1.Effect of positive nursing intervention on quality of life of patients with post-herpetic neuralgia

Objective To discuss the effect of positive nursing intervention on quality of life of patients with post-herpetic neuralgia.Methods 80 cases of patients with post-herpetic neuralgia from January 2011 to June 2012 were chosen and randomly divided into the control group and the treatment group,each with 40 cases.Both groups received neural stem closure therapy,and the treatment group was given positive nursing intervention in addition,including humane care,synchronized implementation of home care guidance by caregivers,hope nursing and telephone interviews.NRS and SF-36 were used to evaluate the curative effect and quality of life,and indexes were compared between two groups.Results The excellence rates 30 and 60 days after treatment were 100% in the treatment group,obviously higher than 85.0% in the control group,and the differences were statistically significant (x2 =5.1,7.6,respectively; P < 0.05).There was no statistically significant difference of SF-26 between two groups before treatment (P > 0.05).30 days after treatment,the score of physical function,body pain and general health was (56 ± 22.5),(79 ± 11.9),(73 ± 17.8) in the treatment group,higher than (44 ± 16.7),(63 ± 21.2),(59 ± 16.7) in the control group,and the differences were statistically significant (t =2.71,4.16,3.63,respectively; P < 0.05).60 days after treatment,the score of physical function,body pain and general health was (68 ± 21.7),(82 ± 14.8),(79 ± 18) in the treatment group,higher than (46.6 ± 14.3),(70 ±22.3),(66 ± 18.2) in the control group,and the differences were statistically significant (t =5.21,2.84,3.21,respectively; P < 0.05).Conclusions Positive nursing interventions can significantly improve the curative effect and quality of life of patients with post-herpetic neuralgia,which can also improve the quality of medical care obviously.
2.Spatio-temporal analysis of the incidence of colorectal cancer in Guangzhou,2010–2014

Introduction: Colorectal cancer (CRC) is a common type of neoplasm. This study examined the spatio-temporal distribution of the CRC incidence in Guangzhou during 2010–2014. Methods: Colorectal cancer incidence data were obtained from the Guangzhou Cancer Registry System. Spatial autocorrelation analysis and a retrospective spatio-temporal scan were used to assess the spatio-temporal cluster distribution of CRC cases. Results: A total of 14,618 CRC cases were registered in Guangzhou during 2010–2014, with a crude incidence of 35.56/100,000 and an age-standardized rate of incidence by the world standard population (ASRIW) of 23.58/100,000. The crude incidence increased by 19.70% from 2010 (32.88/100,000) to 2014 (39.36/100,000) with an average annual percentage change (AAPC) of 4.33%. The AAPC of ASRIW was not statistically significant. The spatial autocorrelation analysis revealed a CRC incidence hot spot in central urban areas in Guangzhou City, which included 25 streets in southwestern Baiyun District, northwestern Haizhu District, and the border region between Liwan and Yuexiu Dis-tricts. Three high- and five low-incidence clusters were identified according to spatio-temporal scan of CRC incidence clusters. The high-incidence clusters were located in central urban areas including the border regions between Bai-yun, Haizhu, Liwan, and Yuexiu Districts. Conclusions: This study revealed the spatio-temporal cluster pattern of the incidence of CRC in Guangzhou. This information can inform allocation of health resources for CRC screening.
4.Evaluation by damped least-square algorithm on target identification ability of intracranial focal bidirectional electrical impedance perturbation

Objective To evaluate the effect of damped least-square algorithm on the identification of focal bidirectional electrical impedance perturbation in the intracranial area, and to analyze the influence of this kind of perturbation on the reconstruction quality of electrical impedance tomography. Methods Focal bidirectional electrical impedance perturbation was built based on the three-dimensional model and damped least-square algorithm was introduced into imaging. The position error and resolution were used to evaluate the imaging performance.Results When the focal electrical impedance perturbation existed,the target whose conductivity varied greatly or volume was large was more likely to be identified in the images while the target with smaller volume or variable conductivity presented in the reconstruction image was not obvious. It's pointed out that it may cause reconstruction images in some cases could not truly reflect the location and change information of the object of primary cerebral hemorrhage.At the same time,it affected the reconstruction precision,causing the position error and resolution with large fluctuation. Conclusion In reconstruction algorithm linearity correct matrix introduced for bidirectional electrical impedance perturbation contributes to enhancing the recognition of bidirectional perturbation, so that the characterization of the electrical impedance imaging can be augmented for clinical intracerebral hemorrhage.
5.Comparison on local dengue fever outbreaks in public place in Shenzhen and Hong Kong in 2018

