2.Effects of capsaicin on lipid metabolism of liver cells in vitro

Objective To observe the effect of capsaicin on lipid deposition in liver cells and the expression of autophagy,to provide new ideas and targets for the study of fatty liver disease.Methods The model of nonalcoholic fatty liver disease (NAFLD) was established by inducing LGF cell line of liver primary cells with oleic acid (40 μg/mL).The experiment was divided into blank control group,oleic acid group,capsaicin group[oleic acid--capsaicin(100 μmol/L)].The level of intracellular lipid was detected by oil red O staining and triglyceride (TG) kit.Western bolt was used to detect the expression level of autophagy marker molecule p62 and LC3,and to calculate the ratio of LC3 Ⅱ/LC3-Ⅰ.Results After treated with 40 μg/mL oleic acid for 24 h,the oil red O staining showed that orange-red lipid droplets were found in the oleic acid group,but there was no obvious orange-red lipid droplet formation in the blank group,which suggesting that the NAFLD cell model was established successfully by using 40 μg/mL oleic acid in vitro.Oil red O staining was observed,the cells in the capsaicin group were significantly less than the oleic acid group,and the content of triglyceride in the liver cells was significantly lower than that in the oleic acid group(P<0.05).Western bolt test results showed that capsaicin group p62 protein levels were significantly lower compared with the oleic acid group(P<0.05),while the LC3-Ⅱ/LC3-Ⅰ ratio was higher than that of oleic acid group(P<0.05).Conclusion NAFLD model was established by inactivating LO2 cell line with oleic acid in vitro.Capsaicin stimulated NAFLD cells,upregulated the autophagy level of NAFLD cells and reduced intracellular lipid deposition,but the specific mechanism need further study.
3.Analysis of association between PTEN gene single nucleotide polymorphism and stomach neoplasms

4.Comparative Study on Computed Radiogram (CR) and Conventional Chest Film in Diagnosing Lung Cancer

Objective To determine the efficacy of computed radiogram (CR) of chest in the diagnosis of pulmonary carcinoma.Methods There were 56 cases of pulmonary carcinoma confirmed by pathology including 34 cases of central pulmonary carcinoma and 22 cases of periphral pulmonary carcinoma in this paper.Four kinds of chest CR images (plain CR,wide CR,HE-CR and LE-CR) and two kinds of chest CR tomogmphic images (plain CR tomogram and HE-CR) were compared with conventional X-ray (CXR) and conventional tomogram (CTomo)respectively in showing normal structures and abnormal shadowes tomogram.Results The normal structure were displayed more clearly by CR than by CXR,except wide CR and HE-CR in showing right inferior pulmonary artery.The central pulmonary carcinoma was displayed more clearly by CR and LE-CR than by CRX in all aspects.The airways were involved displayed much more clearly by wide CR than by CXR.The display of peripheral vessels and inside densiy of neoplasm was similar on wide CR and CXR films,but the mass margins were abscure on wide CR.HE-CR displayed less clearly than CRX in all aspects.In the peripheral lung carcinoma,plain CR and LE-CR played the better in display of neoplasma margin and cavity in tumor than CXR did, but CXR displayed the margin clearly than wide CR did.HE-CR displayed all of the signs less clearly than CXR did. The indexes displayed by CR tomo were all bettre than that done by CXR tomo.Conclusion All kinds of CR (including CR tomo) images of chest have advantages in displaying normal strcture and neoplasma signs, versus CXR and CXR tomo images. Synthesizing the adventages of various CR(including CR tomo) images,the observator can find more signs to improve the accuracy of diagnosis of pulmonary carcinoma.
5.Sorafenib inhibits tumor growth through modulating vasculogenic mimicry in a hepatocellular carci-noma xenograft model

