2.Effects of Shoutaiwan on the STAT Signal Transduction Pathway of CD_4~+ T Cells in Maternal-fetal Interface of Mice with Recurrent Spontaneous Abortion

Objective To study the effects of Shoutaiwan on the STAT signal transduction pathway of CD4+ T cells in maternal-fetal interface of mice with recurrent spontaneous abortion(RSA).Methods CBA/J?BALB/C mating combination and CBA/J?DBA/2 mating combination were used as normal pregnancy and RSA model respectively.Mice with RSA were randomly assigned to model group and Shoutaiwan group.After 14 days of administration,the STAT signal transduction pathway of CD4+ T cells in mice decidual and placental tissues were detected by Springbio 720 antibody microarrays.Results Shoutaiwan could increase the phosphorylation level of STAT3 and STAT6 protein in the STAT signal transduction pathway.At the same time,Shoutaiwan could decrease the phosphorylation level of STAT1 protein.Conclusion The activation of STAT3 and STAT6 signal transduction pathway might play a important role in the therapy of RSA by Shoutaiwan.

Objective:To study the protective effects of Shenmai injection (SMI,参麦注射液) on peritoneal mesothelial cell in rats with 5/6 nephrectomy. Methods:By using light microscopy and scanning electron microscopy the morpholgical structure changes of peritoneal mesothelial cell in rats with 5/6 nephrectomy caused (chronic)renal failure model of after the peritoneal didysis of SMI were observed at the same time the levels of C3 and IgG in peritoneal dialysate were observed also. Results:The SMI could promote the recovery and proliferation of injured peritoneal mesothelial cell increase the immunity function of abdomen in rats. Conclusions:The SMI has protective effect on peritoneal cell in rats with 5/6 nepherctomg the protecive effect related to improving regional defence function in abdomen.
4.Study on Percolation Extraction Process of Panax Notoginseng

Objective To optimize the conditions for the percolation extraction process of Panax notoginseng(Burk.) F.H.Chen.Methods Conditions for the percolation were studied by orthogonal experimental design as guided by the contents of total notoginseng saponin and total amino acids.Results The percolate rate,concentration and quantity of alcohol had significant effects on the process.Conclusion The optimum condition for the extraction of Panax notoginseng was adding 12 times amount of 50% alcohol and percolating at a rate of 1~3 mL/(min?kg).
5.Effect of Fuke Zhixuexiaotong Keli on the Expression of MMP-9 and TIMP-1mRNA in Rats Model of Incomplete Puerperal Involution of Uterus

Objective To explore the mechanism of Fuke Zhixuexiaotong Keli in rats model of incomplete puerperal involution of uterus by detecting the expression of metalloproteinases-9(MMP-9) and tissue inhibitor of metalloproteinase-1(TIMP-1) in the uterine redintegration incomplete model.Methods Seventy-two SD female rats were randomly divided into 6 groups:blank group,model group,Chanfukang group,and high-,middle-,low-dose of Fuke Zhixuexiaotong Keli group.The model of incomplete puerperal involution of uterus was established,and MMP-9,TIMP-1 gene expression level in each group were detected by in situ hybridization detection of uterine tissue.Results The expression of MMP-9 and TIMP-1 could be successfully detected in each group.The expression of MMP-9 in model group was significantly higher than the control group,and the expression of TIMP-1 was lower.The mRNA of MMP-9 gene was significantly decreased in both high-,middle-and low-dose of Fuke Zhixuexiaotong Keli group and Chanfukang group(P

Objective To explore the key way of DUB (dysfunctional uterine bleeding) by traditional and western medicine. Method Analyse the traditional medicine's unique understanding of DUB, the symptoms of DUB and the function of Chinese traditional medicine on DUB. And sum up the modern molecular pathology of DUB. Result Chinese traditional medicine has some regulations in curing DUB. And given the modern molecular biology technique, it maybe play an important part in researching the essential of the syndrome of DUB and the function regulation of Chinese traditional medicine. Conclusion DUB, which is an important disease that is in the person of “menstruation adjustment”, “blooding disease”in traditional Chinese gynecology, must be studied systematically first, so as to improve the combination of traditional medicine and western medicine in gynecology.

Objective To explore the molecular mechanism of elevating placental blood supply by the component from Mongolian Milkurtch and red sage Root. Method Abstract the effective components from Mongolian Milkurtch and red sage Root, apply L-Arginine methy1 ester (L-NAME) to make NO production blocking model, use ELISA to check IL-10 in plasma of 18 d pragant rats, and FISH to check the expression of matricial metal protease-9 (MMP-9) mRNA. Results The plasma level of IL-10 of model group was lower than that of blank group (P 0.05). Conclusion By interving the relationship between IL-10 and MMP-9, the components of Mongolian milkvetch root and red sage root compound may affect the shift and invadation of deciduas, so as to prevent fleet embedding, help building the circulation between mother and embryo, and keep the placental blood supply.
