1.Progress of bariatric surgery on treatment of obesity and type 2 diabetes

Obesity and type 2 diabetes are common chronic diseases which severely threatens human's health and lives.In addition to behavioural and medical approaches,the bariatric surgery is increasingly being valued by medical workers.There are many kinds of surgical method of the bariatric surgery,but by far the most popular ones are Laparoscopic sleeve gastrectomy and Laparoscopic sleeve gastrectomy.The mechanism of the surgery may related to gastrointestinal hormones and fat metabolism,but no final conclusion has yet been reached on this matter.This paper is a literature review on the effect and mechanism of bariatric surgery.
2.Surveillance of bacterial antibiotic resistance in traumatic wound infection: a report of 1006 cases

Objective To monitor the distribution of pathogenic bacteria causing wound infection in trauma patients and their susceptibility to antibiotics in an effort to offer evidence for the rational clinical use of antibiotics.Methods Pathogens were identified by expressive type,but a very few was determined by the molecular methods.Micro-dilution or Kirby-Bauer method for minimum inhibitory concentration (MIC) was adopted for drug susceptibility test.Results A total of 121 species of pathogens numbering 1,257 bacterial strains were isolated from 1,066 patients.Strain number of the top 10 species accounted for 70.09%.Gram-positive cocci accounted for 48.93% and Gram-negative bacilli for 49.72%.Among Staphylococcus aureus and epidermidis,methicillin-resistant bacterial strains accounted for 34.76% and 71.08% respectively,but none were resistant to vancomycin or linezolid.Enterococcus faecalis contained 6.06% vancomycin-resistant enterococcal (VRE) and remained > 90% sensitive to teicoplanin,ampicilin and vancomycin.Enterococcus faecium contained 10.53% VRE and remained >90% sensitive to teicoplanin and > 80% sensitive to vancomycin.Baumanii contained 12.77% multidrug resistant strains and remained > 70% sensitive to imipenem and meropenem.Baumanii/calcoaceticus complex contained 29.72% multi-drug resistant strains and remained > 50% sensitive to imipenem and meropenem.Pseudomonas aeruginosa contained 3.73% multi-drug resistant strains and remained >90% sensitive to amikacin,meropenem,imipenem and piperacillin/tazobactam.No E.coli,klebsiella pneumoniae and enterobacter cloacae resistant to imipenem or meropenem were found.E.coli contained 69.51% extended-spectrum β-lactamases (ESBLs)-producing strains and remained > 90% sensitive to piperacillin/tazobactam and > 80% sensitive to amikacin.Klebsiella pneumoniae contained 53.13% ESBLs-producing strains and remained > 80% sensitive to amikacin.Conclusions Common bacteria are the leading cause of posttraumatic infection.Enterococcus faecalis and enterococcus faecium demonstrate higher susceptibility to teicoplanin; two kinds of acinetobacter demonstrate higher susceptibility to imipenem.
3.Evaluation of clinical efficacy of ultrasonic-harmonic scalpel in open thyroid surgery

Objective To evaluate the clinical efficacy of ultrasonic-harmonic scalpel in open thyroid surgery.Methods Totally 70 patients with thyroid surgery from January 2012 to January 2014 in some hospital were selected as the study objects, who were divided into a control group and an observation group equally. The patients in the control group went through traditional surgery, and the ones in the observation group underwent open thyroid surgery. Then the operative time, blood loss, postoperative hospital stay before and after surgery and body stress-related serum markers of two groups were analyzed and compared.Results The observation group had the operative time, blood loss and postoperative hospital stay significantly better than those of the control group, and the body stress-related serum markers 5 days after operation significantly lower than that of the control group, with P<0.05.Conclusion The short-term clinical effect of ultrasonic-harmonic scalpel in open thyroid surgery is significantly better than that of traditional surgery, which has high safety and lower degree of adverse physical stress.
4.Sleep disorders in patients w ith ischemic stroke:pathophysiological mechanisms and treatment

