1.Expression and clinical significance of P-selectin in colorectal cancer cells

Objective To investigate the expression and clinical significance of P-selectin in colorectal cancer cells. Methods The protein expression of P-selectin was detected by immunohistochemistry in colorectal cancer tissues ( n=116) and corresponding non-tumor normal mucosa(n=57). The differences of P-selectin expression in colorectal cancer tissues, corre?sponding non-tumor normal mucosa and the clinical pathological significance were compared and analysed in two groups. Results The expression of P-selectin protein was found not only in vascular endothelial cells but also in colorectal cancer cells. The positive rate of P-selectin expression was apparently higher in colorectal cancer cells (77.6%, 90/116) than that in normal mucosa (17.5%, 10/57,χ2=56.49, P<0.05). However, there were no significance in positive rates of P-selectin in colorectal cancer cells between tumors with different diameters, different degrees of differentiation, different depths of infiltration and with or without lymph node metastasis (80.6%vs. 74.1%, 79.0%vs. 74.3%, 78.4%vs. 76.9%, 82.7%vs. 73.4%, P>0.05). Conclusion The high expression of P-seletin in colorectal cancer cells is an important characteristic of colorectal cancer.
2.A study of relationships between traditional Chinese medicine syndrome differentiation and serum cystatin C and homocysteine in patients with chronic heart failure

ObjectiveTo investigate the relationships between traditional Chinese medicine(TCM) syndrome differentiation and serum cystatin C(Cys-C) and homocysteine(Hcy) in patients with chronic heart failure(CHF). Methods 115 cases with CHF admitted into the Department of Cardiology of the First Affiliated Hospital of Zhejiang Chinese Medical University were selected in the CHF group, and 30 cases who had taken health examination in the same period were chosen in the healthy control group. According to the TCM syndrome differentiation, the CHF cases were subdivided into four groups with different types of syndrome: 30 cases of deficiency of both Qi and Yin syndrome, 30 cases of Qi deficiency syndrome and blood stagnation syndrome, 30 cases of heart and kidney Yang deficiency syndrome and 25 casesof flooding due to Yang deficiency syndrome. The serum levels of Cys-C and Hcy in different groups were tested, and the relationships between TCM syndrome differentiation and serum Cys-C and Hcy were analyzed by using Spearman rank correlation analysis.Results The serum levels of Cys-C and Hcy in the patients with CHF were significantly higher than those in the healthy control group〔Cys-C(mg/L):1.24±0.34 vs. 0.77±0.22, Hcy(μmol/L):18.66±4.57 vs. 11.65±3.21,bothP<0.05〕. Compared with the healthy control group, the serum levels of Cys-C and Hcy in the above four groups of different syndromes had a tendency of gradual elevation in the sequence as follows: deficiency of both Qi and Yin, Qi deficiency and blood stagnation, heart and kidney Yang deficiency and flooding due to Yang deficiencygroups〔Cys-C(mg/L):1.02±0.27,1.09±0.31,1.32±0.22, 1.59±0.25; Hcy(μmol/L): 14.94±2.20, 17.66±3.04, 19.79±3.48, 22.96±5.31〕, and the elevation in levels of flooding due to Yang deficiency group was the most prominent compared with that in other groups(P<0.05). The correlation analyses showed that different types of TCM syndrome in patients with CHF were positively correlated with the levels of Cys-C and Hcy(r1=0.73,r2=0.79,bothP<0.05).ConclusionThe changes of serum Cys-C and Hcy levels are consistent with the evolution of regular pattern of TCM syndrome differentiation in patients with CHF, and these two markers can be regarded as the objective indicators of TCM syndrome differentiation of CHF.
3.Long-term Toxicity of Jiuxin Fumai Injection in Rats

Objective To observe the long-term toxicity of Jiuxin Fumai Injection and to investigate the safety of clinical medication.Methods The rats were given intramuscular injection with Jiuxin Fumai Injection in large,medium,small dosage(respectively 20,10,5g? kg-1)every day for two weeks,and normal saline group served as the normal control.Two weeks after drug withdrawal,the toxic reaction in rats was observed.Results After two-week continuous administration,all the animals were alive.Some animals were vomiting and getting excited when administered the large dosage and medium dosage injection.The blood sugar elevated,the thoracic gland coefficient lowered,the hepatic cells were cloudily swollen in the animals of large dosage group.Two weeks after drug withdrawal,the above phenomenon vanished.There was no obvious toxic reaction in the small-dosage injection group.Conclusion Long-term administration of Jiuxin Fumai Injection in large dosage shows certain toxic reaction in rats.The safe dosage for intramuscular administration is less than 5 g? kg-1? d-1.

