RORγt, an immune cell-specific isoform of RORγ( retinoic acid receptor-related orphan nuclear receptor gam-ma) , is a key transcription factor for the development of Th 17 cells both in human and mouse .RORγt is required for the induction of IL-17 transcription and for the manifestation of Th 17-dependent autoimmune diseases in mice .RORγt natural ligands are retinoic acid . RORγt is closely implicated in the pathology of numerous autoimmune diseases , infectious diseases and cancer .With the further re-searches about the role of RORγt, we will clarify the mechanism of RORγt in autoimmune diseases , which will provide new ideas to di-agnose and treat autoimmune diseases .

[Objective]To assess the protective effects of Xianlinggubao on articular cartilage and subchondral bone in experimental osteoarthritis induced by anterior cruciate ligament transaction (ACLT)in rabbits. [Methods]Thirty healthy Japanese white rabbits were divided randomly into three groups:sham group,ACLT+XLGB group and ACLT+ NS group. Rabbits in ACLT+XLGB group and ACLT+NS group received anterior cruciate ligament transection(ACLT)on the right knee. Rabbits in sham group received untreatment as control. ACLT+XLGB group received a daily administration of XLGB at a dose of 125 mg/kg of body weight for 6 weeks.In contrast,ACLT+NS group received saline at the same dose. All rabbits were sacrificed at 6 weeks after operation. The right femora were removed,measured for bone mineral density (BMD),and then were observed for the macro-pathologic changes with HE staining and graded by Mankin's scale. Expression levels of BMP-2 and MMP-13 were detected by immunostaining. ObjectiveTo investigate the incidence and risk factors of antibiotic-associated diarrhea (AAD) of pediatric patients with severe bacterial pneumonia.MethodsClinical data of 1086 pediatric patients with severe bacterial pneumonia from January 2010 through January 2014 were recruited. The incidence and risk factors of AAD were retrospectively analyzed. ResultsThe incidence of AAD in 1086 pediatric patients with severe bacterial pneumonia was 36.74%. The incidence of AAD in patients younger than 2 years old were higher than that in those older than 2 years, once or more times of mechanical venti-lation history were higher than that with no arrangements of this treatment, administering of combined antibiotics therapy were higher than that with single antibiotics, and the incidence of AAD due to amoxicillin/clavulanate, piperacillin/tazobactam, cefo- perazone/sulbactam in pediatric patients were 43.55%, 43.75%, and 45.03%, respectively. Three β-lactam/β-lactamase inhibitors above were risk factors of AAD through multivariate Logistic regression analysis.ConclusionThe high incidence of AAD in pediatric patients with severe bacterial pneumonia was associated with some risk factors, including younger than 2 years old, me-chanical ventilation, combined antibiotics therapy and administration of β-lactam/β-lactamase inhibitor (amoxicillin/clavulanate, piperacillin/tazobactam, cefoperazone/sulbactam).

Objective To discuss the relationship between peptic ulcer in children and food-specific IgG, and to evaluate the effect of avoiding the intolerant food on treatment of peptic ulcer. Method Fourteen kinds of food specific IgG in 69 cases of peptic ulcer were tested by ELISA. The patient′s intolerance to different foods and its relationship with age , sex and different ulcer were analyzed. The efficacy of avoiding the intolerant food was assessed after 8 weeks. Results Total positive of food-specific IgG in children with peptic ulcer was 94.2%. For preschool children, the highest intolerance rates were in milk and eggs; for school-age and adolescence children, the highest intolerance rate was in eggs. As the age increases, kinds of intolerant food increased too. Ulcer healing rate in observation group was higher than that in the control group after 8 weeks. Conclusions High positive percentage of food- specific IgG was found in children with peptic ulcer , indicating a possible correlation of peptic ulcer and food-intolerance. Kinds of intolerant food correlates with the age group , but not with sex and ulcer sites. Avoiding the intolerant food may increase ulcer healing rate.

This study is to investigate the inhibitory effect of lidamycin (LDM) and its combination with methotrexate (MTX) on lung metastasis of fibrosarcoma by bioluminescence imaging in athymic mice. A stable luciferase transfected HT-1080 cell line was constructed and the capability to establish experimental lung metastasis in athymic mice was confirmed. The optical imaging system was applied to evaluate the formation of lung metastasis in vivo. In addition, metastatic nodules were counted for the evaluation of inhibition rates. As shown, the fluorescent intensity of luciferase-transfected HT-1080 cells was colinear with the cell population and the minimal detected cell population was 100 cells/well. Optical imaging showed that the fluorescent intensity of treated group was apparently lower than that of the control. The inhibition rates of lung metastasis by LDM alone at 0.025 mg x kg(-1) and 0.05 mg x kg(-1) were 53.9% and 75.9%, respectively, while that of MTX alone at 0.5 mg x kg(-1) was 70.2%. The combination of LDM at 0.025 mg x kg(-1) and MTX at 0.5 mg x kg(-1) showed an inhibition rate of 88.7%. The coefficient of drug interaction (CDI) was 0.82. The results herein demonstrated that LDM alone had strong anti-metastasis effect on human fibrosarcoma HT-1080 and the inhibition efficacy is strengthened when combined with MTX.

Objective:A novel tuberculosis DNA vaccine integrating siRNA,fusional antigen and hIL-12 triple expression units was constructed in our laboratory.Methods:To evaluate the independent expression of the three expression units,two eukaryotic expression plasmid pVAX1-siRNA-PVAE[EGFP]-hIL12 with TB fusional gene Ag85B-ESAT6(PVAE) and enhanced green fluorescent protein(EGFP),and pVAX1-siEGFP-PVAE[EGFP]-hIL12 with siRNA coding sequence targeting EGFP instead of Mcl-1L were constructed.Then two plasmids were used to transfer human embryonic kidney 293 cells.Based on EGFP report gene,it was demonstrated that siRNA expression unit and fusional antigen gene were independently expressed.Results:The hIL12 expression at 48h and 72h post transfection were also detected by ELISA analysis up to 1571.63pg/ml and 2392.25pg/ml respectively in the cell culture fluid.Conclusion:The results demonstrated that the novel DNA vaccine with siRNA,TB fusional antigen and hIL12 three expression units in the same plasmid frame is successfully constructed and independently expressed in eukaryotic cells.It laid a foundation for further animal model study on anti-tuberculosis effects of this novel DNA vaccine.
