Objective To explore the treatment effect and mechanism of A-NK cells on the subcutaneous trans?planted tumor of oral tongue squamous cell carcinoma in nude mice through observing the external growth and hyperplasia of A-NK cells and to provide theoretical evidence for squamous cell carcinoma treatment. Methods A-NK cells and NA-NK cells were both derived from healthy human peripheral blood and cultured in vitro. Cell growth was observed under micro?scope. The squamous cell carcinoma model in nude mice was established through subcutaneous implanting of Tca8113 cells. Then they were randomly assigned into three groups who were injected with either saline solution, or A-NK cells or NA-NK cells paraneoplastically. All animals were sacrificed after 33 days when tumor were isolated then weight and change in tumor size were assessed. Finally curve of tumor growth was drawn. Results Under the microscope, the proliferation of A-NK cells peak in 15 days and NA-NK cells peak in 12 days. After 3 weeks, the number of A-NK cells increased by 39.33 times while the number of NA-NK cells increased by 16.33 times. The Volume of tongue squamous cell carcinoma in saline solu?tion group was larger than that in A-NK cells and NA-NK cells groups, and volume in the NA-NK cells group was larger than that in A-NK cells group. The volume of tongue neoplasms in different groups, time, and interaction effects are statisti?cally significant (P<0.01). The tongue neoplasms weight in the saline solution group was greater than that in the A-NK cells and NA-NK cells group, and the weight in NA-NK cells group was greater than that in A-NK cells group, and the difference are statistically significant (P<0.05). Conclusion A-NK cells and NA-NK cells can significantly inhibit the subcutane?ous transplanted tumors in nude mice and the anti tumor effect of A-NK group is stronger than NA-NK.

Objective: To explore the methodology and efifcacy of transcatheter induced closure in patients with small patent ductus arteriosus (PDA). Methods: A total of 717 PDA patients treated in our hospital from 2005-11 to 2014-08 were summarized and there were 8 patients with small PDA were treated by transcatheter induced closure method including 3 male and 5 female from (1-6) years of age at the mean of (3.9±1.4) years with the body weight of (10-21) kg at the mean of (15.2±3.7) kg. The procedures were performed under local or general anesthesia with right cardiac catheterization and descending aortic arch angiography to observe PDA morphology and to measure PDA diameter at aortic and pulmonary aterial lateral and ductus length. Right catheter along the guide wear was pushed to the narrowest part of PDA and the motion was repeated for several times to stimulate the local area and then, the catheter was kept at PDA aortic lateral about 20 minutes thereafter. Results: Descending aortic arch angiography indicated that no residual shunt at 20 min after catheter partial blockage in all 8 patients, the immediate closure rate was 100%. No patient suffering from re-canalization by 1 year follow-up examination. Conclusion: Transcatheter induced closure of small PDA has minor trauma, no foreign material implantation, with low cost and good effect. It provides a new method for treating such particular type of PDA patients in clinical practice.

Objective:To explore the treatment effect of A-NK cells on the subcutaneously transplanted tumor of tongue squamous cell carcinoma in nude mice.Methods:Tca8113-Tb cells were subcutaneously transplanted into nude mice,14 tumor bearing mice were randomly assigned to 2 groups(n =7).The mice in control group were injected with normal saline,those in the experimental group were injected with A-NK cells.All animals were killed 33 days after tumor cell transplantation,The tumor volume and weight of the mice were measured and compared.Results:A-NK cells significantly inhibited tumor growth in nude mice in terms of the volume (P ＜0.05).After 15 -33 d of treatment the tumor weight(g)in the treated and control mice were 0.96 ±0.38 and 3.74 ±1.22 re-spectively(P ＜0.05).Conclusion:A-NK cells can inhibit the growth of Tca8113-Tb cell induced tumor in nude mice.

Objective To investigate the relation between the expression of toll receptor-4 (TLR-4) and the increase of blood brain barrier (BBB) permeability in the rats with acute pancreatitis.Methods Male Sprague-Dawley rats were randomly divided into 8 groups:control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;acute necrotizing pancreatitis (ANP) 2 h,6 h,12 h,24 h,48 h groups with 8 rats in each group.The BBB permeability,pathological score of pancreas,TLR-4 expression were determined and the relationships between them were analyzed.Results The BBB permeability in control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;ANP 2 h,6 h,12 h,24 h,48 h groups were 1.55±0.29,1.64±0.17,1.69±0.24,1.89±0.12,2.66±0.32,2.91±0.29,2.89±0.69 and 1.84±0.07,respectively;the pathological scores of pancreas were 0,2.38±0.92,3.13±0.64,8.50±1.07,9.75±0.71,10.25±1.28,11.13±1.25 and 10.13±1.13,respectively;there was no significant difference between AEP groups and control group,while there was significant difference between AEP groups and ANP groups (P＜0.05 orP ＜0.01).BBB permeability was correlated with pancreatic injury ( r = 0.626,P ＜0.01).There was no TLR-4 mRNA and protein expression in the control and AEP group,while they were significantly expressed in ANP groups,and the expression were positively related with BBB permeability ( r =0.208,P = 0.027 ).Conclusions BBB permeability was present in the course of ANP.Activation of TLR-4 signal pathway may be involved in the BBB permeability increase caused by ANP.

