2.RHEIN INDUCES APOPTOSIS IN CANCER CELLS AND SHOWS SYNERGY WITH MITOMYCIN

AIM To study the apoptosis-inducing and growth-inhibitory effect of rhein, an herb-derived compound, and its combination with mitomycin C (MMC) on cultured tumor cells. METHODS MTT assays were used to determine the inhibition of proliferation by drugs in several tumor cell lines. Nucleoside transport and DNA synthesis inhibition were determined by [3H] thymidine transport and incorporation assays. Flow cytometry, electrophoresis on agarose gels and morphological assesment were applied to analyze the apoptotic changes. RESULTS The IC50 values of nucleoside transport was 19.1 μg*mL-1 and that of the DNA synthesis inhibited by rhein was 27.4 μg*mL-1. In MTT assay the IC50 values of rhein for KB, hepatoma BEL-7402 and mammary carcinoma MCF-7 cells were 11.5 μg*mL-1, 14.0 μg*mL-1 and 18.4 μg*mL-1 respectively. Synergistic effect of rhein and MMC was found in all the three cell lines. As found, rhein induced apoptosis in KB cells, and the increase of apoptotic cells reached 71.0% at 96 h. The combination of rhein and MMC enhanced the induction of apoptosis significantly. CONCLUSION These results suggest that rhein, as a biochemical modulator, might be potentially useful in cancer chemotherapy.
3.Evaluation of dental plaque by quantitative digital image analysis system

Objective:To analyze the plaque staining image by using image analysis software,to verify the maneuverability,practicability and repeatability of this technique,and to evaluate the influence of different plaque stains.Methods:In the study,30 volunteers were enrolled from the new dental students of Peking University Health Science Center in accordance with the inclusion criteria.The digital images of the anterior teeth were acquired after plaque stained according to filming standardization.The image analysis was performed using Image Pro Plus 7.0,and the Quigley-Hein plaque indexes of the anterior teeth were evaluated.Results:The plaque stain area percentage and the corresponding dental plaque in-dex were highly correlated,and the Spearman correlation coefficient was 0.776 (P<0.01 ).Intraclass correlation coefficients of the tooth area and plaque area which two researchers used the software to calcu-late were 0.956 and 0.930 (P<0.01).The Bland-Altman analysis chart showed only a few spots outside the 95%consistency boundaries.The different plaque stains image analysis results showed that the diffe-rence of the tooth area measurements was not significant,while the difference of the plaque area measure-ments significant (P<0.01).Conclusion:This method is easy in operation and control,highly related to the calculated percentage of plaque area and traditional plaque index,and has good reproducibility.The different plaque staining method has little effect on image segmentation results.The sensitive plaque stain for image analysis is suggested.
4.Curative effect of calcium hydroxide paste combined with root canal in treatment of pulpitis and apicitis

Objective To investigate the clinical curative effect of calcium hydroxide paste combined with root canal in treatment of pulpitis and apicitis.Methods A total of 76 patients with pulpitis and apicitis from Dec.2014 to Aug.2015 were collected and randomly divided into two groups, 38 cases in each group.Control group (65 tooth ) were treated by routine root canal therapy, with zinc oxide iodoform clove oil cement filling root canal;experimental group (71 tooth) were treated by routine root canal therapy, with calcium hydroxide paste filling root canal.Periodontal index, pain during root canal therapy ( IAE ) , serum inflammatory factors and clinical curative effect were observed and compared between two groups pre-and post-treatment.Results Compared with pre-treatment, periodontal index of PLI, BI, PD and GI in two groups were all lower (P<0.05), and IL-6, IL-8, IL-1βand TNF-αwere lower(P<0.05).Compared with control group, PLI, BI, PD and GI of experimental group were lower (P<0.05), IL-6, IL-8, IL-1βand TNF-αwere lower(P<0.05),too.The difference of IAE incidence between two groups was significant (P<0.01) and the clinical curative effect of experimental group was 88.73%, higher than 72.31% of control group (P<0.05).Conclusion Calcium hydroxide paste combine with root canal therapy in treatment of pulpitis and apicitis are effective, which can significantly improve periodontal condition of tooth, reduce the occurrence of pain during root canal therapy.
5.Observation on clinical effects of herbal cake-partitioned moxibustion for knee osteoarthritis