Objective To compare epidemiology characteristics and control measures of dengue fever (DF) outbreaks in public places in Shenzhen and Hong Kong and integrate the advantages for better response to DF outbreaks. Methods Data of DF cases and mosquito vector surveillance in Shenzhen and Hong Kong were collected for descriptive epidemiological analysis and measures of mosquito control were compared. Results A total of 19DF cases were recorded in 13 d among six districts of Hong Kong, the male: female ratio was 1 ∶〗0.9. In Shenzhen, 20 cases were recorded in 33 d, the cases were distributed in three sub-districts, the Male: female ratio was 1 ∶〗0.82; Age difference between Shenzhen and Hong Kong was statistically significant (Z=-2.855, P=0.004). The time interval between date of onset and laboratory confirmation ranging from 5.5 to 8.5 d (median:7d) in Hong Kong, and ranged from 1.5 to 5.4 d (median:2 d) in Shenzhen, the difference was statistically significant (Z=-3.818, P<0.001). On the 4th day of the outbreak, Hong Kong declared 30 d closure for anti-mosquito operations with a range of 500 m, and monthly area ovitrap index (AOI) was used to monitor the territory-wide situation of Aedes albopictus. On the 2nd day of the outbreak, Shenzhen declared source closure for mosquito control with a range of 400 m and emergency vector surveillance was conducted, using Breteau Index (BI) and Human-baited double net trap(HDN) every 3 d. Conclusions The nature, scale, time and place of the two local DF outbreaks in Shenzhen and Hong Kong are similar. The prevention and control system is more sensitive and prompt, the ability of early detection is stronger, mosquito vector surveillance program is more comprehensive and the frequency is higher in Shenzhen. The epidemic information dissemination and risk communication with media is more active, mosquito management and control is more professional in Hong Kong.
6.Induction of bladder lesion by terephthalic acid and its mechanism.

OBJECTIVETo provide more information for rational evaluation of potential risks of terephthalic acid (TPA), we studied the effects of TPA on rats' bladders in 90 days after TPA exposure.
METHODSSprague Dawley rats were subdivided into five groups, ingesting 0%, 0.04%, 0.2%, 1%, and 5% TPA respectively for a sub-chronic feeding study lasting for 90 days. Urine, serum and samples of brain, liver, lung, kidney, bladder, etc. were collected and analyzed.
RESULTSTPA ingesting decreased the value of urinary pH, and increased the contents of Ca2+, Zn2+, Mg2+, Na+, K+ in urine. The volume of 24 h urine was significantly increased in male rats in the 1% and 5% TPA groups. Urinary white sediment was found in both sexes, and its formation in male rats seemed more susceptible than that in female rats. Alpha 2u-globulin (AUG) in serum and urine of male rats was markedly increased in a dose-dependent manner. Fifteen cases of hyperplasia (simple or atypical) were determined in the 5% TPA ingesting group, 14/52 in male rats and 1/23 in female rats. Among them 3 male rats had no stone or calculus. Those with either bladder stones or hyperplasia were accompanied with urinary white sediments.
CONCLUSIONWhite sediment accompanied with elevated urine AUG is the basis of TPA induced urolith formation, and is also associated with TPA induced bladder epithelial cell proliferation. It can act as an early biomarker for the potential toxic effect of TPA.

7.Construction and identification of a eukaryotic expression vector containing human homeobox gene.

In order to investigate the special role of HOXB4 in expansion and self renewal of hematopoietic stem cells, the cDNA of HOXB4 was extracted and cloned from umbilical cord blood mononuclear cells by using RT-PCR. Then the eukaryotic expression bicistronic plasmid vector pIRES2-EGFP/HOXB4 was designed and constructed after cutting HOXB4 and pIRES2-EGFP respectively by restriction enzyme EcoRI and BamHI. The recombinant plasmid was delivered into competent cells of Escherichia coli. The successful construction of plasmid was confirmed by the identification of endonuclease cutting and sequencing. The results showed that the HOXB4 cDNA was cloned successfully from umbilical cord blood mononuclear cells and the recombinant eukaryotic expression bicistronic plasmid vector was constructed, and then introduced it into 293T cells successfully. It is concluded that a pIRES2-EGFP/HoxB1 eukaryotic expression bicistronic plasmid vector has been constructed successfully, which results provide a useful material basis for exploration of HoxB4 function in the proliferation and differentiation of hematopoietic cells.

