1.Effect and mechanism of Imatinib mesilate on intimal hyperplasia of rabbit carotid artery after ballon injury

Objective To investigate the effect and mechanism of Imatinib mesilate (Imatinib) on intimal hyperplasia of rabbit carotid arteries after balloon injury. Methods Thirty adult Newzealand rabbits were randomly divided into three groups:group A, B and C. Their right carotid arteries were injuried then administered with 0, 25 or 50 mg/kg of Imatinib dai?ly for 14 consecutive days when the rabbits were sacrificed. The carotid arteries were harvested and sectioned for HE-stain?ing and immunohistochemisty staining. Real-Time PCR was used to examine transcription levels of PDGF-B and PDGFR-βmRNA. The plasma level of PDGF-BB was assayed by ELISA. Results Arterial intimal hyperplasia and stenosis following balloon injury were seen in three groups. Thickness and area of neointima, ratio of thickness of intima to media, ratio of area of intima to media and mRNA level of PDGF-β are all higher in group A than those in group B than those in group C (P<0.01). By contrast, the mRNA transcription level of PDGFR-β increased significantly in group C than that in group A (1.236±0.356 vs 0.708±0.372;t=2.91;P<0.01). Plasma level of PDGF-BB increased in all three groups after balloon injury than that in the baseline (P<0.01). The transcription level of PDGF-BB is higher in group A than that in group B and in group C (ng/L:23.464±3.542, 19.504±2.454, 16.588±1.207, F=17.322, P<0.05). There was no difference between group B and C. There was positive correlation between mRNA transcription level of PDGF-B and plasma level of PDGF-BB ( r=0.806, P<0.01). Conclusion Vascular injury can cause intimal hyperplasia and increased PDGF-B mRNA transcription. Imatinib mesilate could inhibit the intimal hyperplasia through down regulating PDGF-B mRNA transcription.

4.Ultrasound assisted the enzymolysis of ginsenosides to prepare pare ginseng saponin Compound K.

To prepare ginseng saponin Compound K with ultrasound-assisted total zymolytic ginseng saponins. The conversion rate was taken as the index to detect the pre-treatment factors such as ultrasonic power and ultrasonic time, as well as the impact of enzymatic factors, such as pH value, temperature, concentration of substrate, dosage of enzyme and reaction time, on the conversion rate. The response surface method was used to optimize the preparation conditions. The enzymolytic products were identified with MS, 1H-NMR and 13C-NMR. The results showed that the optimum conditions of the ultrasound-assisted enzymolysis were 250 W for ultrasonic power, 15 min for ultrasonic time, 5.5 for enzymolytic pH, 50 degrees C for enzymolytic temperature, 36 h for enzymolytic time, 4:5 for enzymolytic dosage: substrate and 1.0 g x L(-1) for concentration of substrate. The relative molecular mass of reaction products was 622.4. Therefore, the nuclear magnetic map verified that the reaction product was rare ginseng saponin Compound K. Under the above conditions, based on the total zymolytic ginseng saponins, the conversion rate of rare ginseng saponin Compound K was 6.91% in proportion to the total of ginsenosides. The process features gentle reaction conditions, high conversion rate and simple and reliable process, which is suitable for industrial production.

5.Single nucleotide polymorphisms, inflammation and nutrition of genes.

Inflammation is part of the immune response, and inflammation may also induce or exaggerate some diseases through production of pro-inflammatory cytokines. More evidence have shown that the individual level of cytokine production is affected by single nucleotide polymorphisms in cytokine genes. Furthermore, as several nutrients participate in DNA protection and stabilization, altering gene expression and individual phenotype, nutrition has important interaction with inflammation. The purpose of this review is to give a recent update informations on the interaction of single nucleotide polymorphisms, inflammation and nutrition.

6.Expressions of Let-7 in Normal Breast Tissue and Breast Cancer Tissue

Objective To study the expression characteristics of Let-7 genes in breast cancer.Methods Twenty-eight patients with breast cancer were randomly selected,and their cancer tissue and adjacent normal tissue were collected.TRIzol was used to extract the total RNA and real-time quantitative PCR was used to detect the relative gene expression levels.Results The expression of precursor Let-7 in cancer tissue was (9.65 ± 2.31),which was lower than that in normal tissue (10.05 ± 2.81),P =0.048.Precursor Let-7 had dependence relationship with the long menstruation (b =0.816,P =0.029).The menarche age showed a positive correlation with precursor Let-7 in normal tissue (r =0.502,P =0.048) and a negative correlation in cancer tissue (r =-0.484,P =0.049) Conclusions The expression of precursor Let-7 in cancer tissue is lower than that in normal tissue.The period of menstruation is a protective factor to breast cancer.
7.Study on Syndrome Distribution and Chinese Medicine Composition of AECOPD Based on Complex Networks