Sleep disorder is a common complication after is chemicstroke.If it is nottimely diagnosed and treated, it may affect the rehabilitation of stroke and recurrence. The manifestations of sleep disorders have various forms according to the different locations of infarction. For the treatment of sleep disorder after stroke, the method of the combination of drugs and physical therapy is used in clinical practice at present. In recent years, w ith the w idely use of the polysomnography, poststroke sleep disorders can be diagnosed clearly and carefuly. The treatment for sleep disorders after stroke may significantly improve the prognosis of patients.
5.Biological behaviors of articular chondrocytes on the polydimethylsiloxane with differential surface modifications and stiffnesses

BACKGROUND: Polydimethylsiloxane (PDMS) is widely used in the basic research on cell biology because of its good biocompatibility and ability to be processed at the micro/nano level. However, cell culture on PDMS has been generally compromised by its strong hydrophobicity.OBJECTIVE: To perform a comparative investigation on the influences of different surface modifications as well as stiffness of PDMS on cellular behaviors.METHODS: PDMS films with varying stiffnesses were subjected to various surface modifications, including serum incubation, type I collagen deposition and air plasma treatment. Bovine articular chondrocytes were seeded on PDMS films and cell adhesion, proliferation and matrix production were characterized using F-actin staining, cell counting kit-8,microscopic examination, Sirius red/Safranin-O staining and quantitative determination of glycosaminoglycans,respectively.RESULTS AND CONCLUSION: Serum incubation, type I collagen deposition and air plasma treatment were all found to promote adhesion and proliferation of bovine articular chondrocytes from the results of F-actin staining and cell prolifereration curve, with air plasma treatment the best. Total amount of glycosaminoglycans (GAG) secretion was only increased by air plasma treatment and GAG/DNA was decreased by all modifications. Stiffness also played roles in cell adhesion, proliferation and GAG production, which was found to be dependent on surface modifications. This study would provide guidance for applying PDMS in cell culture.
6.Maternal cell contamination of prenatal samples and the potential effects on prenatal diagnosis results

Objective To assess the frequency and significance of maternal cell contamination (MCC)in the invasive prenatal diagnosis,and to analysis the MCC effect on prenatal diagnosis results.Methods Totally 519 amniotic fluid samples from second trimester pregnancy,57 chorionic villus samples from first trimester pregnancy,and 576 blood samples from corresponded pregnant women were collected and genotyped by Promega PowerPlex 16 system.MCC was determined according to the genotyping results.Karyotypic and molecular diagnosis results were contrasted between MCC and non-MCC specimen of the same fetal.Results MCC presented in 3.1%(16/519)uncultured amniotic fluid,1.3%(7/519)cultured amniotic fluid and 5%(3/57)villi specimens.In the study of fetal karyotype,MCC had no significant effect on normal female fetus;but for male fetus and abnormal female fetus,there were risk of erroneous results of mosaics.As to molecular diagnosis,MCC resulted in more complex effects for the different diagnostic methods.And 10% MCC had led to misdiagnosis.Conclusions For the prenatal cytogenetic tests,MCC should be excluded when there were mosaicism karyotype results or suspicious MCC of chorionic villi samples.The effects of MCC had more seriously impact on prenatal molecular testing,which suggesting the recommend regular identity test for MCC should bc carried out.
7.Inhibitory effect of juglone on migration and invasion of glioma cell line U-87 MG

Objective To investigate the effects and mechanisms of pin1(peptidyl-prolyl isomerase 1) inhibitor ju?glone on migration and invasion in glioma cell line U-87 MG. Methods Glioma cells were treated with juglone at 0, 0.8, 1.6 and 3.2μmol·L-1. Wound-healing assay and invasion assay were performed to examine the inhibitory activity of ju?glone on glioma cell line U-87 MG. Western blot was used to analyze the protein expression of β-catenin, VEGF, MMP-2 and MMP-9. Results The wound-healing assay showed that the wound-healing rate in juglone-treated groups was 46.04%±6.25%and 30.05%±13.35%at concentrations of 1.6μmol·L-1 and 3.2μmol·L-1 , repectively. Juglone treat?ment significantly reduced the wound-healing rate compared with controls (P<0.05). Transwell invasion assay showed that the number of invaded cells in juglone–treated groups was 103.67 ± 5.69 and 77.33 ± 7.77 at the concentrations of 1.6μmol·L-1 and 3.2μmol·L-1 , respectively. Juglone treatment significantly reduced cell invasion compared with con?trols (P<0.05). Treatment with juglone significantly down-regulated the expression levels ofβ-catenin, VEGF, MMP-2 and MMP-9 in U-87 MG cells in a dose-dependent manner. Conclusion The present data suggests that juglone has a significant inhibitory action on cell migration and invasion through down-regulation of theβ-catenin and its downstream VEGF, MMP-2 and MMP-9 protein expressions in glioma cell line U-87 MG.
8.Study on the Quality Standard of Miao Medicine Indigofera stachyoides