Objective To study the frequencies of allele and genotype distribution of alpha-B-crystallin (CRYAB ) gene rs3212227 and rs6894567 single nucleotide polymorphism (SNP) in Chinese guangxi populations ,and to Compare the distribution differences among different ethnic .Methods The CRYAB gene rs3212227 and rs6894567 polymorphisms were detected by the pol-ymerase chain reaction-single base extension (PCR-SBE) technique and DNA sequencing methods in 199 Chinese guangxi popula-tions ,frequencies of allele and genotype of CRYAB gene SNP loci ,rs3212227、rs6894567 were analyzed in guangxi populations com-pared with other the four populations (HapMap-CEU ,HapMap-YRI ,HapMap-JPT and HapMap-HCB) from Human Genome Pro-ject group (Hapmap) data .Results There were CRYAB gene polymorphisms in Guangxi populations .The frequencies of allele and genotype distribution of CRYAB gene rs3212227、rs6894567 polymorphisms had significant difference compared with HapMap-CEU and HapMap-YRI populations (P<0 .05) ,and had no significant difference compared with HapMap-JPT and HapMap-HCB (P>0 .05) .Conclusion The frequencies of allele and genotype distribution of CRYAB gene rs3212227、rs6894567 polymorphisms are significantly difference compared with others ethnic populations ,and this variation might account for a variety of clinical mani-festation and morbidity of of some CRYAB related diseases .
7.Study on anti-invasive effect and apoptosis induction of pentacyclic triterpenoid in human lung cancer cells.

BACKGROUNDTo study the anti-invasive effects and its mechanism and apoptosis induction of pentacyclic triterpenoid including glycyrrhizin (GL), 18β-glycyrrhetinic acid (GA), ursolic acid (UA) and oleanolic acid (OA) in highly potentially metastatic lung cancer cell line (PGCL3).
METHODSThe invasive ability, the adhesive ability, the migration ability and the activity of cathepsin B (CB) of PGCL3 cells treated with the four drugs were determined by the invasion test of reconstituted basement membrane, the laminin adhesion test, the chemotactic migration test and the enzymological method of CB. The apoptosis of the cells was detected with acridine orange-ethidium bromide fluorescent stain (AO/EB) and TUNEL.
RESULTSThe GL,GA, UA and OA could decrease the proliferative ability of PGCL3 cells, and their IC₅₀ values were 1.83 mmol/L, 145.3 μmol/L, 44.73 μmol/L and 40.71 μmol/L respectively. After treatment with 0.5 and 1.0 mmol/L GL, 25 and 50 μmol/L GA, 30 and 40 μmol/L UA, 35 and 45 μmol/L OA for 96 h, the invasive ability of the PGCL3 cells was significantly decreased compared with that of the control groups ( P < 0.01 or P < 0.001). The adhesive and migration ability, the secretion of CB and the colony-formation number in semi solid agar were significantly decreased after PGCL3 cells were treated with the above concentration of the four drugs for 96 h ( P < 0.05, P < 0.01 or P < 0.001), and the inhibition was in a dose-dependent fashion. The percentages of apoptosis of the cells were obviously increased after treatment with the above concentration of the four durgs for 48 h, compared with the control group ( P < 0.05 or P < 0.01).
CONCLUSIONSAll of the four drugs can inhibit the proliferative and invasive ability, and induce apoptosis of the PGCL3 human lung cancer cells. The mechanism of anti invasion may be to inhibit the adhesion, migration, and the CB secretion of the cells.
8.Studies on proliferation inhibition and anti-invasion of retinoic acid and 18β-glycyrrhetinic acid in highly metastasized human lung cancer cell line.

