1.Correlation between Bmi-1 and clinicopathological features of colorectal cancer

BACKGROUND:Bmi-1 (B-cel-specific moloney murineleukemia virus insertion site), a colorectal cancer stem cellgene is of great significance in the regulation of telomere reverse transcriptase activity and transcriptional status as wel as cellaging and carcinogenesis.
OBJECTIVE:To investigate the correlation between Bmi-1 expression and clinicopathological changes of colorectal cancer.
METHODS:The expression of Bmi-1 mRNA in carcinoma cells was detected in 50 cases of colorectal cancer using quantitative real-time PCR. The expression of Bmi-1 protein in cancer tissue was detected by hematoxylin-eosin staining and immunohistochemical staining SP method.
RESULTS AND CONCLUSION:Sex, age, and tumor size showed no different effects on the expression of Bmi-1 mRNA. The expression of Bmi-1 protein showed no significant difference in patients who were different in sex, age, tumor size and Duke staging of tumor. The expressions of Bmi-1 mRNA and protein in patients with lymph node metastasis and depth of invasion deeper than the serosa were much higher than those in patients with no lymph node metastasis and depth of invasion shal ower than the serosa (P<0.05). The expression of Bmi-1 mRNA was much higher in patients with higher Duke staging cancer (P<0.05). The expression of Bmi-1 mRNA and protein are closely related to different pathological features of colorectal cancer, such as metastasis, infiltration, and Duke staging.
2.Construction of Evaluation Model of Rational Use of Drugs of Medical Institutions

Objective To study the evaluation of rational use level of drugs in medical institutions.Methods According to the specific requirements of rational use of drugs in medical institutions,a criteria system is established.And a rational use of drugs evaluation model of medical institutions is to be built on the theory of fuzzy evaluation,group decision making and the analysis method of multi-index evaluation.Results Based on the fuzzy evaluation and group decision making,a rational use of drugs evaluation model of medical institutions is presented in the paper and one example is used to show the feasibility and the effectiveness of the proposed model.Conclusion The rational use of drugs evaluation model based on fuzzy evaluation and group decision making can effectively accomplish the evaluation of rational use of drugs of medical institutions,and build the foundation for improving the rational use of drugs of medical institutions and improving the level of universal health care.
3. Research on the Development Path of Building a World-class Pharmacy Discipline

RESULTS AND CONCLUSION: The ranking status of pharmacy disciplines in China is compared and analyzed, and from the three aspects of the index system, international influence, and innovative talents training, the development path of building world-class pharmacy discipline in China is proposed.
4.Inhibitory Effect of Platycodin D on Proliferation of SGC7901 Cells and Moleclar Mechanism

[Objective] To investigate the effects of platycodin D(PD) on the proliferation and apoptosis of human stomach cancer SGC7901 and the related mechanism.[Methods] SGC7901 was cultured in virto and was treated with 5~20μm·L-1 concentrations of PD.Cell proliferation was examined by MTT assay.Cell apoptosis was detected by Annexin V FITC/PI double staining.The change of mitochondrial trans-membrane potential was measured by JC-1 staining.The potein expression of cleaved caspase-3,cleaved caspase-9,cleaved PARP,bcl-2,bax,p-ERK,ERK,p-JNK,JNK,p-p38 and p38 detected by Western blot.[Results] MTT results showed that PD inhibited the growth of SGC7901 cells in a dose-dependent manner at 24h and 48h.SGC7901 cells treated with PD for 24h showed significantly enhanced apoptosis and weakened mitochondrial membrane potential compared with the control cells.Western blot results showed that PD could up-regulate expression of cleaved PARP,cleaved caspase-3,cleaved caspase-9,bax,p-JNK,p-p38 protein,decreased bcl-2,p-ERK protein,the expression of ERK,JNK,p38 protein did not change significantly.[Conclusion] PD may inhibit the proliferation and induce the apoptosis of SGC7901 cells.These findings indicated that PD inhibited cell proliferation by inhibiting the ERK signaling.PD effect on bax and bcl-2 by activation of JNK and p38 signaling pathway resulted in the decrease of mitochondrial membrane potential and activation of caspase,which induced the apoptosis of cancer cells.
6.The effects of U0126 of different doses On the ability of low frequency magnetic stimulation promoting astrocyte migration

Objective:To investigate the effects of U0126 of different doses on the ability of low frequency magnetic stimulation promoting astrocyte migration and to select suitable dose of U0126.Method:Twenty-four adult healthy SD rats were selected to receive Injection of 0.5ml of 1% ethidium bromide(EB) in PBS into the dorsal spinal cord funiculus on the left side at T10-11 level to make located spinal cord injury models and randomly divided into four groups.The four groups were exposed to magnetic stimulation(1Hz,1.52T.30pulses)at the following dose respectively:Omg/kg U0126(control group).0.1mg/kg U0126(low-dose group), 0.2mg/kg U0126(middle-dose group),0.4mg/kg U0126(high-dose group).On the day 14 after stimulation,the rats were sacrificed and the expressions of glial fibfillary acidic protein(GFAP),microtubule associated protein-2(MAP-2),extracellular signal-regulated kinase1/2(ERg1/2)and the volume of holes were detected with immunohistochemistry.Quantitative analysis 0f the expressions of GFAP,MAP-2 and ERK1/2 were performed with the image analysis system.Result:With the increase of U0126 dose,the volume of hole increased on day 14(p<0.05).In the lesion area,the expressions of GFAP and ERK1/2 could be found,while MAP-2 could not.Significant differences were revealed in the expressions of GFAP、ERK1/2 among the four groups,it Was significantly lower in U0126 groups than that in control greup(P<0.05).while the middle-dose group had similar effect with the high-dose group(P>0.05).Conclusion:U0126 of different doses all could resupinate astrocyte migrations which were coused by low frequency magnetic stimulation,and 0.2mg/kg was the suitable dose.
7.The effect of magnetic stimulation on astrocyte migration and its mechanism

