1.pSilence APE1 inhibits growth of osteosarcoma in animal model

Objective To study the inhibitory effect of APE1 expression vector pSilence APE1 on growth of osteosarcoma in nude mice, and to elucidate the role of APE1 in pathogenesis and development of osteosarcoma. Methods Nude mice model bearing osteosarcoma was reproduced by implanting human osteosarcoma cell line 9901. Twenty mice were randomly divided into two groups: control group in which the mice were treated with lipofectamine, and experimental group in which the mice were treated with pSilence APE1. The tumor inhibition rate was calculated after 12 days of treatment. Meanwhile, the expression of APE1 protein, intratumor microvessel density (MVD), and proliferation index were observed by immunohistochemistry. Apoptosis index was assessed by terminal dUTP nick end labeling (TUNEL) technique. Results Down-regulation of APE1 expression of tumor cells was found in the group treated with pSilence APE1. The tumor inhibition rate was 38.23%. The intratumoral microvessel density (MVD) in experimental groups and proliferation index were significantly lower than the control group, while the apoptosis index was much higher. Conclusion Targeted knock down of APE1 by pSilence APE1 may inhibit the growth of osteosarcoma in vivo.
2.In vitro study of the treatment of human osteosarcoma by pSilence APE1 combined with neutron-ray

Objective Apuinic/apyrimidic endonuclease/redox effector factor-1(APE1/Ref-1,abbreviated as APE1) is a major member of the base excision repair(BER) pathway involved in oxidative DNA damage repair.To knock down APE1 gene expression in HOS cells with pSilence APE1,and explore its effect in combination with 252Cf neutron ray radiotherapy.Methods The constructed APE1 siRNA expression vector pSilence APE1 was transfected into HOS cells by SuperFect Transfection liposome,and it was used to knock down the expression of APE1.The HOS cells and transfected HOS cells were respectively irradiated by 252Cf neutron ray,then the cell survival(D0),DNA single strand breaks(SSB) and cell apoptosis were determined by clone formation assay,alkaline comet assay and flow cytometer.Results The cell-survival curve was plotted by clone formation assay,the D0 value was 2.80 vs.1.89 and Dq value was 2.66 vs.2.00 for the control and transfected HOS cells,respectively,after being irradiated by 252Cf neutron ray.The tail moments at 2,5 and 10 Gy were 6.664?0.648 vs.7.997?0.542,20.322?1.433 vs.25.238?1.185 and 33.909?1.245 vs.39.191?1.052,respectively,for the control and transfected HOS cells,and the cell apoptosis rate at 2,5 and 10 Gy was 4.00 vs.5.68,5.91 vs.7.55 and 9.63 vs.13.51,respectively,for the control and transfected HOS cells.All these findings showed significant difference between the two groups(P
3.Change in Bacterial Flora and Antibiotic Resistance of Bacteria of Burn Patients

OBJECTIVE To investigate and analyze the pathogenic bacteria of burn infection and their drug resistance in recent 5 years from 2001.METHODS The patients were divided into two groups.The group 1 included patients from Jun 1996 to May 2001 and the group 2 was from Jun 2001 to Jun 2006.Burn wound bacteria were cultured,and identified and their drug sensitivity was analyzed.RESULTS In burn wound culture,in group 2 Pseudomonas aeruginosa with the rate of 30% was still in the 1st place.Staphylococcus aureus was in the 2nd place with the rate of 28.2%.The G-bacilli comprised 58.2% and the G+ cocci 40.3 %.Proteus mirabilis,Acinetobacter baumannii,Enterococcus faecalis and Candida albicans increased remarkably.The drug resistant percentage of P.aeruginosa,Enterobacter cloacae and Escherichia coli to third generation cephalosporin increased greatly.CONCLUSIONS The changes of pathogenic bacteria of burn infection and bacteria drug resistance have certain relations with the wide usage of broad spectum antibiotics such as cephalosporin and imipenem.
4.Development of Recognized Software on Multi-tumor Protein Chip Analysis System V1.0

Objective To develop the system of tumor recognized tool software based on the examination results of multi- tumor protein chip. Methods On the basis of the diagnostic recognized equation, the examination results were comparison by HTA combining VB script and Javascript language program. Results The functions of the software include dates input of tumor protein chip,recognized results, dates interrogation and record printing of 10 kinds of common tumor. Conclusion The software can be diagnosed automatically with the sorts of tumor, which is a scientific, convenient and efficient tool for tumor clinician.

Objective To observe the effects of combined generalepidural anesthesia on perioperative myocardial enzymogram and troponin T of the myocardium(cTnT)in old patients undergoing thoracic surgery.Methods Thirty-eight ASAⅠ-Ⅱ patients aged 60~80 years old were randomly divided into two groups.General anesthesia group(group G): anesthesia was induced with fentanly,propofol and vecuronium and maintained with isoflurane,propofol and vecuronium.Combined anesthesia group(group C):general anesthesia was performed as in group G and continuous epidural block with(1.6%) lidocaine was added during operation.Peripheral blood CK,CK-MB,AST,LDH and cTnT were measured before anesthesia,at the end of operation and at 6,24,48 hours after operation.Results Two groups could provide same postoperative analgesia.There was no change for HR,RR and MAP in two groups.Serum CK,AST,LDH and cTnT concentration in group G after operation were significantly higher than those before anesthesia(P
7.Survey of student nurses′acceptance of case-based group assessment