BACKGROUNDTo study the proliferation inhibition and anti-invasion of retinoic acid (RA) and 18β-glycyrrhetinic acid (GA) in highly metastasized lung cancer cell line (PGCL3), and to observe the combined effects of RA and GA.
METHODSThe proliferation inhibitive rate, the colony-formation rate in semi-solid agar, the invasive ability to reconstituted basement membrane, the chemotatic migration ability, the laminin adhesion ability, and the activity of cathepsin B (CB) were tested.
RESULTSTreated with RA and GA, the proliferation of PGCL3 cells were inhibited obviously, and the inhibition degree was related to the dosage of the drugs. IC₅₀ of the proliferation inhibition were 12.58 μmol/L and 145.3 μmol/L respectively. Treated with 5.0 μmol/L RA, 25 μmol/L and 50 μmol/L GA, the invasive ability was decreased significantly (P < 0.01 and P < 0.001), and the inhibition was in a dose dependent manner. In combined treatment with 5.0 μmol/L RA and 25 μmol/L GA, the inhibition of invasion was greater than the sum of them used alone. Treated with GA of above concentrations and 10 μmol/L RA, the adhesion and migration ability and the secretion of CB of the PGCL3 cells were decreased significantly (P < 0.001). Treated with GA of above concentation, the colony formation rate in semi-solid agar was decreased significantly (P < 0.001)..
CONCLUSIONSRA and GA can inhibit the proliferation and invasion of the PGCL3 human lung cancer cells and have the anti-invasion synergism. The mechanism of anti-invasion of RA and GA is to inhibit many points of invasive process.
9.Strengthen the management of intervention on compliance, heart function, life quality and prognosis on the patients with chronic heart failure

Objective To investigate the effect of strengthening intervention management on the compliance, heart func-tion, life quality and prognosis for the patients with chronic heart failure. Methods From September 2013 to September 2014, 100 cases admitted to the Department of Cardiology of General Hospital of Daqing Oil Field in patients with congestive heart failure were selected as the observation objects, they were randomly divided into strengthening the management of intervention group 50 cases and routine management of the intervention group 50 cases, treatment com-pliance and cardiac function index, life quality and prognosis the situation of the patients before and after treatment were compared between the two groups. Results The complete compliance rate of strengthen the management of inter-vention group was 84%, significantly higher than that of the conventional management intervention group (P<0.05). HR, LVEF, CO and E/A of strengthen the management of intervention group significantly improved than the control group, the quality of life scores also improved significantly compared with the conventional management intervention group (P<0.05). And rehospitalization rate of strengthen the management of intervention group was 16.0%, significantly lower than that of the conventional management intervention group, and the mortality rate of strengthen the management of intervention group was 2.0%, was slightly lower than the conventional management intervention group. Conclusion The implementation of strengthening the management of patients with chronic congestive heart failure, can improve the treatment compliance, significantly improve cardiac function and prognosis in patients with heart failure, improve the quality of life of patients with heart failure.
10.Fabrication of three-dimensional microwell patterns and their integration with C17. 2 neural stem cells.

UV photolithography and hydrofluoric acid wet etching were used to produce silicon master molds and polydimethylsiloxane (PMDS)-based soft lithography was adopted to fabricate three-dimensional poly(lactic-co-glycolic acid) (PLGA) and PDMS microwell patterns with high aspect ratio and channel connection. Nine microwell patterns were thus obtained with different structural dimensions. Patterns were treated with oxygen plasma etching and polylysine coating to enhance hydrophilicity and cell compatibility for subsequent culture of C17. 2 neural stem cells. With proliferation during the culture, C17. 2 cells gradually distributed within the microwells, showing an obviously three-dimensional (3-D) growth behavior. The presence of channel structures greatly favored the 3-D growth of C17. 2 neural stem cells on the microwell patterns. Multi-layered scanning with confocal microscopy and 3-D rendering after carboxyfluorescein diacetate succinimidyl ester (CFDA-SE) staining showed that most C17. 2 cells grew within a range of 30 to 90 microm from the microwell bottom. Immunofluorescence staining indicated that C17. 2 cells within 3-D microwell patterns were uniformly nestin-positive on day 2 after cell plating. It could well be concluded that the microwell patterns thus fabricated were suitable for the 3-D culture and subsequent differentiation of C17. 2 neural stem cells. And the cells can be maintained with uniform stemness properties while cultured in these microwell patterns.
