Chinese Journal of Pathophysiology 2015;(6):973-979
[ ABSTRACT] AIM:To investigate the effects of long-term aerobic exercise on the heart and sympathetic neural remodeling ( structure and function remodeling) in heart failure rats induced by myocardial infarction.METHODS:Heart failure model after myocardial infarction was performed by ligating anterior descending coronary artery in the Wistar rats. Four weeks after operation, the rats were randomly divided into sham operation sedentary ( S) group, heart failure sedenta-ry ( H) group and heart failure exercise ( HE) group. The animals in HE group underwent 10-week treadmill running, while those in S group and H group were sustained in a resting state. The cardiac structure and function including left ven-tricular internal diameter at diastole ( LVIDd) , left ventricular internal diameter at systole ( LVIDs) , left ventricular anteri-
or wall diameter at diastole (LVAWDd), left ventricular anterior wall diameter at systole (LVAWDs), left ventricular pos-terior wall diameter at diastole ( LVPWDd) and left ventricular posterior wall diameter at systole ( LVPWDs) , and cardiac function parameters including fractional shortening (FS) and left ventricular ejection fraction (LVEF) were measured by echocardiography. The myocardium was collected for histopathological observation with Masson staining, and the collagen volume fraction ( CVF) was determined. The concentrations of norepinephrine ( NE) in the myocardium and plasma were measured by high-pressure liquid chromatography. The frequency domain analysis was applied for determining the heart rate variability (HRV) via subcutaneous recording electrode involving total power (TP), normalized low power (LFn), nor-malized high power ( HFn) and LF/HF ratio. The mRNA expression of collagen type I ( Col-I) , collagen type III ( Col-III) , atrial natriuretic factor ( ANF) ,α-myosin heavy chain (α-MHC) ,β-myosin heavy chain (β-MHC) , sarcoplasmic endoplasmic reticulum Ca2+-ATPase (SERCA2a) was detected by real-time PCR.The protein levels of nerve growth factor (NGF) and its receptor (TrkA), and tyrosine hydroxylase (TH) were measured by Western blotting.RESULTS:(1) Compared with S group, body weight ( BW) , LVIDd, FS, LVEF, TP, HFn, the mRNA expression ofα-MHC and SER-CA2a, and the protein levels of NGF, TrkA and TH decreased (P<0.05).Left ventricular weight (LVW), left ventricu-lar mass index (LVMI), LVAWDd, LVAWDs, LVPWDd, LVPWDs, CVF, plasma and myocardial NE content, LFn, LF/HF, and the mRNA expression of ANF,β-MHC, Col-I and Col-III increased ( P<0.05) in H group.(2) Compared with H group, LVW, LVMI, LVIDd, FS, LVEF, TP, HFn, the mRNA expression ofα-MHC and SERCA2a, and the pro-tein levels of NGF, TrkA and TH were raised (P<0.05), while CVF, plasma and myocardial NE content, LFn, LF/HF, and the mRNA expression of ANF, β-MHC, Col-I and Col-III decreased ( P<0.05 ) in HE group.CONCLUSION:Long-term aerobic exercise training leads to inhibition of heart and sympathetic neural remodeling and improvement of cardi-ac function and autonomic modulation in the rats after myocardial infarction.

Objective: To inquire into the testis gene change of mouse model with kidney-yang deficiency induced by overfatigue and excessive sexual life, the modern science mechanism of syndrome of kidney-yang deficiency and kidney controlling reproduction can be revealed on the gene level. Methods: The model of kidney-yang deficiency was established by each male mouse with six female mice keeping in the same cage, and all the male mice were forced to swim for 30-40 minutes everyday lasting for four weeks. The testis genes of mice in control group and model group were detected with Mouse OneArrayTM Whole Genome DNA microarray. The differentially expressed genes were screened under the condition of the relative fluorescence intensity ratio of the two groups ≥2 and ≤0.5 and further classified according to gene function by using Molecule Annotation System (MAS) created by CapitalBio Corp. Beijing, China. Results: The mouse model with kidney-yang deficiency was established successfully by the method of overfatigue and excessive sexual life. The scatter plot of gene expression profiles of comparing control group with model group was drawn. Differentially expressed genes were screened, including 2425 up-regulated genes and 3080 down-regulated genes. Among the first one hundred up-regulated genes, 41 genes were known and among the first one hundred down-regulated genes, 62 genes were known. These genes were mainly related to the cellular structure/function, material/energy metabolism, signal transduction/transmission, inflammation/immunization, cell apoptosis, transcription/translation,proliferation/differentiation and cell cycle. Conclusion: The testis gene of kidney-yang deficiency mouse had a wide change. The essence of kidney-yang deficiency syndrome and the theory of the kidney originating and controlling life reproduction were explained on the gene level.

