1.Role of Fuzhisan in reducing Tau protein hyperphosphorylation in cortical neurons through a cyclin-dependent kinase 5 pathway

Aim Fuzhisan ( FZS ) , a Chinese herbal complex prescription that has been used for the treat-ment of AD for over 15 years, is known to enhance the cognitive ability in AD patients. In this study, to in-vestigate whether FZS reduces Aβ25-35-induced Tau protein hyperphosphorylation in neonatal rat cortical neurons by suppressing the cyclin-dependent kinase 5 ( CDK5 ) pathway. Methods Neonatal Wistar rats born within 24 h were selected to separate and purify their cortical neurons for culture in vitro. After 7-day culture of cortical neurons in vitro, 20 μmol · L-1 Aβ25-35 was used to act on them for 24 h. Medication groups were pretreated with FZS ( 20 mg · L-1 ) , CDK5 inhibitor roscovitine ( 15 μmol · L-1 ) and cal-pain preparation calpeptin (20 μmol·L-1 ) for 24 h, followed by reaction with 20 μmol·L-1 Aβ25-35 for 24 h. Tau protein phosphorylation levels at Ser396, Ser202 and Thr231 and the protein level of CDK5 acti-vator proteins p25/p35 were assayed by Western blot. Fluorescence intensity was measured with a fluores-cence microplate reader to reflect calpain activity. CDK5 kinase activity was assayed by immunoprecipita-tion. Results After 20 μmol·L-1 Aβ25-35 acting on cortical neurons for 24h, there were increments in the following: Tau protein phosphorylation levels at Ser396, Ser202 and Thr231, CDK5 kinase activity, CDK5 activator protein p25 level, and calpain activity. In the 20 mg·L-1 FZS treatment group, Aβ25-35 was suppressed markedly, resulting in increments in Tau protein phosphorylation levels at Ser396 , Ser202 and Thr231 , suppression of CDK5 kinase activity and p25 protein level, and elevation in calpain activity. Both CDK5 inhibitor roscovitine and calpain preparation cal-peptin, as positive control drugs, also played a role in suppressing Tau protein hyperphosphorylation. Con-clusion FZS can suppress Aβ25-35-induced Tau pro-tein hyperphosphorylation in cortical neurons through the calpain/CDK5 pathway.
2.Celastrol increases osteosarcoma cells line HOS lysis by γδ T cells through TRAIL way

Objective:To investigate whether celastrol can increase the cytotoxic activity of γδ T cells against osteosarcoma (OS) cells line HOS through TRAIL way. Methods:The death receptors 4/5 (DR4/5) protein levels in the OS cell lines HOS was in-vestigated by Western blot analysis. Zoledronate ( ZOL) was used to induceγδT cells from PBMCs of healthy volunteers.γδT cell cy-totoxicity against HOS cells was investigated by a standard lactate dehydrogenase release assay ( LDH ) . Results:Celastrol increased DR4/5 protein expression in HOS ( P<0. 05 ) .γδ T cells from PBMCs of healthy volunteers showed cytotoxicity against HOS ( P<0. 05). Following co-culture with HOS pre-treated with celastrol (1 μmol/L) for 24 h,γδ T cells showed significantly higher cytotoxicity against HOS (P<0. 05). The induction of DR4/5 molecules increased lysis of HOS by γδ T cells which was abolished by addition of a blocking TRAIL antibody. Conclusion:Celastrol can enhance γδ T cells'cytotoxic activity against OS cells line HOS through TRAIL way.
3.THE APPARENT DIGESTIBILITY OF TWO PREPARATIONS OF CORN AS STAPLES IN MEN

The apparent digestibilities of two preparations of corn as staples in men were estimated during March and April in 1979. Seven healthy adult men, who lived in this region for many years, were accustomed to eat corn as their staple diets. Each of the two preparations from the same batch of corn was used in the experiment. One preparation was "cooked corn meal" and the other was Jian-bing, the latter was considered the better way of cooking. Other ingredients of food in these two test diets were the same. During the test periods, the volunteers ate the test diets and drank boiled water ad lib, but the quantities were recorded. No other foodstuffs and drinks were allowed. The nutritive values of test diets were sufficient to meet the subjects' physiological needs. The 12 days period was divided into adaptative and testing periods of three days each for the two kinds of test diets. During the test periods, the total foodstuffs and the volunteers' faeces were analysed for the contents of water, anhydrates, total ni- trogen, crude fat, ash and fiber in the samples. From the data obtained, the apparent digestibilities of anhydrates, total nitrogen and crude fat ware calculated. Urine nitrogen was analysed and the nitrogen balance was calculated. The body weight at the beginning and the end of test periods were measured.The apparent digestibility (%) of anhydrates was 93.31?0.393 and 94.79 ? 0.578, total nitrogrn 84.60 ? 0.843 and 87.77 ? 1.080 and crude fat 86.32 ? 1.296 and 94.68 ? 0.838 in cooked corn meal diet and the corn flour Jian-bing diet respectively. The apparent digestibility in corn Jian-bing diet was higher statistically than that of cooked corn diet. Body weights fluctuated within the normal range. The nitrogen balances of all subjects were positive and the difference of nitrogen balance studies between the two test diets was not statistically significant.The results of the experiment suggest that, the differences of apparent digestibility between two diets was mainly due to the methods of preparation, Jian-bing being better than cooked corn diet in human nutrition.
4.Progress of clinical pharmacological study on rifabutin