Objective To explore the long-term effect of open reduction and internal fixation for the treatment of displaced acetabular fracture,and analyze the influence of risk factors.Methods The clinical data of 62 patients with acetabular fracture from August 2005 to February 2009 was analyzed retrospectively.All the cases were treated with open reduction and internal fixation.The long-term effect and related risk factors were analyzed.Results All the cases were followed up for 24-52(38.8 ± 2.6) months.According to the Matta standard of replacement of fracture,there were 43 cases with anatomic reduction,14 eases with satisfactory reduction,5 cases with unsatisfactory reduction.Fracture union was obtained in all the patients.Based on the modified Merled' Aubigne -Postel clinical grading system,the result was excellent and good in 50 patients,fair and poor in 12 patients,with excellent rate of 80.65％ (50/62).Postoperative complications including traumatic arthritis were seen in 7 patients,heterotopic ossification in 3 patients and femoral head avascular necrosis in 1 patient.The related risk factors of clinical results of displaced acetabular fracture were age,Letournel-Judet fracture type,operation time,cartilage surface damage of the femoral head and quality of reduction (P ＜ 0.05 ).However,gender,AO fracture type,surgical approach and hip dislocation were not affecting factors of the acetabular fracture(P ＞ 0.05 ).Conclusions Open reduction and internal fixation can result in a satisfactory clinical outcome.Age,Letournel-Judet fracture type,operation time,cartilage surface damage of the femoral head and quality of reduction are independent risk factors affecting postoperative long-term functional outcomes.

Objective To explore the effects of hsa-miR-96 to A549. Method A549 were transfected with miR-96 inhibitor/mimics and shRNA-mTOR. The proliferation were detected by MTT,the expression of p21, cyclin D1,p-4E-BP,S6K were detected via RT-PCR and WB. The luciferase assay were used to anaylse the target of miR-96. Result With or without metformin treated,up-regulated the expression of miR-96 could inhibit A549 proliferation,increase p21 expression and decrease cyclin D1 expression instead. The results may related with G1 arrest which induced by miR-96 up-regulation. The ratio of firefly fluorescence value/Renilla fluorescence value was lower than that in wild type NC or mutant group. Conclusion miR-96 target with mTOR to inhibit the growth of A549.

ObjectiveAim at the problems in the treatments of the children malignant bone tumor of distal femur,we put forward the concept of the dual mobility hemi-knee prosthesis and try to perform the motion parameters analysis and the pilot clinical trials.MethodsBase on the CT data from samples of knee joint in adult,we adopted the Mimics/Geomagic/Pro-E software and computer numerical control milling machine technology to design and produce the dual mobility hemi-knee artificial prosthesis,and then motion parameters was analyzed in vitro test,and at last pilot clinical trial was performed.ResultsIn vitro experiment showed that the displacement of the internal femoral condyle flexion facet center was(2.59±0.43) mm in normal knee group,(2.22±0.52) mm in dual mobility hemi-knee prosthesis group,and (1.18±0.43) mm in total knee arthroplasty group; the displacement of the external femoral condyle flexion facet center was (11.95±6.62) mm in normal group,(11.25±6.19) mm in dual mobility hemi-knee prosthesis group,and (1.26±0.42) mm in total knee arthroplasty group; the maximum relative rotation angle was 13.17°±7.58° in normal knee group,11.69°±6.49° in dual mobility hemi-knee prosthesis group,and 5.40°±1.29° in total knee arthroplasty group.After that,we accomplished the pilot clinical trials successfully for a patient with malignant bone tumor in distal femur.The patient recovered well and the treatment outcome was satisfactory.ConclusionThe motion parameters analysis showed that the motion mode of the dual mobility hemi-knee prosthesis is similar with that of the normal knee.The dual mobility hemi-knee prosthesis provides a whole new concept and method for the treatment of malignant bone tumor of distal femur in children,and the device and concept of ligament reattachment provide a solution to the reconstruction of knee ligament.