Objective To investigate the effects and the mechanism of sorafenib on hepatocellular car-cinoma growth and vasculogenic mimicry (VM)in mice.Methods A subcutaneous implantation mouse model of human hepatocellular carcinoma (HCC)HepG2 cells were established.Mice inoculated with HepG2 cells were randomly divided into the treatment group (sorafenib 30 mg·kg -1 ·d -1 )and the control group by using of paired comparison method.Growth of established tumor xenografts was monitored at least twice weekly by vernier caliper measurements.VMwas assessed by immunohistochemical assay and periodic acid schiff reaction (PAS)histochemical double-staining.The expressions of HIF-1 α,VEGFA,VEGFR-1 and MMP-2 in tumor tissues were also assessed by immunohistochemical assay,Western blotting and real-time quantitative PCR. Results The tumor volume in the sorafenib group was obviously decreased compared with the control group (809.69 mm3 ±208.71 mm3 vs 1 678.00 mm3 ±31 3.29 mm3 ),with a statistically significant difference (t =6.1 03,P =0.030).Haematoxylin and eosin (HE)staining showed that tumor tissues treated with sorafenib were characterized by obvious necrosis,but there were not the same cases in the control group.Sorafenib group significantly reduced the number of tumor functional vessel in HepG2 xenografts compared with the control group,as assessed by tumor vasculature uptake of DiOC7 (4.77 ±0.1 5 vs 8.44 ±0.68,t =9.1 92,P =0.01 3).The number of VMwas significantly decreased by sorafenib (1 .04 ±0.46 vs 2.66 ±0.42,t =4.51 0, P =0.041 ).Relative to controls,CD31 -positive vessels decreased after treatments (3.42 ±0.1 0 vs 1 .26 ± 0.1 4),with a statistically significant difference (t =21 .580,P =0.002).Compared with the control group, the protein levels of HIF-1 α(0.65 ±0.03 vs 1 .00 ±0.00),VEGFA (0.51 ±0.02 vs 1 .00 ±0.00), VEGFR-1 (0.45 ±0.04 vs 1 .00 ±0.00)and MMP-2 (0.69 ±0.02 vs 1 .00 ±0.00)were significantly decreased in the sorafenib group (t =1 9.650,P =0.003;t =40.493,P =0.000;t =23.429,P =0.002;t =26.071 ,P =0.002).Compared with the control group,the mRNA levels of HIF-1 α(0.78 ±0.05 vs 1 .00 ±0.00),VEGFA (0.52 ±0.05 vs 1 .00 ±0.00),VEGFR-1 (0.45 ±0.02 vs 1 .00 ±0.00)and MMP-2 (0.71 ±0.02 vs 1 .00 ±0.00)were also significantly decreased in sorafenib group (t =6.840,P =0.021 ;t =27.71 0,P =0.001 ;t =62.740,P =0.000;t =23.850,P =0.002).Conclusion Sorafenib can inhibit the tumor growth and VMchannels formation,which may be related with the HIF-1 αand VEGFA /VEGFR-1 signa-ling pathway.
6.Effects of endocrine therapy and chemotherapy on stem cells in patients with breast cancer

BACKGROUND:Tumor stem cels are the root of cancer recurrence and metastasis, so clinical researches should focus on the effects of different treatments on tumor stem cels.
OBJECTIVE:To explore the effects of endocrine therapy and chemotherapy on stem cels in patients with breast cancer.
METHODS:After recovery and cultivation of estrogen receptor-positive human breast cancer cel lines MCF-7, passage 3 cels in logarithmic phase were selected and divided into three groups containing control, estradiol and estradiol with tamoxifen groups. The estradiol group was divided into three subgroups: 10-7, 10-8 and 10-9 mol/L estradiol was added into the medium, respectively; the estradiol with tamoxifen group was divided into three subgroups: 10-7, 10-8 and 10-9 mol/L estradiol with 10-6 mol/L tamoxifen were added into the medium, respectively. The same amount of absolute ethyl ethanol was added into the medium of control group. Fifteen female patients with late recurrence and metastasis of breast cancer received chemotherapy as recurrence and metastasis group. Another 15 healthy volunteers were selected as healthy control group.
RESULTS AND CONCLUSION:The proportion of CD44+CD24-/lowcel subsets in the estradiol and estradiol with tamoxifen groups was significantly higher than that of the control group (P < 0.05), and the proportion of CD44+CD24-/low cel subsets in the estradiol group was significantly higher than that of the estradiol with tamoxifen group at the same concentration (P< 0.05). The proportion of CD44+CD24-/lowcel subsets had no significant differences among groups at 10 and 20 days of culture (P < 0.05). The proportion of CD44+CD24-/low cel subsets significantly increased in MCF-7 cels after 24-hour intervention with different chemotherapy drugs. But only the proportion of CD44+CD24-/low cel subsets in the paclitaxel and doxorubicin groups was significantly higher than that of the control group after 20-day intervention (P < 0.05). Besides, the proportion of CD44+CD24-/low cel subsets in the peripheral blood of healthy volunteers was significantly lower than that of the recurrence and metastasis group (P < 0.05). Among 15 patients with late recurrence and metastatic of breast cancer, 9 had stable disease, 5 had partial remission, 1 had failed chemotherapy and cancer progression. Moreover, the proportion of CD45-CD44+CD24-/low cel subsets in the peripheral blood of patients sensitive for chemotherapy was significantly lower than that before treatment (P < 0.05). In conclusion, both endocrine therapy and chemotherapy exert a certain effect on the CD44+CD24-/low cel subsets of breast cancer positive for estrogen receptor. Given that CD44+CD24-/low cel subsets in MCF-7 cels resist chemotherapy drugs, the proportion of CD45-CD44+CD24-/low cels in the peripheral blood of patients sensitive for chemotherapy is decreased.
7.Caudate lobe as the sole remnant liver following anatomical hepatectomy for the treatment of hepatolithiasis

