1.Quantification of vascularization within spherical porous ?-TCP scaffold in vivo

[Objective]To explore the vascularization within spherical porous ?-TCP scaffolds in vivo.[Method]Thirty adult rabbits were selected and divided into 5 groups randomly.Spherical porous ?-TCP scaffolds(the diameter was 2 cm,the pore size was 500~600?m,the interconnection size was 110~120 ?m) were harvested at 1,2,4,8 and 12 weeks after embedded into muscle-fascia lumbodorsalis pouches in each rabbit separately,in order to observe the vascularization by means of morphological and quantificational analysis.[Result]No vessel was detected at one week after surgery,and only a few immature vascular buds could be seen at the 2nd week.The first vascularization peak could be observed at 4 weeks,characterized by the growth of quantity of new blood vessels(P0.05).[Conclusion]The process of vascularization comprises two phases,the quantity increasing in the early stage and the quality enhancing in the later stage.The special architecture and biodegradation characters of scaffolds could influence the vascularization.
2.Analysis on urban-rural differences of reference factors for the identification of students from poor families in university

ObjectiveTo know urban-rural differences of reference factors for the identification of students from poor families in order to provide references for identifying index system.Methods Questionnaires were used among 708 students from poor families.ResultsThere were 78.9% urban and 89.1% rural families whose average earnings were lower than 410 RMB per month.The number of majority urban and rural family (58.8%,75.4% ) was 4 - 6 ; 77.0% ( 86.3% ) urban and 97.3% (97.9%) rural fathers' ( mothers' ) education levels were high school or below; 8.6% ( 23.1% ) urban and 82.3 % ( 85.1% ) rural fathers ( mothers ) were farmers.The proportion of students having computers was 87.2% (urban) and 7 1.9 % (rural) ; 54.9% urban and 63.9% rural students' life was not frugal; 85.2% urban and 79.1% rural students thought campus cards shouldn't be monitored.ConclusionsThere are significant urban-rural differences of reference factors for the idenffication and family residence is the key indicator which should have higher weights.
3.Effect of Flos Daturae Alkaloids on TGF-beta1-induced Epithelial-Mesenchymal Transition of Human Pulmonary Adenocarcinoma A549 Cells.

OBJECTIVETo study the effect of Flos Daturae alkaloids (FDA) on TGF-beta1-1uuuu;U epithelial-mesenchymal transition (EMT) of human pulmonary adenocarcinoma A549 cells.
METHODSA549 cells in vitro cultured were randomly divided into 5 groups, i.e., the blank control group, the TGF-beta1 group, the low dose FDA group, the medium dose FDA group, and the high dose FDA group. The morphologies of A549 cells were observed. Expressions of cytokeratin (CK)-19 and alpha-smooth muscle actin (alpha-SMA) were detected by Western blot and real-time PCR at 24, 48, and 72 h, respectively.
RESULTSA549 cells in the TGF-beta1, group turned from cobblestone to spindle shape gradually. Those in low, medium and high dose FDA groups showed similar shapes to those of the TGF-beta1 group. There was no statistical difference in the morphology of A549 cells among the 3 dose FDA groups (P > 0.05). Western blot showed that, when compared with the blank control group, the expression of CK-19 was down-regulated, but the expression of alpha-SMA was up-regulated in the TGF-beta1 group (P < 0.01). Compared with the TGF-beta1, group, the expression of CK-19 was up-regulated, but the expression of alpha-SMA was suppressed in low, medium and high dose FDA groups (P < 0.01). The CK-19 expression obviously increased, but the alpha-SMA expression was suppressed in high dose FDA group at 72 h (P < 0.01). Real-time PCR results showed, as compared with the TGF-beta1 group, the mRNA expression of CK-19 was increased, but the mRNA expression of alpha-SMA was reduced in low, medium and high dose FDA groups (P < 0.01).
CONCLUSIONSFDA had no effect on EMT morphological changes of TGF-beta1 induced A549 cells. FDA could reverse characteristic markers of A549 cells during EMT to some extent, such as expressions of CK-19 and alpha-SMA. The expression of CK-19 (as the epithelium marker) increased and the expression of alpha-SMA (as the mesenchymal marker) was reduced. Besides, they were most obviously seen in the high dose FDA group at 72 h in a dose- and time-dependent manner.