8.Effects of Flavonoids of Rhizoma Drynariae on Formation of Blood Vessels in Induced Membrane by Masquelet Technique

Objective To explore the effects of flavonoids of Rhizoma Drynariae on the formation of blood vessels in the induced membrane by Masquelet technique. Methods Seventy-two SD rats were randomly divided into 4 groups,namely model group,and high-,middle-and low-dose drug groups,18 rats in each group. Rat model of critical- sized femoral defect was built,and then polymethyl methacrylate (PMMA)bone cement spacer was inserted into the bone defect to induce the formation of membrane. From the first day after surgery , the rats in high-,middle-and low-dose drug groups were given gastric gavage of 0.44,0.22,0.11 g·kg-1·d-1 of Rhizoma Drynariae flavonoids, respectively, and the rats in the model group were given the same volume of normal saline. After 6-week medication,the pathologic features of bone cement- induced membrane were observed by Haematoxilin-Eosin(HE)staining,the contents of transforming growth factor(TGF)-β1 and vascular endothelial growth factor(VEGF)proteins in the induced membrane were tested by enzyme-linked immunosorbent assay (ELISA),and the mRNA levels of TGF-β1 and VEGF in the induced membrane were determined by real-time reverse transcription-quantitative polymerase chain reaction (RT-qPCR). Results More blood vessels in the induced membrane of the high-dose group were found than those of the other groups under the light microscope. The protein and mRNA expression levels of TGF-β1 and VEGF in the induced membrane of the 3 drug groups were much higher than those of the model group(P < 0.05). Except for the VEGF mRNA expression level, the changes of other indexes were dose-dependent. Conclusion Flavonoids of Rhizoma Drynariae are effective on enhancing the protein and mRNA expression levels of TGF-β1 and VEGF in the induced membrane, and can accelerate the vascularization,which promotes the reconstruction of bone defect.
9.Clinical study on mometasone furoate nasal spray in the treatment of non-allergic rhinitis

Objective To evaluate the outcome of mometasone furoate nasal spray (MFNS) used for 3 months on non-allergic rhinitis (NAR). Methods In this multicenter study, NAR patients were enrolled from eight hospitals and received MFNS 200 microgram once daily for 3 months. The patients were followed-up for three times (at baseline, month 1 and month 3) to record the symptom scores and nasal endoscopic appearances. At the same time, the adverse events frequency was recorded and analyzed.Results A total of 188 NAR cases were enrolled in the study. The total nasal symptom score assessment descended significantly at month 1 (1.70 ± 0. 75) and month 3 (0. 95 ± 0. 79) visits versus at baseline (2. 67 ± 0. 68, Z value were from - 11. 603 to - 10. 491, all P < 0. 01). The individual symptoms,including nasal stuffiness, nasal discharge, nasal stuffiness-related dizziness or headache, hyposmia, sleep quality, daily life activity, work or study efficiency, mental status, and whole body fatigue, also showed less scores at month 1 and month 3 visits versus at baseline (Z value were from - 11. 313 to - 6. 802, all P <0. 01). At the same time, nasal mucosal appearances assessed by endoscopy had lower scores at month 1 (1.40±0. 62) and month 3 (0. 75 ± 0. 71) visits versus at baseline (2. 27 ± 0. 73, Z value were from - 11. 484 to - 10. 002, all P <0. 01). Additionally, adverse events were only observed in 5. 3% cases with light rhinorrhagia and nasal dryness. No other side effect was found. Conclusions A 3-months administration of intranasal mometasone can effectively and safely improve NAR patients' clinical symptom and nasal mucosal appearances.
10.Construction and screening of natural immune Fab antibody phage library from patients with colorectal cancer

Objective To construct natural immune Fab fragment phage display library and to screen the antibodies against human colorectal cancer. Methods Total RNA in the metastatic lymph nodes from patients with colorectal cancer was extracted routinely, and reverse transcriptase PCR employed to amplify the heavy chain Fd and light chain κ cDNA. The amplification products were inserted in succession into the vector pComb3 to construct human Fab fragment library, which was screened and identified using phage display technique. Results The heavy chain Fd fragment and light chain κ fragment (650 bp in length) were amplified, and the products were inserted into pComb3 vector with a recombination rate of 40%. The constructed phage display Fab library had a capacity of up to 1.48×106, and was enriched approximately 120 times after 3 cycles of panning. Dot immunoblotting found the expression of Fab fragment which could bind to the antigens of human colorectal cancer. Conclusion The phage display library of the antibody Fab fragment from naturally immunized lymph nodes is successfully established, rendering it possible to screen the antibodies against human colorectal cancer.
