1.Inhibitory effect of aliskiren on LPS-induced angiogenesis of HUVECs

AIM:To investigate the inhibitory effect of aliskiren on the angiogenesis of human umbilical vein endothelial cells ( HUVECs) induced by lipopolysaccharide ( LPS) and to explore its possible mechanism.METHODS:HUVECs were cultured and randomly divided into blank group and renin group.The levels of tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1 (ICAM-1) in the culture supernatant were detected by ELISA.The protein levels of Toll-like receptor 4 ( TLR4) and ICAM-1 in the HUVECs were determined by Western blot.HUVECs were cul-tured and randomly divided into control group, LPS group, low-dose (1μmol/L) aliskiren group, middle-dose (10μmol/L) aliskiren group and high-dose (100 μmol/L) aliskiren group.The proliferation of HUVECs was detected by MTT and BrdU assays.The mobility of HUVECs was measured by Transwell assay.The formation of the vessels was judged by ob-serving the formation of the luminal structure by HUVECs in Matrigel.The levels of TNF-α, ICAM-1 and monocyte chemo-tactic protein 1 ( MCP-1) in the culture supernatant were measured by ELISA.The expression of renin, TLR4, matrix me-talloproteinases-2 (MMP-2) and matrix metalloproteinases-9 (MMP-9) at mRNA and protein levels in the HUVECs was determined by RT-PCR and Western blot.RESULTS:Renin stimulated the expression of inflammatory factors and TLR4 in the HUVECs.Aliskiren inhibited the growth, migration and angiogenesis of HUVECs in a dose-dependent manner, de-creased the levels of TNF-α, ICAM-1 and MCP-1 and the expression of renin, MMP-2 and MMP-9, and inhibited TLR4 expression (P<0.05).CONCLUSION:Aliskiren inhibits LPS-induced angiogenesis of HUVECs, which may be related to the down-regulation of renin expression, the inhibition of TLR4-mediated inflammatory reaction, and the formation of MMP-9 and MMP-2.
2.Experimental study of ballon-dilatation in an establishment of restenosis model in carotid artery of rabbit

Objective To study the mechanism of restenosis for the prevention and treatment by PTA (percutaneous transcather angioplasty) through establishing an model.Methods Under median cervical incision, to eplore right rabbit carotid artery and then clip the segments of ICA at the origination and CCA near the cardiac side of bifucation, balloon catheter with guided wire were finally inserted from proximal cardiac side of the ligated ECA to perform TA after withdrawing the clipper. Histological and morphological analysis with the controll of normal left CCA were carried out during different follow up periods. Results Histopathological and morphometric analysis indicating thrombosis was the main changes in early stage and followed by VSMC from media to intima and irregular proliferation leading to obviously intimal thicking and restenosis at 15 days after PTA.Conclusions This experimental study show low mortality, high practicability and good reproducibility of the model as an ideal presentation for study the mechanism and the prevention of restenosis.
3.The progress of diagnosis and treatment of sepsis in traditional Chinese medicine

Sepsis is a common complication in critically ill patients. The incidence rate was significantly higher in recent years, and it has been as the main cause of death in critically ill patients. The traditional treatment measures failed to significantly improve mortality. In recent years, the treatment of sepsis by traditional Chinese medicine are getting far more attention, the combination of Chinese traditional and western medicine treatment of sepsis bring out new treatment ideas and thoughts,and has achieved good results. This paper reviews the TCM recognition of sepsis from the etiology and pathogenesis, syndrome differentiation, treatments, signgle traditional Chinese medicine and compound preparations and so on,following a brief analysis of existing problems and solutions.
4.Significance produced by examining NSCLC vein formation through enhanced SCT scanning in clinical treatment

Objective To evaluate the examination of non-small-cell lung (NSCLC) vein formation and the dynamic characteristics of blood stream through enforced SCT scanning for the research on and clinical diagnosis of tumor.Methods Double period enhanced SCT scanning was done to 152 NSCLC cases identified pathologically.Makes the color coding the tumour blood stream irrigation chart,the analysis blood stream irrigation characteristic.Compare the enforced morphologic manifestation of cancer focus with histology and analyze their pertinence.Results The enforced CT peak value (PV) of the low differentiation is bigger than that of the medium differentiation,which is bigger than the high differentiation.63 cases are 45-70 HU,78 cases are 20-45 HU and 11 cases are 10-20 HU.67 case are of abnormal arteriola; 23 cases are of abnormal hemal sinus development; 35 cases are pistil-like.Conclusion Accurately examining and quantifying cancer focus vein formation according to SCT double period enforced scanning is of high guiding value in the enactment of the plan for treating NSCLC and the comprehensive treatment of tumors.

