1.The effect of combined treatment of traditional Chinese medicine and western medicine on sequelae of stroke

Objective To study the effect of combined treatment of traditional Chinese medicine(TCM) and western medicine on sequelae of stroke.Methods 120 patients with sequelae of stroke were randomly divided into two groups,the control group(n =60 cases) and the treatment group(n =60 cases).The patients in the control group were treated by western medicine,while the patients in the treatment group were treated by TCM and western medicine.The neural function defect score of two groups was compared after treatment (Fugl-eyer and Barthel).Results The total effective rate was 85.0% in the control group and 96.7% in the treatment group.There was significant difference between the two groups (x2 =4.9041,P < 0.05).Fugl-eyer and Barthel were all improved aftertreatment in both groups(t =9.1823,13.7645,6.5536,9.7977,all P < 0.01).There were significant differences between the two groups (t =1.9440,2.7204,all P < 0.05).Conclusion Combined treatment of TCM and western medicine can significantly improve the prognosis of patients with sequelae of stroke.
2.Clinical effect of Pinggantonyluo Granules on migraine with liver-wind and blood stasis symptom

Objective:To investigate the clinical effects and main pharmacological actions of Pingantongluo Granules (Radix Paeoniae Alba, Rhizoma Gastrodiae, Radix Salviae Miltiorrhizae, Rhizoma Chuanxiong, Concha Haliotidis, etc.) for migraine with liver wind and blood stasis symptom. Methods: Randomized controlled trail, double blind and double dummy method were applied. Yuntongding Tablets was used as a control drug in clinical investigation and Routunding Tablets as a control drug in pharmacological experiment study. Results: The total effective rates of the treatment group and the control group were 95.0% and 83.3% for migraine and 80.0% and 61.6% for the liver wind and blood stasis symptom, respectively. Pinggantongluo Granules began to act on migraine in ( 11.75 ? 6.70 )h, and the control drug did in ( 18.30 ? 8.22 )h. There were a significant difference between two groups ( P
3.Long-term Toxicity of Jiuxin Fumai Injection in Rats

Objective To observe the long-term toxicity of Jiuxin Fumai Injection and to investigate the safety of clinical medication.Methods The rats were given intramuscular injection with Jiuxin Fumai Injection in large,medium,small dosage(respectively 20,10,5g? kg-1)every day for two weeks,and normal saline group served as the normal control.Two weeks after drug withdrawal,the toxic reaction in rats was observed.Results After two-week continuous administration,all the animals were alive.Some animals were vomiting and getting excited when administered the large dosage and medium dosage injection.The blood sugar elevated,the thoracic gland coefficient lowered,the hepatic cells were cloudily swollen in the animals of large dosage group.Two weeks after drug withdrawal,the above phenomenon vanished.There was no obvious toxic reaction in the small-dosage injection group.Conclusion Long-term administration of Jiuxin Fumai Injection in large dosage shows certain toxic reaction in rats.The safe dosage for intramuscular administration is less than 5 g? kg-1? d-1.
4.Fas Ligand gene transfection induces rat mast cells apoptosis

Objective:To show whether the Fas Ligand gene induces mast cells apoptosis.Methods:RT-PCR was used to amplify the gene of rat Fas ligand extracelluar domain and transmembrane domain and cloned it into eukaryotic expression plasmid pcDNA3.1.Transcent transfect RBL-2H3,the expression of Fas ligand RBL-2H3 was detected by RT-PCR、Western blot.The Annexin V FCM was used to detect the RBL-2H3 apoptosis after the transfection of Fas Ligand.Results:It is successful to obtain the gene of rat Fas Ligand extracellular domain and transmembrane segment,cloning it into pcDNA3.1,FasL was expressed on the surface of RBL-2H3 and it's supernatant after the transfection of pcDNA3.1/FasL.The cell start to be apoptosis.Conclusion:Our study reveals that Fas Ligand gene transfection in RBL-2H3 can effectively induced apoptosis.It is a promising strategy for Fas Ligand to be used in the therapy of allergic disease. 5.Expression of VEGF, EGFR, p16 in lip cancers and oral squamous cell carcinomas and their clinic significance.