Based on clinical research and information-sharing system, this article was aimed to study both the syndrome distribution and Chinese medicine composition of acute exacerbation of chronic obstructive pulmonary disease (AECOPD) in the COPD base of our hospital using the complex networks. The analysis was made on the common rules of syndrome distribution and features of key Chinese medicine compositions in the diagnosis and treatment of AECOPD. The clinical research and information-sharing system was used on the AECOPD data study (3000 hospital electronic medical records from August 2012 to January 2015). Data was processed by ETL software. The database was constructed. And the complex networks were used in the data mining. The results showed that the distribution of AECOPD common syndromes was heat-phlegm obstructing lung of 41.94%, phlegm-turbid obstructing lung of 22.97%, phlegm-damp obstructing lung of 10.30%,qi-yin deficiency of 6.31%, phlegm-stasis obstructing lung of 5.72%, lung-kidneyqi-deficiency of 4.01%, lung-spleenqi-deficiency of 1.15%, and phlegm obstructing orifices of 1.15%. Chinese medicine compositions were obviously divided into two large groups, which were the core Chinese medicine group and the associated Chinese medicine group. The research results illustrated that the clinical research and information-sharing system had good analysis and classification effects, which not only analyzing the medication laws of senior Chinese medicine experts and single disease treatment by a hospital, but also using in the large data analysis (data collection of single or multiple diseases on Chinese medicine clinical research nationally). It provided a good scientific research platform for Chinese medicine.
8.Effect of ultra-fine powder of Bletilla striata on experimental gastric ulcer in rats

Objective To compare the pharmacodynamics of common Bletilla striata powder(80 and 200 meshes) with ultra-fine powder of B.striata on experimental gastric ulcer in rats and to clarify how much the dose of B.striata was decreased after superfine comminution.Methods The models of gastric ulcer were set up by anhydrous alcohol and acetic acid to measure the gastric ulcer area and count out the inhibitory rate;The tissue slice of gastric ulcer cicatrix was got far HE staining.Based on the viewpoint of pathology the morbid state was observed.Results B.striata powder could decrease the gastric ulcer area significantly,and the groups of ultra-fine powder were advantaged to the others.Conclusion After superfine comminution,the potency of B.striata can be improved with its dose reducing.It is good for mo-dernization of making dosage forms.
9.Analysis and outlook on teaching status of Molecular Pharmacognosy.

The teaching status of Molecular Pharmacognosy in 28 institutions in China was investigated by questionnaire and the survey data was analyzed by SPSS. Research contents included course beginning years, majors, class hours, characteristics of the course, teaching ways, the theory and practice contents, evaluation modes, selection of teaching material, teaching achievements, teachers and so on for undergraduates and graduates. Research results showed that with 20 years' development, Molecular Pharmacognosy had been offered for both undergraduate and graduate students in at least 20 colleges and universities and Molecular Pharmacognosy education in China showed good development momentum. At the same time, to promote the development of Molecular Pharmacognosy further, investment for it should be increased and practical teaching condition should be improved.

10.Liver transplantation for patients with hepatopulmonary syndrome: long-term follow-up and prognosis analysis

Objeetive To analyze the clinical efficacy of liver transplantation (OLT) for patients with hepatopulmonary syndrome (HPS).Methods From 2008 to 2013,420 adult patients underwent liver transplantation in our hospital.There were 91 patients with,and 329 patients without,HPS.The 5-year survival and mortality rates after OLT for the two groups were retrospectively analyzed.Results There were no significant differences between patients without and with HPS in age,primary disease,Child-Pugh score,MELD score,cold ischemia time and warm ischemia time.However,the differences on serum albumin [(29.6 ± 1.2) g/L vs.(26.4 ± 1.6) g/L] and blood oxygen pressure [(61.0 ±9.0) mmHg (1 mmHg =0.133 kPa) vs.(87.0 ± 6.0) mmHg] were significantly different (P < 0.05).The 1-year cure rate was 65.9% (60/91) in 91 patients with HPS after liver transplantation.The 1,3,5-year cumulative survival rates for patients without HPS were 97.3%,90.9% and 80.3%,respectively,and the main causes of death were primary graft dysfunction,recurrent cardiovascular events and primary disease recurrence or tumors.The 1,3,5-year cumulative survival rates for patients with HPS were 65.9%,59.3% and 56.0%,and the main causes of death were multiple-organ failure,pulmonary infection and cerebrovascular events.Kaplan-Meier survival curve analysis showed that the survival of patients with HPS was significantly lower than that of patients without HPS (P < 0.05).Conclusions Liver transplantation is the most effective treatment for patients with HPS,but the short-term mortality rate is relatively high.We still need to learn more about HPS to improve the survival rate of patients with HPS after liver transplantation.