4.Analysis on reference factors for the identification of students from poor families in the university

Objective To know the problems of reference factors,and then provide evidences for scientific and specific identifying index system for students from poor families.Methods Questionnaires were used among 719 students from poor families.Results There were 86.0% families whose earnings were lower than 410 RMB per month.57.2% families depended on agriculture.70.1% students were from the rural areas.Family member number was 4~6 for the majority ( 70.7% ).91.2% ( 94.5% ) fathers'( mothers' ) education levels were under high middle school.64.0% ( 67.3% )fathers ( mothers ) were farmers.The proportion of students having computers was 76.1%.61.2% students' life was not frugal.There were 80.9% students who thought campus cards shouldn't be monitored.Conclusion Family economic situation should be the key indicator.Most of the basic family information could be reference factors.Basic situation of students at school was inappropriate to be reference factors.
5.Interleukin-17 expression in the injured site of a rat model of complete spinal cord transection

BACKGROUND:Intervention using known inflammatory transmitters has limitations on relieving secondary spinal cord injury. Interleukin-17 is an important proinflammatory cytokine, and is gradual y paid attention in the pathogenesis of central nervous system diseases.
OBJECTIVE:To investigate the altered rule of interleukin-17 mRNA and protein in a rat model of acute spinal cord injury.
METHODS:Healthy male Sprague-Dawley rats were randomly assigned to two groups. In the model group, rats were made into complete spinal cord transaction models. In the sham surgery group, only spinal dura mater was opened, but parenchyma was not injured. Basso, Beattie, Bresnahan locomotor rating scale was used to observe the effects of acute spinal cord injury on limb motor function of rats. Hematoxylin-eosin staining was used to observe histopathological changes at various time points after spinal cord injury. Real-time fluorescence quantitative PCR and western blotting were used to detect interleukin-17 mRNA and protein levels in each group at various time points after spinal cord injury.
RESULTS AND CONCLUSION:Basso, Beattie, Bresnahan locomotor rating scale:Basso, Beattie, Bresnahan scores were 20 to 21 in the sham surgery group. Basso, Beattie, Bresnahan scores were 0 at 1 and 2 days after spinal cord injury. At 7 days, Basso, Beattie, Bresnahan scores were 0 to 3 (P<0.05). Hematoxylin-eosin staining results revealed that compared with the sham surgery group, inflammatory cel infiltration, neuronal and glial cel swel ing, and a reduced number of neuronal processes were observed at 6 hours after spinal cord injury. Gray matter and white matter were loose and vacuolated at 12 hours. Gliocyte proliferation and tissue fibrosis were apparent at 7 days. Real-time PCR results demonstrated that interleukin-17 mRNA appeared at 3 hours, and peaked at 6 hours (P<0.01), and then decreased. Interleukin-17 mRNA levels were similar to the sham surgery group at 7 days. Western blotting results revealed that interleukin-17 expression began to increase at 6 hours and peaked at 12 hours (P<0.05), and then reduced, and reached the levels in the sham surgery group at 7 days. Results indicated that tissue injury was most severe at 12 hours, and showed a time consistency with interleukin-17 expression. It is inferred that interleukin-17 is possibly involved in the process of secondary inflammatory reaction of spinal cord.
6.Change in expression of interleukin-17 in C57 mice′s spinal cord injury area

Objective To investigate the mechanism of expression of interlenkin (IL)-17 in C57 mice′s spinal cord clamp area,and to provide new targets for clinical treatment of spinal cord injury (SCI). Methods Male C57BL/6 mice were randomly divided into three groups. In the spinal cord injury group,mice were made into spinal cord clamp model. In the sham surgery group, the dura was cut without injuring the spinal cord. The IL-17 neutralizing antibody group received IL-17 neutralizing antibody injection through the cadual vein at 1 hour after the spinal cord clamp . Mouse scale for locomotion (BMS)was applied to evaluate the mice's behavior change of hindlimb in 1-7 days,the real time fluorescent quantitative PCR was used to detect the change in the expression of spinal cord injury district TNF-αmRNA each time,HE staining was conducted to detect the morphological changes of spinal cord injury of the sham surgery group,the spinal cord injury group and the IL-17 neutralizing antibody group at the 7th days. Results After spinal cord injury,the mice's BMS score were 9 in the sham surgery group;in the model of spinal cord injury group,the mice's BMS score were 0 on the 1st day,and 2.9 on the 7th day. In the IL-17 neutralizing antibody group,the mice's BMS score were 0 on the 1st day,and 3.5 on the 7th day. The expression of IL-17 mRNA in the injury area peaked at the 3rd hour,which showed statistical difference when compared with sham surgery group (P<0.05),and then decreased. In other times,it had no statistical difference when compared with sham surgery group (P>0.05),and the expression of IL-17 mRNA reduced the lowest levels on the 7th day. The 7th day following spinal cord injury,mice's spinal cord tissue was complete normal in the sham surgery group. In the spinal cord injury group,a large number of mice's nerve cells were necrotic, a lot of cells formed vacuolated. In the IL-17 neutralizing antibody group, part of mice's nuclear neurons were shrinking, cells formed vacuolated, but part of cells remained morphologically complete. Conclusion IL-17 is involved in secondary immune inflammatory process of spinal cord injury, it may be targets for intervention in the treatment of spinal cord injury.
7.Clinical observation of corneoscleral separation for persistent corneal ulcer