6.Effects of endotoxin on phospholipid metabolism and membrane fluidity in liver mitochondria of goats

AIM and METHOD:To study the changes of contents of phosphatidyl inositol (PtdIns),phosphatidylserine (Ptdser),phosphatidylethanolamine (PtdEtn) and phosphatidylcholine(PtdCho) in hepatic mitochondria membrane of goats in vivo at 5 h after administration of E. coli endotoxin(1800 U/kg body weight) with HPLC. The membrane fluidity of the erythrocyte and liver mitochondria of E. coli endotoxin treated group was examined with the fluorescence polarization technique, in which 1,6-diphenyl-1,3,5,-hexatriene was used as a fluorescence probe. Objective To assess and analyze the current status of clinical guidelines for hypertension in the world by using the Appraisal of Guidelines for Research and Evaluation (AGREE) instrument.Methods The clinical guidelines for hypertension were identified and approved by searching China hownet,WANFANG database,PUBMED database,MEDLINE,Embase and related institutions and authorization web site from 1995 to January 2012,and relevant Web sites of agencies and organizations that produce and/or endorse guidelines.Names of the guidelines,published years and organizations,methodology of development and reference number were descriptively analyzed.AGREE instrument was used to evaluate the qualities of latest edited clinical guidelines for hypertension in countries all over the world.Results Nine guidelines were enrolled.The results showed that the hypertension guidelines scored the highest average of 88.4% for clarity of presentation and reliability field; for applicability fields,scored an average of 86.1%; the scope and goal field scored an average of 83.8%; participants field scored an average of 71.7%; editorial independence field scored an average of 64.1% ; rigor of development field scored the lowest average of 62.9%.The overall assessment showed that NICE 2011,Canada 2012,ESC 2009,Australia 2010 editions were the positively recommended guidelines,JNC7,Japan 2009,China Taiwan 2010,China 2010,South Africa 2011 editions were the recommended guidelines (still need to supplement and improve).No recommend or uncertain guide was found.Conclusions The quality of the hypertension guidelines is higher in general,but some common deficiency in the rigor of development and editorial independence in Asian and African guidelines formulated by the states still exists.There still exist certain gaps in evidence-based medical requirement.And the contents and quality are needed for further regulating and enhancing.A set of scientific systemic hypertension clinical guidelines evaluation system should be established.
8.Comparison of coagulation data measured with domestic produced and imported coagulation testing solutions on SD rat

Objective To compare coagulation data measured with domestic produced and imported coagulation testing solutions on SD rat and human by testing prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time(TT), fibrinogen(FIB).Methods Blood samples were obtained from SPF SD rat and human .Domestic produced and imported coagulation testing solutions were applied to test PT , APTT, TT, and FIB.Results Compared to rat data measured with imported coagulation testing solution , data measured with domestic produced coagulation testing solution of PT, APTT, FIB was significantly higher (P<0.05), while, data of TT was statistically lower(P<0.05), and there was no obvious difference in human blood coagulation .Conclusion The data measured with different coagulation testing solution varies on SD rat , so the laboratories are required to establish reference data according to different products .
9.Superficial layer coagulation of trigeminal sensory root in treatment of trigeminal neuralgia

OBJECTIVE To study the clinical value of superficial layer coagulation of trigeminal sensory root in the treatment of trigeminal neuralgia. METHODS The clinical data of 5 cases with trigeminal neuralgia were studied. Head MRI (B-FFE) examinations were made before surgery and operations were performed via retrosigmoidal approach. One case with left facial pain showed 3 arteries with offsets crossing and compressing the left trigeminal nerve. Among 4 cases with right facial pain,3 cases had an artery compressing the right trigeminal and the nerve were distorted and moved from its original site,one case with an artery going with trigeminal nerve sensory root branchlets. All cases were no indications for microvascular decompression and thus were treated with surface coagulation of trigeminal nerve sensory root. RESULTS Pain disappeared instantly after operation and facial sensory were retained. All cases were followed up for 2.4 to 5 years and no recurrences were found. CONCLUSION Superficial layer coagulation of trigeminal sensory root is a safe and effective method for the treatment of trigeminal neuralgia.
10.Effects of Shenmai injection on I?B and TLR4 mRNA expression in lungs of rats exposed to hemorrhagic and LPS shock

AIM: To explore the molecular mechanism of anti-shock effect of Shenmai (SM) injection and to observe its influence on TLR4 and I?B mRNA expression in hemorrhagic shock (HS) plus LPS rat lung. METHODS: Rats were divided into HS, HS+LPS, HS+LPS+ dexamethasone (DEX), HS+LPS+SM group, respectively. HS+LPS injury model of rats were induced by hemorrhagic shock (MAP 40 mmHg for 10 min) followed by intravenous administration of lipopolysaccharide (LPS, 1.5 mg/kg).
