI range of 24 -29. Methods Thirty seven cases of type 2 diabetes mellitus patients undergoing Roux-en-Y gastric bypass operation were studied. Body mass index (BMI), glycosylated hemoglobin ( GHbAlc), fasting glucose ( FPG), fasting insulin (FIns) and C-peptide( FC-p), HOMA-IR, oral glucose tolerance (OGTT) including 2 hour insulin (2hIns) and C-peptide (2hC-p) , plasma levels of total cholesterol (TC), triglycerides(TG), high density lipoprotein( HDL-c)and low density lipoprotein ( LDL-c) were measured preoperatively and on 3 months, 6 months, later postoperatively. Result There was no statistically significant difference between BMI values measured preoperatively and postoperatively (P>0. 05 ). Postoperative levels of HDL-c (1. 2 ± 0. 1, 1. 4 ± 0. 4, 1. 4 ± 0. 2, P<0. 01) ( mmol/L) , 2hIns (17. 2 ±3.4, 26. 3 ±4.7, 28. 6 ±4.1, P<0. 01) (mU/L)and 2hC-p(4. 2 ± 1. 0, 6. 3 ± 1. 5, 6. 2 ± 1.4,P<0. 01 ) ( nmol/L) were significantly higher than that of the pre-operative values ( P<0. 01 ).Conclusions Roux-en-Y gastric bypass significantly improves the metabolism of carbohydrate and lipid in type 2 diabetes patients with BMI 24-29, and the effects are not associated with weight loss.
9.New recognition of the relationship between gallbladder benign lesions and gallbladder cancer

With the development of imaging technology,the detection rate of gallbladder benign lesions has increased year by year.And part of the diseases may evolve into gallbladder cancer through a series of pathophysiologic processes.There are some misunderstandings in the understanding of gallbladder benign lesions for surgeons,so it is difficult for the clinical decision-making.The relationship between gallbladder benign lesions and gallbladder cancer should be correctly understood.Surgeons can neither exaggerate the risk of gallbladder cancer nor miss optimal timing of operation.The key point of the diagnosis and treatment of gallbladder benign lesions should be based on making full use of imaging data and strictly grasp pre-and intraoperative indications,and it is important to identify the malignant transformation of gallbladder benign lesions as soon as possible and carry out standardized treatment.
10.Traditional Chinese Medicine Syndrome Differentiation of AIDS Patients on ARV Complicated with Anemia and Hyperlipidemia

This study was aimed to analyze four diagnostic methods of AIDS patients with HAART complicated with anemia and hyperlipidemia. Characteristics of traditional Chinese medicine (TCM) and common pathogenesis were discussed in this article. This study was supported by Key Project of the National Eleventh-Five Year Research Program of China. This study was conducted through the retrospective analysis. This analysis was made on the four diagnostic methods of AIDS patients with HAART treatment. The results showed that the most frequent symptoms of AIDS patients with anemia or hyperlipidemia are fatigue, facial abnormalities, taste abnormalities, forgetfulness, backache, weakness of back, loss of appetite, insomnia. And the frequent tongue and pulse are white tough coating, pale tongue and fine pulse. The most frequent symptoms of the anemia group are facial abnormalities, backache, dizziness, palpitations, shortness of breath, pale complexion, abnormal state of mind, abnormal sweating, tinnitus, cold hands and feet, spontaneous sweating, white tongue coating, tooth-printed tongue, thin tongue coating and weak pulse (P < 0.05). The most frequent symptom of hyperlipidemia are lassitude, weakness, loss of appetite, abdominal distension, moisture tongue, thick and greasy tongue coating, slippery pulse. It was concluded that syndromes of AIDS patients on ARV complicated with anemia and hyperlipidemia are mainly deficient pattern, which affects the spleen, heart, liver and kidney. The most frequent syndrome type with anemia patients is blood deficiency, which is followed by spleen-kidney deficiency syndrome, liver-kidney deficiency syndrome. The most frequent syndrome type with hyperlipidemia patients is spleen qi deficiency and kidney essence deficiency, which are closely related to the spleen and kidney.