Objective:To observe the clinical effects of herbal cake-partitioned moxibustion in the treatment of knee osteoarthritis (KOA).
Methods:A total of 120 cases with KOA were randomly divided into two groups by the random digital table, 60 cases in each group. The herbal cake-partitioned moxibustion (HCPM) group was treated by herbal cake-partitioned moxibustion, once per day and five days per week, with ten sessions as one course. The Western medication (WM) group was treated by the oral administration of Diclofenac Sodium Sustained-release Tablets. The knee functions of the patients were assessed by Western Ontario and McMaster Universities osteoarthritis index (WOMAC) before and after the treatment. The clinical effects were observed in the two groups after four-week continuous treatment.
Results:The total effective rate was 86.7% in the HCPM group and 73.3% in the WM group. The difference in the total effective rate between the two groups was statistically significant (P<0.05). After treatment, WOMAC scores in the two groups were statistically different than those of the same group before the treatment (bothP<0.05); the difference between the two groups were statistically significant (P<0.05).
Conclusion:Herbal cake-partitioned moxibustion and oral administration of Diclofenac Sodium Sustained-release Tablets can improve the knee functions of KOA patients, but the therapeutic effect was better by herbal cake-partitioned moxibustion than by oral administration of Diclofenac Sodium Sustained-release Tablets.
6.Sounding Resonance Characteristics of Pre-and Post-Tonsillectomy in Patients with Tonsil Hypertrophy

Objective To study the influence of tonsillectomy on resonance characteristics of four resonance cavity in patients with tonsil hypertrophy (II or III degree) .Methods Using multi-channel voice analysis system to test the cavity resonance characteristics and energy distribution of 38 tonsil hypertrophy male patients (including pre- and one month of post-operative) and 40 normal males when they speak /a:/.The frequency spectrum was 4 000 Hz(FR1 ,FR2 ,FR3 ,FR4 ) .Results FR2 ,FR3 energy values of the head cavity and mouth cavity ,and FR2 energy values of the thoracic cavity increased to the level of the normal control group after tonsillectomy(P<0 .05) .FR1 energy values of head ,mouth and thoracic cavity decreased to the level of the normal control group after tonsillecto‐my(P<0 .05) .FR1 ,FR2 ,FR3 ,FR4 energy values of laryngeal cavity maintained the normal level ,including pre- and post - operation .Conclusion Tonsillectomy makes oropharyngeal cavity volume expanded and airflow channel broaden .Tonsillectomy mainly maks high-frequency resonance of head and mouth cavities recovered to the normal level after about one month .

8.Application of Kinesio Taping in Children with Cerebral Palsy (review)

Usually individualized comprehensive treatments are applied to children with cerebral palsy. Among them, Kinesio taping is a simple treatment compared to orthosis, which allows greater rang of motion (ROM) and can more easily be accepted by patients, however, its effectiveness is controversial. This article introduced the mechanism that mainly includes relieving pain, improving joint mobility, increasing strength, relieving edema and increasing proprioception. The application of it in upper limbs, lower limbs and trunk of children with cerebral palsy was discussed. Finally, the advantages and limitations of Kinesio taping, suitable types of cerebral palsy and the therapeutic evaluation were analyzed.
9.Lidamycin inhibits the cancer cell PKC activity induced by basic fibroblast growth factor

Aim To study the mechanism of inhibition of basic fibroblast growth factor (bFGF) related signal transduction by lidamycin in cancer cells. Methods MTT assay was used to determine the growth inhibitory effect of lidamycin (LDM) and adriamycin (ADR) in several cancer cell lines. The inhibition of bFGF bound to its receptor by LDM was measured with [ 125I ]-bFGF binding assay.Intracellular Ca2+ stimulated by bFGF was measured by Fura-3. The formation of bFGF- receptor immune complex and the inhibitory effect of LDM on the activity of PKC isoenzymes induced by bFGF in cancer cells were identified by Western blotting analysis. Results LDM displayed extremely potent growth inhibitory effect on several cancer cell lines in a dose-dependent manner. A comparison of the IC50 values showed that the effect of LDM was 1 000-fold more potent than that of ADR. LDM blocked the specific by anti-FGFR specific antibody, LDM inhibited the formation of bFGF-receptor immune complex. bFGF demonstrated that LDM inhibited the activity of PKC isoenzymes in cancer cells stimulated with bFGF.Conclusion The blocking of bFGF receptors in the signal transduction pathway may be involved in the effect of LDM on cancer cells.
