1.Analysis of 176 Pediatric ADR Reports in Our Hospital

OBJECTIVE:To analyze the characteristics and rules of pediatric adverse drug reaction (ADR) in our hospital, and provide reference for clinical safe drug use. METHODS:Retrospective study was adopted to statistically analyze the ADR re-ports in our hospital from Mar. 2012 to Jun. 2016. RESULTS:In the 176 ADR reports,102 were male(57.95%)and 74 were fe-male (42.05%);children younger than 6 years old had the highest incidence of ADR (81.26%);intravenous administration in-duced 153 cases(86.93%),the top one was antibiotics,which involving 20 antibiotics,totally 106 cases(60.23%);skin and its appendages damage were the main manifestations, totally 127 cases (50.60%). CONCLUSIONS:Pediatric ADR monitoring should be strengthened,dinical rational drug use should be promoted by cooperation in many ways to reduce and prevent the incid-erle of pediatric ADR.
2.Clinical significance of p53 expression in colorectal cancer patients

Purpose:We evaluated the sensitivity and specificity of p53 antibodies (p53-Ab) as a preoperative tumor marker and as a prognosis marker in colorectal cancer patients and we also investigated whether p53-Ab production is dependent on p53 protein overexpression in tumor tissue.Methods:Serum samples from 178 colorectal cancer patients and 40 healthy controls were tested for the presence of p53-Ab by using an ELISA kit. Expression of p53 protein in tumors was determined using mouse anti-human p53-Ab.Results:Serum p53-Ab was detected in 18.54% of patients while all controls were negative. A strong correlation between p53-Ab production and p53 protein expression was observed: 69.7%of p53-Ab positive cases had tumors positive for p53 vs 30.3%of p53-Ab negative cases. There was also a significant predominance of p53-Ab positive cases in Dukes ’ stages B (16/71,22.54%), C (12/55,21.82%)and D(5/30,16.67%)over stage A. No difference in patients’ age, sex and localization was found between the p53-Ab positive and negative groups. Recurrence of cancer did not differ between p53-Ab positive and negative patients after a median follow-up of 13.5 months and also median survival time did not vary significantly.Conclusions:There was a significant correlation between p53-Ab production in serum and p53 protein expression in tumor tissue. Because of their low sensitivity (18.54%) p53-Ab are not suitable as a preoperative tumor marker. However, their high specificity (100%) and their potential for early diagnosis of a tumor relapse makes them valuable for postoperative monitoring during follow-up in p53- Ab positive patients.[
3.Study of Mechanism of Preoperative Oral Carbohydrate Involved in Attenuating Postoperative Insulin Resistance

Objective To assess the tolerance of preoperative carbohydrate-rich beverage,to determine its effect on postoperative insulin resistance and analyze its potential mechanism.Methods Thirty-two patients undergoing elective colorectal cancer resection were recruited to this randomized controlled study and assigned to two groups at random.Patient in control group was fasted before operation,while patient in study group was given oral water.Homeostasis model assessment(HOMA)indexes,activity of PTK,and mRNA and(or)protein expressions of PKB,PI3K and GluT4 were measured before and(or)immediately after surgery.Furthermore preoperative well-beings of patients were studied.Results Among well-beings,feeling of thirst,hunger and anxiety tended to be better in patients receiving carbohydrate-rich beverages compared with fasted ones(P0.05).Conclusion Preoperative consumption of carbohydrate-containing fluids is safe and effective.Provision of carbohydrate energy source prior to surgery may attenuate immediate postoperative insulin resistance.A carbohydrate-rich drink enhances insulin action at the time of onset of anaesthesia or surgery by activating three kinases named PTK,PI3K,PKB which are key enzymes in pathway of insulin signal transduction.It is likely to explain the effects on postoperative insulin resistance.
4.The clinical significance of CK-19 mRNA expression in the axillary drainage fluid of rectal cancer patients with real-time RT-PCR detection

Objective To detect CK-19 mRNA expression by quantitative real-time RT-PCR in axillary drainage fluid of rectal cancer and investigate its clinical significance.Methods Axillary drainage fluids were collected from 59 patients with rectal cancer and 15 patients with benign abdominal lesion from Sep.2010 to Dec.2010.Level of CK-19 mRNA in axillary drainage fluid was detected using specific primers by real-time RTPCR.The data were statistically analyzed to investigate the relationships between CK-19 mRNA level and tumor invasion,lymph node status,tumor stage and tumor differentiation level.Results The positive rate of CK-19 mRNA expression in patients with rectal cancer was 67.8%,which was significantly higher than that in patients with benign abdominal lesion.The expression of CK-19 mRNA was significantly correlated with the depth of tumor invasion,lymphnode status,tumor stage and histopathological differentiation( P < 0.05 or P < 0.01 ).Ck129 mRNA expression was associated with the pathological level,the higher of the lymph node translation level,the higher expression in the axillary drainage fluid after rectal cancer surgery (r =0.674,P =0.021 ).The lower of the lymph node differentiation level,the higher expression in the axillary drainage fluid after rectal cancer surgery (r =-0.741,P =0.014).Conclusion Quantitative detection of CK-19 mRNA in axillary drainage fluid of rectal cancer by RT-PCR could enhance the diagnostic sensibility of colorectal cancer micrometastases.RT-PCR assay is suitable for predicting peritoneal micrometastasis of rectal cancer,which is a reference for postoperative treatment and prognosis prediction.