Rifabutin(RBT) is a rifamycin derivative,like rifampicin(RIF),registered for the prophylaxis and treatment of mycobacterium avium complex (MAC)in patients with AIDS by FDA in 1992.Subsequently,the drug was approved by many other countries.But now,it is used not only in the prophlaxis and treatment of mycobacterium avium complex but also in the treatment of pulmonary tuberculosis and Helicobacter pylori.For its high lipophilic characteristic and weak inducing properties compare to other rifamycin derivative,it can be applied in treatment with many diseases successfully,especially when combine with other antibiotics,and can solve the problem of traditional antibiotics resistance and increase the clinical safety of combined medical treatment.This paper just shows the progress of clinical pharmacological study and related aspects on rifabutin in order to instruct prescription.
5.The reproductive toxicity of rat models implanted with copper-bearing intrauterine device at teratogenic sensitive period by continuous extraction method

BACKGROUND:At present, the copper-bearing intrauterine device, a kind of class III medical devices, is
commonly used in China. However, there is no clear conclusion about whether it has impact on the embryo or
fetus in some cases, such as unexpected pregnancy during long-term implantation and pregnancy in a short time after removing it.
OBJECTIVE: To evaluate the safety of copper-bearing intrauterine device by observing the influence of
copper-bearing intrauterine device extracts on pregnant rats and rat fetuses by tail vein injection in the sensitive period of teratogenesis.
METHODS: A total of 60 fertilized rats were divided into control group, high dosage group, middle dosage group, and low dosage group. The copper-bearing intrauterine device extracts were prepared by the continuous
extraction method. Different concentrations (0.2, 0.1, 0.05 g/mL) of copper-bearing intrauterine device extracts were injected by the tail vein at the 1st day of pregnancy in the latter three groups at a dosage of 0.01 mL/g per day. The control group was given the same amount of normal saline. The injection lasted for 20 days. Then, the pregnant rats were sacrificed to measure body mass, check both sides of the uterus and internal organs, isolate fetal rats, as wel as record the quality of uterus and fetal rats, corpus luteum, implantation numbers, the number of
stilbirths, then number of live births and the number of fetal absorption. The fetal rats were determined in the folowing aspects: body mass, body height, tail length, the ossification degree and appearance of the occipital bone, bone and visceral anomalies.
RESULTS AND CONCLUSION: The number of births, implantation numbers, the number of live births, the number of
corpus luteum, the percentages of live births and stilbirths, the number of resorbed fetuses, and the weight of uterus and fetal rats in the control group showed no difference from those in the other three groups (P > 0.05). No malformation in the internal organs occurred. Compared with the control group, the high, middle and low dosage groups showed no
difference in the height, tail length, body mass, and ossification degree of the occipital bone of fetal rats (P > 0.05). No malformation in the appearance, skeleton and internal organs occurred in the fetal rats. These findings indicate that there were no maternal toxicity, abnormal embryonic growth or rat fetus anomalies after injecting copper-bearing intrauterine device extracts into pregnant rats in sensitive period of teratogenesis.
6.Effects of Compound Musk Injection Combined with Edaravone on Coagulation and Neurological Func-tion of Patients with Traumatic Brain Injury

OBJECTIVE:To investigate the effects and safety of Compound musk injection combined with edaravone on coag-ulation and neurological function of patients with traumatic brain injury. METHODS:A total of 84 patients with traumatic brain in-jury in our hospital during Apr. 2015 to Apr. 2016 were divided into observation group and control group,with 42 cases in each group. Control group was given Edaravone injection 30 mg+0.9% sodium chloride injection 100 mL,bid,ivgtt;observation group was additionally given Compound musk injection 20 mL+0.9% sodium chloride injection 250 mL,qd,ivgtt. Both groups were treated for 4 weeks. NIHSS score,GCS score,the levels of NSE,CRP,PT,APTT and D-dimer,GOS score,the occurrence of ADR were compared between 2 groups before and after treatment. RESULTS:After treatment,NIHSS score of 2 groups decreased significantly,compared to before treatment;the observation group was significantly lower than the control group,with statistical significance (P<0.05). GCS score of 2 groups increased significantly,compared to before treatment;and the observation group was significantly higher than the control group,with statistical significance(P<0.05). The serum levels of NSE,CRP,PT,APTT and D-dimer in 2 groups decreased significantly,compared to before treatment;and the observation group was significantly lower than the control group,with statistical significance(P<0.05). GOS score of observation group was significantly higher than that of control group,with statistical significance(P<0.05). No obvious adverse reaction was found in 2 groups during treatment. CON-CLUSIONS:Compound musk injection combined with edaravone can improve coagulation and neurological function of patients with traumatic brain injury,relieve traumatic brain injury and improve prognosis.
7.A biomechanical comparison of conventional versus an anatomic plate and compression bolts for fixation of intra-articular calcaneal fractures.