OBJECTIVETo detect the expression of vascular endothelial growth factor (VEGF), epidermal growth factor receptor (EGFR), p16 protein in lip cancers and oral squamous cell carcinoma (OSCC) as well as their clinicopathological significance.
METHODSImmunohistochemisty for expression VEGF, EGFR, P16 were carried out in 69 cases of lip cancers and OSCC.
RESULTSExpression of VEGF, EGFR, p16 protein in OSCC and lip cancers was respectively 71.01%, 46.37%, 28.98% and there were no significance between their positive expressions (P > 0.05) as well as in different sites of them (P > 0.05). Expression of VEGF was respectively 71.01% in cancers and 10.00% in non-tumor tissues, there was statistic significance among those (P < 0.05).
CONCLUSIONSThe results show that there is no correlation to the expression of VEGF, EGFR and P16 protein in OSCC and lip cancers. It is suggests that the expression of VEGF might become one of the useful markers for OSCC.

6.Chondrogenesis of synovial mesenchymal stem cells co-cultured with chondrocytes on the three-dimensional scaffold

BACKGROUND:Articular chondrocytes with the ability of autocrine and paracrine can provide the growth factors and microenvironment for synovial mesenchymal stem cels differentiating into the chondrocyte. The
three-dimensional scaffold could provide space for cels adhesion, proliferation and differentiation.
OBJECTIVE: To study the ability of chondrogenesis by co-culturing synovial mesenchymal stem cels and chondrocytes under the three-dimensional condition.
METHODS:The synovial membrane and articular cartilage were harvested from rat knee joint. The synovial
mesenchymal stem cels and chondrocytes were obtained through the method of enzyme digestion. The passage 3 synovial mesenchymal stem cels and passage 2 chondrocytes were co-cultured in the chitosan/I colagen
composite scaffolds at the ratio of 1:2. Then, the cels/scaffold composite was harvested to be examined
morphologicaly, histologicaly and immunohistochemicaly after being cultured 21 days. The confocal laser was also employed to detect the cels distribution in the scaffold.
RESULTS AND CONCLUSION: After being cultured 72 hours, it could be observed from the cels/scaffold composite examined through the scanning electron microscope that the cels adhered on the surface of the
scaffold and extracelular matrix surrounding the cels was seen on the scaffold. After being cultured 21 days, it could be found through the confocal laser scanning that the cels were wel-distributed on the scaffold, and cels decreased gradualy. Type II colagen was positive in the extracelular matrix immunohistochamicaly. It
suggested from this study that the synovial mesenchymal stem cels could be co-cultured with chondrocytes in the chitosan/I colagen composite scaffolds and have the ability of chondrogenesis differentiation.
7.Chondrogenic differentiation of rat synovial-derived mesenchymal stem cells

BACKGROUND:Compared with other sources of mesenchymal stem cells, synovial-derived mesenchymal stem cells have significant characteristics of chondrogenesis and cloning. Therefore, synovial-derived mesenchymal stem cells are one of the most promising seed cells in cartilage tissue engineering.
OBJECTIVE:To isolate and culture synovial-derived mesenchymal stem cells of Sprague-Dawley rats, identify the multipotential differentiation and the potential ability of chondrogenic differentiation in three-dimensional culture condition.
METHODS:The synovium tissue was harvested from Sprague-Dawley rats. The synovial-derived mesenchymal stem cells were isolated with typeⅠcol agen enzyme digestion method and cultured in vitro. The passage 3 cells were detected with giemsa staining, the cellcycle, adipogenic and osteogenic differentiation were determined. The passage 3 cells were centrifuged as pel ets and cultured in the chondriogenic medium for 21 days. And the pel ets were examined by toluidine blue staining, typeⅡcol agen immunohistochemical staining and RT-PCR.
RESULTS AND CONCLUSION:The mesenchymal stem cells isolated from the synovium tissue of rats have the characteristics of mesenchymal stem cells, and exhibit fibroblast-like morphology after cultured in vitro. The multilineage differentiation potentials were also revealed. After the cellwere cultured in chondrogenic medium for 21 days, chondroid tissue was found, type II col agen and aggrecan could be detected positively by toluidine blue staining, typeⅡcol agen immunohistochemical staining, and expressed by RT-PCR examination. Therefore, synovial mesenchymal stem cells have a chondrogenic differentiation potential.
8.The clinical efficacy of sivelestat sodium combined with ulinastatin for sepsis-induced acute respiratory dis-tress syndrome