Objective To investigate the effect of magnetic stimulation on astrocyte migration and its mech-anism. Methods Twenty-four adult, healthy Spragne-Dawley rats were injected with 0.5 μl of 1% ethidium bro-mide (EB) in the left side of the dorsal spinal cord funiculus at the T_(10-11) level to make a local spinal cord injury mod-el. They were then randomly divided into four groups and exposed to 30 pulses of magnetic stimulation at 1 Hz and the following intensities: O T (Group A);1.9x40% T (Group B); 1.9x80% T (Group C); 1.9x100% T (Group D). On the 14th day after stimulation, the rats were sacrificed and the expression of glial fibrillary acidic protein (GFAP), microtubule associated protein-2 (MAP-2) and extracellular signal-regulated kinase 1/2 (ERK 1/2) were detected, and the volume of holes in the injured area of the spinal cord was measured. Quantitative analysis of the GFAP, MAP-2 and ERK1/2 expression was performed using immunohistochemistry and an image anal-ysis system. Results The volume of holes in the injured area of the spinal cord decreased with increasing stimula tion intensity. In the reduced area of the holes, the expression of GFAP and ERK 1/2 could be seen, but not MAP-2. Significant differences were revealed in the expression of GFAP and ERK 1/2 among the four groups, but it was always significantly higher in the magnetic stimulation groups than in the controls. Conclusions After magnetic stimulation, astrocytes migrate into the injured spinal cord's holes. Astroeyte migration increases with increased mag-netic stimulation intensity, which may be associated with high expression of ERK 1/2.
8. The trends of female breast cancer incidence and mortality in Beijing, Shanghai, Linzhou and Qidong in China

Objective: To analyze the trends of female breast cancer incidence and mortality in Beijing, Shanghai, Linzhou and Qidong in China between 1988 and 2007. Methods: The data of female breast cancer incidence and mortality and the data of corresponding population during 1988-2007 were collected from four eligible cancer registries which could provide complete sets of historical data. The pooled analysis and time-trend analysis of the data were performed. Results: During 1988-2007, the APCs (annual percent changes) of age-standardized incidence rates (by China's population in 1982) of female breast cancer in Beijing, Shanghai, Linzhou and Qidong were 2.49%, 2.55%, 7.04% and 4.16%, respectively. Although the average annual growth of APC of age-standardized mortality was 4.10% in Linzhou, there were no significant changes in Beijing, Shanghai and Qidong. Conclusion: During 1988-2007, the breast cancer incidence and mortality of the urban women in Beijing and Shanghai were significantly higher than those of the rural women in Linzhou and Qidong. The incidence rate of female breast cancer in these four areas showed an obvious rising trend, and this increase was more significant in rural areas. The mortality rate of female breast cancer in Linzhou also showed a rising trend, but the mortality rates in the other three areas remained steady. Copyright © 2012 by TUMOR.
9. Incidence and mortality of female breast cancer in China - A report from 32 Chinese cancer registries, 2003-2007

Objective: To describe the incidence and mortality of female breast cancer in China during 2003-2007. Methods: The incidence and mortality data of female breast cancer and the corresponding population data from 32 eligible cancer registries in China during 2003-2007 were collected and analyzed. Results: The combined crude incidence rate of female breast cancer during 2003-2007 was 41.64 per 100 000, which was the highest among female cancer incidence rates. The combined crude mortality rate of female breast cancer was 9.63 per 100 000, which was ranked as the sixth among the mortality rates of all female cancers. The incidence rate was 3.04 times higher and the mortality rate was 1.92 times higher in urban areas than those in rural areas. Compared to the incidence and mortality rates of 184 regions provided from GLABOCAN 2008 database, the female breast cancer incidence in China was ranked as the one hundred and tenth, and the mortality was ranked as the one hundred and seventy-second. Conclusion: Breast cancer has been becoming one of the most common cancers among Chinese women. The incidence and mortality rates of female breast cancer in urban areas are significantly higher than those in rural areas, although the overall incidence and mortality rates of breast cancer in Chinese women are still at a medium or low level in the world. © 2012 by Tumor.
10.Immobilization of ?-glucosidase with Nano SiO_2 and Its Application in Soybean Isoflavone Hydrolysis in Two-phase-system

Nano materials are popular with its interesting properties. Large specific surface area, high mechanical strength, good absorbability make them good potencial carriers of catalyst. With the help of chemical modification, nano SiO2 particals were used as carrier to immobilize ?-glucosidase with covalent binding. It is found that the load of nano particals can reach 1/8 of its weight, and the recovered activity of enzyme is 70%. The resistence of immobilized enzyme to organic solvent wase improved obviously, which was then used in Ethyl Acetate-Water-two-phase-system to hydrolyzed soybean isoflavones.