Objective To explore students nurses′acceptance of case-based group assessment. Method A total of 100 student nurses participated in the survey by a self-designed questionnaire to evaluate the perception and acceptance of the student nurses. Results The score on the acceptance of case-based group assessment was (2.44 ± 0.46). The items with higher acceptance included teamwork spirit, clinical reasoning and decision-making ability, and the items with lower acceptance were improving of nurse-patient communication and the nursing skills. Conclusions The student nurses have a good acceptance of the case-based group assessment and think this assessment method can contribute to development of teamwork spirit and competency of clinical reasoning and decision-making. On the other hand, they suggest we should strengthen the ability in nurse-patient communication and optimize the links in ability assessment.
8.Comparison of efficacy of flurbiprofen axetil combined with fentanyl administered using different modes for postoperative analgesia: a prospective, multicenter, randomized, double-blind, control, parallel-group study

Objective To compare the efficacy of flurbiprofen axetil combined with fentanyl administered using different modes for postoperative analgesia.Methods This was a prospective,multicenter,randomized,double-blind,control,parallel-group study.ASA Ⅰ or Ⅱ patients,aged 14-91 yr,weighing 35-95 kg,scheduled for orthopedic,thoracic or hepatobiliary surgery under general anesthesia from January 2010 to October 2010,were randomly divided into A,B and C groups.The three groups received patient-controlled intravenous analgesia (PCIA) after surgery.In group A,flurbiprofen axetil 100 mg was injected immediately after the end of surgery and then PCIA was performed with fentanyl 1.0 mg in 100 ml of normal saline.In group B,PCIA was performed with flurbiprofen axetil 200 mg and fentanyl 0.6 mg in 100 ml of normal saline.In group C,flurbiprofen axetil 100 mg was injected immediately after the end of surgery and then PCIA was performed with flurbiprofen axetil 200 mg and fentanyl 0.6 mg in 100 ml of normal saline.The PCA pump was set up with a 2 ml bolus dose,a 10 min lockout interval and background infusion at a rate of 2 ml/h.VAS scores at rest and during activity and sedation score were recorded at the end of surgery and 4,8 and 24 h after surgery.The effective analgesia,excessive sedation,nausea and vomiting,dizziness,somnolence and respiratory depression were recorded within 24 h after surgery.Samples from the PCIA bump were taken to do microbe culture experiment at 24 and 48 h after surgery.Results Two thousand five hundred and ninety-six cases completed this trial (875 cases in group A,946 cases in group B and 775 cases in group C).Compared with group A,VAS scores at rest and during activity at the end of surgery and 4,8 and 24 h after surgery and sedation score were significantly decreased in group B,VAS scores at rest and during activity were significantly decreased at the end of surgery and 4 and 8 h after surgery and sedation scores were significantly increased at 4 and 8 h after surgery in group C,the rate of effective analgesia was increased in groups B and C,the incidence of excessive sedation was decreased in group B,while increased in group C,the incidence of postoperative nausea and vomiting was significantly decreased in groups B and C,and the incidence of postoperative dizziness was significantly decreased in group C (P < 0.05).Compared with group B,no significant change was found in the VAS scores at rest and during activity,rate of effective analgesia,and incidences of nausea and vomiting,and somnolence (P > 0.05),sedation scores were significantly increased at the end of operation and 4 and 8 h after surgery,the incidence of excessive sedation was increased,and the incidence of postoperative dizziness was decreased in group C (P < 0.05).Neither bacterium nor fungus was found in the PCIA pump samples.Conclusion PCIA with flurbiprofen axetil 200 mg and fentanyl 0.6 mg (background infusion at a rate of 2 ml/h,2 ml bolus dose,10 min lockout interval) provides better efficacy and the occurrence of sides effects is low for the patients undergoing moderate or major operations.
9.Clinical significance of multi-tumor markers detection in the early diagnosis of hepatocellular carcinoma

Objective To investigate the value of combined detection of multi-tumor markers in the diagnosis of primary hepatocellular carcinoma (HCC) and to establish the discriminant equation. Methods Using a protein chip, 12 tumor markers in the serum from 98 patients with HCC and 67 patients with benign liver diseasewho had been admitted to Daping Hospital from November 2003 to April 2006, and 46 healthy individuals during he same period were analyzed. A discriminant equation was established to discriminate primary HCC from benign liver diseases. All the data were processed by variance analysis and chi-square test. Results The positive rates of the tumor markers were 89% (87/98) in patients with primary HCC, 19% (13/67) in patients with benign liver disease and 4% (2/46) in healthy individuals. There was statistical difference in the serum level of alpha-fetoprotein (AFP), eareinoembryonic antigen (CEA), ferritin (FER), CA19-9 and CA125 among the 3 groups (F =59.530, 40.472, 31.708, 75. 897, 153.066, P <0.05). Combined detection of AFP, CEA, FER, CA19-9 and CA125 improved the diagnostic accordance rate to 89%, which was significandy higher than the diagnostic accordance rate (64%) when only AFP was detected (X2 = 16.362, P <0.05). The accuracy of the discriminant equation was 90%. Conclusions Combined detection of multi-tumor markers is superior to AFP detection. Combined detection of multi-tumor markers can be used in screening of the HCC patients in HCC high risk population and in the early diagnosis of primary HCC.