Objective To observe the clinical efficacy of sivelestat sodium combined with ulinastatin in the treatment of sepsis-induced acute respiratory distress syndrome(ARDS).Methods One hundred and four patients with sepsis-induced ARDS had admitted to our hospital from January 2020 to May 2023 were selected and randomly divided into a control group(routine treatment plus sivelestat sodium)and combination group(routine treatment plus sivelestat sodium and ulinastatin)by a computer random number generator,52 in each group.Murray lung injury score(MLIS),sequential organ failure assessment(SOFA)score,extravascular lung water index(ELWI),arterial blood oxygen partial pressure/fraction of inspired oxygen(PaO2/FiO2),white blood cell count(WBC),neutrophil percentage(NEUT%),and levels of endothelial cell specific molecule-1(ESM-1),soluble urokinase-type plasminogen activator receptor(suPAR)and interleukin-6(IL-6)were compared between the two groups before and after treatment.The recovery speed,prognosis and adverse reactions were compared between the two groups.Results T-test showed there were no significant differences in MLIS score,SOFA score,ELWI,PaO2/FiO2,WBC,NEUT%,ESM-1,suPAR and IL-6 levels between the control group and the combination group before treatment(P>0.05).After treatment,MLIS score,SOFA score,ELWI,WBC,NEUT%,ESM-1,suPAR and IL-6 levels in the combination group were lower than those in the control group(P<0.05),and PaO2/FiO2 was higher than that in the control group(P<0.05).Time to mechanical ventilation and length of ICU stay in the combination group were shorter than those in the control group(P<0.05),and the 28-day mortality rate was lower than that in the control group(P<0.05).No serious adverse reactions occurred in both groups during the treatment period.Conclusion Sivelestat sodium combined with ulinastatin can reduce lung injury and inflammatory response,accelerate recovery speed,improve lung function and prognosis in patients with sepsis-induced ARDS,and the therapy has higher safety.
9.Effect of Dingkundan in Treatment of Qi Stagnation and Blood Stasis Syndrome in Patients with Chronic Obstructive Pulmonary Disease at Stable Phase

ObjectiveTo explore the effect of Dingkundan on Qi stagnation and blood stasis syndrome in patients with chronic obstructive pulmonary disease (COPD) at a stable phase. MethodA randomized controlled clinical design method was adopted, and 60 patients who were diagnosed with Qi stagnation and blood stasis syndrome in COPD at a stable phase in the outpatient and inpatient departments of the respiratory department of Guang' anmen Hospital of China Academy of Chinese Medical Sciences from June 2019 to December 2019 were divided into observation group and control group according to 1∶1. During the study period, there was no dropout, loss of follow-up, or exclusion between the two groups. On the basis of both groups receiving traditional Chinese medicine (TCM) lung rehabilitation training, the observation group took Dingkundan 7 g/time orally, twice a day. The control group received oral administration of the same specification of Dingkundan starch simulator of 7 g/time, twice a day. Both groups have a treatment period of 12 weeks. The COPD Assessment Test (CAT), modified Medical Research Council (mMRC), fatigue scale-14 (FS-14), self-rating anxiety scale (SAS), self-rating depression scale (SDS), 6-minute walk distance (6MWD), and pulmonary function before and after treatment were evaluated. ResultAfter treatment, both groups showed improvements in CAT, mMRC, FS-14, SAS scores, and 6MWD (P<0.05). The observation group also showed improvements in SDS scores and lung function indicators (P<0.05). Compared with the control group after treatment, the observation group showed more significant improvement in CAT, FS-14, SAS, SDS scores, and 6MWD (P<0.05). ConclusionDingkundan has a clear therapeutic effect on Qi stagnation and blood stasis syndrome in patients with COPD at a stable phase. It can reduce symptom burden, enhance exercise capacity, and improve psychological status and has the potential to improve lung function.