6.Model establishment and finite element planning for suturing manipulation in corneal surgery

Suturing is the key to the success of corneal transplant surgery.By analyzing the suturing-process of corneal,a model for suturing was established.A suturing theorietical trajectery was proposed without considering the viso-elasticity of comeal.Then,a suturing simulation was done with the ANSYS software,the manipulating trajectory was planned for corneal soft-tissue with viso-elasticity,correspondingly,and the position and posture of needle in suturing were obtained.The study can be used for suturing manipulation in cornea suture surgical.
7.Expression of VEGF and VEGFR-2 in patients with operable advanced laryngeal cancer treated with induction chemotherapy

Objective To assess the prognostic values of the immunohistochemical expression of vascular endothelial growth factor (VEGF) and vascular endothelial growth factor receptor-2 (VEGFR-2) in a cohort of patients with operable advanced laryngeal cancer who have been treated with induction chemotherapy. Methods VEGF and VEGFR-2 expression in the Forty-nine patients was quantified by an enzyme immunosorbent assay in a retrospective series.Results The moderate to high VEGF expression in 34 patients were found and low expression in 15 patients. VEGFR-2 expression was moderate to high in 31 patients and was low expression in 18. The probability of a complete response to induction chemotherapy was significantly higher in patients with none to low VEGF expression.Conclusion VEGF expression seems to be a significant predictor of complete response to induction chemotherapy.
8.The Efficacy and Safety of 2μm Laser Vapoenucleation of the Prostate

Objective To evaluate the safety and effectiveness of 2μm Thulium laser vaporization enucleation (Thu-VEP) in the treatment of benign prostate hyperplasia (BPH). Methods A total of 145 patients with BPH were randomized into two groups including patients underwent ThuVEP and patients underwent standard transurethral resection of the pros-tate (TURP). The intraoperative blood loss,flushing fluid quantity,operation time,bladder irrigating time,catheterization time,the international prostate symptom score (IPSS),quality of life score (QOL),the maximum flow rate (Qmax),the post-void residual (PVR) and complications were observed in two groups. Results There were no significant differences in pa-tient age, preoperative duration, prostate weight, IPSS score,QOL score,Qmax and RUV between two groups. Patients in two groups were performed surgery successfully. The values of blood loss, bladder irrigating time and catheterization time were significantly less in ThuVEP group than those of TUEP group (P<0.05). There were no postoperative complications in pa-tients of ThuVEP group. There were 5 cases of complications in TUEP group after operation. The values of IPSS score,QOL score,Qmax and RUV were significantly different after 3-12-month follow up than those before operation(P<0.01),but no significant difference between two groups. Conclusion ThuVEP is a safe and more effective treatment than that of TURP for patients with BPH. ThuVEP can significantly improve the quality of life, and reduce complications in patients with BPH.

Objective The purpose of the present study is to illustrate the effect of olfactory ensheathing cells(OECs) on the survival and neurite outgrowth of GABAergic neurons in vitro. Methods OECs were dissociated from olfactory bulb and neurons from spinal cord of E12 mouse. On the sixth day in vitro,the Millipore cultue blank with OECs was transferred to the neuron culture mediam and continue the co-culture for another 6 days.The cultured neurons were stained with anti-GABA antibody.The neurite of neurons was observed with an image system.The number of GABAergic positive neurons was counted under the microscope. 10.Expression of NM-23,PCNA,p53 and S-100 in colorectal carcinoma in the elderly and their relationship

Objective To study the expression of NM-23 ,PCNA,p53 and S-100 in colorectal cancer tissues and their relationship.Methods Three hundred and seventy eases of elderly eolorectal cancer were labelled with NM-23,PCNA,p53 and S-100 and staged by pTNM classification.Results The positive rates of NM-23,PCNA,p53 and S-100 were 74.9% ,60.3% ,58.1% ,68.1% respectively.In elderly group,the expression of NM-23 had a neg-ative correlation with that of PCNA (r = -0.101 ,P = 0.026).However the expression of NM-23 had a positive corre-lation with that of p53 (r =0.111 ,P =0.016 ),the expression of S-100 had a positive correlation with that of p53 (r=0.112,P = 0.015 ),the expression of S-100 had a positive correlation with that of PCNA (r =-0.229,P =0.000).Conclusions In the elderly patients,NM-23,p53 and S-100 were significantly correlated with the inva-sion,metastasis and prognosis of eoloreetal cancer.PCNA may be a stimulative factor in the invasion and metastasis of colorectal cancer.NM-23 ,PCNA,p53 and S-100 can serve as effective markers in reflecting the invasion,metas-tasis and prognosis of colorectal carcinoma in the elderly.
