1.Analysis of measurement results of 25-hydroxy vitamin D among 0 - 3 years old infants in Nantong area

Objective To understand the levels of 25‐hydroxy vitamin D [25‐(OH)D] in 0 - 3 years old infants in Nantong area to provide the theoretical basis for the scientific and rational guidance for infants′ vitamin D supplement .Methods The level of fin‐gertip peripheral blood 25‐(OH)D was detected in 3 689 infants and young children undergoing the conventional health examination in the child health care clinic of our hospital from January to June 2014 .The 25‐(OH)D levels and deficiency rate were compared a‐mong different months old and different months .Results The mean level of serum 25‐(OH)D in 3 689 detected infants and young children was (67 .45 ± 35 .01)nmol/L ,the deficiency rate was 39 .9% ,the insufficient rate was 28 .9% and the sufficient rate was 31 .2% ,the deficiency rate and insufficient rate were added up to 68 .8% ;0 - < 2 months old infants had the highest 25‐(OH)D , followed by 2 - < 6 months old infants ,which in 6 - < 12 ,12 - ≤ 36 months old groups was gradually decreased in turn ,the 25‐(OH)D level had statistical differences among different months old infants and young children (P< 0 .05) ;the 25‐(OH)D level had no statistical differences between male and female infants (P> 0 .05) .The deficiency rate and insufficient rate of 25‐(OH)D in 0 - <2 months old infants and 2 - < 6 months old infants had the statistical difference between the first quarter and the second quarter (χ2 = 74 .477 ,P= 0 .000 ;χ2 = 29 .211 ,P= 0 .000) .Conclusion The deficiency rate and insufficient rate of 25‐(OH)D in 0 - 3 years old infants and young children are higher in Nantong area ,especially in infants and young children over 6 months old .
2.Determination of Dehydrocostus Lactone in Dahuoluo Pill by TLC Scanning

Objective To establish a determination method for dehydrocostus lactone in Dahuolu Pill.Methods TLC-Scanning was used.Results The average recovery was 98.30 %and RSD was 1.90 %.Conlusion The method is simple,accurate,repeatable,and can be used for the reference of the quality control to Dahuoluo Pill.

Transport of gossypol by rat cauda epidldymis has been studied using microperfusion technique. It was found that gossypol entered ep-ididymal duct at very much limited rate in spite of its high plasma level. There was no difference between the aqueous & liposome formulations of gossypol to get into the epididymal duct from the blood. Since it was indicated in a previous paper that the transport rates of the 2 gossypol formulations via the blood-testis barrier were quite different. It is suggested that the transfer mechanism in test-is & epididymis may be different.

5.Effect of precursor on growth and accumulation of alkaloids of Lycoris radiata suspension cells.

In order to investigate the effects of phenylalanine, tyrosine and tyramine on the growth of Lycoris radiata suspension cells and the accumulation of alkaloids, the growth quantity of the cells as well as the content of alkaloids in cells were determined, which were treated with above three kinds of precursors alone and phenylalanine combined with tyrosine respectively. The results indicate that the addition of phenylalanine alone and addition of phenylalanine on the basis of tyrosine at high concentration (200 micromol/L) had no significant effect on the growth of Lycoris radiata suspension cells and the content of alkaloids in cells; whereas tyrosine and tyramine promoted the growth of the cells and alkaloids accumulation. Treated with tyrosine at high concentration (200 micromol/L), the content of alkaloids of the cells was 2.56-fold higher than that of the control group, the amounts of lycoramine (3.77 mg/g) and galanthamine (4.46 mg/g) were 6.61-fold and 6.97-fold higher than that of the control group, respectively. When treated with tyramine (200 micromol/L), the amount of alkaloids in Lycoris radiata suspension cells was 2.63-fold higher than that of the control group, and the amounts of lycoramine (4.45 mg/g) and galanthamine (5.14 mg/g) were 9.08-fold and 9.18-fold higher than that of the control group, respectively. The above results demonstrate that adding tyrosine and tyramine in the media significantly promoted the growth of the Lycoris radiata suspension cells and alkaloids accumulation in the cells.