AIM: To evaluate the efficacy and safety of corneoscleral separation in the treatment of persistent corneal ulcer.METHODS: Collected 22 cases (22 eyes) with deep corneal ulcer were treated with drugs.These patients underwent corneoscleral separation or corneal interstitial filling of sterile gelatin sponge,by retrospectively observing the cornea ulcer healing,anterior chamber reaction and separation of new blood vessels grow between the layers,and so on,the surgical indications,surgical techniques,postoperative complications and treatment were evaluated.RESULTS: Patients were followed up 3 to 6mo,of which 18 eyes cornea were seen a small amount of new blood vessels,anterior chamber empyema after 7-9d.Ulcers gradually narrowed,infection controlled,the surrounding edema relieved,visual acuity improved by varying degrees after 12-15d.Postoperative 4 eyes for the mixed infection,2 of the 4 eyes were of the rapid progress,neovascularization did not grow into the lesion,ulcers on the verge of perforation and then done penetrating keratoplasty.CONCLUSION: The aim of this study is to introduce deep limbal vessels into the surrounding of ulcer foci,which to combine with drug therapy is one of the effective methods to treat deep corneal infection.
9.The prognostic and predictive value of plasma D-dimer in children with neuroblastoma: a 7-year retrospective analysis at a single institution

Purpose:
Elevated plasma D-dimer level is a poor prognostic factor for many solid tumors. However, limited research has been conducted on D-dimer in children with neuroblastoma (NB), and its clinical significance remains unclear. The present study investigated the clinical and prognostic significance of D-dimer in pediatric NB patients.
Methods:
A retrospective analysis of all newly admitted NB patients was conducted from January 2014 to December 2020.Baseline clinicopathological features, preoperative laboratory parameters, and follow-up information were collected. Univariate and multivariate analyses were performed to determine the relationship between D-dimer level, clinical features, and the prognostic value.
Results:
Among 266 patients, the median value of D-dimer was 2.98 ng/mL, of which 132 patients showed elevated D-dimer levels before surgery (>2.98 ng/mL). Univariate analysis revealed that elevated D-dimer was significantly associated with age, hemoglobin, neutrophil-to-lymphocyte ratio, neuron-specific enolase, 24-hour vanillylmandelic acid, overall survival, and so on (P < 0.05). Patients with elevated D-dimer levels had shorter median overall survival time when compared with normal D-dimer levels (P = 0.01). The prognosis was better in patients with normal D-dimer levels when combined with lower age, ganglioneuroblastoma tumor type, lower stage on International Neuroblastoma Staging System, low-risk group, and without bone metastasis or bone marrow metastasis. The continuous increase of D-dimer level after treatment indicated tumor recurrence or progression.
Conclusion
A high D-dimer level is associated with low overall survival, and an elevated D-dimer level after treatment indicates tumor recurrence and progression. D-dimer can be used as one of the evaluation factors for NB treatment or prognosis.
10.Reconstruction of pulmonary artery by autologous pericardium or azygous venae for non-small cell lung cancer

Objective To review the clinical experience of reconstruction of pulmonary artery(PA) by a pateh of autologus pericardium or azygous venae for non-small cell lung cancer. Methods Between March 1992 and August 2009, 62 patients with locally advanced central lung cancer received sleeve resection and reconstruction of PA. According to PTNM classification, 4 patients were in stage Ⅱb, 46 in stage Ⅲa and 12 in stage Ⅲb. 17 patients had induction chemothoerapy. Sleeve lotrate the PA, the surgical procedures included partial PA tangential resections and reconstructions by a pateh of autologous azygous venas in 18 cases , a patch of autologous pericardium in 38 cases and a complete PA sleeve resection reconstructios by a custom-made autologous pericardial conduit interposition in 6 cases. Partial superior vena cava tangential resctions and reconstructions were performed in 5 patients by a patch of autologous pericardium or azygous venae. 47 patients received pstoperative chemotherapy and 19 had radiotherapy. Results There was 2 early postoperative deaths(3.2%). The cause of death was bronchial anastomotic leak led to respiratory failure in 1 case and severe arrhmia led to heart arrest in 1. No cancerous tissue of all resection margins are checked by frzen section histology and examination of resection specimens in the surgical pathology laboratory. The postoperative complications occurred in 11 patients(17.7%) and all of them recovered uneventfully.Roentgenography, flexible bronchoscopy and echocardiography were in normal range in the remaining 60 patients with no bronchial anastomosis stenosis or vascular thombosis before discharge and at 2-6 months after surgery. The mean follow up time was 49.5 months (6-210 months). The overall 1, 3, 5 and 10 yerr survival rates were 80.2%, 44.7% ,31.4% and 23. 1%, respectively. Conclusion Reonstruction of PA by autologous pericardial patch or autologous azygos vein patch is a safe and effective technique for locally advanced lung cancer.For extended circumferential defects of PA,the autologous pericardial conduit interposition could bue used for reconstruction.