OBJECTIVE:To establish the quality standard for Mian medicine Indigofera stachyoides. METHODS:Medicinal properties and microscopic characteristics were identified;TLC was used for the qualitative identification;HPLC was conducted for content determination of epicatechin and 2α,3α-epoxy-5,7,3′,4′-tetra-hydroxy-flavan-(4β→8)-epicatechin(reference A):the col-umn was Comatex TM C18 with mobile phase of acetonitrile- water at a flow rate of 1.0 ml/min,and detection wavelength was 210 nm,column temperature was 30 ℃,injection volume was 5 μl. RESULTS:It showed clear microscopic identification map. I. stachyoides TLC had clear spots and well separated. The linear range was 0.274 5-4.392 0 μg (r=0.999 6) for epicatechin,and 0.103 0-1.648 3 μg for reference A(r=0.999 4),RSDs of precision,stability and reproducibility tests were lower than 3%,recov-eries were 99.76%-104.82%(RSD=2.42%,n=6) and 97.98%-104.99%(RSD=2.75%,n=6) respectively. CONCLUSIONS:The established standard can be used for the quality control of I. stachyoides.
9.Prediabetes and ischemic stroke

Prediabetes is a glucose metabolism status between normal glucose metabolism and diabetes.It can not only increase the risks of occurrence and recurrence of stroke, but also affect stroke outcomes.Prediabetes causes ischemic stroke mainly through the insulin resistance and blood-brain barrier damage.Non-drug or drug intervention in patients with prediabetes can delay progression from prediabetes to diabetes and reduce the risks of occurrence and recurrence of stroke.
10.Application of FBC conditioning regimen in HLA haplotype peripheral blood stem cell transplantation.

OBJECTIVETo observe the influence of decreasing conditioning regimen intensity on the engraftment of HLA haplotype peripheral blood stem cell transplantation.
METHODTwelve patients with leukemia, including 4 in complete remission, whose HLAs were full matched with donors, and 8 with refractory leukemia, whose HLAs were mismatched, were transplanted with G-CSF mobilized allogeneic peripheral blood stem cells after conditioned with a regimen consisting of fludarabine (30 mg/m(2) x 6 days), busulfan (4 mg/kg x 2 days) and cyclophosphamide (30 approximately 60 mg/kg x 2 days) (FBC). Donor lymphocytes were infused at day + 30, + 60 and + 90 after transplantation, respectively. Hematopoietic reconstitution was observed. Engraftment was documented by the analysis of short tandem repeats with polymerase chain reaction (STR-PCR).
RESULTPatients in HLA haplotype group received a mean number of 4.87 x 10(8)/kg donor mononuclear cells (MNC), with CD(34)(+) cells of 4.58 x 10(6)/kg and patients in HLA identical group a mean number of 4.85 x 10(8)/kg MNC with CD(34)(+) cells of 4.47 x 10(6)/kg. The mean time of white blood cell count more than 1.0 x 10(9)/L was 14 (10 approximately 18) days in HLA matched patients and 29 (11 approximately 90) days in HLA haplotype group. One three locus mismatched patient failed to engraft, but auto-hematopoiesis was recovered on day + 50. Full donor chimerism was observed in all patients except one with mixed chimera. The mixed chimera was converted into full donor chimera after three times donor lymphocyte infusion. One each died from severe acute GVHD, severe VOD and severe chronic GVHD in HLA haplotype group, and one from chronic GVHD in HLA identical group.
CONCLUSIONPatients survived engraftment was not influenced by decreasing conditioning intensity as in this regimen. Haplotype stem cells could be engrafted durable in recipients by this regimen combined with donor lymphocyte infusion.