Objective To investigate the safety and short-term effect of anatomical hepatectomy for the treatment of hepatolithiasis with the caudate lobe as the sole remnant liver.Methods The clinical data of 1 patient with hepatolithiasis combined with liver atrophy-hypertrophy syndrome who was admitted to the Hunan Provincial People's Hospital in April 2014 were retrospectively analyzed.The stones were located in the left and right liver, the involved liver became fibroatrophy, and the hepatic caudate lobe not containing stones became hypertrophy.The body surface area of the patient was 1.65 m2 , standard total liver volume was 1 167.63 mL.According to the result of CT, expected residual liver volume after hepatectomy was 706.12 mL, and the ratio of residual liver volume over the standard total liver was 60.47%.The radio of residual liver volume over the body mass index was 1.21%.The patient received the second exploration of common bile ducts, hepatectomy with the caudate lobe as the sole remnant liver and T tube drainage.The follow-up including recurrence of calculus was performed by outpatient examination and telephone interview up to April 2015.Results The patient underwent caudate lobe as the sole remnant liver following anatomical hepatectomy successfully without blood transfusion.The operation time and volume of intraoperative blood loss were 380 minutes and 350 mL.The peritoneal drainage tube was removed at postoperative day 2 and the patient was discharged at postoperative day 8 with a good recovery of liver function.The postoperative pathological examination showed that there were focal biliary epithelial papillary hyperplasia combined with light-medium atypical hyperplasia and no canceration.The T tube cholangiography two month later showed that there were unobstructed lower bile duct and no residual intra-and extra-hepatic stones.The liver function was normal.Then T tube was removed and patient resumed normal life.During the 1-year follow-up, no chills and fever, jaundice and abdominal pain occurred, no calculus was detected by B-ultrasonography, and computed tomography reexamination showed that remnant liver volume was increased and no intra-and extra-hepatic bile duct stones were detected.Conclusion Anatomical hepatectomy for the treatment of hepatolithiasis with the caudate lobe as the sole remnant liver is safe and feasible, with a good curative effect.
8.CT Diagnosis of Renal Leiomyosarcoma

Objective To discuss the CT manifestations of renal leiomyosarcoma. Methods 9 cases of renal leiomyosarcoma proved by pathology were analyzed retrospectively. Results CT imaging showed homogeneous hyperattenuation in two cases. Heterogeneous in density with necrosis inside was observed on the CT scan in six cases. After contrast medium administration heterogeneous enhancement with cystic and necrotic areas could be revealed in all cases on both CT scan. Conclusion Large renal mass with intra-tumor necrosis,cystic and hemorrhagic change is the characteristic manifestations of renal leiomyosarcoma. But differential diagnosis from renal carcinoma by CT clinical setting is difficult before surgery, and final diagnosis should depend on pathology examination.
9.Regulatory mechanism of PESV on tumor-infiltrating natural killer cells in liver orthotopic transplantation tumor

Objective:To investigate the regulatory mechanism of PESV on tumor-infiltrating natural killer ( NK) cells in a mice model with H22 orthotopic transplantation tumor .Methods:Suspensions of H22 cells were injected into the lobe of liver on C 57BL/6 mice for establishing liver orthotopic transplantation tumor model ,then the mice were randomly divided into four groups:normal group , control group ,PESV low dose group ( PESV-L ) and PESV high dose group ( PESV-H ) .Mice were either sacrificed for mechanistic studies or survival followed 14 days of therapy.The volume and weight of the tumor were measured .The proportion of infiltrating NK cells was measured by flow cytometry and the expression of NK 1.1(NK) cells was investigated by immunohistochemistry method .The expression of perforin and granzyme B were further investigated by real-time PCR.Results: In contrast to control group , the tumor inhibition rate was 15.38%and 30.77% in PESV-L group and PESV-H group respectively.The survival showed that PESV-H could significantly prolong the survival time of mice ,and life extension rate was 34.06%,(P<0.05).Histological analysis revealed significant pleomorphism of the neoplastic cells and invasive extendion in control group ,while there were more necrosis and less degree of atypia in PESV-L and PESV-H.The level of tumor-infiltrating NK cell was significantly higher in PESV-H than in tumor-bearing control group [(5.91±0.49)%vs.(3.69±0.50)%,P<0.05],and NK cells were infiltrating in peritumoral lesions.The mRNA of perforin and granzyme B in PESV-H were respectively 3.62 and 5.82 times than that of control group ( P<0.05 ) .Conclusion: These findings suggest that the treatment of PESV might increase the infiltration of natural killer cells in the orthotopic transplantation tumor and contribute to NK cells migration to the tumor , which induct and maintain the activities of natural killer cells against tumor cells by expressing perforin and granzyme B in vivo .