Objective :
To investigate the AIDS-related knowledge and behaviors and health service demand among secondary vocational school students in Shanghai Municipality, so as to provide the evidence for formulating the HIV/AIDS health education and behavioral interventions among secondary vocational school students.
Methods :
Three secondary vocational schools were randomly sampled in Shanghai Municipality using a stratified cluster sampling method from September to November in 2019, and grade was stratified in each school. Then, a class was sampled from each grade, and all students in this class were sampled. Participants' demographics, AIDS-related knowledge, sexual behaviors, and health service needs for AIDS control were collected using questionnaires and descriptively analyzed.
Results :
Totally 2 078 questionnaires were allocated, and 2 022 questionnaires were recovered, including 2 013 valid questionnaires (99.55%). The respondents had a median age of 16 years (interquartile range, 2 years), and included 1 366 men (67.86%). A low awareness rate was found for “Self-testing may understand HIV/AIDS infection status” (40.83%) and “Mosquito bites fail to transmit HIV” (37.95%). There were 182 respondents with sexual behaviors (9.04%), including 47 respondents with use of condoms during each sexual behaviors (25.82%), 20 respondents with multiple sexual partner behaviors simultaneously (10.99%), and 21 respondents that had received HIV testing (11.54%). There were 807 respondents with questions about sex and reproductive health (40.09%), and 852 respondents that wished to acquire answers from professional network platforms and professionals (42.32%).
Conclusions
The awareness of HIV/AIDS transmission route and use of condoms is low among secondary vocational students in Shanghai Municipality, and there are high-risk behaviors for HIV/AIDS among students. The secondary vocational school students are most eager to obtain the answers from professional institutions and professionals if there are questions about sex and reproductive health.

BACKGROUND:Bone ultrastructural destruction is an important cause for bone strength decrease, bone friability and bone fracture incidence increase in osteoporosis.OBJECTIVE:To observe the ultrastructural changes of distal femur cancellous bone in glucocorticoid (GC)-treated rats by scanning electron microscopy (SEM).METHODS:A total of 32 female Sprague-Dawley rats, aged 3.5 months old, were respectively treated with methylprednisolone 3.5 mg/kg per day by subcutaneous injection to induce osteoporosis and normal saline. At 4 and 9 weeks, the distal femurs were coronary sectioned and rinsed with distilled water, dehydrated in graded ethanol, coated with gold, and observed by SEM.RESULIS AND CONCLUSION:Compared to the control, the number of bone trabeculae in the GC group was significantly decreased, and the bone trabeculae became thin, fragile, discontinuous;network structures of bone trabeculae were destroyed, and bone resorption surface increased, with disorderly arranged collagenous fibers and increased micro-damage.Rats treated with GC for 4 weeks maintained better bone ultrestructure compared with rats treated with GC for 9 weeks. Results show that GC can induce bone mass lost, destroy network structures of bone trabeculae, accelerate bone resorption in rat cancellous bone, and accordingly lead to increased bone brittleness and decreased bone functional of biodynamics.

Research on novel pullulanase has major significance on the domestic industrialization of pullulanase and the breakdown of foreign monopoly. A thermophilic bacteria LM 18-11 producing thermostable pullulanase was isolated from Lunma hot springs of Yunnan province. It was identified as Anoxybacillus sp. by 16S rDNA phylogenetic analysis. Full-length pullulanase gene was cloned from Anoxybacillus sp. LM18-11. The optimum temperature of the pullulanase was between 55 and 60 degrees C with a half-life as long as 48 h at 60 degrees C; and its optimum pH was between 5.6 and 6.4. V(max) and K(m) of the pullulanase was measured as 750 U/mg and 1.47 mg/mL, which is the highest specific activity reported so far. The pullulanase crystals structure showed a typical alpha-amylase family structure. The N-terminal has a special substrate binding domain. Activity and substrate binding were decreased when the domain was deleted, the V(max) and K(m) were 324 U/mg and 1.95 mg/mL, respectively. The pullulanase was highly heterologous expressed in Bacillus subtilis by P43 promoter. The extracellular enzyme activity was 42 U/mL, which increased more than 40 times compared to the initial strain. This pullulanase has good application prospects.