The purpose of this study was to compare the biomechanical stability obtained by using our technique featured an anatomical plate and compression bolts versus that of the conventional anatomic plate and cancellous screws in the fixation of intraarticular calcaneal fractures. Eighteen fresh frozen lower limbs of cadavers were used to create a reproductive Sanders type-III calcaneal fracture model by using osteotomy. The calcaneus fractures were randomly selected to be fixed either using our anatomical plate and compression bolts or conventional anatomic plate and cancellous screws. Reduction of fracture was evaluated through X radiographs. Each calcaneus was successively loaded at a frequency of 1 Hz for 1000 cycles through the talus using an increasing axial force 20 N to 200 N and 20 N to 700 N, representing the partial weight bearing and full weight bearing, respectively, and then the specimens were loaded to failure. Data extracted from the mechanical testing machine were recorded and used to test for difference in the results with the Wilcoxon signed rank test. No significant difference was found between our fixation technique and conventional technique in displacement during 20-200 N cyclic loading (P=0.06), while the anatomical plate and compression bolts showed a great lower irreversible deformation during 20-700 N cyclic loading (P=0.008). The load achieved at loss of fixation of the constructs for the two groups had significant difference: anatomic plate and compression bolts at 3839.6±152.4 N and anatomic plate and cancellous screws at 3087.3±58.9 N (P=0.008). There was no significant difference between the ultimate displacements. Our technique featured anatomical plate and compression bolts for calcaneus fracture fixation was demonstrated to provide biomechanical stability as good as or better than the conventional anatomic plate and cancellous screws under the axial loading. The study supports the mechanical viability of using our plate and compression bolts for the fixation of calcaneal fracture.
8.Mediating effect of anxiety between sleep disturbance and quality of life in perimenopausal women

Objective To explore the mediating effect of anxiety in perimenopausal women between sleep disturbance and life quality. Methods A total of 516 perimenopmenopausal women in Shenyang,Lia-oning province and Jinzhou city were investigated using the Pittsburgh sleep quality index(PSQI),the self-rating anxiety scale(SAS) and the WHO quality of life( WHOQOL-BREF). Results The scores of sleep disorder,anxiety and quality of life were (14. 31±2. 75),(49. 94±9. 58) and (54. 02±4. 50) in perimeno-pausal women. There was a positive correlation between anxiety and sleep disorder (r=8. 32,P<0. 01),and a negative correlation between anxiety,sleep disorder and quality of life (r=-5. 37,-5. 62,P<0. 01). Anxiety had a mediating effect between sleep disorders and quality of life. The intermediate effect value was-0. 201, accounting for 32. 4%. Conclusion Anxiety has some mediating effects between sleep disorders and quality of life in perimenopausal women,and regulating anxiety can relieve sleep disorders and reduce their impact on quality of life.

10.Effects of sulfhydryl compounds on pancreatic cytoprotection in acute necrotic pancreatitis.

OBJECTIVETo observe sulfhydryl compound variation in the injury of pancreatic cells and the effects of external sulfhydryl compounds on cytoprotection.
METHODSMale Wistar mice were divided randomly into three groups: groups A and B served as animal models (retrograde duct infusion with 5% sodium taurocholate), in group A, 45 animals were treated with normal saline therapy, in group B, 45 animals were treated with Tiopronin therapy; and group C, 15 animals, were designated as normal control. Animals were killed at 2, 4, 6, 12 and 24 h, and pancreatic tissue was analyzed for total sulfhydryl (TSH), nonprotein sulfhydryl (NPSH) and malondialdehyde (MDA). Histopathology, serum amylase (Sam) and C reactive protein (CRP) were assessed as well.
RESULTSLevels of Sam and CRP increased in both group A and group B, with corresponding pathological changes of acute nerotic pancreatitis (ANP). Levels of TSH, NPSH and protein sulfhydryl (PSH) in group A decreased markedly during pancreatitis (P < 0.01), but MDA increased significantly (P < 0.01). The depletion of NPSH in group B was markedly ameliorated at 4 h or 6 h, when Tiopronin was prophylactically administered (P < 0.05), after which the level of MDA showed very little increase when compared to group A (P < 0.01). Histopathological damage was attenuated to a certain extent, in regards to serum amylase and CRP.
CONCLUSIONSAll sulfhydryl compounds decreased significantly during ANP; external sulfhydryl compound could protect the pancreatic cells most likely as a type of scavengers of oxygen free radicals, which are critically involved in the pathophysiology of ANP. Sulfhydryl plays an important role in the action of pancreatic cytoprotection.