6.Biocompatibility research of the collagen-polymer as human implants

BACKGROUND: A new type of materials, Collamer, is polymerized with type V collagen and HEMA by Staar company, which is a good material for the establishment of intraocular contact lens(ICL). There is no similar material in our country.OBJECTIVE: To evaluate the biocompatibility of ICL made mainly by collagen-polymer in animals.DESIGN: A randomized controlled trial.SETTING: Testing Center of Shanghai Institute of Biomaterial.MATERIALS: Our study was conducted in the Testing Center of Shanghai Institute of Biomaterial from January 2000 to April 2000. Cell strain: hearty L-929 cells(fibroblast of mouse) after 48-72 hours of passage; Twenty white guinea pig of either gender aged between 1 month and 3 months with a body mass between 300 g and 500 g; Seven healthy adult New Zealand rabbit of either gender(female rabbits were not required pregnant for pyrogenic reaction test) with a body mass between 1.7 kg and 3.0 kg or between 2.5 kg and 3.5 kg(experimental animals were obtained from Shanghai experimental animal center, ordinary grade).INTERVENTIONS: Collagen was used to perform the following biological tests: ① cytotoxicity test (cell growth rate); ② anaphylaxis test; ③ pyrogenic reactions test; ④ subcutaneous implant test. The data were analyzed and evaluated according to criteria.MAIN OUTCOME MEASURES: ① Growth and proliferation of fibroblast in mice; ② Reactions of erythema and edema of every provocation site and every observatory time after the induction of intradermal injection and local patch test; ③ Increase of body temperature of the rabbit after the injection of the extraction of the material into auricle marginal vein; ④ Reactions around the materials after 4 weeks of collagen implantation in rabbits subcutaneously.RESULTS: ① Cytotoxicity test: relative growth rate of collagen group was 99% to 106%, and the toxicity grade was level 0 - 1. ② Anaphylaxis test:there was no erythema or edema reaction at every provocation site at each time point of collagen group. The skin reaction to collagen-polymer was level 0. ③ Pyrogenic reactions test: the temperature increased in rabbit was below 0.6 ℃ and the total increase of the temperature was below 1.4 ℃ in rabbits. ④ Subcutaneous implant test: there was very little lymphocyte infiltration around the sample in both collagen group and control group with Level I inflammatory reaction. And the evaluation of the formation of fiber cyst was level I.CONCLUSION: Results of material biological evaluation tests demonstrate that this collagen-polymer has high biocompatibility, and can become an ideal material for ICL.
7.Establishment of a miniature pig model of mandibular osteoradionecrosis.

OBJECTIVETo establish a miniature pig model of mandibular osteoradionecrosis (ORN).
METHODSA single dose of irradiation (IR25 Gy, 28 Gy) was delivered via an electronic linear accelerator to the right mandible of 24 miniature pigs by utilizing three dimensional conformal radiotherapy (3D-CRT). The first molar of the mandible was extracted 2 months after radiation. Mandibular ORN was diagnosed through gross observation, X-ray film, CT, and histopathologic examination.
RESULTSAt 3-4 months after radiation, all animals suffered from mandibular ORN after dental extraction. The symptoms of ORN were more severe in the 28 Gy group than in the 25 Gy group.
CONCLUSIONA miniature pig model of mandibular ORN can be established through 25 Gy irradiation + dental extraction.

8.MicroRNA in the regulation of nervous system development.

Several types of molecules, including the small non-coding RNAs, are involved in the precision regulation of neural development. The expression of microRNAs appears in a specific spatialtemporal sequence during the neural development, and abnormal expression may lead to neural dysplasia. MicroRNAs also play important roles in the regulation of cell biological behaviors in terms of survival, proliferation and differentiation. Neural stem cells are the mother cell of the nervous system. The proliferation, differentiation and migration of neural stem cells are tightly controlled so as to generate appropriate number and phenotype of daughter cells to ensure normal neural development. MicroRNAs are involved in the regulation of neural stem cell proliferation and differentiation via affecting the expression and function of their target mRNAs.

9.Impact of different posture angles on the pain of patients with early esophageal cancer after endoscopic submucosal dissection

Objective:To explore the effect of different posture angles on the pain and comfort in patients with early esophageal cancer after endoscopic submucosal dissection (ESD), so as to provide a basis for patients to choose the best position after ESD.Methods:This study was a randomized controlled trial. One hundred and twenty patients with early esophageal cancer who underwent ESD in the Department of Gastroenterology, Nanjing Drum Tower Hospital from March 2021 to March 2022 were selected as the study subjects, and they were randomly divided into 4 groups of 30 patients each according to the randomized numerical method. Group A was the conventional group, which was in the free position after the operation, and groups B, C, and D were the experimental groups, with group B in the head-high-feet-low 30° position, group C in the head-high-feet-low 45° position, and group D in the head-high-feet-low 60° position. The pain scores after returning to the room after the operation, at 8, 16, 24 h after the operation and comfort scores at 24 h after the operation of the patients in the four groups were evaluated by Numeric Rating Scale (NRS) and General Comfort Questionnaire (GCQ).Results:There were 17 males and 13 females in group A; there were 20 males and 10 females in group B; there were 22 males and 8 females in group C; there were 19 males and 11 females in group D. All patients were aged 30-85 years old. The time main effect, grouping main effect, and interaction effect of postoperative pain NRS score among four groups of patients were all statistically significant ( F=618.13, 12.14, 6.75, all P<0.01). There was no significant difference in the NRS scores of patients after returning to the room after the operation among the four groups ( P>0.05). The NRS scores in group D at 8 and 16 h after the operation were (1.93 ± 0.64), (0.60 ± 0.47) points, lower than the (2.87 ± 1.14), (1.97 ± 1.22) points, (2.17 ± 0.83), (1.97 ± 1.61) points, (2.30 ± 0.75), (0.80 ± 0.61) points in groups A, B, and C, the differences were statistically significant ( t values were 0.79-4.72, all P<0.05). The NRS scores in group B at 24 h after the operation was (0.23 ± 0.18) points, lower than the (1.53 ± 1.08), (0.30 ± 0.21), (0.46 ± 0.25) points in groups A, C, and D, the differences were statistically significant ( t= 5.32, 1.34, 1.37, all P<0.05). The GCQ total scores at 24 h after the operation were (96.96 ± 3.05), (99.77 ± 3.21), (93.53 ± 3.76), (92.20 ± 3.69) points in group A, B, C, D, the difference was statistically significant ( F= 29.59, P<0.05). Moreover, the GCQ total scores at 24 h after the operation in group B were higher than those in groups A, C, and D, and the differences were statistically significant ( t=3.15, 7.01, 8.52, all P<0.05). Conclusions:Targeted body position management can effectively reduce postoperative pain and improve patient comfort in early esophageal cancer patients undergoing ESD.
10.IDEAL-IQ Technique in the Quantitative Assessment of Muscle Fat Infiltration in Lower Limbs of Idiopathic Inflammatory Myopathies

Purpose To evaluate the fatty infiltration of lower limbs by using iterative decomposition of water and fat with asymmetry and least squares estimation quantitative fat imaging(IDEAL-IQ)technique in idiopathic inflammatory myopathy(IIM)patients,and to analyze the correlation between muscle fat fraction(FF)and clinical assessments.Materials and Methods Thirty-two patients with IIM were diagnosed by muscle biopsy and 32 age-,gender-matched healthy volunteers(control group)were recruited.T1WI,T2WI in axial view and IDEAL-IQ sequence of thighs were scanned on each subject.FF values of the anterior,interior and posterior thigh muscles were measured on the FF image generated in the IDEAL-IQ sequence,and medical research council scale score of the IIM group were collected.The difference of muscle FF value between the IIM group and control group was compared,the correlation between FF value and muscle strength of thigh muscles was also analyzed.Results The mean FF values of anterior,interior and posterior thigh muscles in IIM group were 16.60±3.67,6.77±4.92 and 17.32±4.01,respectively,which were significantly higher than those in control group(2.58±2.57,1.40±0.64 and 1.57±0.19),with statistically significant differences(t=-7.29,-6.91,-4.85;all P<0.05).Spearman test showed that the mean FF value was significantly correlated with course of disease(r=0.587,P<0.001).The mean FF values of anterior,interior and posterior thigh muscles were significantly correlated with muscle strength(r=-0.885,-0.761,-0.594;all P<0.001).Conclusion The IDEAL-IQ technique can quantitatively and objectively analyze the severity of muscle fat infiltration in IIM patients,and its degree is correlated with the muscle strength,which has significant clinical application value.
